      • Oridonin Suppresses Proliferation of Human Ovarian Cancer Cells via Blockage of mTOR Signaling

        Xia, Rong,Chen, Sun-Xiao,Qin, Qin,Chen, Yan,Zhang, Wei-Wei,Zhu, Rong-Rong,Deng, An-Mei Asian Pacific Journal of Cancer Prevention 2016 Asian Pacific journal of cancer prevention Vol.17 No.2

Oridonin, an ent-kaurane diterpenoid compound isolated from the traditional Chinese herb Rabdosia rubescens, has shown various pharmacological and physiological effects such as anti-tumor, anti-bacterial, and anti-inflammatory properties. However, the effect of oridonin on human ovarian cancer cell lines has not been determined. In this study, we demonstrated that oridonin inhibited ovarian cancer cell proliferation, migration and invasion in a dose-dependent manner. Furthermore, we showed oridonin inhibited tumor growth of ovarian cancer cells (SKOV3) in vivo. We then assessed mechanisms and found that oridonin specifically abrogated the phosphorylation/activation of mTOR signaling. In summary, our results indicate that oridonin is a potential inhibitor of ovarian cancer by blocking the mTOR signaling pathway.

Do ACR TI-RADS scores demonstrate unique thyroid molecular profiles?

        Rong Xia,Wei Sun,Joseph Yee,Sheila Sheth,Chrystia Slywotzky,Steven Hodak,Tamar C. Brandler 대한초음파의학회 2022 ULTRASONOGRAPHY Vol.41 No.3

Purpose: The present study aimed to examine the molecular profiles of cytologically indeterminate thyroid nodules stratified by American College of Radiology Thyroid Imaging Reporting and Data System (TI-RADS) categories and to determine whether certain ultrasonographic features display particular molecular alterations. Methods: A retrospective review was conducted of cases from January 1, 2016 to April 1, 2018. Cases with in-house ultrasonography, fine-needle aspiration Bethesda System for Reporting Thyroid Cytopathology (TBSRTC) diagnoses, molecular testing, and surgery were included. All cases were diagnosed as TBSRTC indeterminate categories. The ultrasound studies were retrospectively reviewed and assigned TI-RADS scores (TR1-TR5) by board-certified radiologists. The final diagnoses were determined based on the surgical resection pathology. Binary logistic regression analysis was used to study whether demographic characteristics, TI-RADS levels, and TBSRTC diagnoses were associated with ThyroSeq molecular results. Results: Eighty-one cases met the inclusion criteria. RAS mutations were the most common alteration across all TI-RADS categories (TR2 2/2; TR3 10/19, TR4 13/44, and TR5 8/16), and did not stratify with any particular TI-RADS category. Only TR4 and TR5 categories displayed more aggressive mutations such as <i>BRAF</i>V600E and <i>TERT</i>. ThyroSeq results were positively correlated with thyroid malignancy when non-invasive follicular thyroid neoplasm with papillary-like nuclear features (NIFTP) was categorized in the malignant category (odds ratio [OR], 6.859; P<0.01), but not when NIFTP was removed from the malignancy category. Echogenicity scores were found to be negatively correlated with ThyroSeq results in thyroid nodules (OR, 0.162; P<0.01).Conclusion: Higher-risk molecular alterations tended to stratify with the higher TI-RADS categories.

Self-assembled micelles of the natural medicine ginsenosides for cancer metastasis therapy

        Xia-Rong Tan,Chao-Li,Ke-Ke Feng,Jing-Qing Le,Jiang-Wen Shen,Jing-Wei Shao 한국공업화학회 2022 Journal of Industrial and Engineering Chemistry Vol.116 No.-

The nanocarrier-based drug delivery systems have become an alternative strategy for cancer treatment. Whereas, increasing studies find that the utilization of drug carriers may result in poor biocompatibility, lower drug loading capacity and unpredictable side reactions. Therefore, we herein reported selfassembledmicelles based on herbal product ginsenosides, which are the main active ingredients ofPanax ginseng C.A. The ginsenosides-based micelle system exerts excellent biosafety, better stability, highdrug capacity, and lower cytotoxicity compared to free ginsenosides. Meanwhile, ginsenosides micellesexhibits superior inhibitory effects on cancer cell adhesion activity, especially the expression of intercellularadhesion molecule-1, which is the critical factor of cancer metastasis. Importantly, ginsenosidesmicelles results in remarkably therapeutic efficacy on lung metastasis of liver cancer. All these resultshighlight the potential utilization of ginsenosides micelles in the clinic. Meanwhile, the carrier-freenano-drugs self-delivery system of ginsenosides showed great promise to become an emerging approachfor cancer metastasis therapy.

