      • 마우스 Sp2/0 세포의 형질전환에 관한 연구

        Transfection, gene transfer into mammalian cells, is used to study gene structure and function. In this study, transfection of Sp2/0 cells using LK4LAC plasmid DNA(containing β-galactosidase gene) with various parameters was investigated. The transformants were identified by the β-galactosidase staning method. Optimal transfection conditions of Sp2/0-Ag14 cells were studied with two methods, calcium phosphate(CaPO_4) precipitation method and liposome method. In calcium phosphate precipitation method, the highest transfection efficiency was obtained when 20㎍ of DNA was transfected for 12hours and incubated for 16hours. In case of liposome method, the optimal. condition was achieved when 25㎍ of lipofectin amount was transfected for 18hrs. The optimal drug concentration of selection with G418 was 400㎍/㎖ in the 20 cells/dish of Sp2/0-Ag14 cell line.

      • Zn를 첨가한 고온초전도체 Y Pr Ba₂Cu₃O 의 Raman 분석

        고온초전도제 Y Pr Ba₂Cu₃O 에 Zn를 첨가(x=0.00, 0.05, 0.10, 0.15, 0.20)하여 전이온도 측정, X-ray회절 실험과 Raman 실험을 하였다. Raman 측정결과 다섯 개의 시료가 일반적인 YBCO 초천도체의 Raman 스펙트럼에서 존재하는 다섯 개의 대칭활성모드(115cm¹,150cm¹.340cm¹,440cm¹,504cm¹)가 관측되었다. 이것은 Cu 와 관련된 대칭활성모드의 주파수가 모두 저주파 변위를 나타내어 Zn가 Cu(1)과 Cu(2)에 치환되었음을 의미한다. 저주파 변위 정도는 Zn에 거의 무관했으나, Raman 스펙트럼의 각 피이크의 선폭은 치환된 Zn에 의해 증가했다. 이 결과는 Zn가 Cu자리에 어느 한계 이상은 치환되지 못하고 시료 내에서 불순물상을 형성함을 나타내는 것이다. We measured transition temperature, X-ray diffraction pattern and Raman spectra of Zn added(x=0.00, 0.05, 0.10, 0.15, 0.20) high-T superconductor Y Pr Ba₂Cu₃O All samples have been observed five symmetric activity modes(115cm,150cm¹.340cm¹,440cm¹,504cm¹)which exist in Raman spectrum of general YBCO superconductor. This means Zn is substituted for Cu(1) and Cu(2) due to the fact that symmetric activity mode frequency related with Cu shows low-frequency shift. The degree of low-frequency shift was nearly independent on Zn but line width of Raman spectra peaks were increased by the substituted Zn. This results shows that Zn does not be substituted for Cu-site more than limit value and formes impurity phase in the sample.

      • HPLC 시스템을 이용한 大氣中 多核芳香族 炭化水素의 定量에 關한 硏究

        In this study, an analytical procedure was evaluated for the determination of atmospheric PAH, both in gaseous and particulate phases. The method involves Soxhlet extraction of the filters and Tenax adsorbent, followed by a clean-up stage using a silica mini column prior to analysis by reversephase HPLC with wavelength programmable fluorescence and UV detection. A total of 18 PAH were identified and quantified, all of which have been of environmental concern. In order to validate the methodology and to ensure compatibility of the results, the analytical method used for the determination of PAH was evaluatd with respect to the efficiencies of extraction and clean-up procedure, HPLC separation, and lower limits of detection. In addition, substrate dependency of PAH recovery was also investigated for the two type of filter(glass fiber and PTEF filters) and Tenax adsorbent.

        재래종과 개량종 돼지고기의 영양성분 비교 연구

        The purpose of this study was to investigate the nutritional composition of native and hybrid pork in Korea. Three different parts (ham, loin and belly) of both native and hybrid pork were used The results were as follows : The moisture content from Korean native pork was about 60.28%, while that from the loin of hybrid one was 69% and decreased in the order of ham, loin, and belly. The highest protein content of 19.71% was found in Korean native pork loin, and Korean native pork ham had a significant amount of protein of 17.80% and hybrid one had 13.14% (p< .05). The highest crude lipid, 34.44%, was found in hybrid pork belly, Korean native pork ham had a significant amount of 5.43% and hybrid pork had 2.33% (p<.05). The highest carbohydrate content of 13.28% was found in the Korean native pork belly. The amount of ash was in the order of loin, ham and belly in Korean native pork. Among the minerals, K was found the most in Korean native pork ham (654.82㎎) and hybrid one (747. 35㎎) (p<.05). Fe was higher in the Korean native pork ham (23.03㎎), loin (15.86㎎) and belly (10.80㎎) compared to the hybrid pork ham (19.04㎎), loin (11.63㎎) and belly (7.61㎎). That was significant ham, loin(p< .01) and belly(p< .05). The main free amino acids of the native and the hybrid pork in Korea were alanine, aspartic acid and lysine. While the cholesterol content was found to be high in the order of ham, belly, and loin in the Korean native pork, in the order of belly, fresh ham and loin in the hybrid pork. The cholesterol contents in ham were significantly different between the Korean native pork(789.32㎍) and the hybrid pork (538.84㎍) (p< .01).

