        The Constituents Isolated from Peucedanum japonicum Thunb. and their Cyclooxygenase (COX) Inhibitory Activity

        Five coumarins, psoralen (1), scopoletin (2), isoimperatorin (4), (+)-marmesin (5) and xanthotoxin (6), three chromones, cimifugin (3), hamaudol (7) and sec-O-glucosylhamaudol (10), one sterol, daucosterol (8) and one aliphatic alcohol, galactitol (9) were isolated from the root of Peucedanum japonicum. Their chemical structures were identified by the physicochemical and spectroscopic data by comparing literature values. Among them, compounds 9 and 10 were isolated for the first time from this plant. The anti-inflammatory effects of isolated compounds were examined on cyclooxygenase (COX), compounds 1, 2 and 7 showed inhibitory activity on COX-1 with IC50 values of 0.88, 0.27 and 0.30 mM, respectively. In the test for COX-2 activity, only compound 7 showed significant inhibitory activity with the IC50 value of 0.57 mM. The other compounds exhibited weak inhibitory or no inhibitory activity.

        Differential Effects of Tautomycetin and Its Derivatives on Protein Phosphatase Inhibition, Immunosuppressive Function and Antitumor Activity

        In the present work, we studied the structure-activity relationship (SAR) of tautomycetin (TMC) and its derivatives. Further, we demonstrated the correlation between the immunosuppressive fuction, anticancer activity and protein phosphatase type 1 (PP1) inhibition of TMC and its derivatives. We have prepared some TMC derivatives via combinatorial biosynthesis, isolation from fermentation broth or chemical degradation of TMC. We found that the immunosuppressive activity was correlated with anticancer activity for TMC and its analog compounds, indicating that TMC may home at the same targets for its immunosuppressive and anticancer activities. Interestingly, TMC-F1, TMC-D1 and TMC-D2 all retained significant, albeit reduced PP1 inhibitory activity compared to TMC. However, only TMC-D2 showed immunosuppressive and anticancer activities in studies carried out in cell lines. Moreover, TMC-Chain did not show any significant inhibitory activity towards PP1 but showed strong growth inhibitory effect. This observation implicates that the maleic anhydride moiety of TMC is critical for its phosphatase inhibitory activity whereas the C1-C18 moiety of TMC is essential for the inhibition of tumor cell proliferation. Furthermore, we measured in vivo phosphatase activities of PP1 in MCF-7 cell extracts treated with TMC and its related compounds, and the results indicate that the cytotoxicity of TMC doesn’t correlate with its in vivo PP1 inhibition activity. Taken together, our study suggests that the immunosuppressive and anticancer activities of TMC are not due to the inhibition of PP1. Our results provide a novel insight for the elucidation of the underlying molecular mechanisms of TMC’s important biological functions.

        Residual stress of cold-formed thick-walled steel rectangular hollow sections

        This paper presents the experimental and numerical study on the distribution of transverse and longitudinal residual stresses in cold-formed thick-walled structural steel rectangular hollow sections manufactured by indirect technique. Hole-drilling method is employed to measure the magnitude of the transverse and longitudinal surface residual stress distribution, and the effects of the residual stresses are evaluated qualitatively by sectioning method. It is shown that compared to normal cold-formed thin-walled structural hollow sections (SHS), the coldformed thick-walled SHS has similar level of residual stress in the flat area but higher residual stresses in the corner and welding areas. Both the transverse and longitudinal residual stresses tend to open the section. In order to predict the surface residual stresses in the corners of the cold-formed thick-walled SHS, an analytical model is developed. 2D finite element simulation of the cold bending process is conducted to validate the analytical approach. It is shown that in analyzing bending for thick-walled sections, shifting of neutral axis must be considered, since it would lead to nonlinear and non-symmetrical distribution of stresses through the thickness. This phenomenon leads to the fact that cold-formed thick-walled SHSs has different distribution and magnitude of the residual stresses from the cold-formed thin-walled SHSs.

