      • Early Growth Response Protein-1 Involves in Transforming Growth factor-β1 Induced Epithelial-Mesenchymal Transition and Inhibits Migration of Non-Small-Cell Lung Cancer Cells

        The zinc finger transcription factor EGR 1 has a role in controlling synaptic plasticity, wound repair, female reproductive capacity, inflammation, growth control, apoptosis and tumor progression. Recent studies mainly focused on its role in growth control and apoptosis, however, little is known about its role in epithelial-mesenchymal transition (EMT). Here, we aim to explore whether EGR 1 is involved in TGF-${\beta}1$-induced EMT in non-smallcell lung cancer cells. Transforming growth factor (TGF)-${\beta}1$ was utilized to induce EMT in this study. Western blotting, RT-PCR, and transwell chambers were used to identify phenotype changes. Western blotting was also used to observe changes of the expression of EGR 1. The lentivirus-mediated EGR 1 vector was used to increase EGR 1 expression. We investigated the change of migration to evaluate the effect of EGR 1 on non-small-cell lung cancer cells migration by transwell chambers. After stimulating with TGF-${\beta}1$, almost all A549 cells and Luca 1 cells (Non-small-cell lung cancer primary cells) changed to mesenchymal phenotype and acquired more migration capabilities. These cells also had lower EGR 1 protein expression. Overexpression of EGR 1 gene with EGR 1 vector could decrease tumor cell migration capabilities significantly after adding TGF-${\beta}1$. These data s howed an important role of EGR 1 in the EMT of non-small-cell lung cancer cells, as well as migration.

        Control Design for Stochastic Nonlinear Systems with Full-state Constraints and Input Delay: A New Adaptive Approximation Method

        In this paper, the full state constraints and input delay of stochastic nonlinear systems are studied. A new adaptive control algorithm is proposed using backstepping approach and multi-dimensional Taylor network (MTN) method. Firstly, the input delay problem is dealt with by introducing a new variable using the Padé approximation with Laplace transform. Secondly, MTNs are employed to approximate unknown nonlinear functions, and the barrier Lyapunov functions (BLFs) are constructed to deal with the state constraints. Based on this, a new approximation-based adaptive controller is proposed. Thirdly, it is proved that the proposed control method can ensure that all signals in the closed-loop system are semi-global ultimately uniformly bounded (SGUUB) in probability and the tracking error converges to a small neighborhood of the origin. Finally, two simulation examples are given to illustrate the effectiveness of the proposed design method.

      • Thermal Insulation of Wet Empire Style Dress and Spencer Jacket by Thermal Manikin Method

        Empire style dress in the late 18th and early 19th attracted the hugely popular among women. High waisted and graceful styles are known as the Empire style. The volume in the skirt is great and breast is exposed a lot. Underwear such as the corset and panier did not wear in the dress. Women wore the dress in winter as well as in summer, however, they just wore spencer jacket or shawl over the dress. Furthermore, this fashion gained popularity not only a young lady but also older women. At the time when wearing this clothing for daily life or work, in particular, a lot of women in the winter felt cold. When women continue to cough, they look beautiful because of delicate femininity, but health was deteriorated and they even died. Even they wore wet robe to render the erotic image of see-through effect. Because of thin and wet dress, they would feel the terrible sense of dissatisfaction. We want to investigate that these environment is how much risk to health. No one knows thermal insulation of the empire style dress, specially wet condition. We find out thermal insulation and layer effect of empire style dress and jacket from thermal manikin method. In particular, when women wearing the dress got into perspiration or in rain environment, they felt unpleasant.

        Statistical-based evaluation of design codes for circular concrete-filled steel tube columns

        This study addresses the load capacity prediction of circular concrete-filled steel tube (CFST) columns under axial compression using current design codes. Design methods given in the Chinese code CECS 28:2012 (2012), American code AISC 360-10 (2010) and EC4 (2004) are presented and described briefly. A wide range of experimental data of 353 CFST columns is used to evaluate the applicability of CECS 28:2012 in calculating the strength of circular CFST columns. AISC 360-10 and EC4 (2004) are also compared with the test results. The comparisons indicate that all three codes give conservative predictions for both short and long CFST columns. The effects of concrete strength, steel strength and diameter-to-thickness ratio on the accuracy of prediction according to CECS 28:2012 are discussed, which indicate a possibility of extending the limitations on the material strengths and diameter-to-thickness ratio to higher values. A revised equation for slenderness reduction factor in CECS 28:2012 is given.

      • The Effect of Font Type and Font Size on the Legibility in Smart Watch Usage of the Elderly People

        Objective: The aim of this study is to investigate the effect of character size and font type on legibility of elderly in smart watch usage, and to suggest a design guideline for smart watch - dedicated for legibility enhancement of the elderly people. Background: As the elderly population increases, the demand for health care and relevant costs for elderly are increasing correspondingly. Hence, there have been researches to implement wearable device onto the health monitoring system for elderly. In such manner, smart watch is one of the wearables popularly studied. However, poor legibility due to the small display of smart watch is considered to be a problem for the elderly to fully use it. Thus, legibility should be improved to allow elderly to use the smart watch more conveniently. Method: The task of selecting a target stimulus within two simultaneously given stimuli on a smart watch display was given to participants. A total of 10 sets of stimuli was given, and they were in combination of single font size and type. All subjects performed the task for a total of 8 stimuli sets combining 4 font sizes and 2 font types. The dependent variable is the time to select stimulus and the error occurrence frequency: the number of interference stimulus chosen. Results and Conclusions: In Gothic type, there was no significant differences in recognition time between font sizes. However, in the case of Myung, 9pt, 10pt is better than 11pt, and 12pt. In the case of error frequency, it is shown that 10pt and 11pt results better compared to other two font sizes in case of Gothic. There was no difference in Myung. Application: The results of the study can be the useful guideline to improve legibility for the elderly.

