      • 바이칼린 함유 생약의 제제화 및 생체 이용률 (제 2보) : 황금 및 황련 공침물의 장내 흡수 및 항균 효과 Gastro-Intestinal Absorption and Antibacterial Effect of Coprecipitated Product of Scutellariae Radix and Coptidis Rhizoma

        Precipitation was formed during the preparation of decoction from mixure of Scutellariae Radix and Coptidis Rhizoma or Phellodendri Cortex according to the prescription of Hwang-ryean-hae-dog-tang. Baicalin and berberine, the active ingredients of the two herbal medicine were identified in coprecipitated product. Pills were prepared using the coprecipitated product and various binders. The dissolution rate of baicalin and berberine from pills was increased in at pH1.2 when acacia or tragacanth was used. The absorption rate of baicalin from the coprecipitated product was raster than the from Scutellaria extract, but the absorption of berberine from CPP was slower in stomach, duodenum and jejunum of rats compared with Coptis expract. The time equired for the maximum serum concentration (Cmax) of baicalin and berberine from CPP in mice were 150 and 200 min after oral administration. respectively. The maximum serum concentration of baicalin from CPP in mice was higher than Scutellaria extract, but the concentration of berberine was lower compared with Coptis extract. The minimum inhibitory concentration of CPP was below 50 ug/ml against gram positive bacteria, and was higher than that against gram negative bacteria. The antibacterial activity of CPP was lower than the of berberine.but was more potent than Scutellaria extract. It was found that the ingibition rates of growth by CPP against S. cpidermidis, K. pneumoniae, B cereus and S. aureus were 60.1, 5.11, 45.4, and 39.9%. respectively.

        "지황 1호"를 이용한 숙지황 제조기술 연구

        재래종 지황과 신육성품종 지황 1호를 이용하여 절단 및 원형으로 숙지황을 제조한 결과는 다음과 같다. 1. 가압에 의한 숙지황 제조시간이 재래종은 254∼264시간, 제품수율이 71.42∼71.48%였고, 지황 1호는 제조시간이 260∼276시간 소요되었고, 수율은 71.50∼71.51%였으며 중량은 감소하는 경향이었다. 2. 상압에 의한 숙지황 제조에서도 가압에 의한 제조와 비슷한 경향을 보였다. 3. 가압과 상압에 의한 숙지황 제조 후 유리당 조성은 재래종, 지황 1호 전처리구에 63∼65% 이상이 fructose였으며, 30% 이상이 glucose였고, sucrose, maltose가 함유되었다. 4. 가압 및 상압에 의한 숙지황 제조시 catalpol 함량 변화는 종자회수가 많아질수록 caltalpol 함량이 낮아지는 경향이었으나 완전히 없어지지는 않았다. 5. 숙지황 제조 후 포장재별로 포장하여 상온에 저장한 결과 제품에 곰팡이 발생이 없어 저장에 문제가 없었다. New processing method for R. Radix preparata was developed by using the landrace variety as a control and "Jiwhang 1" (Rehmannia glutinosa Liboschitiz). Sliced Rehmanna glutionsa Liboschitiz was steam heated by normal and high pressure conditions for the round-shaped R. Radix preparata processing. About 254 to 264 hours were required for the processing of domestic variety, and about 260 to 280 hours were required for "Jiwhang 1", but their yields were ranged about 71.42∼71.6%. Fructose and glucose were major free sugar of R. Radix preparata, and their composition rates were 63∼65% and 30%, respectively, but sucrose and maltose were traced only small amount. The Catalpol content was decreased as increasing the frequencies of steam heating repetion. But the catalpol was not completely removed. After processing, R. Radix preparata packed with paper-bag or PE-bag, and storaged at common temperature was keeping a best quality.