        Model based state-of-energy estimation for LiFePO<sub>4</sub> batteries using unscented particle filter

        Lithium-ion batteries have been treated as the most efficient energy storage candidates in electric vehicles. However, range anxiety hinders its popularity in daily transportation. When compared with the extensively discussed state-of-charge, state-of-energy (SoE) is a more appropriate indicator of remaining driving mileage by considering the voltage degradation in the discharging process. In this paper, an equivalent circuit model (ECM) based SoE estimator is proposed where a pseudo power definition is utilized to exclude the internal loss influence, and an unscented particle filter is employed to address system nonlinearities and noises. Additionally, to accommodate battery time-variant characteristics, the ECM parameters and variables are decoupled in complex-frequency domain, upon which the recursive least square algorithm is utilized for the on-line identification of parameters. Finally, a series of validation experiments on a LiFePO<sub>4</sub> battery verify that the proposed methodology can track the real SoE closely with an error of less than 1.8%.

      • Experimental study of welding effect on grade S690Q high strength steel butt joint

        This study experimentally reveals the influence of welding on grade S690Q high strength steel (HSS) butt joints from both micro and macro levels. Total eight butt joints, taking plate thickness and welding heat input as principal factors, were welded by shielded metal arc welding. In micro level, the microstructure transformations of the coarse grain heat affected zone (CGHAZ), the fine grain heat affected zone (FGHAZ) and the tempering zone occurred during welding were observed under light optical microscopy, and the corresponding mechanical performance of those areas were explored by micro-hardness tests. In macro level, standard tensile tests were conducted to investigate the impacts of welding on tensile behaviour of S690Q HSS butt joints. The test results showed that the main microstructure of S690Q HSS before welding was tempered martensite. After welding, the original microstructure was transformed to granular bainite in the CGHAZ, and to ferrite and cementite in the FGHAZ. For the tempering zone, some temper martensite decomposed to ferrite. The performed micro-hardness tests revealed that an obvious “soft layer” occurred in HAZ, and the HAZ size increased as the heat input increased. However, under the same level of heat input, the HAZ size decreased as the plate thickness increased. Subsequent coupon tensile tests found that all joints eventually failed within the HAZ with reduced tensile strength when compared with the base material. Similar to the size of the HAZ, the reduction of tensile strength increased as the welding heat input increased but decreased as the thickness of the plate increased.

        Inhibitory Constituents against Cyclooxygenases from Aralia cordata Thunb

        Seven diterpenes, four polyacetylenes, a lipid glycerol, and two sterols were isolated from the methylene chloride fraction of the root of Aralia cordata. Their chemical structures were determined as (-)-pimara-8(14), 15-dien-19-oic acid (2), pimaric acid (3), (-)-kaur-16-en-19-oic acid (4), 17-hydroxy-ent-kaur-15-en-19-oic acid (9), $7{\alpha}$-hydroxy-(-)-pimara-8(14), 15-dien-19-oic acid (10), $16\alpha$, 17 -dihydroxy-(-)-kauran-19-oic acid (11), 16-hydroxy-17-isovaleroyloxy-ent-kauran-19­oic acid (12), falcarindiol (5), dehydrofalcarindiol (6), dehydrofalcarindiol-8-acetate (7), falcarin­diol-8-acetate (8), alpha-mono palmitin (13), stigmasterol (1), and daucosterol (14) by the spectral evidences. These compounds were tested with COX-1 and COX-2 inhibition assays. This study found that compounds 2, 4, 5, 6, 7, 8, and 10 inhibited COX-1 dependent conversion of the exogenous arachidonic acid to $PGE_2$ in a dose-dependent manner with $IC_{50}$ values of $134.2{\mu}M$, $121.6{\mu}M$, $170{\mu}M$, $50.4{\mu}M$, $11.7{\mu}M$, $99.6{\mu}M$, and $69.6{\mu}M$, respectively. But, most of these compounds weakly inhibited COX-2 dependent $PGE_2$ generation. Among them, only compound 4 showed relatively significant inhibitory activity $(IC_{50}\;:\;127.6{\mu}M)$.
