        The Diversity of BoLA-DRB3 Gene in Iranian Native Cattle

        This study describes genetic variability in the BoLA-DRB3 gene in Iranian native cattle (Bos Indicus and Taurus) and relationships between these breeds. This is the first study of genetic polymorphism of the BoLA-DRB3 gene in Iranian native cattle. We examined exon 2 of the major histocompatibility complex (MHC) class II DRB3 gene from 203 individuals in four populations of Iranian native cattle (52 Sarabi, 52 Najdi, 49 Sistani, 50 Golpayegani cattle) using the hemi-nested PCR-RFLP method. We identified the 36 previously reported alleles and one novel pattern (*eac). Analysis of the frequencies of the various BoLA-DRB3.2 alleles in each breed indicated that DRB3.2*52 in Sarabi cattle (23%), DRB3.2 *14 and *24 alleles in Najdi cattle (13%), DRB3.2 *8 allele in Sistani cattle (22%) and DRB3.2*16 allele in Golpayegani cattle (14%), were the most frequent alleles. Allelic frequencies ranged from 1 to 23% among the 36 alleles and there were some alleles that were found only in Iranian cattle. Effective number of alleles in the four breeds was estimated to be 7.86, 11.68, 7.08 and 3.37 in Sarabi, Najdi, Sistani and Golpayegani, respectively. Observed heterozygosities were the highest in Sarabi (94%) and Najdi (94%). A population tree based on the frequency of BoLA-DRB3.2 alleles in each breed suggested that Najdi, Sarabi and Golpayegani cattle clustered together and Najdi and Sarabi were the closest breeds. Sistani cattle differed more from these three breeds. These new data suggest that allele frequencies differ between Iranian cattle breeds.

        Identification of Bovine Lymphocyte Antigen DRB3.2 Alleles in Iranian Golpayegani Cattle by DNA Test

        The bovine lymphocyte antigen (BoLA)-DRB3 gene encodes cell surface glycoproteins that initiate immune responses by presenting processed antigenic peptides to CD4 T helper cells. DRB3 is the most polymorphic bovine MHC class II gene which encodes the peptide-binding groove. Since different alleles favour the binding of different peptides, DRB3 has been extensively evaluated as a candidate marker for associations with various bovine diseases and immunological traits. For that reason, the genetic diversity of the bovine class II DRB3 locus was investigated by polymerase chain reaction-restriction fragment length polymorphism method (PCR-RFLP). This study describes genetic variability in the BoLA-DRB3 in Iranian Golpayegani Cattle. Iranian Golpayegani Cows (n = 50) were genotyped for bovine lymphocyte antigen (BoLA)-DRB3.2 allele by polymerase chain reaction and restriction fragment length polymorphism method. Bovine DNA was isolated from aliquots of whole blood. A two-step polymerase chain reaction followed by digestion with restriction endonucleases RsaI, HaeIII and BstYI was conducted on the DNA from Iranian Golpayegani Cattle. In the Iranian Golpayegani herd studied, we identified 19 alleles.DRB3.2${\times}$16 had the highest allelic frequency (14%), followed by DRB3.2${\times}$7 (11%). Six alleles (DRB3.2${\times}$25, ${\times}$24, ${\times}$22, ${\times}$20, ${\times}$15, ${\times}$3) had frequencies = 2%. Although additional studies are required to confirm the present findings, our results indicate that exon 2 of the BoLA-DRB3 gene is highly polymorphic in Iranian Golpayegani Cattle.

        Association of Leptin Polymorphism with Production, Reproduction and Plasma Glucose Level in Iranian Holstein Cows

        The objective of this study was to evaluate the association of genetic differences in the bovine leptin gene and milk yield, reproduction, body condition score (BCS), and plasma glucose level in Iranian Holstein cows. In total, two hundred and thirty eight cows were used and genotyped for a restricted fragment length polymorphism at the leptin gene locus. Two genotypes, AA and AB, have been distinguished which have the frequencies of 0.89 and 0.11, respectively. The genotypes were distributed according to the Hardy - Weinberg equilibrium ($x^2$ = 0.733). During the first 12 wk of lactation, milk yield and composition, live weight, BCS and plasma glucose were measured in 50 cows. Data were analyzed based on a repeated measures ANOVA. During this period, milk yield and composition, live weight, BCS and plasma glucose level were similar among the genotypes. The first cumulative 60-d milk yield, 305-d milk yield, days to first breeding, days open and days from first breeding to conception using previous lactation records were also analyzed using Standard Least Square within mixed models. Fixed effects were year, season, parity and age at calving, and sire. For the reproductive traits the cumulative first 60-d milk yield was also added to the model. Animal was fitted as a random effect. A significant association was detected between the RFLP-AB genotype and 305-d milk yield (p<0.05). The first 60-d cumulative milk yield was similar for the two genotypes (p = 0.21) and tended to be higher in the heterozygous cows. The heterozygous genotypes at the above mentioned locus had a trend to better reproductive performance than the homozygous. The results demonstrate that the RFLP B-allele can yield a higher 305-d milk production with a trend to better reproductive performance.

