      • 운동과 적혈구 노화와의 관계

        본 연구는 노화의 정도에 따라 적혈구를 분리하고 장시간의 운동 스트레스가 노화에 미치는 영향을 알아 보고자, 고도로 훈련된 남자 마라톤 선수 3명을 대상으로 HRmax 80% 수준에서 지속적인 런닝을 실시하였다. 전혈에서 RBC, WBC, MCV, Hb, Hct를 측정하고, 적혈구 내에 존재하는 Protein Carboxyl O-Methyltransferase(PCM)을 정제하여 그 activity를 측정하고, 적혈구 막을 분리하여 그 구성 단백질에 대한 PCM 의 Methylatability를 측정하여 운동과 적혈구 노화와의 관계를 설명하고자 한다. 각 시간별, Rest(0분), Running(30, 60, 90, 120분), Recovery(10, 30분)에 혈액을 채혈하여, 전혈에서 RBC, WBC, MCV, Hb, Hct의 변화를 조사하고 (Hematology Analyzer 5), 적혈구를 분리하여 50% Prcoll과 0.154M Hypaque에 넣고 22,000??g에서 40분간 원심분리하면 11 -15개 정도의 적혈구 층이 노화된 정도에 따라 분류된다. 적혈구 세포내의 PCM을 정제하고, 적혈구 막(ghost)을 분리하여 방사성 동위원소 (??C-SAM)를 methyl donor로 하여 PCM activity와 적혈구 막에 대한 Methylatability를 측정하였다. 런닝 30분에 RBC수가 증가하였으며, WBC와 Hb은 운동하는 동안 높은 수준을 유지하였다. MCV와 Hct는 변화가 없었다. Percoll과 Hypaque를 이용하여 노화된 정도에 따라 적혈구를 분류하였으나, 각 층에 대한 세포수의 측정이 뒤따르지 못하여 적혈구 수의 증가 혹은 감소와의 관계를 직접 볼수 없었다. 적혈구 내에서 정제된 PCM activity 는 운동 스트레스에 의한 변화를 보이지 않음으로써 항상 full을 유지하고 있음을 알 수 있다. 적혈구 막에 대한 Methlatability변화는 운동전 26.77±0.32 units/mg.min에서 런닝 30분에 34.29±0.55 units/mg.min로 큰 증가를 보였으며 런닝하는 동안 높은 수준을 유지하였다. 이는 운동 스트레스가 적혈구 막단백질을 구성하는 아미노산에 영향을 미쳐 비정상의 L-isoaspartyl 잔기를 만들었음을 말해주는 것이며, 이것이 노화된 단백질의 수복기전과 일치한다. 즉 PCM이 비정상의 L-isoaspartyl 잔기를 메칠화시켜 정상의 L-aspartyl잔기로 수복시킴으로써 운동 스트레스에 의한 영향을 극복하고자 하는 것이다. 그러므로 장시간의 운동 스트레스가 주는 영향이 노화에 의한 현상과 일치함을 알수 있다. Our studies showed the separation method of erythrocyte according to aging process and the effect that influenced a long-time exercise stress to aging. Highly trained marathon runners (n=3) performed a prolonged running(2hours) at the level of HRmax 80% RBC,WBC,MCV, Hct was measured in a whole blood.Protein Carboxyl O-Methyltransferase(PCM) in the erythrocyte was purified from the cytosol and the activity was measured. Methylatability of erythrocyte membrance protein was measured during marathon performance and recovery phase. Therefore, we interpreted the relation of exercise and erthrocyte aging. The count of RBC increased at the running 30 min. WBC and Hb maintained at the high level during marathon running. MCV and Hct did not changed. Erythrocyte divided into 11-15bands by Percoll and Hypaque according to aging. Because the erythrocyte did not count numerically in different age-related fraction of human erythrocyte, we cannot show the change of erythrocyte count. PCM activity purified in erythrocyte did not change by exercise stress, this suggests that PCM is full at all times. Methylatability of erythrocyte membrane protein changed from 26.77±0.32 units/mg.min before exercise to 34.29±0.55 units/mg.min at 30min after exercise and maintained a high level of methylatability during a running. This suggests that abnormal L-isoaspartyl residues came into being in the membrane protein composition as a result of exercise stress and are repaired by the same mechanism as aging repair that is, PCM methylated abnormal L-isoaspartyl residues in membrane protein and repaired into normal L-aspartyl residues, which removed the effect of exercise stress. Therefore, this study showed that the effect by exercise stress is the same as that of aging process.

