      • 근원세포의 융합과 연관된 표면 단백질의 확인

        배양 근원세포 분화에 따른 세포융합에 직접적으로 관여하는 단백질을 찾기 위해 48 시간 배양한 근원세포를 항원으로 BALB/C mouse에 면역시켜 얻은 spleen cell과 myeloma cell(P3U-1)을 융합시켜 근원세포와 반응하는 항체를 분비하는 hybridoma를 얻었다. 세 차례 한계 희석 후 계대배양하여 분리한 clone들이 분비하는 항체를 근원세포 배양액에 첨가하여 근원세포의 융합을 억제하는 효과가 있는 단일클론항체인 MAb MII-3J3l을 얻었다. MAb MII-3J3l의 효과는 2 ㎍/ml의 농도에서 세포분열에 영향이 없이 근원세포의 융합을 완전히 억제하였으며 이 효과는 적어도 48 시간 지속되었고 항체를 제거할 경우 융합이 재개되어 가역적인 억제효과를 나타내었다. MAb MII-3J31이 인지하는 단백질은 38 kDa로 근원세포 표면을 trypsin 처리하였을 때 잘리워 나감으로 표면단백질로 추정된다. The present study describes the production of monoclonal antibodies against cultured chick myoblast to pursue a protein may play a critical function in muscle cell fusion. Among a panel of monoclonal antibodies, MAb MII-3J3l inhibits myoblast fusion completely, while cell growth proceeds normally and its effect is reversible. The antigen reactive with MAb MII-3J31 isolated by radioimmunoprecipitation and detected by immunoblot was found a 38 kDa protein. For trypsin-treated cell lost its antigenic determinant, the protein may be located in the cell surface.

        기저세포암 및 편평세포암에서 미세혈관밀도와 p53 단백 발현에 관한 연구

        Basal cell carcinoma(BCC) and squamous cell carcinoma (SCC) are very prevalent neoplasms of the human skin. Ultraviolet radiation in sunlight is a well-established mutagen of the p53 gene and is one of basal cell carcinoma and squamous cell carcinoma. The newly-formed vascular network is important for neoplasms to grow beyond a size of about 1 ㎣. Recent reports have suggested the hypothesis that a mutant p53 protein is closely related with capillary density. Immumohistochemistry for p53 protein and CD34 was performed in 20 cases of BCCs and 14 SCCs to evaluated the relationship between p53 protein and capillary density. The results were as follows:1. The microvessels stained by CD34 were mainly located in the interface of tumor cells and stroma. 2. There was no difference in the microvessel density according to the histologic types and age of the patients, but a higher microvessel density was noted in male patients. 3. The aggressive BCCs and the less-differentiated SCCs showed higher p53 immunostaining. 4. The mean microvessel density of cases showing strong positive immunostaining of the p53 gene(54.73±17.75) was higher than that of others(39.75±18.30). These results suggested that p53 protein expression and microvessel density are not related to the histologic types and age of the patients, but that differentiation and biologic behavior such as the infiltrating property of tumors and the microvessel density are closely related to p53 protein expression.

      • 다단변동/충격하중하의 탄소섬유강화 복합재료의 피로강도 특성 및 잔류강도 저하거동에 관한 연구

        Under the multi-step loading and impact loading, similar to real loading, fatigue tests have been carried out to examine the characteristics of a fatigue strength and a fatigue life and the degradation of the residual strength in carbon-epoxy composite materials. Two different shape of test specimen and test methods were used and the theory of cumulative damage and the residual strength degradation was applied to the test data of two parameter Weibull distribution. The static ultimate strength of the test piece used here in is observed to be relatively higher than that of existing composite materials, while fatigue life is shorter than that of estimated by residual strength degradation model when the stress level above 0.62. The fatigue life of the materials tested were observed the one of the high-low multi-step was longest and the one of the low-high next compared by Miner's value. It is proven that the prediction of fatigue life by the analysis of the theory of cumulative damage and degradation is resonable.

      • 기계시각을 이용한 강구 자동 검사장치

        Metal balls are commonly used in mechanical elements and systems including bearings. If there is a defect in a metal ball, it can cause a serious machine failure. By this reason every balls produced are required to be inspected, and the inspection is usually done by human eyes. However, the inspection by human eyes is always with the risk of mistakes, and a need to automate the inspection process is arisen. This paper describes a design of an automatic inspection system for detecting defects on ball surface. A machine vision system is used, and design process and techniques are described in detail. 강구는 베어링을 비롯한 기계 요소 및 장치에서 널리 사용되는 부품이다. 만약 강구에 결함이 있을 시, 이를 사용하는 기계에서 치명적인 결함이 초래될 수 있다. 따라서 제조된 강구에 대해서는 전량검사가 요구되고 있고, 검사과정은 사람의 육안에 의한 검사가 주를 이루고 있다. 그런데 사람에 의한 검사에는 항상 실수의 가능성이 있으므로, 이를 자동화하는 것이 필요하다. 본 논문에서는 강구의 표면 결함을 자동적으로 검사할 수 있는 장치를 설계한다. 기계시각 장치가 사용되었고, 설계에 따른 구체적 절차와 기술들이 서술된다.

