Therapeutic efficiency of adipose-derived mesenchymal stem cells in healing of experimentally induced gastric ulcers in rats

        Safaa A. Hassan,Amal Taha Abou Elghait,Zainab S. Abdelqader,Fatma Y. Meligy 대한해부학회 2021 Anatomy & Cell Biology Vol.54 No.3

Gastric (peptic) ulcer is a major gastrointestinal disorder with high morbidity and mortality. While several drugs have been used to treat gastric ulcers, such as proton pump inhibitor-based triple therapy for Helicobacter pylori eradication, but hey result in adverse side effects. Therefore, development of new alternative therapies is desirable. Many recent studies have shown that mesenchymal stem cells (MSCs) might have an enhancing effect on the ulcerated gastric mucosa. The aim of this study is to evaluate the efficacy of MSCs in the treatment of indomethacin-induced gastric ulcer, and to compare it with the normal ulcer autohealing. This work was performed on 36 adult male albino rats, divided into four groups: Group I (control group), Group II (ulcer group), Group III (autohealing group), and Group IV (stem cells-treated group). The histological changes of gastric mucosa were examined in sections stained with H&E using light microscope for expression of vascular endothelial growth factors (VEGF) and proliferating cell nuclear antigen (PCNA) in immunohistochemical stained sections using image analyzer. The results from MSCs-treated group revealed restoration of the normal architecture of the gastric mucosa with comparison to the autohealing group which showed excessive granulation tissue and heavy cellular infiltration with disorganized architecture of the fundic mucosa. Immunohistochemical examination showed strong expression of both VEGF and PCNA in the MSCs-treated group. So it was concluded that MSCs accelerate gastric ulcer healing when injected intraperitoneally, compared to autohealing process which showed delayed healing.

        ( Safaa A. S. Al-qaysi ),( Halah Al-haideri ),( Sana M. Al-shimmary ),( Jasim M. Abdulhameed ),( Othman I. Alajrawy ),( Mohammad M. Al-halbosiy ),( Tarek A. A. Moussa ),( Mohamed G. Farahat ) 한국미생물생명공학회(구 한국산업미생물학회) 2021 Journal of microbiology and biotechnology Vol.31 No.5

Levan is an industrially important, functional biopolymer with considerable applications in the food and pharmaceutical fields owing to its safety and biocompatibility. Here, levan-type exopolysaccharide produced by Pantoea agglomerans ZMR7 was purified by cold ethanol precipitation and characterized using TLC, FTIR, 1H, and 13C NMR spectroscopy. The maximum production of levan (28.4 g/l) was achieved when sucrose and ammonium chloride were used as carbon and nitrogen sources, respectively, at 35°C and an initial pH of 8.0. Some biomedical applications of levan like antitumor, antiparasitic, and antioxidant activities were investigated in vitro. The results revealed the ability of levan at different concentrations to decrease the viability of rhabdomyosarcoma and breast cancer cells compared with untreated cancer cells. Levan appeared also to have high antiparasitic activity against the promastigote of Leishmania tropica. Furthermore, levan had strong DPPH radical scavenging (antioxidant) activity. These findings suggest that levan produced by P. agglomerans ZMR7 can serve as a natural biopolymer candidate for the pharmaceutical and medical fields.

Camellia sinesis leaves extract ameliorates high fat diet-induced nonalcoholic steatohepatitis in rats: analysis of potential mechanisms

Mohamed Safaa H.,Shahat Abdelaaty A.,Mohamed Mohamed Ragaa,Khalil Wagdy K. B.,Salem Ahmed M.,Farrag Abdel Razik H.,Ahmed Hanaa Hamdy 한국약제학회 2021 Journal of Pharmaceutical Investigation Vol.51 No.2

