Glucagon-like peptide-1 (GLP-1) was a potential therapeutic drug for type II diabetes, mainly because of the stimulatory effect on insulin secretion under condition of high blood glucose. We used PCR to obtain a recombination gene, GGH, in which two GLP-1 (GLP-1A2G) mutants were connected in series and then fused to the N terminal of human serum albumin. The fusion gene was inserted into pGAPZaA plasmid with Saccharomyces cerevisiae α- factor secretion signal sequence, and was expressed by the glyceraldehyde-3-phosphate dehydrogenase (GAP) promoter. The engineered strain was constructed by integrating the recombinant plasmid pGAPZαA/GGH into the genome of Pichia pastoris GS115. Genome PCR and western blot showed that the recombinant P. pastoris successfully expressed the fusion protein GGH. The yield of GGH reached 78 mg/L after 72 h fermentation in a flask, using glucose as the optimal carbon source. Fed-batch fermentation was investigated in a 5 L bioreactor, and the expression level of GGH reached 246 mg/L in 52 h. The fusion protein GGH was purified in four steps, and the final purity was 96.1%. The in vitro bioactivity of GGH was the same as that expressed in P. pastoris by the AOX1 promoter. This study described an efficient way to express GGH fusion protein in P. pastoris using GAP promoter, fermentation was easier to control without carbon source change and fermentation time was 20 h less than AOX1 promotercontrolled GGH fermentation.

<P>Magnetic van der Waals (vdW) materials have emerged as promising candidates for spintronics applications, especially after the recent discovery of intrinsic ferromagnetism in monolayer vdW materials. There has been a critical need for tunable ferromagnetic vdW materials beyond room temperature. Here, we report a real-space imaging study of itinerant ferromagnet Fe3GeTe2 and the enhancement of its Curie temperature well above ambient temperature. We find that the magnetic long-range order in Fe3GeTe2 is characterized by an unconventional out-of-plane stripe-domain phase. In Fe3GeTe2 microstructures patterned by a focused ion beam, the out-of-plane stripe domain phase undergoes a surprising transition at 230 K to an in-plane vortex phase that persists beyond room temperature. The discovery of tunable ferromagnetism in Fe3GeTe2 materials opens up vast opportunities for utilizing vdW magnets in room-temperature spintronics devices.</P> [FIG OMISSION]</BR>

Based on the actual operating environment of transformer, the aging tests of nitrile butadiene rubber (NBR) were conducted systematically under four conditions: in air, in transform oil, under compression in air and under compression in transform oil to studythe effect of high temperature, transform oil and compression stress simultaneously on the thermal aging behaviors of nitrile butadiene rubber and predict the lifetime. The effects of liquid media and compression stress simultaneously on the thermal aging behaviors of nitrile butadiene rubber were studied by using characterization methods such as IR spectrosc-opy, thermogravimetric measurements, Differential Scanning Calorimetry (DSC) measurements and mechanical property measurements. The changes in physical properties during the aging process were analyzed and compared. Different aging conditions yielded materials with different properties. Aging at $70^{\circ}C$ under compression stress in oil, the change in elongation at break was lower than that aging in oil, but larger than that aging under compression in air. The compression set or elongation at break as evaluation indexes, 50% as critical value, the lifetime of NBR at $25^{\circ}C$ was predicted and compared. When aging under compression in oil, the prediction lifetime was lower than in air and under compression in air, and in oil. It was clear that when predicting the service lifetime of NBR in oil sealing application, compression and media liquid should be involved simultaneously. Under compression in oil, compression set as the evaluation index, the prediction lifetime of NBR was shorter than that of elongation at break as the evaluation index. For the life prediction of NBR, we should take into account of the performance trends of NBR under actual operating conditions to select the appropriate evaluation index.

