        마그네슘 풍부 해양미네랄 용액이 hairless 마우스의 아토피성 피부염에 미치는 영향

        Atopic dermatitis (AD) is a chronically relapsing inflammatory skin disease that often has asthma and allergic rhinitis. Magnesium salts, the important component of minerals in Dead Sea water, are known to exhibit beneficial effects in inflammatory disease. Favorable effects of magnesium ions and sea water treated to the skin of patients with contact dermatitis have been reported. But histological and immunological investigations are insufficient. This study was performed to examine the inhibitory effect of magnesium-rich sea mineral water on the development of AD-like skin lesions in hairless mice. AD-like skin lesions are induced by the repeated application of 2,4-dinitrochlorobenzene (DNCB). Local application of magnesium-rich sea mineral water on hairless mice skin applied with DNCB inhibited the development of AD-like skin lesions as exemplified by a significant increase in skin hydration (p<0.01), and a decrease in epidermal water loss (p<0.01). Serum IgE level was also significantly decreased (p<0.01). These results suggest that magnesiumrich sea mineral water inhibits the development of DNCB-induced AD-like skin lesions in hairless mice. These observations indicate that magnesium-rich sea mineral water may be alternative and assistant substances for the management of AD. 아토피성 피부염은 주로 천식과 비염 등을 동반하는, 주위에서 흔히 볼 수 있는 만성 염증성 피부질환으로 유전학적, 환경적, 면역학적 요인이 복잡하게 연관되어 발병한다. 해수에 포함된 마그네슘염은 피부에 작용하여 피부장벽을 보호하는 것으로 알려지고 그에 대한 면역학적인 연구와 조직학적 연구는 아직 부족한 실정이다. 이번 연구에서는 피부염을 인위적으로 일으키는 hapten 형성물질인 DNCB를 hairless mice에 도포하여 아토피 피부염 동물 모델로 만든 후, 마그네슘이 다량 함유된 해양 미네랄수를 처리한 후 피부장벽에 미치는 영향을 관찰하였다. DNCB로 피부염을 유발한 hairless mice에 해양미네랄수를 국소적으로 도포하였을 때 유의한 피부수분함량이 증가와 경피수분손실의 감소를 확인하였다 (p<0.01). 피부측정에서 피부거칠기(skin roughness, p<0.05)와 스케일생성 (skin scaliness, p<0.01)은 실험군에서 유의한 개선효과를 나타내었으며 조직학적 검사에서도 피부손상지수의 유의한 감소 (p<0.01)와 비만세포와 (p<0.01) 호산구의 감소(p<0.05) 소견을 보였고 또한 혈청 IgE의 감소를 관찰할 수 있었다(p<0.01). 이상과 같이 마그네슘이 다량 함유된 해양 미네랄수 도포는 피부장벽의 손상을 줄이고 피부수분손실을 효과적으로 줄임으로 아토피성 피부염 증상 유발을 억제할수 있음을 확인하였다. 현재까지 아토피성 피부염의 관리를 위하여 세라마이드나 식물성 오일의 보습제가 주로 활용되고 있는 상황에서 부가적인 피부장벽의 보호를 위하여 탈염 해양 미네랄수의 활용이 가능할 것으로 판단되며 장기적으로 아토피 피부염치료의 대체, 혹은 보조적 물질로 활용될 수 있을 것으로 기대된다.

        Gastrodia elata (GE) is an oriental medicinal herb which has been used traditionally for the treatment of var-ious brain diseases including convulsion and epilepsy. The purpose of this study was to determine pharmacokinetic param-eters of 4-hydroxy-3-methoxybenzaldehyde (HMBA) and p-hydroxybenzaldehyde (PHBA), constituents of GE, in rats. Male rats were cannulated in the femoral vein, femoral artery, bile duct and ureter. They received a single i.v. bolus dose of either HMBA or PHBA through the femoral vein. The concentration of HMBA or PHBA in plasma, bile and urine sam-ples were analyzed by reversed-phase HPLC. HMBA and PHBA have very short half-lives, i.e. 4.03 and 2.26 minutes respectively. Most of HMBA and PHBA were thought to be eliminated through metabolism as the metabolized fraction approaches unity. Derivatives of HMBA or PHBA with longer biological half-lives should be designed to develop better anticonvulsants and more complete qualitative and quantitative understanding of the overall pharmacokinetic fate of these compounds awaits further investigation.

