        Alteration of the Fatty Acid Profile of Pork by Dietary Manipulation

        This work was undertaken to study the effect of dietary fat source on the fatty acid profile of pork, and to evaluate the effect of inclusion of vitamin E in pig diets on lipid oxidation of pork tissue and processed pork products. Fifty-six pigs were allocated to four treatments, that included two dietary fat sources and two levels of vitamin E inclusion. Dietary fat was derived from either tallow, a source of saturated fatty acids (SFA), or from a mixture of soybean and linseed oils, which contain polyunsaturated fatty acids (PUFA). Vitamin E was included at either 0% or 0.011% of the diet. Growth and carcass characteristics were not affected by the dietary treatments. Dietary fat source affected the fatty acid profile of the longissimus muscle and subcutaneous fat tissue, with the PUFA diet resulting in significantly more polyunsaturated fatty acids in the tissues, and more favourable ratios of SFA to PUFA and C18:2 to C18:3 in terms of human health considerations. Lipid oxidation was significantly greater in tissues and processed products from PUFA-fed pigs. Inclusion of vitamin E in the diets, however, reduced the extent of lipid oxidation in the meat and meat products. Dietary manipulation of the fatty acid profile of pigs is an effective means of altering the fat composition of pork in order to provide human consumers with a healthy product. Vitamin E is effective as an antioxidant agent, particularly where processed products are concerned.

        Effects of Particle Size of Barley on Intestinal Morphology, Growth Performance and Nutrient Digestibility in Pigs

        A growth trial and a digestibility trial were conducted to examine the effect of feed particle size on the performance, nutrient digestibility, gastric ulceration and intestinal morphology in pigs fed barley-based diets. Barley was processed through a hammer mill to achieve four diets varying in particle size (average particle $size{\pm}standard $deviation): coarse ($1,100{\pm}2.19\;{\mu}m$), medium ($785{\pm}2.23\;{\mu}m$), fine ($434{\pm}1.70\;{\mu}m$) and mixed (1/3 of coarse, medium and fine) ($789{\pm}2.45\;{\mu}m$). Sixty-four entire male pigs were used in the growth trial and the diets were fed ad libitum between 31 kg and 87 kg live weight. Following slaughter, stomach and ileal tissues were scored for integrity (ulceration or damage) and histological measurements taken. Twenty-four entire male pigs were used in the digestibility trial, which involved total faecal collection. Over the entire growth phase, there were no differences (p>0.05) in average daily gain and feed conversion ratio between pigs fed diets of different particle size. Pigs fed the coarse and medium diets had lower (p<0.05) stomach ulceration scores (0.20 and 0.25, respectively, on a scale from 0 to 3) than those fed the mixed (0.69) or the fine diets (1.87). The stomachs of all animals fed the fine diet had lesions and stomach ulcerations were present only in this group. Pigs fed the fine diet had thicker (p<0.001) ileal epithelial cell layer with no differences (p>0.05) being observed for villous height or crypt depth. Faecal digestibility coefficients of neutral and acid detergent fibre were the highest (p<0.05) for the mixed diet, intermediate for the fine and coarse diets and the lowest for the medium diet. A similar numerical trend (p = 0.103) was observed for the apparent faecal energy digestibility coefficient. It is concluded that, with barley based diets, a variation in average particle size between $400{\mu}m$ and $1,100{\mu}m$ had no effect on pig performance but the fine dietary particle size affected the integrity of the stomach, as well as the structure of the small intestine, thus compromising overall gut health. Our data also demonstrate that changes in particle size distribution during the digestion process, rather than average particle size or particle size variation, are related to apparent faecal digestibility.

        Effect of Non-starch Polysaccharides and Resistant Starch on Mucin Secretion and Endogenous Amino Acid Losses in Pigs

        Generally, dietary fibre (DF) includes lignin, non-starch polysaccharides (NSP) and resistant starch (RS). In monogastric species, low levels of dietary fibre in the diet are associated with various diseases and high levels reduce nutrient digestibilities. In this study, the effects of different types and levels of NSP (soluble: $\beta$-glucan, insoluble cellulose) and resistant starch on mucin secretion and endogenous nitrogen and amino acid losses in pigs were investigated. A total of 25 five-week-old weaner pigs (9.5 kg${\pm}$1.5 kg), were randomly allocated to each of five experimental diets. Different levels of purified barley $\beta$-glucan (BG) extract (5 or 10% of $Glucagel^{(R)}$ $\beta$-glucan, providing 4 or 8% of BG in the diet), and resistant starch (RS) (8.3 or 16.6% of Hi-$Maize^{TM}$, providing 5 or 10% RS in the diet) were substituted for wheat starch in a purified diet in which enzymatically-hydrolysed casein was the sole source of protein. The diets were fed for 21 days. No statistically significant difference between treatments (p>0.05) was observed for growth performance and organs weights. No difference in ileal starch digestibility was observed between pigs on the cellulose or $\beta$-glucan diets. However, as the level of resistant starch in the diet increased the ileal starch digestibility decreased (p<0.05). The inclusion of resistant starch in the diet (5 or 10%) did not increase mucin production when compared with the cellulose-only diet. However, as the level of beta-glucan in the diet increased, both crude mucin in the digesta dry matter and per kg dry matter intake increased (p<0.05). Pigs fed the diet containing 8% of beta-glucan had higher endogenous loss flow than those fed the diets including 5 or 10% of resistant starch or 4% of $\beta$-glucan. In conclusion, dietary inclusion of resistant starch increased the level of starch reaching the large intestine without any effect on mucin secretion, or endogenous nitrogen or amino acid losses content in the small intestine. The addition of $\beta$-glucan to a diet containing cellulose increases both mucin secretion and endogenous amino acid and nitrogen losses in the small intestine.