Purpose Our research aims to address and determine the effect of Camellia sinensis extract in the management of nonalcoholic steatohepatitis (NASH) in rats. Methods Forty adult female albino rats were divided into four groups. Group 1 (G1) served as the control group, while the other three groups received high-fat diet for 32 weeks to induce NASH and then were later assigned to the following groups: (G2) NASH-afflicted group which was left untreated, (G3) NASH-afflicted group treated with Camellia sinensis extract in a dose of 400 mg/kg, and (G4) NASH-afflicted group treated with Camellia sinensis extract in a dose of 200 mg/kg. Results Significant elevation in serum alanine aminotransferase, albumin, bilirubin (total and direct), cholesterol, low density lipoprotein, triglycerides, leptin, Cox-2, and CD40 values was recorded. Moreover, overexpression of hepatic tumor necrosis factor alpha and hepatocyte growth factor genes were recorded, whereas blood platelet count and serum high density lipoprotein concentration revealed significant depletion, which was paralleled by significant downregulation of hepatic adiponectin gene expression level in NASH group versus the control group. On the opposite side, treatment of NASH groups with two different doses of Camellia sinensis extract reversed the values of the measured biochemical parameters and the targeted gene expression levels when compared with the NASH group. Optical micrograph of liver tissue sections of rats treated with Camellia sinensis extract showed the observed improvement in the studied biochemical and genetic markers. Conclusion This study provides a clear evidence for the promising therapeutic potential of Camellia sinensis extract against NASH. This could be ascribed to its hepatoprotective activity, hypolipidemic effect, and anti-inflammatory potency.

      • Enhanced MPR Selection Strategy for Multicast OLSR

Matter, Safaa S.,Al Shaikhli, Imad F.,Hashim, Aisha H.A.,Ahmed, Abdelmoty M.,Khattab, Mahmoud M. International Journal of Computer ScienceNetwork S 2022 International journal of computer science and netw Vol.22 No.10

Wireless community networks (WCNs) are considered another form of ownership of internet protocol (IP) networks, where community members manage and own every piece of equipment in a decentralized way, and routing for traffic is done in a cooperative manner. However, the current routing protocols for WCNs suffer from stability and scalability issues. In this paper, an enhanced routing protocol is proposed based on the optimized link state routing (OLSR) protocol to meet the standards of efficiency in terms of stability and scalability. The proposed routing protocol is enhanced through two phases: multicasting expansion and multipoint relay (MPR) selection based on an analytical hierarchical process (AHP). The experimental results demonstrate that the proposed routing protocol outperforms the OLSR protocol in terms of network control overhead and packet delivery ratio by 18% and 1% respectively.

Lead Removal from Aqueous Solution by Green Solid Film Based on Cellulosic Fiber Extracted from Banana Tree Doped in Polyacrylamide

        Amr Abdelkhalek,Safaa S. M. Ali,Zhanwu Sheng,Lili Zheng,Mohamed Hasanin 한국섬유공학회 2022 Fibers and polymers Vol.23 No.5

In the present study, we investigated the lead ions removal on the solid form (as strips) by adsorption on greencellulosic fiber/polyacrylamide (GCFP). Strips were prepared in a solid form, not a hydrogel, for performing the adsorptionprocess without squandering of water. Then, the prepared film and its original materials were characterized using attenuatedtotal reflection infrared spectroscopy (ATR-FTIR), and scanning electron microscopy with energy dispersive electronspectroscopy (SEM-EDX). The experiments were conducted under different operating conditions, such as contact time, initial Pb concentration, adsorbent dose, and pH value. The adsorption process mechanism was tested by applying twokinetic models to the experimental data, which are the pseudo-first order and the pseudo-second order, and intraparticlediffusion models. The equilibrium results were fitted to three isotherm models, namely Langmuir, Freundlich, and Dubinin-Radushkevich (D-R) models. We found a Pb removal yield of 98 % (approximately 128 mg/g) at pH=7, an adsorbent doseof 0.4 g, an initial Pb concentration of 50 ppm, and a contact time of 60 min. The kinetic study results showed that thepseudo-second-order kinetic model provided superior correlation for the adsorption of Pb ions. Moreover, Dubinin-Radushkevich isotherm model had better-fit adsorption data. The obtained results revealed the high efficiency of GCFPfilms in lead ions adsorption from water. Additionally, the components of these films, represented here by banana pseudostemwaste, are green sources, inexpensive and easy to obtain, whereas this waste is a burden on the farmers due to itsdifficult disposal.