      • Genes and SNPs Associated with Non-hereditary and Hereditary Colorectal Cancer

        Background: Colorectal cancer is the third most common cancer in both men and women in the world and the second leading cause of cancer-related deaths. The incidence of colorectal cancer has increased in Iran in the past three decades and is now considered as a serious problem for our society. This cancer has two types hereditary and non-hereditary, 80% of cases being the latter. Considering that the relationship between SNPs with diseases is a concern, many researchers believed that they offer valuable markers for identifying genes responsible for susceptibility to common diseases. In some cases, they are direct causes of human disease. One SNP can increase risk of cancer, but when considering the rate of overlap and frequency of DNA repair pathways, it might be expected that SNP alone cannot affect the final result of cancer, although several SNPs together can exert a significant influence. Therefore identification of these SNPs is very important. The most important loci which include mutations are: MLH1, MSH2, PMS2, APC, MUTYH, SMAD7, STK11, $XRCC_3$, $DNMT_1$, MTHFR, Exo1, $XRCC_1$ and VDR. Presence of SNPs in these genes decreases or increases risk of colorectal cancer. Materials and Methods: In this article we reviewed the Genes and SNPs associated with non-hereditary and hereditary of colorectal cancer that recently were reported from candidate gene y, meta-analysis and GWAS studies. Results: As with other cancers, colorectal cancer is associated with SNPs in gene loci. Generally, by exploring SNPs, it is feasible to predict the risk of developing colorectal cancer and thus establishing proper preventive measures. Conclusions: SNPs of genes associated with colorectal cancer can be used as a marker SNP panel as a potential tool for improving cancer diagnosis and treatment planning.

        Immunological Responses of Broiler Chicks Can Be Modulated by Dietary Supplementation of Zinc-methionine in Place of Inorganic Zinc Sources

        Male broiler chicks were fed graded levels of organic zinc (zinc-methionine) supplementation to investigate the effects of partial or complete substitution of the organic zinc source for inorganic ones on the development of lymphoid organs and immunological responses. A total of 450 day-old male broilers were distributed into groups of 10 chicks and randomly assigned to nine experimental diets during a 42-day feeding trial. Dietary treatments consisted of two basal diets supplemented with 40 mg/kg added zinc as feed-grade Zn sulfate or Zn oxide in which, Zn was replaced with that provided from zinc-methionine (ZnMet) complex at the levels of 25, 50, 75 or 100%. Two randomly-selected birds from each pen replicate were bled and then slaughtered by cervical cutting on the final day of the trial to measure leukocyte subpopulations and relative weights of lymphoid organs. Among lymphoid organs, only thymus weight was affected (p<0.05) by dietary treatments. The sulfate-supplemented birds were heavier (p<0.01) in relative weight of thymus than oxide-supplemented birds. The 10 days of age-assessed cutaneous hypersensivity reaction was stronger in chicks fed ZnMet-containing diets. Dietary ZnMet supplementation caused (p<0.05) an increase in proportion of lymphocytes and consequently a decrease in heterophil to lymphocyte ratio. Diet fortification by zinc-methionine complex increased (p<0.01) Newcastle antibody titer at 19 days of age. Also, a similar response was observed in antibody titers at 6 and 12 d after infectious bronchitis vaccine administration. There was no significant effect of replacement of dietary zinc on antibody titer against infectious bursal disease virus (IBDV) at the 6th d post-vaccine inoculation; however, at d 12 after vaccination, ZnMet-fortified diets improved antibody titer against IBDV. Although dietary inclusion of ZnMet had no marked effect on primary antibody titer against sheep erythrocytes, effective responses were observed during secondary reaction from the viewpoint of both total antibody and immunoglobulin Y (IgY) titers. From the present findings, it can be concluded that dietary supplementation with organic zinc improves both cellular and humoral immune responses. It is necessary to replace 75% of supplemental inorganic zinc with organic ZnMet complex to achieve the optimum immunological responses in broiler chicks.

