Double-Face Activity of Resveratrol in Voluntary Runners: Assessment of DNA Damage by Comet Assay

Voluntary runners are subjected to a massive increase in reactive oxygen/nitrogen species production, which can promote different oxidative stress–related diseases such as premature aging, neurodegenerative disorders, and cancer. The aims of this work were to evaluate the following in peripheral blood cells of voluntary runners: (i) DNA status; (ii)susceptibility to the in vitro insult induced by hydrogen peroxide (H2O2) as a breaking agent; (iii) capabilities of 3,5,40-trihydroxystilbene (RESV) in counteracting DNA damage. Twenty-five male voluntary runners were compared with 20sedentary men, as age-matched controls, and DNA status was evaluated with different versions of comet assay: alkaline,neutral, and Fpg enzyme–modified version to measure 8-OH-deoxyguanosine (8-oxo-dG) levels. The H2O2 and/or RESV treatments were performed directly on agarose-embedded cells (atypical comet assay). The results evidenced DNA damage and levels of 8-oxo-dG higher in runners than in sedentary control subjects. The runners’ DNA was more prone to the in vitro–induced oxidative insult (200 lM H2O2) than that of the control group. Resveratrol (100 lM), depending on the individual basal DNA status, was able to switch from antioxidant to pro-oxidant. Our results, on the one hand, validated the proposed in vitro experimental protocol in order to measure individual DNA status. On the other hand, our data point out the importance of monitoring the athletes’ redox status before subjecting them to dietary supplementation treatment.

Aerobic exercise increases free radical production as a consequence of enhanced oxygen consumption. If free radical formation exceeds antioxidant capacity, lipids, proteins, and DNA may be oxidized. Oxidative stress is widely recognized as a factor in many degenerative human diseases. The role of dietary antioxidants in protection against disease is a topic of continuing interest. In fact, there is epidemiological evidence correlating a higher intake of nutrients possessing antioxidant abilities with a lower incidence of various human diseases. This study was directed at investigating whether changes in plasma antioxidant capacity and oxidative stress markers occur in voluntary wheel runners, before and after oral supplementation with lycopene and isoflavones. For this purpose, plasma antioxidant capacity and oxidative stress markers were assessed in long distance runners at the end of a 60-minute run. Comparisons were made between runners before and after 60 days of supplementation with lycopene and isoflavones. DNA damage in blood cells of the same samples was also evaluated by comet assay. This investigation shows that oral supplementation with lycopene and soy-derived isoflavones significantly reduced lipid peroxidation and enhanced plasma nonproteic antioxidant defense.

Model of implementation Mechanization for Rice Production System is done in Sidowayah Village, Klaten district, Central Java Province. The research method is institutional approach with the rationale of corporate development in developing integrated agricultural area by applying Economies of scale, which the principle is the wider the more efficient, in term of business cost which includes plant management, production facility cost, transportation costs and marketing costs of farm products. Through the cooperative there will be consolidation of business that facilitate access of information, capital, and bergaining poistion in the market. The essence of the corporate concept is to build cooperation through a single management system, by merging small farms into a large scale business that meets economies of scale to become more efficient, high productivity, homogeneous products, and higher production quality that are prerequisites for improving competitiveness, added value and improve production quality. Institutional factors is an important element in the successful implementation of mechanization, technical assistance and management is necessary to improve the performance of gapoktan ( group of farmer group) institutional. Reformion to empower gapoktan as the main driver in rice production system, from the land preparation to harvesting and marketing activities. Institutional strengthening includes technical and management, from technical aspect of operator training and basic maintenance of machines. As for the management, the method is meetings, socialization, discussion and comparative studies. Strengthening gapoktan institutions in synergy with BUMDES (village owned corporation) empowering gapoktan to handle rice production system from supplying farm input, land preparation, transplanting, weeding, harvesting and selling the paddy. And this new system increase farmer income Rp 1.5 to 2 million per stakes (1 stakes = 1/3 ha). So the implementing rice mechanization is directly give profit for farmers.
Review Essay: What Happened to the Study of China in Comparative Politics?

