        Antioxidant Activity of Various Extracts from an Edible Mushroom Pleurotus eous

        Pleurotus eous is a pink edible oyster mushroom cultivated in Coimbatore. The aim of present study was to evaluate antioxidant potential of ethyl acetate, methanol,and hot water extracts from P. eous using various in vitro ROS/RNS generated chemical and biological models. Results demonstrated that the graded-dose (2-50 mg/mL)of various extracts markedly scavenged the DPPH, ABTS,and hydroxyl radicals, and showed metal chelating ability,reducing capacity in Fe3+/ferricyanide complex, and ferric reducing antioxidant power assays. In biological models,different extracts were found to inhibit lipid peroxidation in brain and liver homogenate. Total antioxidant activity was high in methanolic extract. Hot water extract showed higher amount of total phenol and methanolic extract showed higher amount of flavonoid as compared to other extracts. Based on EC50 values it can be concluded that the various extracts from P. eous were good in antioxidant properties with methanol and ethyl acetate extracts being more effective. The results of this study showed that various extracts could serve as natural antioxidants owing to their significant antioxidant activity.

        Antidiabetic and antioxidant effect of methanol extract of edible plant $Talinum$ $portulacifolium$ (Forssk) in Streptozotocin induced diabetic rats

        The aim of this study is to evaluate antidiabetic and antioxidant effects of methanol extract of $Talinum$ $portulacifolium$ (FORSSK) leaves (Portulacaceae) (METP) in streptozotocin induced diabetic rats. The acute and sub acute antihyperglycemic effect of two different doses (200 and 400 mg/kg,) of METP were investigated. Blood glucose level, serum insulin level, lipid profiles and biomarker enzymes were evaluated in normal and diabetic rats. Liver glycogen content, kidney and liver antioxidant enzymes were evaluated. Supplementation of METP by oral gavage significantly reduces the fasting blood glucose level. This supplementation significantly increases the glycogen level in the liver ($32.33{\pm}1.453$ mg/gm) as compare to diabetic control group ($5.66{\pm}0.881$ mg/gm) ($p$ <0.001). Elevated serum glutamate oxaloacetate transaminase (SGOT), glutamate pyruvate transaminase (SGPT), and alkaline phosphatase (SALP) were decreased significantly by the treatment of METP with respect to diabetic group. The antioxidant activity was studied in liver and kidney tissues by measuring malondialdehyde (MDA), reduced glutathione (GSH) and catalase levels. In the METP treated groups, the HDL/LDL ratio was improved and cholesterol and triglycerides levels also decreased significantly. Histopathological studies of the pancreas of these animals showed comparable regeneration by METP. Glibenclamide was used as a standard at a dose of 0.5 mg/kg p.o.

        The aim of the study is to investigate the antioxidant activity through, reducing power, 2, 2-diphenyl-1-picryl hydrazyl radical (DPPH), nitric oxide radical (NO), superoxide radical, hydrogen peroxide radical ($H_2O_2$) scavenging activity and the amount of total phenolic compounds of chloroform, ethyl acetate, methanol and aqueous extracts of the leaves of Talinum portulacifolium. Chloroform extract of leaves of T. portulacifolium showed highest antioxidant activity, with a direct relationship between activity and concentration of extracts ($15-240\;{\mu}g/mL$). Among all the extracts, the highest amount of the total polyphenolic compounds was found in the chloroform extract. Chloroform extract of T. portulacifolium showed an important free radical scavenging activity towards the DPPH, NO, Superoxide and $H_2O_2$ radicals, with $IC_{50}$ values of 133.26, 165.75, 156.34 and $135.29\;{\mu}g/mL$, respectively. In the lipid peroxidation assay, extracts of chloroform and ethyl acetate showed a remarkable inhibitory activity. The extracts showed significant activity in all the experiments but lower than the standard antioxidant, ascorbic acid.

