      • 1-(5-bromo-2-hydroxy-4-methoxyphenyl)ethanone [SE1] suppresses pro-inflammatory responses by blocking NF-κB and MAPK signaling pathways in activated microglia

        <P><B>Abstract</B></P><P>Unregulated activation of microglia is a key risk factor contributes to neurodegenerative diseases and suppression of this phenomenon is considered as a potential therapeutic target. The compound isolated from sea horse <I>Hippocampus kuda</I> Bleeler; 1-(5-bromo-2-hydroxy-4-methoxyphenyl)ethanone [SE1] was characterized for its ability in suppressing LPS mediated activation of murine BV-2 cells. Despite the presence of various active molecular groups in the structure, SE1 has not well explored for its biological activities. The outcome of this study clearly indicated that SE1 inhibited the production of inflammatory mediators; nitric oxide, prostaglandin E<SUB>2</SUB> and pro-inflammatory cytokines. Furthermore, it inhibited the protein and gene expression levels of inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis factor-α, interleukin-1β and interleukin-6. The responsible signaling mechanisms leading to these inhibitions were identified as SE1 mediated blocking of phosphorylation of mitogen activate protein kinase (MAPK) molecules; C-jun-N-terminal kinase (JNK), p38 and nuclear translocation of nuclear factor-κB (NF-κB) p65 and p50 subunits. These results suggest that SE1 has the potential to be further developed as therapeutic against neuro-inflammation.</P> <P><B>Graphical abstract</B></P><P><ce:figure id='f0045'></ce:figure></P>

        Effects of Interleukin-1β, Tumor Necrosis Factor-α and Interferon-γ on the Nitric Oxide Production and Osteoclast Generation in the Culture of Mouse Bone Marrow Cells

        Nitric oxide(NO) is a labile, uncharged, reactive radical that functions as a sensitive mediator of intercellular communication in diverse tissues. It has been reported that NO is produced by osteoblast and these results may suggest that NO is integrally involved in the regulation of osteoclast formation and osteoclast resorption activity by osteoblastic cells. We examined the effect of cytokines on NO release by mouse bone marrow cell. We also examined the effects of cytokokines and sodium nitroprusside(SNP) on the formation of osteoclast-like cell from mouse bone marrow cells in culture. Cytokines stimulated NO production of mouse bone marrow cells, and N-nitro-L-arginine methyl ester, a specific inhibitor of NO synthase, suppressed the cytokine-induced NO production. SNP showed dual action in the generation of osteoclasts. The addition of 30μM SNP inhibited the formation of tartrate resistant acid phosphatase(TRAP)(+) multinucleated cell, whereas lower concentration(3μM) of SNP enhanced it. Althogh the precise action of NO remains to be elucidated in detail, the action of NO in osteoclast generation in our studies seems to be associated, at least in part, with bone matabolism and bone pathophysiology.