        알칼리환원수 음용이 급성 염증성장질환 생쥐 모델에 미치는 영향

        알칼리환원수 (Alkaline Reduced Water: ARW)는 아시아를 비롯한 여러 나라에서 음용수로 사용되고 있으며 항산화효과와 장내이상발효 개선효과 등을 중심으로 안전성과 유효성의 검증이 이루어지고 있다. 본 연구에서는 알칼리환원수가 급성 염증성대장질환 (Inflammatory Bowel Disease: IBD) 동물모델에 미치는 영향을 알아보기 위해, ARW를 2주 동안 급이 시킨 후 4% DDS로 염증성 대장염을 유발시키고 대장의 길이와 조직병리학적 변화를 관찰하였다. 그 결과 대장의 길이와 육안적, 현미경적 질환점수의 수치가 대조군과 비교하여 유의한 차이를 나타내지 않았다. 이 결과는 DSS로 유도된 급성 염증성장질환 모델에서는 2주 동안의 ARW 급이가 개선효과를 나타내지 않았음을 말해준다. 그러나 ARW 가 장내환경을 개선시키고 위장관의 질환을 개선시키는 효과가 있음을 고려해볼 때 급성 IBD 동물모델이 ARW의 효과를 증명하기에 적합하지 않았거나 MARW의 급이기간이 장내환경을 개선할 만큼 충분히 길지 않았을 가능성을 추측해 볼 수 있다. 알칼리환원수의 장내이상발효 개선효과는 확인되고 있으나 그 기전은 아직까지 구체적으로 확인되어지지 않고 있다. 이번 연구결과에 의하면 인위적으로 유발시킨 급성 염증성 동물모델에서 ARW가 유의한 영향을 미치지 않았지만, 다른 장질환 모델을 이용한 효과 검증과 ARW의 장기 급이에 따른 효과, 그리고 ARW 가 장내환경에 미치는 작용기전에 대한 연구가 더 깊이 있게 이루어져야 할 것으로 사료된다. Alkaline reduced water (ARW) has been used for drinking widely in several countries of Asia. The safety and clinical effects of ARW has been reported including anti-oxidative effect and intestinal abnormal fermentation. To confirm the effect of ARW on DSS-induced acute inflammatory bowel disease (IBD) mouse model, we observed the length of total large intestine and the histopathological changes after supplying mineral induced-ARW (MARW) for 2 weeks and oral administration of 4% DSS (dextran sulfate sodium). As the result, the length of total large intestine and the disease scores by macro and microscopical access in the ARWsupplied group showed no significant differences compared with those in the control group. This result suggests that the supply of ARW for 2 weeks exerted no effect on amelioration in the DSS-induced acute IBD model. However, in consideration of the effect of ARW on the improvement of intestinal environment and gastrointestinal disease, this result seems that acute IBD animal model is not suitable or the period of ARW supply is not enough to prove the effect of ARW. The ameliorative effect of ARW on the intestinal abnormal fermentation has been confirmed by some researchers, but the precise mechanism also remain unclear. In conclusion, although MARW had no effect on the DSS-induced acute experimental colitis model, further studies on the verification of the effects of ARW by using other intestinal disease model and by long-term supply of ARW will be required. Also, It needs to clear the mechanism of ARW on the intestinal environment.

        Construction of antibacterial nano-silver embedded bioactive hydrogel to repair infectious skin defects

