      • Casein micelle 의 조성에 관한 연구 : I. Micelle 크기 별에 따른 각 casein 의 구성비율 I. The relative amounts of individual caseins in the different casein micelles

        本 實驗은 casein miceile의 크기에 따른 각종 casein의 構成成分을 究明하고자 超遠心分離機를 利用하여 casein micelle을 크기 別로 分類하고 DEAE-cellulose column chromatography를 이용하여 각 casein의 構成比率을 조사하였다. 그 結果를 要約하면 다음과 같다. 1. 脫脂乳를 100,000×g에서 각각 30分, 60分, 90分동안, 200,000×g에서 60分동안 遠心分離하였을 때 沈澱되는 casein micelle 量은 45.00%, 61.43%, 80.00% 및 95.00%였으며 沈澱되지 않는 serum casein量은 55.00%, 38.57%, 20.00% 및 5.00%였다. 2. 脫脂乳를 100,000×g에서 時間 差에 따라서 遠心分離하였을 때 각각 沈澱되는 casein micelle量은 10分間에는 全 casein量의 23.13%, 11∼30分間에는 30.26%, 31∼60分間에는 28.11%였으며, 60分問의 遠心分離에 의해서 沈澱되지 않는 serum casein은 17.79%였다. 3. 時問 差로 遠心分離하여 얻어진 casein micelle중 각 casein의 構成比率을 測定한 結果, α_s-casein, β-casein, κ-casein 및 γ-casein의 상대적인 量은 100,000×g, 10分의 pellet에서 각각 54%, 29%, 11% 및 6%였고, 100,000×g, 11∼30分의 pellet에서는 각각 54%, 28%, 13% 및 5%였고, 100,000×g, 31∼60分의 pellet에서는 각각 51%, 28%, 16% 및 5%로 나타났으며, serum casein은 44%, 29%, 19% 및 8%로 각각 나타났다. This experiment was carried out to study on the protein composition of casein micelles. Three micellar casein pellets and a serum casein were isolated at diferent time of ultracen trifugation. And the composition of individual caseins for the fractions was determined by DEAE-cellulose column chromatography. The results are summarized as follows: 1. When skimmilk was centrifuged for 30 min, 60 min and 90 min at 100,000g and for 60 min at 200,000g, the values for the sedimented casein micelles were 45.00%, 61.43%, 80.00% and 95.00% respectively and that of serum casein was 55.00%, 38.57%, 20.00% and 5.00% respectively. 2. When skimmilk was centrifuged for different time at 100,000g, the values for the sedimented casein micelles were 23.18% for 10 min.(pellet 1), 30.26% from 11 to 30 min.(pellet 2), 28.11% from 31 to 60 min.(pellet 3) and that of serum casein which was not sedimented at 60 min was 17.79% of the total casin. 3. The relative amounts of α_s-, β-, k- and γ-caseins in casein micelles obtained after different time of centrifugation were 54%, 29%, 11% and 8% in pellet 1, 54%, 28%, 13% and 5% in pellet 2, 51%, 28%, 16% and 5% in pellet 3,44%, 29%, 19% and 8% in serum casein respectively.

      • 정읍지역의 다양한 지표변위 인자들을 이용한 연약지반 리스크 평가

        연약지반의 지반침하는 고속철도의 열차 탈선, 심하게는 건물의 붕괴 등 대형재난을 야기할 우려가 있다. 그렇기 때문에 지반침하를 사전에 파악해 재난을 예방하기 위한 다양한 연구가 진행 중이다. 최근 광범위한 지역의 지표변위를 분석할 수 있는 InSAR(interferometric SAR)기법을 이용한 연구가 활발히 진행되고 있다. InSAR 기법 중 PS-InSAR(Permanent Scatterer Interferometric SAR)기법이란 SAR영상에서 긴밀도가 높은 고정산란체(PS:Permanent scatterer)를 이용하여 지반침하를 분석하는 기법으로 다른 InSAR 기법에 비해 대기에 의해 발생하는 오차가 적으며, 보다 정확한 지표 변위값을 도출할 수 있다. 또한 장기간에 걸친 시계열적 지표 변위 분석에 용이하다. 본 논문에서는 독일의 Terra SAR-X 위성 영상을 이용하여, 정읍지역의 지표변위를 분석하였다. 지반침하 데이터를 누적 지표 변위. MIN-MAX값, 그리고 최종침하량을 종합적으로 분석하여 보다 정확하게 결과를 도출하고자 하였다. 연구지역인 정읍시는 평야지역과 산지의 구분이 뚜렷하게 나타나는 지역이며, 연약지반인 평 야지역의 지반침하를 발견할 수 있었다. 연구 결과 정읍 신태인 지역은 최대 -11mm/year, 평균-3mm/year로 지반이 침하되고 있음을 확인할 수 있었다. 정읍지역의 지반침하 대비를 위해 다양한 노력들이 필요할 것으로 사료된다.