        Nutritional Evaluation of Full-fat Sunflower Seed for Broiler Chickens

        Two experiments were conducted to evaluate the use of various levels of full-fat sunflower seeds (FFSS) on broiler performance and carcass characteristics. In the first experiment, FFSS was included in a basal diet at 70, 140, and 210 g/kg and the $AME_n$ values of the experimental diets were determined. The linear regression equation of $AME_n$ values on rate of inclusion was calculated. Extrapolation value for the $AME_n$ of FFSS at 100% inclusion was 14.22 MJ/kg. In the second experiment, diets containing various levels (0, 70, 140, and 210 g/kg) of FFSS were given to broilers (Ross strain) from 0 to 49 d. At 28 days of age, blood parameters and digestive enzyme activities were determined and carcass parameters were evaluated at 49 days of age. Weight gain, feed intake and feed conversion ratio (FCR) were improved (p<0.05) when broilers were fed various levels of FFSS in the starter and finisher diets. Breast, thigh, gastrointestinal tract and gizzard weight percentages were not affected by dietary treatments; however, liver weight percentage was decreased significantly (p<0.05) and weight of abdominal fat decreased but this effect was not significant. The activities of digestive enzyme (protease and ${\alpha}$-amylase) were not influenced by the treatments. Activity of alkaline phosphatase, concentrations of calcium, phosphorus, glucose, triglyceride, protein, high density lipoprotein (HDL) and low density lipoprotein (LDL) were not affected by incorporation of FFSS in the broiler diet. Although concentration of HDL increased and LDL decreased, these effects were not significant. The results of this study indicate that FFSS can be used at up to 21% in broiler diets without adverse effects on performance or other parameters of chickens.

      • Hereditary Genes and SNPs Associated with Breast Cancer

        Breast cancer is the most common cancer among women affecting up to one third of tehm during their lifespans. Increased expression of some genes due to polymorphisms increases the risk of breast cancer incidence. Since mutations that are recognized to increase breast cancer risk within families are quite rare, identification of these SNPs is very important. The most important loci which include mutations are; BRCA1, BRCA2, PTEN, ATM, TP53, CHEK2, PPM1D, CDH1, MLH1, MRE11, MSH2, MSH6, MUTYH, NBN, PMS1, PMS2, BRIP1, RAD50, RAD51C, STK11 and BARD1. Presence of SNPs in these genes increases the risk of breast cancer and associated diagnostic markers are among the most reliable for assessing prognosis of breast cancer. In this article we reviewed the hereditary genes of breast cancer and SNPs associated with increasing the risk of breast cancer that were recently were reported from candidate gene, meta-analysis and GWAS studies. SNPs of genes associated with breast cancer can be used as a potential tool for improving cancer diagnosis and treatment planning.

      • Identification of Germline BRCA1 Mutations among Breast Cancer Families in Northeastern Iran

        Background: The purpose of this study was to evaluate the prevalence of BRCA1 (MIM: 113705) founder mutations in familial breast cancer (BC) patients with high risks in Iran. BRCA1 is among the cancer susceptibility genes best known for high penetrance mutations. BRCA1 genotyping is now used to determine patient counseling, management decisions, and prognosis of this syndrome. Materials and Method: Thirty nine patients with clinical BC and 29 high risk healthy women, related to the patients, participated in the study. DNA from blood samples was extracted and analyzed by PCR and SSCP methods in order to find 185delAG and 5382insC founder mutations. In addition, a 251bp fragment of BRCA1's exon 11 was amplified and analyzed for determination of new mutations. Results: The data indicated the presence of 185delAG and 5382insC founder mutations in both groups studied. Two out of 39 BC patients (5.1%) and one out of 29 relatives (3.4%) were suspected to be carriers of 185delAG mutations. However, we found only one patient (2.6%) to be a carrier of a 5382insC mutation. Also, 2 women (5.1%) of the patient group and 3 n (10.3%) of relatives group were identified as carriers of unclarified mutations in the 251bp fragment of the BRCA1 gene. The carriers of BRCA1 founder mutations have a high lifetime risk of breast cancer. Conclusions: Therefore, these data are useful in counseling of individuals with a significant family history of breast cancer.
