        Production of a Recombinant Laccase from Pichia pastoris and Biodegradation of Chlorpyrifos in a Laccase/Vanillin System

        The recombinant strain P. pastoris GS115-lccC was used to produce laccase with high activity. Factors influencing laccase expression, such as pH, methanol concentration, copper concentration, peptone concentration, shaker rotate speed, and medium volume were investigated. Under the optimal conditions, laccase activity reached 12,344 U/L on day 15. The recombinant enzyme was purified by precipitating and dialyzing to electrophoretic homogeneity, and was estimated to have a molecular mass of about 58 kDa. When guaiacol was the substrate, the laccase showed the highest activity at pH 5.0 and was stable when the pH was 4.5~6.0. The optimal temperature for the laccase to oxidize guaiacol was 60℃, but it was not stable at high temperature. The enzyme could remain stable at 30℃ for 5 days. The recombinant laccase was used to degrade chlorpyrifos in several laccase/mediator systems. Among three synthetic mediators (ABTS, HBT, VA) and three natural mediators (vanillin, 2,6-DMP, and guaiacol), vanillin showed the most enhancement on degradation of chlorpyrifos. Both laccase and vanillin were responsible for the degradation of chlorpyrifos. A higher dosage of vanillin may promote a higher level of degradation of chlorpyrifos, and the 2-step addition of vanillin led to 98% chlorpyrifos degradation. The degradation of chlorpyrifos was faster in the L/V system (kobs = 0.151) than that in the buffer solution (kobs = 0.028).

        Prevalence, Risk Factors, and Psychological Effects of Primary Nocturnal Enuresis in Chinese Young Adults

        Purpose: This study aimed to investigate the prevalence, risk factors, and effects of primary nocturnal enuresis (PNE) on physical and mental health in young adults in mainland China. Methods: An anonymous questionnaire was used to collect information including the sociodemographic characteristics, history of PNE, family history, daytime voiding symptoms, Pittsburgh Sleep Quality Index (PSQI) scores, Self-Esteem Scale (SES), and Self-Rating Depression Scale (SDS). A total of 22,500 university students from 23 provinces and 368 cities in mainland China were included. Results: In total, 21,082 questionnaires were collected, and 20,345 of them qualified for statistical analysis. The PNE prevalence was 1.17%, and the distribution of monosymptomatic nocturnal enuresis (MNE) and nonmonosymptomatic nocturnal enuresis (NMNE) was 66.1% and 33.9%, respectively. In total, 28% of respondents with PNE reported bedwetting daily, 31.6% between 1 and 7 times weekly, and 40.4% between 1 and 4 times monthly; 80% of PNE cases had no history of treatment. The prevalence of PNE in patients with a family history, frequency, urgency, urinary incontinence, and recurrent urinary tract infections was significantly higher than in those without these conditions (P<0.001). PNE was significantly correlated with the PSQI total score (sleep quality) (P=0.011). The SES score was lower and the SDS was higher (P<0.001) in the PNE group than in those without PNE. Conclusions: In mainland China, the PNE prevalence among young adults was found to be high, and PNE had significant effects on physical and mental health. Risk factors included a family history, daytime voiding symptoms, and lack of treatment.

        — Invited Review — Understanding the functionality of the rumen microbiota: searching for better opportunities for rumen microbial manipulation

        Rumen microbiota play a central role in the digestive process of ruminants. Their remarkable ability to break down complex plant fibers and proteins, converting them into essential organic compounds that provide animals with energy and nutrition. Research on rumen microbiota not only contributes to improving animal production performance and enhancing feed utilization efficiency but also holds the potential to reduce methane emissions and environmental impact. Nevertheless, studies on rumen microbiota face numerous challenges, including complexity, difficulties in cultivation, and obstacles in functional analysis. This review provides an overview of microbial species involved in the degradation of macromolecules, the fermentation processes, and methane production in the rumen, all based on cultivation methods. Additionally, the review introduces the applications, advantages, and limitations of emerging omics technologies such as metagenomics, metatranscriptomics, metaproteomics, and metabolomics, in investigating the functionality of rumen microbiota. Finally, the article offers a forward-looking perspective on the new horizons and technologies in the field of rumen microbiota functional research. These emerging technologies, with continuous refinement and mutual complementation, have deepened our understanding of rumen microbiota functionality, thereby enabling effective manipulation of the rumen microbial community.