        Circ_0001666 affects miR-620/WNK2 axis to inhibit breast cancer progression

        Background Circular RNAs (circRNAs) play important roles in the progression of various cancers, including breast cancer (BC). However, the role of circ_0001666 in BC remains unclear. Objective To explore the role of circ_0001666 in the progression of BC and reveal its potential molecular mechanism. Methods Real-time polymerase chain reaction was conducted to determine the expression of circ_0001666, miR-620 and with-no-lysine kinase 2 (WNK2). Cell counting kit 8 assay, fow cytometry and transwell assay were used to measure cell proliferation, apoptosis, migration and invasion. Western blot was utilized to examine the level of protein. Dual-luciferase reporter assay and RNA immunoprecipitation assay were used to verify the interaction between miR-620 and circ_0001666 or WNK2. Mice xenotransplantation models were built to explore the efect of circ_0001666 on BC tumor growth in vivo. Results Circ_0001666 was downregulated in BC tumor tissues and cells. Overexpressed circ_0001666 inhibited the proliferation, migration, invasion, while promoted apoptosis and tumor growth of BC in vitro or in vivo. Furthermore, circ_0001666 could serve as a sponge of miR-620. MiR-620 inhibitor hindered BC cell progression, which was similar to the efect of circ_0001666 overexpression. WNK2 was a target of miR-620, and circ_0001666 could sponge miR-620 to positive regulate WNK2. The knockdown of WNK2 reversed the efect of circ_0001666 overexpression on BC progression. Conclusion Circ_0001666 hindered the progression of BC via miR-620/WNK2 axis.

      • DNA Repair Gene Associated with Clinical Outcome of Epithelial Ovarian Cancer Treated with Platinum-based Chemotherapy

        Objective: The nucleotide excision repair (NER) and base excision repair (BER) pathways, two DNA repair pathways, are related to platinum resistance in cancer treatment. In this paper, we studied the association between single nucleotide polymorphisms (SNPs) of involved genes and response to platinum-based chemotherapy in epithelial ovarian cancer. Method: Eight SNPs in XRCC1 (BER), XPC and XPD (NER) were assessed in 213 patients with epithelial ovarian cancer using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and primer-introduced restriction analysis-polymerase chain reaction (PIRA-PCR) techniques. Results: The median progression-free survival (PFS) of patients carrying the Lys/Lys and Lys/Gln+Gln/Gln genotype of the XPC Lys/Gln polymorphism were 25 and 12 months, respectively (P=0.039); and the mean overall survival (OS) of patients was 31.1 and 27.8 months, respectively (P=0.048). Cox's multivariate analysis suggested that patients with epithelial ovarian cancer with the Gln allele had an increased risk of death (HR=1.75; 95% CI=1.06-2.91) compared to those with the Lys/Lys genotype. There are no associations between the XPC PAT+/-, XRCC1 Arg194Trp, Arg280His, Arg399Gln, and XPD Asp312Asn, Lys751Gln polymorphisms and the survival of patients with epithelial ovarian cancer when treated with platinum-based chemotherapy. Conclusion: Our results indicated that the XPC Lys939Gln polymorphism may correlate with clinical outcome of patients with epithelial ovarian cancer when treated with platinum-based chemotherapy in Northern China.

        Background: Bee venom (BV) has long been used as an oriental traditional medicine to relieve pain and to treat chronic inflammation. Objectives: However, clinical effect of BV in treating AD hasn`t been investigated in depth. This study aimed to address both clinical efficacy and mechanism of BV concerning the anti-inflammatory effect on AD-like skin lesions in Nc/NgA mice. Methods: AD-like skin lesions were induced by repeated application of the extract of Dermatophagoides farinae, and NC/Nga mice were randomly allocated into four treatment groups; non-treatment (AD), bee venom treatment (BV), vehicle treatment (Vehicle), and methylprednisolone aceponate treated group (MethylP). Each solution was applied to the AD-like skin once a day for 14 days. Results: The clinical dermatitis score was significantly improved in BV group than in AD group. Histological analysis showed that BV had both reduced hypertrophy, hyperkeratosis in the epidermis and the infiltration of inflammatory cells in the dermis. RT-PCR revealed that IL-4 and TSLP decreased in the skin of BV group, compared to AD control group and vehicle group. However, IL-17 and IFN-γ was increased in BV group. Conclusion: In conclusion, BV appears to be safe and effective to treat AD-like skin lesions in Nc/NgA mice, and may be an alternative topical medicine for topical steroid. Furthermore, BV may affect modulating immune functions such as Th2 and Th17 related cytokines in the skin of AD-like skin lesions.