      • 구강점막 상피성 종양의 케라틴 분포에 관한 면역조직화학적 연구

        The keratins are a group of water-insoluble proteins that form 10nm desmosome-associated tonofilaments in all epithelia and used as a sensitive marker for epidermal differentiation. Keratin expressions are may affected by various factors and the pattern of expression was concerned with the distribution of keratins in benign and malignant lesions. In this study, immunohistochemical examination was performed to detect cytokeratin patterns in epithelial neoplasms of oral regions, known to be associated with human papillomavirus and results obtained are as follows. 1. AEl antibody was diffusely stained in all layers of inverted papilloma and verrucous carcinoma, but in papilloma, AEl antibody was restricted in suprabasal layer. In squamous cell carcinoma, cells with variable staining intensity were present in highly disorganized fashion. 2. CK19 antibody was restricted to the basal layers in some of papilloma and inverted papilloma in that were found koilocytes in suprabasal layers. From the above results, we observed that the distribution of AEl antibody was correlated with benign and malignant differentiation, CK19 antibody would prove helpful for confirmation of virus infected tumors.

      • 골격성 Ⅲ급 부정교합 환자에서 양악 수술후 연조직 변화에 대한 연구

        The purpose of this study was to assess the soft tissue changes using twenty skeletal class Ⅲ malocclusion patients who treated with bimaxillary surgery for the correction of dentofacial deformities. Patients were devided into two groups. One was impaction and advancement of maxilla with mandibular set-back (Group 1), the other was downward and advancement of maxilla with mandibular set-back (Group 2). Preoperative and postoperative one year cephalometric data were analyzed and compared. Results obtained were as follows : 1. The ratio of horizontal changes of soft tissue to hard tissue at Nt to ANS, Ls to UI, Li to LI, sPog to Pog were 1:0.60, 1:0.79, 1:0.47, 1:0.63 in group 1 respectively, and 1:0.59, 1:0.48, 1:0.83 1:1.09 in group 2 respectively. There were highly predictable in upper lip, lower lip, and chin. 2. The ratio of vertical changes of soft tissue to hard tissue at Nt to ANS, Li to LI were 1:0.72, 1:0.06 in group 1, and others showed no statistically significant differences. 3. The ratio of horizontal changes of Ls to hard tissue movements at LI(h) was 1:-0.82 in group 1 and at UI(h), LI(h) were 1:0.48, 1:0.01 in group 2. These ratios of group 1 were greater than those of in group 2. 4. The direction of horizontal change of Li was the same as that of hard tissue change. The ratio of horizontal changes of Li to LI was 1:0.47 in group 1 and others showed no statistically significant difference. 5. The changes of upper lip thickness and length were -1.6 mm, -1.4 mm in group 1, and -1 mm, -2.7 mm in group 2. 6. The ratios of thickness of upper lip to ANS, Ul, LI were 1:-0.83, 1:-0.37, 1:0.11 in group 1. There was similar trend in group 2, but there were no statistically significant difference. These results suggest that prediction of changes in soft tissue of upper lip, lower lip, and chin were 79%, 47%,and 63% in group 1, and 48%, 83%, and 109% in group 2 after bimaxillary surgery. There was a tendency of decrease in thickness and increase in length of the upper lip.