        Background: Hydrogels loaded with antimicrobial agents have been widely used for treating infected wound defects. However, hydrogels derived from a porcine dermal extracellular matrix (PADM), containing silver nanoparticles (AgNPs), have not yet been studied. Therefore, we investigated the therapeutic effect of an AgNP-impregnated PADM (AgNP–PADM) hydrogel on the treatment of infected wounds. Methods: An AgNP–PADM hydrogel was synthesized by embedding AgNPs into a PADM hydrogel. We examined the porosity, moisture retention, degradation, antibacterial properties, cytotoxicity, antioxidant properties, and ability of the PADM and AgNP–PADM hydrogels to treat infected wounds in animals. Results: The PADM and AgNP–PADM hydrogels were pH sensitive, which made them flow dynamically and solidify under acidic and neutral conditions, respectively. The hydrogels also exhibited porous network structures, satisfactory moisture retention, and slow degradation. Additionally, the AgNP–PADM hydrogel showed a slow and sustained release of AgNPs for at least 7 days without the particle size changing. Thus, the AgNPs exhibited adequate antibacterial ability, negligible toxicity, and antioxidant properties in vitro. Moreover, the AgNP–PADM hydrogel promoted angiogenesis and healed infected skin defects in vivo. Conclusions: The AgNP–PADM hydrogel is a promising bioderived antibacterial material for clinical application to infected wound dressings

        Thermal Conductivity of Diamond Packed Electrospun PAN-Based Carbon Fibers Incorporated with Multi Wall Carbon Nanotubes

        <P>Multi wall carbon nanotubes (MWCNTs) and diamond are renowned as superlative material due to their relatively high thermal conductivity and hardness while comparing with any bulk materials. In this research, polyacrylonitrile (PAN) solution incorporated with MWCNTs at an alteration of mass fractions (0 wt%, 0.6 wt%, 1 wt%, 2 wt%) were fabricated via electrospinning under optimized parameters. Dried composite nanofibers were stabilized and carbonized, after which water base polytrafluorethylene (PTFE) mixed with nano diamond powder solution was spin coated on them. Scanning electron microscopy, Raman spectroscopy, X-ray scattering and Laserflash thermal conductivity were used to characterize the composite nanofiber sheets. The result shows that the thermal conductivity increased to 4.825 W/m K from 2.061 W/mK. The improvement of thermal conductivities is suggesting the incorporation of MWCNTs.</P>

        Robust Low Complexity Predictive Control of Constrained Piecewise Affine Systems with Bounded Disturbances

        A robust low complexity model predictive control (MPC) scheme, referred to as robust one-step control, is proposed for constrained piecewise affine (PWA) systems with bounded disturbances. First, the maximal robust stabilizable set is added into the MPC formulation to guarantee the robust feasibility and low complexity. Second, the robust stability is analyzed via linear matrix inequalities (LMI). Extensive numerical examples illustrate the low complexity of the proposed robust one-step control.

        Identifying a Cinnamoyl Coenzyme A Reductase (CCR) Activity with 4-Coumaric Acid: Coenzyme A Ligase (4CL) Reaction Products in Populus tomentosa

        A cinnamoyl coenzyme A reductase (CCR, EC 1.2.1.44), one of the key enzyme involved in lignin biosynthesis, was cloned from Populus tomentosa (Chinese white poplar). At the same time, a 4CL1 gene was cloned from P. tomentosa, too. The two genes were subcloned in pQE31 vector and expressed in Escherichia coli M15. Both of them were purified by Ni-NTA. Purified CCR protein was digested by trypsin and analyzed by HPLC-MS; the peptide segments had 27% similarity with the sequence of the CCR protein. 4CL was thought to be a neighbor enzyme of CCR in lignin biosynthesis. In this paper, a 4CL1 from P. tomentosa was cloned, and its enzyme reaction products were extracted for the substrates of CCR. Three 4CL1 enzyme reaction products were monitored by HPLC-MS and then the CCR enzyme reaction was detected by GC-MS. In the CCR reaction, the three corresponding aldehyde (p-coumaraldehyde, caffealdehyde, and coniferaldehyde) were detected and identified by Frontier3 software. The results showed that the CCR that we cloned from P. tomentosa had affinities with 4CL1 enzyme reaction products and a ptCCR that was cloned from aspen (Li et al., Plant Cell Physiol 46(7):1073–1082, 2005) only had affinity with feruloyl-CoA. The different results maybe depend on the different study method. The method of exacting 4CL enzyme products as the substrates of CCR in the paper was reliable and can be used in lignin biosynthesis network to detect the enzymes in the neighborhood that depended on the polarity of the substrates and products. This CCR gene had eight homology sequence CCR gene when a BLAST was conducted in Populus trichocarpa genome database. The CCR homology genes in Populus suggested that some CCRs may take part in the lignin biosynthesis, too. The gene family would be the hot spot in the future study.
