The role of Sohlh2 during the meiotic process

The spermatogenesis and oogenesis-specific helix-loop-helix transcription factor 2 (Sohlh2) is exclusively expressed in germ cells of male and female gonad. Sohlh2 acts as a transcriptional factor via its specific DNA binding site, E-box to regulate target genes such as Lhx8, Zp genes, Ngn3. Sohlh2 localize in the female oocyte and in the male spermatogonia. In recent studies, Sohlh2 knockout (KO) mice occurs abnormal spermagoenesis resulting in sperm defect. Sohlh2 KO male mice were infertility due to disruption of numerous gene expression. However, the gene profiles of Sohlh2 KO testes were not characterized and the regulatory mechanism of Sohlh2 was poorly understood. In this study, we analyzed the gene profiles and examined the possible mechanism of Sohlh2 in the spermatogenesis. First, we performed histological analysis such as Hematoxylin and eosin stain, Tunel assay, and Immunohistochemistry to show the onset of disruption of Sohlh2 KO testes. These results showed that Sohlh2 KO testes have atrophic seminiferous tubule due to increased apoptosis at 2 weeks old. And then we analyzed the whole gene profiles in the Sohlh2 KO testes at 2 weeks old. We found that 91 genes were regulated at least 5-fold in knockout testes. Among these, several genes are involved in meiotic process. Quantitative-PCR results are shown that several meiotic factors are significantly down-regulated in 2-weeks-old Sohlh2 KO testes compared with that of wild type mice. Through chromosome spreading assay, we observed that the formation of synaptonemal complex of homologous chromosome during the meiosis in Sohlh2 KO testes was not completed. These suggest that Sohlh2 is critical for regulation of numerous factors including meiotic factors either directly or indirectly. Therefore, mis-regulation of meiotic factors at prophase I of meiosis during spermatogenesis leads to disruption of spermatogenesis in Sohlh2 KO testes. Further studies are needed to look at the mechanism of Sohlh2 for regulation of target genes in detail.

Making Future Cities calls for understanding and evaluating the contemporary urban social system, its recent history, its present dynamics, and its possibilities. Recently, Crisis and uncertainty are increasing surrounding the Korean and global society, and policy forecasting based on future research should be needed and becomes more and more important. The socioeconomic polarization and social exclusion has been the most important social challenges in contemporary cities. Cities are called upon to appropriately respond to this economic, social and political restructuring. Seoul Metropolitan City has faced same challenges. In the past decades, Seoul has experienced dramatic changes derived from unprecedented technological innovation and unfettered economic globalization. These changes undoubtedly have delivered substantial improvements in the lives of peoples in Seoul. Seoul has developed 10 times in economic growth of GRDP during the past 40 years. In the same period, the social and economic polarization also has increased rapidly. Social exclusion, generational unemployment, material poverty, wealth inequalities and ageing population has continued to deteriorate. Fast development brought economic wealth to Seoul, but it gradually disintegrated the kinship neighborhood, shared local identities, and communal traditions as well. What are the policies currently applied and how can there be a more comprehensive, resource-minded urban development? The main aim of this study was to highlight the main challenges for Seoul over the following two decades and to identify the main objectives, instruments and methods Seoul can deploy when tackling them. These questions have been discussed within three methods with the participatory citizen survey(townhall meeting survey), expert survey and empirical data analysis on Seoul’s future. We cover the 20 years of Seoul’s social and economic features, so the temporal scope of this study is by 2030 and its metropolitan area is the spacial extent. In chapter 2, we review the methodology of future study and history of future research. In chapter 3, we analyse future study of cities including Tokyo and United State and recent future trends in Korea. In chapter 4, we propose four mega-trends affecting Seoul, such as population reconfiguration, generation conflict, the mega city region and the climate changes. In chapter 5, we analyse the Mayor’s inaugural address with the text analysis method and issued the enhancing of social quality as the aim of Seoul’s future. In chapter 6, we conduct the citizen participatory survey using the town hall meeting and conduct the expert survey based on the frame of social quality. Finally, we propose the scenarios of social quality in Seoul at the year of 2030. These are the Social Tribe, Me Gray Senior, Nomadingles and Passive Silver.