        치자 엑스의 가수분해에 의한 피부투과 및 항염효과

        Geniposide and its related iridoid compounds have been used in traditional herbal medicine for the treatment of Jaundice hepatic diseases and various inflammatorys. For the purpose to increase trandsdermal absorption, the hydrolyzed products of Gardeniae Furcuts were identified and assayed of active ingredients and investigated trandsdermal absorption and anti-inflammatory effects. Geniposide was hydrolyzed to genipin by β-glucosidase and it was suggested that genipin was more suitableform than geniposide for transdermal absorption by its lipophilic property. Using Franz type diffusion cell and the skin of hairless mouse, the permeation rate of hydrolyzed products and their emulsion preparation were determined. Genipin have more increased absorption ratio through the skin hairless mouse than geniposide. Also, the emulsion of hydrolyzed products of extracts showed higher permeability than that of nonhydrolyzed preparations. After 9 hours 280.85 ㎍/㎠ of genipin was absorbed and 193.52 ㎍/㎠ in case of geniposide. The Js of geniposide and genipin were 26.27±4.11 ㎍/㎠/hr and 40.35±5.04 ㎍/㎠/hr respectively. After carrageenan injection, the swelling was increased rapidly to 24 hr and maintained as a plateau. But emulsion group were reached about 2.5 mL and the swelling decreased successively from 24 hr to 72 hr. The anti-inflammation effects of extracts and hydrolyzed products emulsion were increased with significant difference with control group after 24 hr, 48 hr and 72 hr. In carrageenan induced edema, inhibition of swelling was increased in case of hydrolyzed product emulsion compare with nonhydrolyzed group at 24 hr, 48 hr and 72 hr after swelling. In histological study, the anti-inflammatory effects of hydrolyzed products were remarkable at 48 hr and 72 hr compare with nonhydrolyzed. Hydrolyzed products of Gardeniae Frucuts extract containing genipin would be a suitable preparation to increase the transdermal absorption and anti-inflammatory effects.

        The circuit substrate was made from the LTCC that a ε_(r) was 7.8. Accumulated Varactor and the low noise transistor which were a Surface Mount Device-type element on LTCC substrate. Let passive element composed R, L, C with strip-line of three dimensions in the multilayer substrate circuit inside, and one structure integrated band-pass filter, resonator, a bias line and a matching circuit. Used Screen-Print process, and made Strip-line resonator. A design produced and multilayer-type VCO and recognized a characteristic with the Spectrum Analyzer which was measurement equipment. Measured multilayer structure VCO is oscillation frequency 1292[MHz], oscillation output -28.38[dBm], harmonics characteristic -45[dBc] in control voltage 1.5[V]. A phase noise is -68.22[dBc/Hz] in 100 [kHz] offset frequency. The oscillation frequency variable characteristic showed 30[MHz/V] characteristic, and consumption electric current is approximately 10[mA].

      • 폐쇄성 황달에 대한 내시경적 비 담도 배액법(ENBD)의 효과

        To evaluate the clinical effects of ENBD in patients with biliary obstruction, 18 patients were included in this study. The results were as follows ; 1) The causes of obstructive jaundice in 18 patients, with performed ENBD were distal CBD cancer (3 cases), CBD stone with cholangitis (14 cases) and CBD stone without cholangitis (1 case). 2) The duration of ENBD was 12.5 days (8-20 days). 3) The positive results of bacteriologic culture via ENBD tube revealed 8 cases among 13 (61.5%). 4) Amory 15 cases with CBD stones, Basket removal (2 cases) and Mechanical lithotripsy (1 case) perfomed in 3 cases after EST and 4 cases had an operation after ENBD. Three patients with CBD cancer underwent ERBD. In conclusion, ENBD is a safe and effective noninvasive biliary drainage method for initial decompression of malignant or benign biliary obstruction with or without cholangitis (especially with).

      • Felodipine 함유 Eudragit microsphere의 생체이용율

        Microencapsulations of felodipine which is one of the calcium channal blocker, using the mixture of Eudragit RL®, L® and E® and celullose on the base of Eudragit RS® were investigated. The average diameters of microspheres were from 40 to 55㎛. The incorporation ratios of felodipine into the microspheres were higher than 90 %. After 7h, about 60 % of felodipine was released. Dissolution rate of felodipine from microsperes was increased by the addition of Cremopher®. After oral administration of microspheres, the Cmax were reduced and incresed the bioavailities as up to 50 %.

