        Fusarium Species from Sorghum in Thailand

        Sorghum is the fifth most important cereal worldwide, spreading from Africa throughout the world. It is particularly important in the semi-arid tropics due to its drought tolerance, and when cultivated in Southeast Asia commonly occurs as a second crop during the dry season. We recovered Fusarium from sorghum in Thailand and found F. proliferatum, F. thapsinum and F. verticillioides most frequently, and intermittent isolates of F. sacchari and F. beomiforme. The relatively high frequencies of F. proliferatum and F. verticillioides, suggest mycotoxin contamination, particularly fumonisins and moniliformin, should be evaluated. Genetic variation within the three commonly recovered species was characterized with vegetative compatibility, mating type, Amplified Fragment Length Polymorphisms (AFLPs), and female fertility. Effective population number ($N_e$) was highest for F. verticillioides and lowest for F. thapsinum with values based on mating type allele frequencies higher than those based on female fertility. Based on AFLP genetic variation, the F. thapsinum populations were the most closely related, the F. verticillioides populations were the most distantly related, and the F. proliferatum populations were in an intermediate position. The genetic variation observed could result if F. thapsinum is introduced primarily with seed, while F. proliferatum and F. verticillioides could arrive with seed or be carried over from previous crops, e.g., rice or maize, which sorghum is following. Confirmation of species transmission patterns is needed to understand the agricultural systems in which sorghum is grown in Southeast Asia, which are quite different from the systems found in Africa, Australia, India and the Americas.

        High Plasma Sphingosine 1-phosphate Levels Predict Osteoporotic Fractures in Postmenopausal Women: The Center of Excellence for Osteoporosis Research Study

        Background: Higher sphingosine 1-phosphate (S1P) plasma levels are associated with decreased bone mineral density (BMD), and increased risk of prevalent vertebral fracture. So, we hypothesized that postmenopausal women with increased baseline plasma S1P levels have a greater risk for future incident fracture (osteoporosis-related fractures [ORFs]). Methods: This study was conducted in a prospective longitudinal cohort of 707 women recruited in 2004 and followed up annually for a mean period of 5.2±1.3 years. They were postmenopausal (aged ≥50 years). The primary outcome measure was the time to the first confirmed ORF event using radiographs and/or a surgical report. Results: The plasma S1P levels (μmol/L) were significantly higher in the women with incident fracture (7.23±0.79) than in those without ORFs (5.02±0.51; P<0.001). High S1P levels were strongly associated with increased fracture risk. After adjustment for age and other confounders, the hazard ratio (HR) was 6.12 (95% confidence interval [CI], 4.92-7.66) for each 1-standard deviation increase in plasma S1P levels. The women in the highest quartile of S1P levels had a significant increase in fracture risk (HR, 9.89; 95% CI, 2.83-34.44). Results were similar when we compared plasma S1P levels at the 1-year visit. Conclusions: The associations between plasma S1P levels and fracture risk were independent of BMD and other confounders. These findings demonstrate that high plasma S1P level at baseline and at years 1 to 5 is a strong and independent risk factor for future [ORFs] among postmenopausal women and could be a useful biomarker for fracture risk assessment in this population.

        Gene Knockout Identification for Metabolite Production Improvement Using a Hybrid of Genetic Ant Colony Optimization and Flux Balance Analysis

        The increasing demand of biochemical supply for various industries has spurred the development of metabolic engineering to find the optimal design of the microbial cell factories. Traditional method of chemical synthesis using the natural producer leads to the production far below their theoretical maximums. Gene knockout strategy is then introduced to improve the metabolite production. To aid the process, many computational algorithms have been developed to design the optimal microbial strain as cell factories to increase the production of the desired metabolite. However, due to the size of the genome scale model of the microbial strain, finding the optimal combination of genes to be knocked out is not an easy task. In this paper, we propose a hybrid of Genetic Ant Colony Optimization (GACO) and Flux Balance Analysis (FBA) namely GACOFBA to find the optimal gene knockout that increase the production of the target metabolite. Using E. coli and S. cerevisiae genome scale model, we test our proposed hybrid algorithm to increase the production of four different metabolites. By comparing with the results from existing method OptKnock as well as the conventional Ant Colony Optimization (ACO), the results show that our proposed hybrid algorithm able to identify the best set of genes and increase the production while maintaining the optimal growth rate.

        The Role of Brand Knowledge in Explaining Relationship between Brand-Centered Communications and Brand Commitment: Evidence from Public Banks Pakistan

        Purpose – The purpose of this study is to examine the extent to which internal brand communication (IBC) impacts on brand commitment (BC). Research design, data, and methodology – A structured questionnaire has been built and distributed on a total sample of 650 banking staff working in five public banks in Pakistan. Results – The findings of PLS-SEM revealed that besides the indirect effect of internal brand communication (IBC) on brand commitment via brand knowledge, the study also exposes a direct positive effect of IBC on brand commitment. This study also reveals that brand commitment is affected by brand knowledge and internal brand communication practices. The result of mediation reveals that brand knowledge partially mediated the relationship between independent and dependent variables. Practical implications- This study empirically validates that internal brand communication and brand knowledge are prerequisites for brand commitment. Therefore, managers, particularly in-service enterprises, should provide sufficient IBC practices and appropriate brand knowledge to enhance employees’ brand commitment. Conclusions – The comprehensive analysis of the impact of brand knowledge on the proposed relationship. It further adds up to the branding literature, which is mostly qualitative and lacks empirical validation.

        Effect of supplementary lighting on nutrient recovery of Ocimum basilicum and fishes in a polyculture aquaponic system

        The effects of prolonged photoperiod (additional night lighting) were investigated on the production of Ocimum basilicum (lemon basil) and fish/crayfish raised in a nutrient film technique aquaponic system. Hybrid lemon fin barb and red claw crayfish juveniles were co-cultured with O. basilicum and subjected to 12 h of ambient natural daylight and additional 0, 4, 8, and 12 h of night lighting for 14 weeks (two batches of 7 weeks each). The water quality parameters and the performance characteristics of the fish/crayfish/plant were evaluated, and the nutrient uptake (nitrogen, phosphorus, and potassium) in the system by the three organisms was also measured. The fish growth performance showed no significant differences (p > 0.05) in all the growth parameters measured across the treatments. The body proximate and nitrogen, phosphate, and potassium (NPK) composition of the fish and crayfish were not significantly (p > 0.05) different across the treatments. Similarly, there was no significant difference (p > 0.05) in the NPK uptake by the fish and crayfish across the treatments. The plant growth performance showed that there were no significant (p > 0.05) differences in the plant growth parameters measured, except for % plant height gained in the 12 h-light treatment (403.2 ± 26.0%) which was significantly (p < 0.05) higher than other treatments and cycles. Hence, this study demonstrated that varying supplementary night lighting has no significant effect on the growth performance of O. basilicum, hybrid lemon fin barb, and red claw crayfish. This study, therefore, suggests that supplementary night lighting should not be considered for O. basilicum production as it does not significantly improve the performance characteristics of the plant nor the fish co-cultured with it in a polyculture aquaponic system.