Zinc-Tin-Oxide (ZTO) thin films were fabricated using a simple and eco-friendly sol-gel method and their application in thin film transistors (TFTs) was investigated. Annealing temperature has a crucial influence on the structure and electrical properties of sol-gel ZTO thin films. The ZTO thin films annealed at 300–600 °C revealed smooth and uniform surfaces with amorphous state, in addition, a high optical transparency over 90% of the ZTO films in the visible range was obtained. The electrical performance of ZTO TFTs showed obvious dependence on annealing temperature. The ZTO TFTs annealed at 500 °C showed a high carrier mobility of 5.9 cm2 /V, high on/off current ratio (Ion/off) of 106 -107 , and threshold voltage (Vth) of 1.03 V. To demonstrate the application of sol-gel ZTO films in low-power display fields, we also fabricated ZTO TFTs with a solution-processed high-permittivity (high-k) ZrTiOx dielectric layer. The ZTO/ZrTiOx TFTs showed high mobility of 17.9 cm2 /V and Ion/off of 105 -106 at a low operation voltage of 3 V, indicating that Indium-free ZTO thin films would be potential candidates for low cost, high performance oxide TFT devices.

<P><B>Abstract</B></P> <P>Nuclear receptors regulate gene expression by differentially binding to coactivators or corepressors in a ligand-dependent manner, which further recruits a set of epigenome-modifying enzymes that remodel chromatin conformation. Histone acetylation is a major epigenomic change controlled by histone acetyltransferases (HATs) and histone deacetylases (HDACs). HDAC3 is the only HDAC that confers the enzymatic activity to the complexes nucleated by nuclear receptor corepressors NCoR and SMRT. To address the metabolic function of HDAC3, we have deleted it specifically in mouse skeletal muscles. We have performed the following omics profiling in skeletal muscles of these mice: (1) RNA-seq profiling of total RNA; (2) Global nuclear run-on (GRO-seq) analysis of nascent RNAs; (3) Chromatin immuno-precipitation (ChIP-seq) of HDAC3 at both early evening and early morning; (4) proteomics profiling with mass spectrometry; (5) snap-shot metabolomics profiling of water-soluble metabolites at the basal condition; (6) snap-shot metabolomics profiling of lipid species at the basal condition; (7) kinetic fluxomics analysis of glucose utilization using 13C6-glucose <I>In vivo</I> during treadmill running exercise. These approaches have provided several novel insights into how nuclear receptors regulate circadian rhythm of skeletal muscle fuel metabolism, which has been published elsewhere. Silica supported $Cs_{2.5}H_{0.5}PMo_{12}O_{40}$ catalyst was prepared through sol-gel method with ethyl silicate-40 as silicon resource and characterized by X-ray diffraction, infrared spectroscopy, scanning electron microscopy, nitrogen adsorption-desorption and potentiometric titration methods. The $Cs_{2.5}H_{0.5}PMo_{12}O_{40}$ particles with Keggin-type structure well dispersed on the surface of silica, and the catalyst exhibited high surface area and acidity. The catalytic performance of the catalysts for benzene liquid-phase nitration was examined with 65% nitric acid as nitrating agent, and the effects of various parameters were tested, which including temperature, time and amount of catalyst, reactants ratio, especially the recycle of catalyst was emphasized. Benzene was effectively nitrated to mononitro-benzene with high conversion (95%) in optimized conditions. Most importantly, the supported catalyst was proved has excellent stability in the nitration progress, and there were no any other organic solvent and sulfuric acid were used in the reaction system, so the liquid-phase nitration of benzene that we developed was an eco-friendly and attractive alternative for the commercial technology.
Testing serial correlation for partially nonlinear models
Zhan-Gong Zhou,Wei-Min Qian,Chen He 한국통계학회 2010 Journal of the Korean Statistical Society Vol.39 No.4
Partially nonlinear models, as extensions of partially linear models are extensively used in statistical modeling. This paper considers the spline empirical log-likelihood ratio for testing serial correlation in partially nonlinear models. It is shown that the proposed empirical log-likelihood ratio converges to the standard chi-square distribution under the null hypothesis of no serial correlation. Some simulations are conducted to estimate the rejection probabilities under the null hypothesis and serial correlation. An example of application is also illustrated.
