Suppressive effects of fructus of Magnolia denudata on IL-4 and IL-13 expression in T cells.

<P>Magnolia species have been used for the treatment of allergic diseases in Asia as folk medicine; however, the cellular and molecular mechanisms of its anti-allergic effects have rarely been investigated. In this study, we demonstrated that a methanolic extract of the fructus of Magnolia denudata has suppressive effects on Th2 cytokine production such as IL-4 and IL-13, but not IFN-γ and IL-17, produced by both phorbol 12-myristate 13-acetate/ionomycin (PI)- and CD3/CD28-stimulated EL-4 T cells. Moreover, the mRNA expression of Th2 cytokines was significantly inhibited, and luciferase activity in cells transiently transfected with IL-4 or IL-13 promoter reporter plasmids was suppressed by M. denudata, indicating that M. denudata may regulate these expression at the transcriptional level. Western blot analysis for transcription factors involved in the cytokine gene expression indicated that the activation of c-Jun was significantly downregulated in the nucleus of cells, while the activations of nuclear factor of activated T cells, nuclear factor kappa B and c-Fos, were not affected. Furthermore, the mRNA expression and nuclear translocation of GATA-binding protein 3, a key transcriptional factor for Th2 commitment and Th2 cytokine expression, but not T-bet and RORγt, were dramatically downregulated by M. denudata. Treatment with M. denudata suppressed the phosphorylation of p38 mitogen-activated protein kinase; however, the PI-induced phosphorylation of extracellular signal-related kinase and c-Jun N-terminal kinase was unaffected. Taken together, our study indicated that M. denudata inhibited IL-4 and IL-13 expression, possibly through regulation of p38 mitogen-activated protein kinase phosphorylation and selective transcription factors, such as GATA-3 and c-Jun, in EL-4 T cells.</P>

To elucidate the molecular mechanisms for the suppression of LPS-induced nitric oxide (NO) production by a dehydrocostus lactone (DL) from Saussurea lappa, we examined the preventive effect of this compound on $NF-{\kappa}B$ activation in LPS-treated RAW 264.7 macrophages and U937 human monocytic cells. The results suggest that the suppression of NO production is mediated by the inhibitory action on the i-NOS gene expression through the inactivation of $NF-{\kappa}B$ and this sesquiterpene lactone can act as a pharmacological inhibitor of the $NF-{\kappa}B$ activation.
Suppression of Spontaneous Dermatitis in NC/Nga Murine Model by PG102 Isolated from Actinidia arguta

Atopic dermatitis (AD) is a chronic inflammatory skin disease, which requires safe and effective pharmacological therapy. We previously found that two preparations from Actinidia arguta, PG102T, and PG102E, could modulate Th1/Th2 pathways and suppress IgE biosynthesis. This study was performed to assess the therapeutic effects of PG102T and PG102E on the development of dermatitis in NC/Nga mice, characterized by the spontaneous onset of AD along with an elevated level of IgE under conventional conditions. PG102T or PG102E administration significantly reduced dermatitis severity as well as scratching tendency in conventional mice. The suppression of dermatitis by PG102 was accompanied by a decrease in the plasma level of IgE, IgG1, and IL-4 and also by an increase in that of IgG2a and IL-12. The splenic level of IL-4, IL-5, and IL-10 was downregulated, whereas that of IFN-γ and IL-12 was increased. The number of eosinophils and the expression of eotaxin and thymus and activation-regulated chemokine were decreased by PG102T or PG102E. Histological findings also indicated that the thickening of epidermis/dermis and the dermal infiltration of inflammatory cells including mast cells were greatly inhibited. <P><B>Background</B></P><P>Various allergic responses are thought to result from the unbalanced development of T<SUB>H</SUB>1 and T<SUB>H</SUB>2 pathways and, subsequently, the overproduction of IgE. Therefore the modulation of T<SUB>H</SUB>1 and T<SUB>H</SUB>2 responses is a rational strategy for the treatment of allergic diseases.</P><P><B>Objective</B></P><P>The present study was performed to investigate the immune-modulating activities of PG102 preparations from <I>Actinidia arguta</I> in ovalbumin-sensitized murine models.</P><P><B>Methods</B></P><P>Two preparations from <I>A arguta</I>, PG102T and PG102E, were chosen for animal experimentation on the basis of their ability to regulate the production of IgE in U266B1 cells. The changes in splenic levels of cytokines and plasma levels of immunoglobulin isotypes were examined. The effects of PG102 on subcellular composition (CD4<SUP>+</SUP>IL-4<SUP>+</SUP> or CD19<SUP>+</SUP>IgE<SUP>+</SUP> cells), IgE production in B cells, and selective transcription factors were analyzed.</P><P><B>Results</B></P><P>Oral administration of PG102T and PG102E significantly decreased the level of selective T<SUB>H</SUB>2 cytokines, whereas it increased the level of T<SUB>H</SUB>1 cytokines. The differential effects of PG102T and PG102E on T<SUB>H</SUB>1 and T<SUB>H</SUB>2 cytokines were accompanied by a decrease in the plasma levels of IgE and IgG1 and by an increase in the plasma level of IgG2a. The percentages of both IL-4–producing T cells and IgE-producing B cells were decreased. The concentration of IgE produced within B cells also appeared to be reduced. Finally, PG102T and PG102E downregulated the level of GATA-binding protein 3, while inducing that of T-box transcription factor and nuclear factor of activated T cells c2.</P><P><B>Conclusion</B></P><P>PG102T and PG102E have great potential as orally active immune modulators for the therapy of various allergic diseases.</P>