        FORMATION OF II–VI SEMICONDUCTOR NANOCRYSTALS WITH TUNABLE VISIBLE EMISSION IN AQUEOUS SOLUTION PROMOTED BY HYDRAZINE

        II?VI Semiconductor nanocrystals (NCs) with tunable visible emission, such as CdS, CdSe and CdTe, were synthesized in aqueous solution using thiols as capping molecules. Hydrazine was found to promote the growth of NCs through a special mechanism. In only a few hours, the synthesis process was completed at room temperature. Under moderate conditions, the capping molecules not only changed the growth rate of NCs simply by varying the concentration, but also altered the spectral properties of NCs. The capping molecules with amino groups were propitious to the growth of CdS NCs, whereas the kinetic growth of CdS NCs was more a®ected by the surface passivation e±ciency of NCs than by steric hindrance in the system. The fastest growth of the CdS NCs was achieved when glutathione was used as a capping molecule, while the emission of CdS and CdSe NCs were shown to remain steady and tunable using the same capping molecule. The growth rate of 3-mercaptopropionic acid-capped CdS and CdSe NCs slowed down signi¯-cantly, while CdTe NCs were obtained with excellent emission properties when capped with the same molecule. Furthermore, our approach will also be useful for the study of the kinetic growth of NCs in aqueous solution.

        Inhibition of ERK and Activation of p38 are Involved in Diallyl Disulfide Induced Apoptosis of Leukemia HL-60 Cells

        We investigated the effects of diallyl disulfide (DADS) on the induction of apoptosis in human Leukemia cell line HL-60 and explored the roles of mitogen-activated protein kinase (ERK and p38 MAPK) pathways in the growth inhibition and apoptosis induced by DADS. MTT assay was used to determine the DADS induced cell growth inhibition in HL-60 cells. Flow cytometry and DNA fragmentation were used to examine the roles of apoptosis in DADS-mediated cell death. Western blot analysis of the expression of phospho-MAPKs (ERK and p38) was employed to elucidate the possible mechanisms of DADS induced apoptosis. We found that growth inhibition of HL-60 cells treated with DADS exhibited a dose-dependent response (P<0.05) and DADS induced significant apoptosis. DADS at the concentration of 10 mg/L persistently activated p38 and simultaneously reduced ERK activity. PD98059, an inhibitor of ERK upstream activators MAPK kinase MKK1 and MKK2, promoted cytotoxicity and apoptosis in HL-60 cells treated with DADS. In contrast, SB203580, an inhibitor of p38, decreased cytotoxicity and apoptosis induced by DADS. Therefore, DADS can effectively inhibit the proliferation and induce apoptosis of human leukemia cell line HL-60. Inhibition of ERK signaling pathways and activation of p38 signaling pathways are likely involved in DADS induced apoptosis in HL- 60 cells.

        <▼1><P>Well dispersed MnCO3 quantum dots (∼1.2 nm) decorated on Ni(HCO3)2–MnCO3 as a cathode electrode for high performance supercapacitors.</P></▼1><▼2><P>We have developed a high performance supercapacitor cathode electrode composed of well dispersed MnCO3 quantum dots (QDs, ∼1.2 nm) decorated on nickel hydrogen carbonate–manganese carbonate (Ni(HCO3)2–MnCO3) hedgehog-like shell@needle (MnCO3 QDs/NiH–Mn–CO3) composites directly grown onto a 3D macro-porous nickel foam as a binder-free supercapacitor electrode by a facile and scalable hydrothermal method. The MnCO3 QDs/NiH–Mn–CO3 composite electrode exhibited a remarkable maximum specific capacitance of 2641.3 F g<SUP>−1</SUP> at 3 A g<SUP>−1</SUP> and 1493.3 F g<SUP>−1</SUP> at 15 A g<SUP>−1</SUP>. Moreover, the asymmetric supercapacitor with MnCO3 QDs/NiH–Mn–CO3 composites as the positive electrode and graphene as the negative electrode showed an energy density of 58.1 W h kg<SUP>−1</SUP> at a power density of 900 W kg<SUP>−1</SUP> as well as excellent cycling stability with 91.3% retention after 10 000 cycles, which exceeded the energy densities of most previously reported nickel or manganese oxide/hydroxide-based asymmetric supercapacitors. The ultrahigh capacitive performance is attributed to the presence of the high surface area core–shell nanostructure, the well dispersed MnCO3 quantum dots, and the high conductivity of MnCO3 quantum dots as well as the synergetic effect between multiple transition metal ions. The superior supercapacitive performance of the MnCO3 QDs/NiH–Mn–CO3 composites makes them promising cathode materials for high energy density asymmetric supercapacitors.</P></▼2>