        Gao, Shao-Rong,Li, Lu-Ming,Xia, Hai-Ping,Wang, Guang-Ming,Xu, Hong-Yan,Wang, Ai-Rong Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.9

Objective: To explore the clinical efficacy and toxic and side effects of recombinant human endostatin (rhendostatin/endostar) combined with chemotherapy in the treatment of advanced gastric cancer. Materials and Methods: A total of 70 patients with advanced gastrointestinal adenocarcioma confirmed by histopathology and/or cytological examination were divided into group A (37 patients) and group B (33 patients). Patients in group A were given intravenous drip of 15 mg endostar added into 500 mL normal saline, once every other day until the cessation of chemotherapy or patients' maximal tolerance to chemotherapy. Patients in group B received chemotherapy alone. Two groups selected the same chemotherapy regimens. FOLFIRI scheme: 90-min intravenous drip of $180mg/m^2$ irinotecan, intravenous drip of $200mg/m^2$ calcium folinate (CF) and $400mg/m^2$ 5-fluorouracil (5-Fu) on d1, and continuous intravenous pumping of 2 $400mg/m^2$ 5-Fu for 46 h. FOLFOX4 scheme: intravenous injection of $85mg/m^2$ oxaliplatin (L-OHP), $200mg/m^2$ calcium folinate (CF) and $400mg/m^2$ 5-FU on d1 for 2 h, and then continuous intravenous pumping of 2 $400mg/m^2$ 5-Fu for 46 h. XELOX scheme: oral administration of 1 $500mg/m^2$ xeloda (or tegafur 50~60 mg) in twice during d1~14 and intravenous drip of $135mg/m^2$ L-OHP on d1 for 2 h. The modified FOLFOX scheme: intravenous injection of $135mg/m^2$ L-OHP on d1 for 2 h, $200mg/m^2$ CF and 1.0 g tegafur during d1~5. Whereas, control Group B received chemotherapy regimens which were same as Group A, but no addition of endostar. Before chemotherapy, patients were given intravenous injection of 8 mg ondansetron, intramuscular injection of 10 mg metoclopramide and 20 mg diphenhydramine for prevention of vomiting, protection of liver and stomach as well as symptomatic supportive treatment. One cycle was 21 d, 4~6 cycles in total. The efficacy was evaluated every 2 cycles. Results: 32 patients in Group A could be evaluated, and the response rate (RR) and disease control rate (DCR) were 59.38% and 78.13%, respectively. 31 patients in Groups could be evaluated, and the RR and DCR were 32.26% and 54.84%, respectively. The differences between 2 groups were significant. The toxic effects include myelosuppression, gastrointestinal reaction, fatigue, cardiotoxicity and peripheral neurotoxicity. Conclusions: Preliminary observations show that endostar (once every other day) combined with chemotherapy is effective in the treatment of advanced gastrointestinal cancer, with low toxic effects, good tolerance, deserving further study.

      • Computer vision-based displacement measurement with m-sequence target

        Yi-Ding Hu,Qi Xia,Rong-rong Hou,Yong Xia,Jian-yi Yan 국제구조공학회 2021 Smart Structures and Systems, An International Jou Vol.27 No.3

The development of image sensors enables the application of vision-based techniques to the non-contact displacement measurement of large-scale structures. The features of the physical targets are critical to the accuracy, stability and anti-interference of the displacement measurement results. In this study, a novel m-sequence target and the associated circular correlation processing technique are developed for real-time displacement measurement. The properties of the m-sequence as a pseudo-random sequence are introduced. The vision-based displacement calculation method is then derived from the correlation property of the m-sequence. The algorithms and measurement systems are integrated in the LabVIEW environment. To verify the anti-interference performance of the developed system, static and dynamic experimental tests are carried out with various forms of interference, such as partial occlusion, uneven illumination, out of focus and smoke effect. Experimental results indicate that the developed system cannot only accurately measure structural displacement, but also has outstanding antiinterference performance, even if 30% of the target is masked.