<P>The oral consumption of capsicum has been reported to increase interleukin (IL)-2 and interferon (IFN)-γ production in Peyer's patches (PP); however, the active components responsible for these effects have not been completely identified. The beneficial biological effects of green peppers cultivated under environmentally friendly farming conditions (ECP), without the use of chemical pesticides, have rarely been compared with those of green peppers cultivated under conventional farming conditions (CCP). Oral administration of ECP extract significantly induced the production of IL-2 and IFN-γ in concanavalin A-treated cells from PP ex vivo; their levels were much higher than those in the CCP extract-treated group. A comparative analysis of the HPLC profiles indicated a 1.7-fold increase of a peak, named EF-1, at 415?nm in the ECP extract. The major component of EF-1 was identified as pheophytin a, which is a chlorophyll a molecule lacking a central Mg(2+) ion, as determined from NMR data. Intake of pheophytin a and chlorophyll a significantly increased IL-2 and IFN-γ production, and the percentage of IL-2- and IFN-γ-producing CD4+ T-cells in PP. Taken together, our data suggest that ECPs produce a higher content of pheophytin a than CCPs, and pheophytin a and chlorophyll a are immune-modulating components in green vegetables.</P>

In the previous study we reported the isolated compounds from Lentinus lepideus as biological response modifiers, especially B cell mitogen. For the characterization of the compounds we performed anthrone test, Lowry-Foline test, hexosamine test, gas liquid chromatography and amino acid analysis. The results showed the acidic polysaccharide fraction, named lepidan, consists of protein bound polysaccharide which has galactose, xylose and mannose as monosaccharides. As one apprach to understanding the molecular mechanisms of this action, we investigated effects of the compound on cellular transcription factors which are known to control the proliferation of immune cells. Using transient transfection we found that lepidan significantly activated NF-kB in 70Z/3 pre B cell line, but not AP-1. Also activation of NF-kB by lepidan blocked by the protein kinase inhibitor H7. It suggested that the actions of lepidan on NF-kB was activated through the PKC pathway.

The paper proposes a diagnostic function to prevent over charging and discharging of the Li-Ion batteries as well as to keep connections with generator and motors of electric vehicles. Both operating conditions normal and abnormal are taken into account in terms of communications. Especially to cope with a situation that communication from the battery side has been cut off, an additional protected function is introduced. The diagnostic function has one generic logic based on the voltage threshold and two layered logics for the unexpected communication situations. First generic logic simply cuts batteries off by monitoring cell voltages. The logic is simply triggered by an upper and lower bound which are the boundary values of the cells" operating range. The other logics, the layered logics are operating when an abnormal communication of the cell voltage monitoring system is detected. The layered logics are based on the battery cell voltage estimation with the first order Thevenin circuit model. In addition, the pack voltages which is measured periodically from a separate circuit board are used to double protect batteries as a redundancy. To apply this estimation logics to the real industrial automotive field, influenceable factors including sensor accuracy and an estimation error are considered on the calculation process. Simulation result is presented to show the effectiveness of the diagnostic function and the estimated cell voltages are compared with real cell voltages as well to prove modeling accuracies.

<P>Conclusion: Based on various experimental studies, traditional herbs and their bioactive constituents regulate the cellular processes involved in the initiation of DN owing to their significant pharmacological activities; however, the efficacy in animal models and humans has not yet been explored. Therefore, studies should be performed to evaluate the nephroprotective effects of medicinal plants in preclinical animal models and in humans.</P>