        첫 경련이 간질 중첩증으로 발현된 소아의 특성

        목 적 : 간질 중첩증은 신속한 치료를 요하는 신경학적 응급 질환으로 때로는 심각한 신경학적 후유증이 발생할 수 있으므로 원인 질환과 예후에 영향을 미치는 인자들에 대해 정확한 이해가 필요하다. 이에 저자들은 첫 경련이 간질 중첩증으로 발현된 소아의 원인, 임상 양상, 치료, 예후 및 신경학적 후유증의 발생 빈도의 연관 관계를 알아보고자 이 연구를 시행하였다. 방 법 : 1996년 7월부터 2002년 6월까지 인하대학교병원 소아과 및 응급실에서 첫 발작이 간질 중첩증으로 진단받은 1개월에서 5세 사이의 환아 25명을 대상으로 나이에 따른 분포, 원인 질환, 경련 형태, 지속 시간, 사용 약물, 뇌파 및 MRI의 소견, 예후를 기록지를 통하여 후향적으로 분석하였다. 결 과 : 1) 대상 환아의 남녀의 비는 2.1:1이었고 대상 환자 25명 중 남아 17명(68%), 여아 8명(32%)이었으며 연령은 20±1.6개월로 대부분 3세 이상(80%)에서 발생하였다. 2) 경련 형태는 전신 강직-간대형이 60%, 전신 강직형 16%, 부분 간대형이 16%, 부분 발작형이 8%이었다. 3) 원인 질환과 예후는 증후성인 경우가 64%였으며 특발성인 경우가 36%였다. 증후성의 경우 발열이 40%, 중추 신경계 감염이 20%, 허혈성 손상이 4%이었으며 원인 질환이 있는 경우와 그렇지 않은 경우와의 통계학적 유의성은 없었다. 4) 경련의 지속시간은 30분-1시간이 17례(68%), 1-2시간이 4례(16%), 2시간 이상이 4례(16%) 이었다. 지속 시간이 짧을수록 예후가 좋은 경향을 보였다. 5) 사용 약물은 phenobarbital, phenytoin, midazolam, valium, valproic acid이었다. 약물 한 가지로 조절된 경우 13례(52%), 두 가지로 조절된 경우 11례(44%), 세 가지로 조절 된 경우 2례(4%)이었으며 세 군간의 예후를 비교해 보았을 때 한 가지로 조절된 군이 다른 군 보다 예후가 좋은 경향으로 나타났다. 6) 뇌파는 20례에서 시행하였는데 정상 7례(35%)이었고, 13례(65%)는 비정상이었다. 비정상은 전기적인 지속경련을 보인 경우 6례(30%), 배경파 이상을 보인 경우 6례(30%), 국소적인 간질파를 보인 경우 1례(5%)이었다. MRI는 19례에서 시행하였는데 정상 소견 10례(53%), 이상 소견 9례(47%)이었다. 7) 원인이 발열인 경우에서의 예후는 정상을 보인 경우가 6례, HHE를 보인 경우가 2례, 복합 부분 발작을 보인 경우가 1례, 정신 지체를 동반한 뇌성 마비가 1례였다. 결 론 : 첫 경련이 간질 중첩증으로 발현된 소아는 대부분 3세 이하였다. 경련 시간이 1시간 이내, 한 가지 항경련제로 경련이 조절되는 경우는 예후가 좋았고, 경련 시간이 1시간 이상이거나, 여러 가지 항경련제를 사용하거나, 원인 질환이 있는 경우는 예후가 나빴다. Purpose : Status epilepticus(SE) is a pediatric and neurologic emergency associated with significant morbidity and mortality. Prompt recognition and management are needed for successful outcomes. We evaluated, clinical manifestations, treatment and prognosis of the patients whose first seizure attacks were presented as SE. Methods : According to the department of pediatrics and the emergency room in Inha University Medical Center, there were a total of 25 children between 1 month and 5 years old who were diagnosed as status epilepticus from July 1996 to June 2002. Also, their medical records were reviewed and analyzed interms of age distribution, accompaning diseases, the types and duration of convulsion, medications, EEG and MRI findings and prognosis. Results : Twenty-five cases were studied. The mean age at the time of the diagnosis was 20±1.6 months and 80% of the patients were less than 3 years old. The most common type of the status epilepticus was generalized tonic clonic seizure comprising 15 cases(60%). 64% of the patients were symptomatic: fever(40%), CNS infection(20%), ischemic injury(4%) while 36% were idiopathic. Seizure attacks were terminated within an hour in seventeen patients(68%) and they controlled by one antiepileptic drug in thirteen patients(52%). Of the 20 EEGs, abnormal findings were shown in 13 cases(65%); namely, electrical seizure(30%), abnormal background(30%), and focal epileptiform discharge(5%). Of the 19 brain MRIs, abnormal findings were shown in 9 cases(47%). Conclusion : The children whose first seizure attack were presented as SE were less than 3 years old. The prognosis is good in that most of the seizure attacks were terminated within an hour and controlled by one epileptic drug. However those seizure attacks with longer duration, multiple antiepileptic druge and underlying causeare had poor prognosis.