Identification and Characterization of LHX8 DNA Binding Elements

Lhx8 (LIM homeobox 8) gene encodes a LIM homeodomain transcriptional regulator that is preferentially expressed in germ cells and critical for mammalian folliculogenesis. However, Lhx8 DNA binding sequences are not characterized yet. We aimed to identify and characterize a cis-acting sequence of germ-cell specific transcriptional factor, Lhx8. To identify Lhx8 DNA binding element, Cyclic Amplification of Sequence Target (CAST) Analysis was performed. Electrophoretic Mobility Shift Assay (EMSA) was processed for the binding specificity of Lhx8. Luciferase assay was for the transcriptional activity of Lhx8 through identified DNA binding site. We identified a putative cis-acting sequence, TGATTG as Lhx8 DNA binding element (LBE). In addition, Lhx8 binds to the LBE with high affinity and augments transcriptional activity of luciferase reporter driven by artificial promoter containing the Lhx8 binding element. These findings indicate that Lhx8 directly regulates the transcription of genes containing Lhx8 binding element in oocytes during early folliculogenesis.

노인요양시설 중간관리자의 근무경험에 대한 질적연구

본 연구의 목적은 노인요양시설의 효과적인 조직운영과 관리를 위해 핵심적 역할을 하는 중간관리자의 직무경험 의미를 파악하고 이해하는 것이다. 노인요양시설의 중간관리자는 실무 전반을 담당하면서 직원과 자원을 관리하고, 돌봄서비스 활동을 감독, 조정, 평가하며, 시설장에서부터 서비스 직접제공인력에 이르기까지 긴밀한 소통과 연계를 하는 등 조직에서 중요한 역할을 수행한다. 그럼에도 불구하고 노인요양시설 중간관리자의 직무경험에 대한 관심은 매우 부족하다. 본 사례연구의 연구참여자는 연구 안내 자료를 발송한전국의 노인요양시설 837개소에서 연구 참여 의사를 밝힌 중간관리자 24명 중 연구의 목적과 취지에 적합한 대상자 8명을 유의 표집(purposive sampling)하여 선정하였다. 자료수집은 2021년 5월부터 7월까지 코로나19 재난 상황에서 화상프로그램 Zoom을 이용한 심층면담을 통해 이루어졌고, 면담 내용은 Stake(1995)가 제안한 범주적 집합(categorical aggregation)과 직접적 해석(direct interpretation) 방법을사용하여 분석하였다. 연구결과, 중간관리자의 핵심 역할과 관련해서 20개의 하위범주와 9개의 상위범주, 4개의 핵심주제가 도출되었고, 그 핵심주제는 ‘시설장과 직원들의 연결고리로서 원(one)팀 만들기’, ‘어르신 및 보호자의 주된 소통창구 역할 해내기’, ‘중간자로서가치 혼란 속에서 균형 잡기’, ‘제도·환경 변화에 유연하게 대응하기’로 나타났다. 본 연구결과를 토대로 노인요양시설의 중간관리자를지원하기 위한 실천적, 정책적 함의를 제시하였다 The purpose of this study is to understand the meaning of the working experience of middle managers who play a key role for effective organizational operation and management of nursing homes. Middle managers of nursing homes are in charge of the overall operation, manage staff and resources, supervise, coordinate and evaluate care services, and communicate and link closely from the head of the nursing homes to the direct service providers. Nevertheless, there is very little interest in the job experiences of middle managers of nursing homes. The participants of this case study were 8 middle managers of nursing homes, which were extracted, by purposive sampling, from 24 middle managers who expressed their intention to participate among 837 nursing homes nationwide. Data collection was conducted through in-depth interviews using the video program Zoom during the COVID-19 pandemic situation from May to July 2021, and was analyzed using direct interpretation and categorical aggregation methods suggested by Stake(1995). As a result of the study, 20 sub-categories, 9 super-categories, and 4 core themes were derived in relation to the core role of middle managers. ‘Creating one team as a link between the facility manager and employees’, ‘Acting as the main communication channel for residents and their family’, ‘Balance in value conflict’, and ‘responding flexibly to changes in the system and environment . Based on the results, this study offered practical and policy implications for supporting middle managers of nursing homes.