Short and Long RC Columns with Internal WWM Reinforcement under Concentric and Eccentric Compression

        Ahmed M. El-Kholy,Safaa F. Abd El-Rahman,Mohamed M. El-Assaly 한국콘크리트학회 2023 International Journal of Concrete Structures and M Vol.17 No.3

Recently, various types of steel meshes were used as additional internal reinforcement for improving the confinement, ductility, and strength of the reinforced concrete (RC) columns. In this experimental study, a welded wire mesh (WWM) layer was used as an internal reinforcement in addition to the traditional steel reinforcement (longitudinal bars and transverse ties) for short and long RC columns under concentric and eccentric compression. Thirty-six square RC columns with two slenderness ratios λ (height to width ratio) of 9 and 18 were tested under compression with eccentricity ratios e/t (eccentricity to section thickness ratio) of 0, 0.13, and 0.26. The reference columns were traditionally reinforced with longitudinal steel bars and transverse ties with a reference volumetric ratio ρr of 0.44%. The other columns comprised a WWM layer wrapped outside the ties whose volumetric ratio ranged from 0.22% to 0.44%. The results demonstrated that the columns reinforced with a WWM layer in addition to traditional reinforcement showed an improvement in ductility and strength compared to those reinforced with longitudinal bars and transverse ties only. A WWM layer increased the ultimate load of the columns comprising ρr ties by approximately 16% and 9% for short and long columns, respectively. These improvements were proportional to the ties volumetric ratio.

Histological study on the effect of nicotine on adult male guinea pig thin skin

        Sohair A. Eltony,Safaa S. Ali 대한해부학회 2017 Anatomy & Cell Biology Vol.50 No.3

obacco smoking has been identified as an important factor in premature skin aging to detect the histological changes occurred in adult male guinea pig thin skin under the influence of low and high doses of nicotine; which constitutes approximately 0.6%-3.0% of the dry weight of tobacco. Fifteen adult male pigmented guinea pigs were equally divided into three groups: group I, control; group IIA, low dose nicotine treated; 3 mg/kg subcutaneously for 4 weeks; and group IIB, high dose nicotine treated; 6 mg/kg subcutaneously for 4 weeks. Specimens from the back thin skin were processed for light and electron microscopy. Nicotine administration revealed flattened dermo-epidermal junction and reduced rete ridges formation. Collagen bundles were disorganized with increased spaces between them. A reduction in the amount of elastic fibers in the dermis were also observed compared to group I. Ultrastructurally, keratinocytes had hyperchromatic nuclei, intracytoplasmic vacuoles, disruption of desmosomal junctions, irregular tonofilaments distribution, and increased inter-cellular spaces. These changes were more pronounced with high dose nicotine administration. The epidermal thickness was reduced in low dose nicotine administration. But, high dose nicotine administration revealed increased epidermal thickness compared to the control group. Nicotine induced structural changes of adult male guinea pig thin skin. These changes were more pronounced with high dose nicotine administration.

Use of DNA adduct and histopathological defects as indications for bio-persistence potency of zinc oxide nanoparticles in gastropod, Monacha cartusiana (Mǜller) after short-term exposure

        Khaled Yassin Abdel-Halim,Safaa Ramadan Osman,Atef Mohamed Khedr Nassar,Alaa Khozimy,Heba Mohamed El-Danasoury 환경독성보건학회 2022 환경독성보건학회지 Vol.37 No.3

The cytotoxic effects of metallic nanoparticles (MNPs) might be revealed in genomic and histopathological defects. Therefore current study aimed to assess the bio-persistence and adverse effects potency of zinc oxide nanoparticles (ZnONPs) in the gastropod, Monacha cartusiana. Gastropods were exposed to 74 μg/mL for 14 d then the DNA adduct and histopathological defect profiles were assessed. The results demonstrated significant decline in the estimated genomic template stability (GTS%) in haemolymph and digestive gland ranging from 10.0 to 42.9% in treated animals compared to controls. In the treated and recovered snails, randomly amplified polymorphic (RAPD)-DNA showed the appearance and/or disappearance of DNA bands, indicating DNA damage due to the cytotoxicity of ZnONPs on gastropods. Significant defects in microvilli (MV), nucleus (N), mitochondria (M), and execratory glands (EXG) were noticed in the treated individuals with respect to controls. The remaining live animals were subjected to a recovery period (14 d, without treatment) and slight recovery profiles were reported for both measures compared to the control group. The recovery pattern was recognized in the nucleus/cytoplasm ratio with 0.186 and 0.428 in the treated and recovered groups concerning their control (0.176). The studied parameters reported herein might be reliable tools to assess accumulation and bio-persistence patterns of NPs in the organisms for short-term exposure indicating the cytotoxic and genotoxic effects. Also, gastropods may provide simple models for evaluating the ecotoxicological effects of nanomaterials.