        In order to develope an effective production method of Atrophic Rhinitis (A. R.) vaccine, the physiological characteristics of Bordetella bronchiseptica, causative pathogen of A. R. in swine, were investigated by using the immunological techniques. The optimum culture conditions for the cell growth and epitome production of Bordetella bronchiseptica were monosodium glutamate 2.0%, yeast extract 1.0%, magnesium chloride 0.1%, and riboflavin 0.003%, initial pH 7.4 for 48hrs.

        Franz Kafka erkla¨rt in seinem Tagebuch die ju¨dische Literatur in Warschau oder in Prag mit dem Begriff der dleinen Literatur. Die kleine Literatur ist nicht die Literatur einer kleinen Sprache, sondern die einer minderheit, die sich einer groBen Sprache bedient. Nach deleuze und Guattari hat die kleine Literatur drei Merkmale, " Deterritoriaiisierung der Sprache", "Koppelung des Individuellen ans unmittelbar Politische", und "kollektive Aussageverkettung". Kiese Merkmale dienen Zum einheitlichen Zusammenhalten des nationalen BewuBtseins. Das wird nicht etwa durch die einhejtliche Greschichte oder durch die intersubjektive Vereinigung erreicht. Die kleine Literatur versteht Klfka vielmehr als Tagebuchfuhren einer Nation, das etwas ganz anderes ist als Geschichtsschreibung. Damit betont Kafka die Vielfalt der Teile statt eines einheitlichen Ganzen. Dieser Entwurf kommt in Kafkas spa¨teren Erza¨hlungen zum Ausdruck, Beim Bau der Chinesischen Mauer und Josefine, die Sa¨ngerin oder das Volk der Ma¨use, in denen Legenden oder Sagen eine groBe Rolle spielen. Diese Erza¨hlformen wird durch ein diskontinuierliches Nebeneinander der Teile gekennzeichnet, wie das Tagebuch. In Beim Bau der Chinesischen Mauer thematisiert Kafka das gewaltige Mauerwerk Chinas und das Kaisertum. Die Mauer, die durch das System des Teilbaus wegen des unermesslich groBen Raums gebaut wird, produziert immer wieder eine neue Legende. Der Inhalt der Legende ist weder nachpru¨fbar noch feststellbar. Trotzdem haben die mannigfaltigen kleinen Erza¨hlungen u¨ber den Mauerbau die funktion, die Selbstu¨berwindung des Einzelnen zu ermo¨glichen und das Volk zur Einheitlichkeit zu fu¨hren. Der Kaiser existiert auch nur in der Legende, in der die Hierarchie zwischen Oben und Unten praktisch nicht funktioniert. Die Folg ist ein gewissermaBen freies, unbeherrschtes Leben. In Josefine, die Sa¨ngerin oder das Volk der Ma¨use entwirft Kafka eine Sagenwelt, in der alle Mitglieder des Volkes ranggleich sind. Die Erza¨hlung konzentriert sich auf die Frage, wie das einzelne Pfeifen, das jeder als eine charakteristische Lebensa¨uBerung hervorbringt, denn als eine eigensta¨ndige Kunst, wie Josefine es behauptet, aufzunehmen sei, Josefines Behauptung widerspricht anscheinend dem Umstand, daB alle gleich sind. Aber Gleichheit impliziert, daB jeder die potentielle Mo¨glichkeit hat, ein Held oder ein Ku¨nstler zu sein. Im BewuBtsein des Volkes ist Josefines Musik auf die gro¨Bte mo¨gliche Nichtigkeit, das pfeifen, reduziert. Obwohl Josefine die Einzigartigkeit ihrer Stimme behauptet, liegt ihre Leistung vielmehr in der Allgemeingu¨ltigkeit ihres Pfeifens. Selbst wenn Josefine als das zeichen eines Protestes verschwindel, ist ihre Musik weder einmalig noch letztmalig. Sie ist nur eine kleine Episode in der ewigen Geschichte, die einem der zahllosen Helden geho¨rt.