Design and Preparation of Magnetic CTAB/Montmorillonite Nanocomposite for Phenols Removal

        Rong Shen,Yichang Yu,Yue Wang,Zhining Xia 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2018 NANO Vol.13 No.10

The cetyltrimethyl ammonium bromide (CTAB)-modified montmorillonite (MMT) was synthesized via a novel "dissolution and reassembly" method. To determine the optimal formula, the adsorption of C.I. Reactive Red 2 (X3B) with CTAB/MMT was investigated. The optimal CTAB/MMT nanocomposite was used to remove 2,6-dichlorophenol and p-nitrophenol from aqueous solutions. The adsorption results can be described by Langmuir isotherm, and the adsorption capacities were 200 mg/g and 125 mg/g for 2,6-dichlorophenol and p-nitrophenol, respectively. To realize the quick separation and recycle, the magnetic CTAB/MMT was further strategized and synthesized. The adsorption equilibrium time was 15 min for both contaminants; the ions' strength showed a little bit of influence on the adsorption performance. In addition, compared with acidic condition, neutral condition was more beneficial to the adsorption reaction. Due to the addition of Fe3O4, the adsorption capacities of this magnetic nanocomposite for 2,6-dichlorophenol and p-nitrophenol were a little bit decreased, which were 170 mg/g and 91 mg/g, respectively. However, the magnetic nanocomposite can be separated within 30s under an external magnetic field, which would be useful in the practical application.

        Xia, Yi-Qun,Wei, Xiao-Yan,Li, Wu-Lan,Kanchana, Karvannan,Xu, Chao-Chao,Chen, Da-Hui,Chou, Pei-Hong,Jin, Rong,Wu, Jian-Zhang,Liang, Guang Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.16

Curcumin and its analogues have been reported to exert anti-cancer activity against a variety of tumors. Here, we reported A501, a new curcumin analogue. The effect of A501 on cell viability was detected by MTT assay, the result showed that A501 had a better inhibiting effect on the four non-small cell lung cancer (NSCLC) cells than that of curcumin. Moreover, Colony forming experiment showed A501 significant restrained cell proliferation. Flow cytometry displayed A501 can cause G2/M arrest and induce apoptosis. Western blotting showed that A501 decreased the expression of cyclinB1, cdc-2, bcl-2, while increased the expression of p53, cleaved caspase-3 and bax. In conclusion, curcumin analogues A501 played antitumor activity by inhibiting cell proliferation and inducing apoptosis of NSCLC cells. And it was likely to be a promising starting point for the development of curcumin-based anticancer drugs.

      • Roles of Signaling Pathways in the Epithelial-Mesenchymal Transition in Cancer

        Liu, Xia,Yun, Fen,Shi, Lin,Li, Zhe-Hai,Luo, Nian-Rong,Jia, Yong-Feng Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.15

The epithelial-mesenchymal transition (EMT) is a cellular process though which an epithelial phenotype can be converted into a phenotype of mesenchymal cells. Under physiological conditions EMT is important for embryogenesis, organ development, wound repair and tissue remodeling. However, EMT may also be activated under pathologic conditions, especially in carcinogenesis and metastatic progression. Major signaling pathways involved in EMT include transforming growth factor ${\beta}(TGF-{\beta})$, Wnt, Notch, Hedgehog and other signaling pathways. These pathways are related to several transcription factors, including Twist, Smads and zinc finger proteins snail and slug. These interact with each other to provide crosstalk between the relevant signaling pathways. This review lays emphasis on studying the relationship between EMT and signaling pathways in carcinogenesis and metastatic progression.