        Surveillance of Burkholderia spp. in Bukit Merah Orang Utan Island (BMOUI), Perak, Malaysia

        Melioidosis is an infection caused by Burkholderia pseudomallei and it represents one of the most common infectious diseases in zoo and farm animals giving rise to high mortality, leading to biological diversity loss. We use the bacteriological and biochemical methods such as culture, biochemical test, Gram-staining and oxidase test to examine the presence of B. pseudomallei in 50 soils treated and non-treated with quicklime powder collected from Bukit Merah Orang Utan Island, (BMOUI), Perak, Malaysia. We observed mixed bacterial colony growth with various colony morphologies. Six of the isolates labelled as P2(25), P5(2), P5(3), P5(5), P6(1), and P6(6)2 showed pinkish purple colour with wrinkled characteristics similar to B. pseudomallei colony morphology after incubation for 48 h. The results showed that all putative soil-isolated organisms were Gram-negative bacteria, positive for oxidase test and negative for catalase test. In the VITEK® ID system and PCR sequencing analysis, no B. pseudomallei was identified. All six putative organisms were identified as in B. cepacia complex i.e.: B. stagnalis (four cases), B. contaminans (one) and B. multivorans (one). These preliminary findings demonstrate that a few species of B. cepacia group were successfully isolated from the soil of BMOUI using selective media for B. pseudomallei.

        A common coding variant in CASP8 is associated with breast cancer risk

        The Breast Cancer Association Consortium (BCAC) has been established to conduct combined case-control analyses with augmented statistical power to try to confirm putative genetic associations with breast cancer. We genotyped nine SNPs for which there was some prior evidence of an association with breast cancer: CASP8 D302H (rs1045485), IGFBP3 −202 C → A (rs2854744), SOD2 V16A (rs1799725), TGFB1 L10P (rs1982073), ATM S49C (rs1800054), ADH1B 3′ UTR A → G (rs1042026), CDKN1A S31R (rs1801270), ICAM5 V301I (rs1056538) and NUMA1 A794G (rs3750913). We included data from 9–15 studies, comprising 11,391–18,290 cases and 14,753–22,670 controls. We found evidence of an association with breast cancer for CASP8 D302H (with odds ratios (OR) of 0.89 (95% confidence interval (c.i.): 0.85–0.94) and 0.74 (95% c.i.: 0.62–0.87) for heterozygotes and rare homozygotes, respectively, compared with common homozygotes; P<SUB>trend</SUB> = 1.1 × 10<SUP>−7</SUP>) and weaker evidence for TGFB1 L10P (OR = 1.07 (95% c.i.: 1.02–1.13) and 1.16 (95% c.i.: 1.08–1.25), respectively; P<SUB>trend</SUB> = 2.8 × 10<SUP>−5</SUP>). These results demonstrate that common breast cancer susceptibility alleles with small effects on risk can be identified, given sufficiently powerful studies.

        혈중 Paraquat 농도가 Paraquat 중독환자의 생존율에 미치는 영향

        N/A Objectives: Assessment of the severity and prognosis of paraquat poisoning and the need to institute active therapy has been based on the quality alleged to have been taken and the intensity of the blue colour obtained on testing the urine with alkali/dithionite. Such arbitrary methods are fat from satisfactory and may be led to unnecessary vigorous treatment of some petients and, possibly, failure to treat others who might. have benefited. Material: 19 patients for whom the time of paraquat ingestion was known with reasonable certainty were admitted to Soonchyang University Chunan hospital, were investigated, Urine test with alkal/dithionite was positive in all of patients.Venous blood samples were collected on admission to the emergency room and repeatedly every 6hours during 12-18 hours. All patients has treated with same protocol in the text. Methods: Plasma paraquat concentrations were measured by radioimmunoassay. Urine paraquat concentratioons were measured by alkali/dithionite method. Prognostic significanse of plasma paraquat concentrations and the relation to urine concentration were described. Results: Plasma paraquat concentrations was not detected in 5 out of 19 patients and all of these patients survived. Plasma paraquat concentratopn was detected in 14 patients. It was distributed very widly. Among these 14 cases, 3 cases wer survivors and 11 cases died. In died patients, the peak levels of plasma parasquat ranged between 0.16mg/ml-3.13mg/ml (mediam;0.87mg/ml). Plasma paraquat concentration of the 3 survivors decreased to near zero levels in 12 hours. There was a significance relation between plasna parquat conentrations measurd by radioimmunoassay and ILD urine paraquat concentrations measured by alkali/dithionite method. Conclusion: Eliminatjon rate of plasma paraquat seems to be a more significant factor for the survival rate than the intiial plasma concentration of the paraqrat. Because of its simplicity, and low cost, urine paraquat conecentrations measured by alkali/ dithiontit method is coplementary to the plama paraquat conentrations measured by radioimmunoassay.