      • 등방 난류 유동장내 난류 예혼합 화염 전파의 통계적 시뮬레이션

        등방 난류 유동장내를 전파하는 난류 예혼합 화염에 관한 이론적인 연구를 수행하였다. 통계적 시간 급수 방법을 사용하여 화염 표면을 따라 미연 가스 영역의 난류 유동장을 모사 (simulate) 하고 2차원 화염면 운동 알고리즘을 사용하여 화염 전파에 따르는 화염면의 성질을 수치적으로 모사하였다. 본 연구의 목적은 난류 예혼합 화염의 기하학적 구조와 전파 속도를 효율적으로 예측할 수 있는 계산법을 개발하고 검증 하는데 있다. 계산 결과는 다양한 난류 강도와 레이놀즈수를 갖는 수소-공기 혼합 가스에 대하여 측정된 기존의 실험 결과와 비교하였다. 본 연구에서 제안된 2차원 계산은 화염 반경 성장율, 화염 반경 변형율, 프랙탈 차수등에서 측정치와 유사한 경향을 보였으며, 이 방법이 보다 복잡한 형태의 난류 화염 전파에 효율적으로 적용될 수 있음을 확인하였다. A theretical investigation of turbulent premixed flames propagating in isotropic turbulence is presented. Flame surface properties were numerically simulated using a two-dimensional flame propagation algorithm combined with statistical time series simulation of unburned gas velocities along the flame surface. The objective is to develop and test an efficient technique to predict the propagation speed of flame as well as the geometric structure of the flame surfaces. Predictions were compared with existing experimental data for flames propagating in a closed vessel charged with hydrogen/air mixture with various turbulence intensities and Reynolds numbers. Two dimensional time dependent simulation resulted in correct trends of the measured flame data. Comparisons were made in flame radius growth rate, rms flame radius fluctuations, and average perimeter and fractal dimensions of the flame boundaries. The reasonable behavior and high efficiency proves the usefulness of this method in complex problems of flame propagation.

        유용성 도료와 수용성 도료의 유해성 비교에 관한 연구 : 자동차 보수용 도료를 중심으로

        The purpose of this study is to substitute water-based painting materials for the current solvent-based ones used in motor-repairing process to minimize the exposure of organic solvents to the painters. This study assessed the exposure of organic solvents to the painters using water-based and solvent-based painting materials and compared compositions, painting processes and the health hazards of the application of these alternative painting materials. The results of this study are as follows. 1. solvent-based painting materials used in motor-repairing process consist of various organic solvents, which consist primarily of toluene, xylene, ethyl benzene, ethyl methyl bezene, trimethyl bezene, ethyl acetate, butyl acetate, methyl isobutyl ketone, 2-ethoxy ethanol, 2-ethoxy ethyl acetate and toluene-2.4-diisocianate and the others. These organic solvents are know as health-hazardous substances. But water-based painting materials are high-solid, low-solvent ones and consist of such two organic solvents as 2-butoxy ethanol and 2-heptanone and the others. 2. The painters using solvent-based painting materials in motor-repairing process are exposed to various organic acetate, methyl isobutyl ketone, trimethyl benzene, 2-ethoxy ethanol, and 2-ethoxy ethyl acetate. But the painters using solvent-based ones are only exposed to 2-butoxy ethanol and 2-heptanone. 3. By using water-based painting materials in stead of solvent-based painting materials containing health-hazardous organic solvents, the exposure of such organic solvents in the painter's breathing zone can be largely prevented. 4. This study recommends water-based painting materials as substitutes for the current solvent-based ones used in motor-repairing process to minimize the exposure of organic solvents to the painters.