      • 77K와 상온에서 CO(NH₂)₂의 ¹⁴N Pulsed NQR 연구

        77K와 상온에서 CO(NH₂)₂분말 시료의 ??N Pulsed NQR실험을 하여 위와 아래 공명 진동수, 핵 사중극 결합상수, 비대칭 인자, 스핀-스핀 완화시간, 결정 격자 진동에서의 평균 비틀림 진동수, 진동자의 관성 모먼트 등을 구하였다. 공명 진동수의 온도 의존성은 전형적인 Bayer이론의 격자 진동에 의한 주파수 변화를 보였다. Bayer-Kushida이론의 고온근사식을 실험값에 적용하여 OK에서 공명 진동수 ν??와 ν??를 구했으며, 77K와 상온에서의 스핀-스핀 완화시간 T₂를 관측했다. 상온에서 T₂의 감소는 결정 내의 reorientation motion과 관련이 있을 것으로 보인다. Upper and lower resonance frequencies, nuclear quadrupole coupling constants, asymmetry parameters, spin-spin relaxation time, average torsional frequency in the crystal lattice motion and moment of inertia in the oscillator are found for CO(NH₂)₂powder sample by using ??N Pulsed NQR(Nuclear Quadrupole Resonance) technique at 77K and room temperature. Temperature dependence of resonance frequencies showed frequency variation in agreement with the typical Bayer's theory in lattice motion. Resonance frequencies ν?? and ν?? at OK were obtained by fitting the high temperature approximation of Bayer-Kushida theory to the experimental data and spin-spin relaxation time (T₂) was observed at 77K and room temperature. Decrease of T₂at room temperature was explained by the effect of reorientational motion in the crystal lattice.

      • 數種의 增量劑 粒度와 增量比率에 따른 尿素樹脂合板의 接着 特性

        The purpose of this research was to evaluate and to examine the substitutional feasibility of paper sludge extender instead of wheat flour. As the extending materials, taro flour, paper sludge, wood flour and wheat were selected and dried at 103±2℃ during 24 hours in the drying oven and pulverized into three levels(-50∼+100, -100∼+200, -200∼mesh). The extenders were mixed with urea formaldehyde resin in the ratio of 10, 20 and 30%. After plywoods were manufactured by the above extended ratios, dry and wet shear strength were examined. The results obtained at this research were summarized as follows : 1) In 10% extionsion on -50∼+100 mesh particle size, paper sludge showed the highest dry and wet shear strength among the extenders. 2) As 20% extension, wheat showed the highest value. 3) the shear strength was affected by the particle size and extending ratios, and the shear strength extended by paper sludge powder was sensitively varied with particle sizes and extending ratios. 4) The shear strength extended by taro and wheat flour were less affected by the particle size of extender. Therefore, the particle size of extender would be considered along the extention ratios and extender types.

        Since 1990s, Korea has been developing surprisingly the area of the sports club and now the area is a state of almost bringing to completion. Many researchers, engaged men in the business, and the club members related with sports club have gotten interested with the following questions. When is the sports club going to hold its development? When will be the state of the top? Besides the sports business environment with the sports club which is grown up is very different from those of the past Its part is also very necessary to a variety of marketing activities not only for promoting to keep and increase the members of the sports club, but also for competing intensively. This kind of changes in the market environment has been different according to no vacancy resulted in the increase of the sports club’s members, the changes of the customers’ consciousness, the demand increase of the outdoor sports activities progressing from the past indoor sports, the increase of spare time because of working five days a week, and participation in the sports activities according to making use of people’s spar time. Unfortunately national market environment of the sports club has been declined due to the uncertain market environment throughout domestic and foreign countries, a war between the U.S.A. and Iraq, and the nuclear crisis of the North Korea. However crisis is the very opportunity. It is time to make a new market paradigm in order to live in the difficult situations, preparing to equip some facilities and tools for sports club, developing more competitive programs, offering services for customer satisfaction. In the study, in the target for the managers of the sports club and executives, some research was carried out previously anticipated prospects and future sports club’s positive changes in order to survive as a competitive sports club which is for anticipating the environment of the sports club more exactly and for preparing those situations.