      • Mg_(x)Zn_(1-x)FeAIO₄의 결정학적 구조 및 Mo¨ssbauer 효과

        Mg_(x)Zn_(1-x)FeAlO₄ spinel ferrite에 대하여 X-선 회절과 Mossbauer 스펙트럼을 조사하였다. X-선 회절상으로부터 Mg_(x)Zn_(1-x)FeAlO_(4)는 면심입방구조를 가지는 spinel ferrite임을 확인하였고 Mg 함유량 x가 증가함에 따라 격자상수는 선형적으로 감소하여 8.3496A - 8.4128A의 값을 보이고 있다. Mossbauer 스펙트럼은 모든 시료가 초미세 자기장에 의한 공명흡수선이 나타나고 Mg 함유량 x가 증가됨에 따라 초미세 자기장은 20K와 300K에서 모두 증가하고 있다. I.S.값이 0.2569mm/sec에서 0.3564mm/sec사이에 존재하는 것으로 Fe^(3+)로 이루어져 있으며, Q.S.값은 zero값에 가까운 값이므로 대칭분포 구조를 갖는다.

        There are developing many remote medical systems according to the advanced of communication application technology and network. However, such of development is concentrated in the western medical system, not for oriental medical system. Therefore in this paper, we implement Remote Electronic Pulse Diagnosis Services System for oriental medical examination, diagnosis and treatment using network between remote places. Remote Electronic Pulse Diagnosis Services System is to support Oriental Medicine Services, for instance, Pulse Diagnosis, Inquiring, and Patients Information Management etc. in real time using communication network from remote place. Hence, it provides infrastructure for saving the cost on medical services and supports high level realtime medical services using developed medical network.

      • 계배 배양 근원 세포 항원에 대한 단일 클론 항체의 생산

        Monoclonal antibodies raised in mouse BALB/c against cultured chick myoblast were found to exhibit reactivity toward myoblast proteins of 109 kDa, 19 kDa, 18 kDa and 15 kDa and others. Especially the levels of 109 kDa, 18 kDa and 16 kDa proteins decreased during the course of myogenesis in culture and between embryo and adult muscle. These antibodies delayed the differentiation and enhanced the cell division of myoblast when added to culture media. Their effects were saturated by adding one fiftieth of media and accompanied with the increments of 200 kDa, 29 kDa and 15 kDa myoblast proteins, the decrement of 25 kDa protein, and the new synthesis of 57 kDa protein.

        Enforcing network security in telemedicine systems is necessary to ensure security of the information exchanged. This paper deals with the need for security in application programming based on the information superhighway. A secure network is critical for the survival and success of applications. In this paper we discuss some features of well-known security mechanisms such like Kerberos, NetSP, SPX and SESAME. We also propose a software design strategy of telemedicine systems using SESAME developed by ECMA.

      • 황정(黃精) 추출물의 화학구조결정에 관한 연구(Ⅱ)

        본 연구에서는 황정 속에 포함되어 있는 생리활성 물질을 hexane, CHCl₃과 n-butanol층에서 각각의 성분들을 추출하였고, hexane층에서 분리된 화합물 I과 Ⅱ 중에서, 화합물 I의 봐학구조를 ¹H-nmr, ??, DEPT135, COSY, HMQC, HMBC 스펙트럼 및 MS 스펙트럼 등의 분광학적인 방법에 의해 결정하였다. 화합물 I의 구조는 9,12-(9E,12E) -octadecadienoyloxy) propanoic acid 임을 확인하였다. In this Paper, biologically active compounds were extracted using organic solvents as hexane. CHC1₃, n-butanol to give each component. Chemical structure of compound I was characterized using ¹H-nmr, ??, DEPT135, COSY, HMQC, HMBC spectrum and MS-spectrum, in separated compound I and Ⅱ from hexane layer. Finally, chemical structure of compound I was determined as 9,12-(9E,12E)-octadecadienoyloxy)propanic acid.

        백합가루 첨가량에 따른 백합병(百合餠)의 관능적·텍스쳐 측정

        The purpose of this study was to determine gain the most desirable mixture ratio for the ingredients of Backhapbyung. Sensory examination and mechanical test were conducted on the texture, water contents and color degree of Backhapbyung with various ratios of liliibulbs powder and sugar added to nonglutinous rice flour. The results of sensory evaluation showed that Backhapbyung composed of put 8% (24g) liliibulbs powder, 40g sugar, 40ml water and 276g nonglutinous rice flour had the highest overall score in terns of acceptability, after-swallowing and taste of flavor, and overall quality preference. Overall among the recipes tested, that with 6% (24g) liliibulbs powder, 40g sugar, 40m1 water and 282g nonglutinous rice flour showed the highest marks in springiness, cohesiveness, gumminess and chewiness. The moisture content was 33.54% nonglutinous rice flour, and 11.8% liliibulbs powder. The overall quality of sensory examination for bitterness was positively correlated with the hardness of sensory examination and the mechanical examination for adhesiveness. The overall quality of sensory examination for chewiness was positively correlated with the moistness of sensory examination and the mechanical examination for cohesiveness. From the above results, the best mixture ratio of Backhapbyung, with a moisture content of 37.18%, is 8% (24g) of liliibulbs powder, -10g sugar, 40ml water, and 3g salt and 276g nonglutinous rice flour.