        치자엑스 가수분해물 제제의 피부투과 및 생체 피부조직과의 교차결합에 관한 연구

        Gardeniae Fructus is consisted of geniposide and its derivatives. For the purpose of treatment of skin disease, geniposide and hydrolyzed products (HP) of Gardeniae Fructus were studied on skin permeation and crosslinking with biological tissue. The hydrolyzed products (HP) and active ingredients of Gardeniae Fructus were identified and investigated about skin permeability. Genipin has provided low cytotoxic crosslinking reagents and formed stable and biocompatible crosslinked products. The permeation enhancing effects of geniposide and genipin under the hydrolyzed products of cream and hydrogel preparations were tested using Franz type diffusion cell and the skin of hairless mouse. The remaining pro-portions of geniposide and genipin were measured in the hydrolyzed products of cream and hydrogel preparations. The crosslinking of epidermic and endodermic tissue with genipin under the hydrolyzed prodcuts of cream and hydrogel preparation was observed using light microscopy. Increased absorption ratio of the skin of hairless mouse about genipin was higher than that of geniposide. Loads at break, tensile strengths and skin permeation rate of the hydrolyzed products (HP) of cream and hydrogel preparations were higher than the nonhydrolyzed products (NHP). The hydrolyzed products (HP) of cream and hydrogel of Gardeniae Fructus Extracts were proper preparations and crosslinking agents to increase the trans-dermal absorption with epidermic and endodermic tissue.

In this process they raise various climate appearance. And that various climate appearance influence the whole lives on the earth. This paper is concerned with the classification of them.

        Abstract - in this study, Polymide(PI) Thin-films of MIS structure and MIM structure is deposited on Si wafer and AI Thin-films based on spincoating method. Using material is SE5291(PAA) of NISSAN CHEMICAL. Spincoating velocity is 4000[rpm] for 5[min] The uniform thin films of PI formed by curing at 150[℃] for 30[min](Soft baking) and 350[℃] for 60[min](Hard baking). Thickness of PI Thin-films is 150-170 [nm]. For Electrical characteristics of PI Thin-films measured Ⅰ-Ⅴ properties, C-V properties and f-C properties. The result analysis is like below: Ⅰ-Ⅴ characteristic using HP 4156A, C-V is HP 4280A f-C is HP 4294A

        베타-글루쿠로니다제에 의한 황련ㆍ황금 공침물의 가수분해 및 생체이용률 증가

        During the preparation of decoction from the mixture of Coptidis Rhizoma and Scutellariae Radix, insoluble copreciptate was formed. The coprecipitated product(COP) was composed of berberine and baicalin which was the active ingredient of Coptidis Rhizoma and Scutellariae Radix, respectively. COP was slightly soluble in water and could not be well absorbed after oral administration. This poor bioavailibility might be associated with its poor aqueous solubility. With the purpose of increasing the solubility and bioavailibility of COP, hydrolysis of COP by β-glucuronidase was carried out. Hydrolyzed products (HOP) of COP were identified and assayed for active ingredients. The partition coefficient study, in situ absorption test, and pharmacokinetic study after oral administration were also performed. COP was found to be consisted of berberine and baicalin with molecular ratio or 1 to 1. This compound was hydrolyzed to berberine and baicalin by β-glucuronidase. The rate of hydrolysis was higher at higher temperature up to 50℃ and higher concentration of β-glucuronidase up to 2500 unit under our experimental conditions. Baicalein, which is more liphophilic than baicalin, showed greater absorption in small intestine than baicalin did. The plasma concentrations of berberine and baicalein after oral administration of HOP were significantly higher than those of COP. The possible mechanism of increased bioavailibility of berberine and baicalein could be the hydrolysis of COP by β-glucuronidase. On the basis of the above results, it might be said that HOP should be a suitable preparation for increasing the bioavailibility of Coptidis Rhizoma and Scutellariae Radix.