      • 백서의 피하에 이식한 치수에서의 mineralization에 대한 형태학적 연구

        The dental pulp is the soft connetive tissue that chiefly composed of odontoblasts, fibroblasts, undifferentiated mesenchymal cells, and macrophages. The odontoblast is important in the formation of dentin, and its function is similar to that of the osteoblast in intramembranous bone formation. It was established that odontoblasts strongly resemble to osteoblasts after calcification of dentin and, when cultured in vitro, they are similar to each other morphologically and cytochemically. Bone and dentin are closely related in both orgnaic and inorganic composition. The present study was undertaken to observe the morphologic changes of transplanted pulp cells in rats. The results were as follows : 1. Light microscopically, at the early stage(1-3 days) of transplantation, hyalinized tissues and calcospherites around of odontoblasts and fibroblasts of the transplanted tissue were observed. Osteoid tissues and spongy bone surrounded by osteoblasts were observed at the middle stage(5-11days). With the time lapsed, mineralization of the bones were advanced. More advanced mineralization in bone tissue was observed but osteoid tissues were not observed at the late stage(2-4 weeks). 2. Electron microsopically, at the early stage of transplantation, the cells having ecentric nucleus with abundant cytoplasm, well developed rough endoplasmic reticulum and round shaped of mitochondria and resembled osteoblasts. A few of collagen fibers were observed. At the middle stage, osteoid and osteblasts were observed. Osteoid tissures were consist of irregularaly arranged of collagen fibers and osteoblasts disclosed well developed rough endoplasmic reticulum with cisternal dilatations. Calcifying globules and calcium deposits were observed in osteoid tissue. From the above results, it is suggested that pulps transplanted subcutaneously, were observed formed the bone tissues and as bone formations and maturations are various accoring to the changes of environment and durations. It is thought that odontoblasts, fibroblasts and undifferentiated mesenchymal cells are invloved in bone formation.

        악골에 발생한 악성 섬유성조직구종 2례

        Malignant fibrous histiocytoma rarely occurs in the jaws. Although the histogenesis of this tumor remains controversal, it is best regarded as a primitive and pleomorphic nature reflected by collagen production and occasional phagocytosis. It is common for metastatic foci to appear in lung and regional lymph node. There are variable treatments such as radiotherapy, surgical excision or combination, therapy of surgical excision, chemotherapy and radiotherapy. With the review of literatures, the authors report the clinical study of two cases of malignant fibrous histiocytoma of the jaw.

      • 분광색채계측기를 이용한 Vita Shade 광중합형 복합레진의 색상 비교

        To get a satisfactory result in the composite resin restorations, it is necessary to choose correct shade. At present, most of the commercial composite resins are based on the Vita Lumin shade guide or Bioform shade guide, but color differences might be expected even using the same shade in various materials. In the study, five kinds of light-cured composite resins with A2 and B3 shade were used to measure and compare the color each other while one porcelain served as a control. All composite resins (Spectrum TPH (SP), VeridonFil- Photo (VE), Z100 (Z100), Charisma (CH), Prodigy (PRO) were filled in to the metal mold (12 mm diameter, 2 mm depth), followed by compression, polymerization and polishing with wet sandpaper. The specimens of porcelain were fabricated by using the refractory mold for porcelain. After 24 hours, the specimens were placed on the spectrocolorimeter and spectral reflectance were measured under CIE illuminant D65. After measuring the values of L^*,a^*,b^* and ΔE^*, following results were obtained; 1. The L^*, a^* and b^* values of both shade of porcelain specimens showed significantly higher than those of resin specimens(p<0.05). 2. In comparing the resin specimens of the A2 shade, differences were significant except L^* values of SP-CH and PRO-VE, a^* values of the VE-SP and b^* values of the VE-Z100 and SP-PRO(p<0.05). 3. In comparing the resin specimens of the B3 shade, differences were significant except L^* values of PRO-SP, a^* values of the SP-PRO and Z100-VE and b^* values of the PRO-SP(p<0.05). 4. In comparing the resin specimens of the A2 shade, color differences between materials(ΔE^*) showed the lowest value of 1.66, and the highest was 5.16. ΔE^* values of the materials of VE-PRO, CH-PRO, SP-PRO, SP-Z100 and SP-CH were lower than 3.3. 5. In comparing the resin specimens of the B3 shade, the lowest value of the ΔE^* was 0.57 and the highest was 5.92. ΔE^* values of Z100-CH and SP-PRO were lower than 3.3. The present study revealed there was perceptible color difference between materials even if they have the same designated shade based on Vita shade guide. The results of the present study suggested that it would be necessary to establish the reproducible and constant color specification system for an esthetic restoration.