In 1986, Kenneth Lieberthal observed that the study of China in the United States had had little effect on the evolution of political science. Twenty years later, its impact on the core debates in comparative politics seems to have been no more significant. Why have some of the most influential books in the study of contemporary Chinese politics not been significant in the discipline of comparative politics? Based on a quantitative overview of forty-two comparative politics syllabi, my argument is twofold. First, China scholarship has isolated the study of Chinese politics by primarily publishing in area journals, building analyses around debates exclusive to Chinese politics, and generating knowledge with limited contemplation of its potential for generalization outside China. Second, comparative politics seems to have been caught in a “democratic prism,” which has impeded scholars’ ability to adapt some of the debates to empirical changes associated with China’s rise and development.

<P><B>Rationale</B></P> <P>The P2Y<SUB>12</SUB> receptor inhibitor clopidogrel is widely used in patients with acute coronary syndrome, percutaneous coronary intervention, or ischemic stroke. Platelet inhibition by clopidogrel shows wide interpatient variability, and high on-treatment platelet reactivity is a risk factor for atherothrombotic events, particularly in high-risk populations. <I>CYP2C19</I> polymorphism plays an important role in this variability, but heritability estimates suggest that additional genetic variants remain unidentified. The aim of the International Clopidogrel Pharmacogenomics Consortium (ICPC) is to identify genetic determinants of clopidogrel pharmacodynamics and clinical response.</P> <P><B>Study design</B></P> <P>Based on the data published on www.clinicaltrials.gov, clopidogrel intervention studies containing genetic and platelet function data were identified for participation. Lead investigators were invited to share DNA samples, platelet function test results, patient characteristics, and cardiovascular outcomes to perform candidate gene and genome-wide studies.</P> <P><B>Results</B></P> <P>In total, 17 study sites from 13 countries participate in the ICPC, contributing individual patient data from 8,829 patients. Available adenosine diphosphate–stimulated platelet function tests included vasodilator-stimulated phosphoprotein assay, light transmittance aggregometry, and the VerifyNow P2Y<SUB>12</SUB> assay. A proof-of-principle analysis based on genotype data provided by each group showed a strong and consistent association between <I>CYP2C19</I>*2 and platelet reactivity (<I>P</I> value=5.1 × 10<SUP>−40</SUP>).</P> <P><B>Conclusion</B></P> <P>The ICPC aims to identify new loci influencing clopidogrel efficacy by using state-of-the-art genetic approaches in a large cohort of clopidogrel-treated patients to better understand the genetic basis of on-treatment response variability.</P>

The Green Mark eco-label constitutes an essential program introduced by Taiwan’s Environmental Protection Administration to support environmental protection. In the manufacturing industry, the Green Mark eco-label is intended to enable differentiating between manufacturers that apply green processes and those that do not. This paper presents an assessment model for identifying manufacturers’ initial intention to apply the Green Mark eco-label. The model was derived from the certification process flow. A total of 200 structured questionnaires were distributed to manufacturers in Taiwan, and 172 responses were retrieved. The model was analyzed by applying structural equation modeling, and the correlation of review process and end certification process exhibited the strongest correlation. Improving the document verification stage is highly likely to increase the overall initial intention ofmanufacturers to participate in the Green Mark eco-label program. The overall certification process contributed 16% to the total initial intention of manufacturers to apply the Green Mark eco-label to their products.
Correlation between events with different safety significance in nuclear power plants

The main purpose of collecting and analyzing operating experience events is to look for insights to improve safety. The underlying assumption is the correlation and causality between events with different safety significances. The analysis of this assumption could be valuable to optimize operating experience feedback programs and to enhance safety. This paper analyses the correlation between events with different safety significances. Groups of events from six nuclear energy related databases are considered. The findings are that a correlation exists but with various levels and not as large or consistent as might be expected across different databases. These results might be the basis for further work to analysis causality, to find out how a similarity in causes influences the correlation, and finally to improve the operating experience program.