        Effect of Non-starch Polysaccharides on Mucin Secretion and Endogenous Amino Acid Losses in Pigs

        This study was undertaken to examine the influence of soluble non-starch polysaccharides on growth performance, mucin secretion, and endogenous amino acid flows in weaner pigs. Different levels (0, 4 and 7.5%) of purified corn arabinoxylan (AX) or barley $\beta$-glucan extract (BG) were substituted for cellulose in a purified diet based on starch, sucrose and enzymatically hydrolyzed casein. All diets contained titanium oxide as an indigestible marker. Each experimental diet was fed to five, 6-wk old weaner pigs for 21 days. Average daily gain (p<0.05) and feed conversion ratio (p<0.01) were improved with dietary inclusion of 7.5% AX and BG, indicating high degradation rates of AX and BG in pigs. Crude mucin contents and endogenous nitrogen flow were increased (p<0.05) with increased levels of AX, but not with BG. Numerical increases in endogenous amino acid flow (EAAF) were observed with increased levels of AX but no definite trend with BG. Endogenous amino acid flow in pigs fed mixed NSP diets (4% BG and 3.5% cellulose) was significantly higher (p<0.05) than those fed 7.5% BG diets. Among diets containing pure sources of soluble non-starch polysaccharides, endogenous amino acid flows were highest in 7.5% AX (p<0.05), intermediate in BG, and lowest in control diet. Increased flows (p<0.01) of threonine, proline and serine in pigs fed 7.5% AX diets are consistent with the increased flow of crude mucin determined in this treatment. In conclusion, mucin and endogenous amino acid flows were increased with dietary inclusion of AX, which could be related to its physicochemical property, particularly its high water-holding capacity. In contrast, $\beta$-glucan, due to its high degradation rate in pig, may be considered as unimportant factor in inducing mucin and endogenous amino acid secretions, at least at levels such as those used in this study.

        Evaluation of Three Pork Quality Prediction Tools Across a 48 Hours Postmortem Period

        Numerous reports have evaluated the predictive ability of carcass probes for meat quality using measurements taken early postmortem or near 24 h. The intervening time period, however, has been largely ignored. In this study, the capacity of three probes [pH, electrical conductivity (EC), and grading probe light reflectance (GP)] to predict pork longissimus muscle quality (drip and cooking losses, Warner-Bratzler shear, L*, n = 30) was evaluated at 45 min, 90 min, 3, 6, 12, 24, and 48 h postmortem. The strongest relationships were observed between cooking loss and 6 h EC and GP (R2 = 0.66, 0.72), and L* and GP (R2 = 0.57-0.66, 12-48 h). pH was most valuable early postmortem (R2 = 0.63, 90 min with cooking loss). GP at 6 h most effectively (R2 = 0.84) predicted a two factor (cooking loss+L*) meat quality index. Results emphasize the predictive value of measures taken between 3 and 12 h postmortem.

        Immune metabolism regulation of the germinal center response

        The humoral immune response requires germinal centers to produce high-affinity antigen-specific antibodies that counter pathogens. Numerous studies have provided a better understanding of how metabolic pathways regulate the development, activation and functions of immune cells. Germinal centers are transient, highly dynamic microanatomic structures that develop in lymphoid organs during a T-cell-dependent humoral immune response. Analysis of germinal centers provides an opportunity to understand how metabolic programs control the differentiation and function of highly specialized germinal center B cells and follicular helper CD4+ T cells. Targeting immunometabolism during the germinal center response may afford the possibility to improve vaccine design and to develop new therapies to alleviate autoimmunity. In this review, we discuss the major metabolic pathways that are used by germinal center B and T cells, as well as the plasma cells that they produce, all of which are influenced by the microenvironment of this unique structure of the adaptive immune system.

        Dietary Bovine Colostrum Increases Villus Height and Decreases Small Intestine Weight in Early-weaned Pigs

        This experiment examined the effect of dietary spray-dried bovine colostrum on intestinal histology and organ weights in early-weaned pigs. In a randomised complete block design, twelve 14-day-old weaner pigs were offered a diet containing either 5% spray-dried bovine colostrum or no colostrum (control). Diets were formulated to contain 14.8 MJ/kg DE, 1.26% available lysine and to meet or exceed requirements for other nutrients. Piglets were offered the diets for a period of 14 days. No effect of diet on growth rate or feed intake was observed (p>0.10). Small intestine weight was reduced by 12% in piglets consuming dietary bovine colostrum (p< 0.05). Villous height and crypt depth were increased and decreased, respectively, in the proximal jejunum, mid jejunum and distal ileum of pigs consuming dietary bovine colostrum (p<0.05). Mid-jejunal lamina propria $CD4^+$ and $CD8^+$ T lymphocyte density was increased by 28 and 37%, respectively, in piglets consuming dietary bovine colostrum (p<0.05). Diet did not affect thickness of tunica muscularis externa or tunica submucosa (p>0.10). Collectively, these results suggest a positive effect of dietary bovine colostrum on intestinal morphology and immune status in early-weaned pigs.
