        Dynamic changes in carbohydrate metabolism and endogenous hormones during Tulipa edulis stolon development into a new bulb

        The stolon is the main asexual reproductive organ of Tulipa edulis (Miq.) Baker. It has a special morphology and can develop into a new bulb for propagation. In the current greenhouse experiment, the dynamic changes in carbohydrates and related enzymes, protein and endogenous hormones during T. edulis stolon development were investigated. The results showed that soluble sugar levels were basically declining, whereas starch and protein content rose continuously during stolon development. The adenosine diphosphoglucose pyrophosphorylase (AGPase) activity peaked in the initial swelling stage and stayed a relative high level in the middle swelling stage; sucrose synthase (SS), soluble starch synthase (SSS) and granule-bound starch synthase (GBSS) activities followed the same law that showed rising trends during stolon development. SS activity was significantly inversely related to sucrose content but had significantly positive relations with starch content, SSS and GBSS activities. Gibberellin (GA), indole-3-acetic acid (IAA) and zeatin riboside (ZR) peaked in the initial swelling stage and maintained high levels in the middle swelling stage; they then decreased significantly in the later swelling stage. A substantial increase was observed in abscisic acid (ABA) content until the middle swelling stage, followed by a significant reduction in the later swelling stage. The ratios of ABA to IAA, GA and ZR reached their lowest levels in the initial swelling stage. In conclusion, T. edulis stolon development is a process of new bulb morphogenesis along with the starch accumulation catalyzed by AGPase, SSS and GBSS, using the product of sucrose cleavage caused by SS. Initial low ABA content and low ratios of ABA to IAA, GA and ZR, together with the GA, IAA and ZR of high-content, soluble sugars worked more efficiently to induce new bulb formation.

        Optimal design of micro-nano displacement driving mechanism for obtaining mechanical properties of micro structure

        The mechanical structures of micro-electro-mechanical systems (MEMS) are composed of different types of microstructures, and their mechanical properties are very important for the realisation and reliability of the system performance. One of the key problems in measuring the mechanical properties is the design and implementation of micro-nano displacement driving mechanisms. This paper describes a mechanism that adopts a two-level loading strategy, fast approach, and precise bending displacement loading structures, and has a theoretical analysis and optimal design based on optimal targets of resistance and displacement. The results show that the relative error is 6.98 % for the fast-approaching structure experiment and its optimal simulation and 4.26 % for the precise bending displacement loading structure (PBLS) experiment and its optimal simulation. The optimised micro-nano displacement loading mechanism can achieve optimal output performance under existing constraints.

        Authentication of an Endangered Herb Changium smyrnioides from Different Producing Areas Based on rDNA ITS Sequences and Allele- Specific PCR

        The rDNA ITS region of 18 samples of Changium smyrnioides from 7 areas and of 2 samples of Chuanminshen violaceum were sequenced and analyzed. The amplified ITS region of the samples, including a partial sequence of ITS1 and complete sequences of 5.8S and ITS2, had a total length of 555 bp. After complete alignment, there were 49 variable sites, of which 45 were informative,when gaps were treated as missing data. Samples of C. smyrnioides from different locations could be identified exactly based on the variable sites. The maximum parsimony (MP) and neighbor joining (NJ) tree constructed from the ITS sequences based on Kumar’s two-parameter model showed that the genetic distances of the C. smyrnioides samples from different locations were not always related to their geographical distances. A specific primer set for Allele-specific PCR authentication of C. violaceum from Jurong of Jiangsu was designed based on the SNP in the ITS sequence alignment. C. violaceum from the major genuine producing area in Jurong of Jiangsu could be identified exactly and quickly by Allele-specific PCR.

        An in vitro Actinidia Bioassay to Evaluate the Resistance to Pseudomonas syringae pv. actinidiae

        Pseudomonas syringae pv. actinidiae (Psa) is by far the most important pathogen of kiwifruit. Sustainable expansion of the kiwifruit industry requires the use of Psa-tolerant or resistant genotypes for the breeding of tolerant cultivars. However, the resistance of most existing kiwifruit cultivars and wild genotypes is poorly understood, and suitable evaluation methods of Psa resistance in Actinidia have not been established. A unique in vitro method to evaluate Psa resistance has been developed with 18 selected Actinidia genotypes. The assay involved debarking and measuring the lesions of cane pieces inoculated with the bacterium in combination with the observation of symptoms such as callus formation, sprouting of buds, and the extent to which Psa invaded xylem. Relative Psa resistance or tolerance was divided into four categories. The division results were consistent with field observations. This is the first report of an in vitro assay capable of large-scale screening of Psa-resistance in Actinidia germplasm with high accuracy and reproducibility. The assay would considerably facilitate the breeding of Psa-resistant cultivars and provide a valuable reference and inspiration for the resistance evaluation of other plants to different pathogens.

        A comprehensive analysis and comparison between vacuum and electric oven drying methods on Chinese saffron (Crocus sativus L.)

        The red stigmas of saffron are one of the mostexpensive spices in the world and serve as a traditionalChinese medicine. More saffron has been cultivated inChina, and different drying technologies have been studied. However, a comprehensive and comparative analysis ofdifferent drying approaches has not been well studied. Inthis study, we compared electric oven and vacuum ovendrying approaches on saffron. We found saffron was driedquicker under high vacuum drying mode with high temperatureand the quicker drying rate provided, the moreopen microstructural interstices on the saffron surface. Both methods were best fit to Midilli and Kucuk model. Besides, the coloring, aroma and bitterness strength afterdrying showed the similar results. In sum, our data suggestedthe optimal drying temperature was 100 C for20 min for two evaluated methods, however consideringthe machine cost, the electric oven drying would be the firstchoice.