        중국 주택가격 형성요인에 관한 연구

        칭다오시(靑島市)는 산동성(山東省)의 부성급도시(副省级市)로서 산동성의 주요 도시 중 하나이며 항구도시로서 한국과의 교류가 활발하게 이루어지는 도시이다. 본 연구는 칭다오시의 주택시장을 분석하고 칭다오시의 주택가격 형성요인을 분석하는 것을 목적으로 한다. 칭다오시 주택가격형성요인을 분석하기 위한 모형은 헤도닉 가격모형을 사용하였다. 자료의 공간적 범위는 스난구. 스베이구. 리창구. 라오상구. 청양구. 황다오구의 6개 구를 대상으로 하며 시간적 범위는 2014년부터 2016년까지 거래된 주택을 대상으로 한다. 칭다오 주택가격에 영향을 미치는 주택특성을 지역특성, 단지특성, 세대특성, 입지특성으로 구분하였으며 주요 분석결과는 다음과 같다. 먼저 지역특성에서 스난구나 스베이구나 리창구, 라오산구에 위치한 주택의 가격은 다른 지역의 주택보다 가격이 높은 것으로 나타났다. 이들 지역은 칭다오시의 주요 중심지역이거나 고급 주거지역으로 주택 수요자들의 주거선호 도가 높기 때문이다. 둘째, 단지특성에서는 단지의 총세대수가 많을수록, 경과연수가 짧을수 록, 주택을 건설한 시행사가 유명 시행사일 경우에 주택의 가격이 높아지는 것으로 나타났다. 셋째, 세대특성에서는 공급면적이 클수록, 세대의 향이 남향이거나 남북향인 경우 가겨이 높게 나타났다. 마지막으로 입지특성에서는 공원 또는 버스정류장과의 거리가 짧거나 지하철과 인접하여 위치하는 주택의 가격이 높은 것으로 나타났다. Qingdao City is main city of Shandong Province along with Jinan City, one of Shandong Province's major cities. As the interest of the housing market is increasing, studies on Chinese real estate are also being actively conducted. However, the reserch on the housing price in Qingdao is relatively insufficient. The purpose of this study is to analyze the housing market in Qingdao and to analyze the formation factors for housing price in Qingdao. Hedonic price model was used as a model to analyze the housing price formation factors in Qingdao City, and data were collected from China's major housing trading site, Soupang Mang. The subjects of the study were six districts: Shinan, Shibei, Licang, Laoshan, Chengyang and Huangdao District. and the time range is for houses traded from 2014 to 2016. Housing characteristics affecting Qingdao housing prices are divided into regional characteristics, complex characteristics, household characteristics, and location characteristics. The main analysis results are as follows. First, in the regional characteristics, the prices of houses located in Shinan, Shibei, Licang, and Laoshan district were higher than those in other regions. Second, in the characteristics of the complex, the more the total number of households in the complex, the shorter the elapsed years, the more famous the contractor who built the house, the house has a higher price. Third, in the characteristics of the household, the larger the supply area, the higher the price when the household's scent is south or south. Lastly, in terms of location characteristics, it was found that the distance from the park or bus stop is short, or the prices of houses located close to the subway are high.

      • Roles of E-Cadherin (CDH1) Genetic Variations in Cancer Risk: a Meta-analysis

        E-Cadherin (CDH1) genetic variations may be involved in invasion and metastasis of various cancers by altering gene transcriptional activity of epithelial cells. However, published studies on the association of CDH1 gene polymorphisms and cancer risk remain contradictory, owing to differences in living habits and genetic backgrounds. To derive a more better and comprehensive conclusion, the present meta-analysis was performed including 57 eligible studies of the association between polymorphisms of CDH1 gene promoter -160 C>A, -347 G>GA and 3'-UTR +54 C>T and cancer risk. Results showed that these three polymorphisms of CDH1 were significantly associated with cancer risk. For -160 C>A polymorphism, -160A allele carriers (CA and CA+AA) had an increased risk of cancer compared with the homozygotes (CC), and the similar result was discovered for the -160A allele in the overall analyses. In the subgroup analyses, obvious elevated risk was found with -160A allele carriers (AA, CA, CA+AA and A allele) for prostate cancer, while a decreased colorectal cancer risk was shown with the AA genotype. For the -347 G>GA polymorphism, the GAGA genotype was associated with increased cancer risk in the overall analysis with homozygous and recessive models. In addition, results of subgroup analysis indicated that the elevated risks were observed in colorectal cancer and Asian descendants. For +54 C>T polymorphism, a decreased risk of cancer was found in heterozygous, dominant and allele models. Moreover, +54T allele carriers (CT, CT+TT genotype and T allele) showed a potential protective factor in gastric cancer and Asian descendants.