      • 치근단 낭종에서 염증세포와 콜레스테롤 결정의 분포양상

        Apical periodontal cysts correlate to dental caries, and derived from the epithelial rests of apical granuloma. The lesions are histologically characterized by infiltrates of monocytes/macrophages, lymphocytes, plasma cells and mast cells. In addition, cholesterol crystals and foam cells are found. The aim of this study was to investigate the relationship between chronic inflammatory cells, cholesterol crystals and foam cells. The results obtained were as follows : 1. In all apical periodontal cysts, mild to severe number of inflammatory cells could be observed, while foam cells and cholesterol crystals were found in varying number. 2. The cholesterol crystals were not always associated with foam cells. 3. The cholesterol crystals appeared as long narow spaces with pointed tips and cluster formation. 4. The foam cells and choesterol crystals surrounded by lymphocytes, plasma cells, and erythrocytes, hemosiderin pigments and giant cells were observed. 5. Vast number of the cholesterol crystals and foam cells congregated in connective tissue, less frequently crystal mass eroded the epithelial lining and empties the crystal into the cystic cavity. From above the results, it was found that cholesterol crystals and foam cells are characteristic findings and correlated to inflammatory reaction in apical periodontal cyst. It suggested that inflammation closely related to cholesterol crystals and foam cells.

      • 앵글씨 분류에 의한 성인 골격구조 및 하악운동량 평가

        The purpose of this study was to identify the difference of vertical movement of mandible according to Angle's molar relationship and by skeletal factors affect to vertical movement of mandible. 172(age ranged from 20 to 30) subjects who go to college within territory of Kwangju city without any experience of temporomandibular disorder, extraction and orthodontic treatment. were selected for this study. The subjects were classified into class Ⅰ(male :30, female:49), class Ⅱ(male:18, female:24) and class Ⅲ(male:18, female:33) according to Angle's molar relationship. The distance was measured between incisal edge of maxillary and mandibular central incisor and between bottom of central fossa of maxillary and mandibular 1st molar with ruler. The arch length and width were measured on diagnostic cast. Cephalometrics were taken and then traced. Landmarks were identified and analyzed. The following results were obtained: 1. Maximal interincisal opening of male is larger than that of female in class Ⅰ, class Ⅱ and class Ⅲ. Among each group maximal interincisal distance is the largest in class Ⅲ. Maximal intermolar distance of male is superior to that of female in class Ⅰ, class Ⅱ, and class Ⅲ, but there is no siginficant difference among them. 2. On maximal opening movement of Angle's classification class Ⅰ and class Ⅱ, total mandibular length, mandibular ramal length, madibular inferior border length and upper arch width were important variables and facial length, upper arch length and lower arch length had negative relationship to that. On maximal opening movement of Angle's class Ⅲ, the upper arch length, the lower arch length and anterior facial length were important variables especially when compared with class Ⅰ and Ⅱ, and upper arch width had negative relationship. These results suggest that maximal opening movement is affected by facial morphology in all of Angle's class Ⅰ, class Ⅱ and class Ⅲ, but each group is affected by different facial skeletal variables. Accordingly, facioskeletal variables are considered for diagnosis and treatment to improve the amount of mouth opening.