        가자의 즉각형 알레르기 반응 억제 효과

        We investigated the effects of methanol extract of Fructus Chebula (MEFC) on local and systemic anaphylaxis. Passive cutaneous anaphylaxis inhibited to 61.42 12.8 % by oral administration of MEFC(I.0 glkg). MEFC inhibited compound 48/80-induced anaphylactic shock 100% with doses of 0.05-1.0 glkg . Administrations of MEFC (1.0 glkg) 60 min before, 5 min after, and 10 min after the compound 48/80 treatment were shown the mortality rates as 0%. MEFC (0.01-1.0 glkg) inhibited the histamine release significantly from the rat peritoneal mast cells by compound 48/80. These results indicate that MEFC inhibits mast cell mediated anaphylactic reactions by inhibition of mast cell degranulation in vivo and in vitro.

        Simultaneous Determination of Amphetamine-Type Stimulants and Cannabinoids in Fingernails by Gas Chromatography-Mass Spectrometry

        A gas chromatography-mass spectrometric (GC-MS) method was developed and validated for the simultaneous detection and quantification of four amphetamine-type stimulants (amphetamine (AP), methamphetamine (MA), 3,4-methylenedioxyamphetamine (MDA) and 3,4-methylenedioxymethamphetamine (MDMA)) and two cannabinoids (Δ9-tetrahydrocannabinol (Δ9- THC) and 11-nor-Δ9-tetrahydrocannabinol-9-carboxylic acid (THCCOOH)) in fingernails. Fingernail clippings (30 mg) were washed with distilled water and methanol, and then incubated in 1.0 M sodium hydroxide at 95oC for 30 min. The compounds of interest were isolated by liquidliquid extraction followed by derivatization with N-methyl-N-trimethylsilyltrifluoroacetamide (MSTFA) at 70oC for 15 min. The derivatized compounds were analyzed by GC-MS in the selective ion monitoring (SIM) mode. The linear ranges were 0.1-15.0 ng/mg for AP, 0.2-15.0 ng/mg for MDA, Δ9-THC and THCCOOH, and 0.2-30.0 ng/mg for MA and MDMA, with good correlation coefficients (r2 > 0.9991). The intra-day, inter-day, and inter-person precisions were within 10.6%, 6.3%, and 5.3%, respectively. The intra-day, inter-day and inter-person accuracies were between -6.1 and 5.0%, -6.2 and 5.7%, and -6.4 and 5.6%, respectively. The limits of detection (LODs) and quantification (LOQs) for each compound were lower than 0.056 and 0.2 ng/mg, respectively. The recoveries were in the range of 74.0-94.8%. Positive GC-MS results were obtained from specimens of nine suspected MA or cannabis abusers. The concentration ranges of MA, AP, and THCCOOH were 0.10-1.41, 0.12-2.64, and 0.20 ng/mg, respectively. Based on these results, the method proved to be effective for the simultaneous qualification and quantification of amphetamine-type stimulants and cannabinoids in fingernails.

        가토에서 Vecuronium bromide 의 용량에 따른 근육이완 효과

        The introduction of muscle relaxants was an epoch making event in the development of clinical anesthesia and their use has hecame essential to anesthesia practice. Vecuronium, one of the newest muscle relaxants, many advantages; it is .apid acting, non-accumulating, has minimals side reactions and is promptly antagonized by anticholinesterases. However, its pharmacological effect, like other conventional muscle relaxants depends upon the dose and is influenced br many factors including body temperature, blood flow, interaction with other drugs, antibiotics, diuretics and so forth, To obtain a dose·response relationship which is important for clinical use, the effect of the vecuronium in various doses on the neuromuscular block in rabbits was investigated. The results were as follows. 1) In the vecuronium 0.025 mg/kg group, maximal twitch depression was varied between 75 to 92% with a mean value of 84,9%. The spontaneous recovery index was 141.0 seconds (2.35 min). 2) In the vecuranium 7.75 mg/kg group, thmean maximum twitch depression and spontaneous recovery index were 98.4% and 218.6 seconds (3.64 min.) respectively. 3) In the vecuronium 7.1 mg/kg group, maximum twitch depression was 100% and the spontaneous recovery index was markedly prolonged to 627.8 seconds (10.35 min).

        Simultaneous Determination of Amphetamine-Type Stimulants and Cannabinoids in Fingernails by Gas Chromatography-Mass Spectrometry

        소동물 폐종양의 정량적 개선을 위한 내부 움직임 평가

        이 연구에서는 폐종양의 정량적 개선을 위하여 분자체를 이용하여 내부 움직임을 측정하고 평가된 데이터를 기반으로 소동물 PET 영상내의 폐종양을 국소화하고자 하였다. 소동물 폐 영역의 내부 움직임은 방사성물질을 흡착한 분자체를 이용하여 소동물 폐 영역에 부착함으로써 구현하였다. 폐 영역의 내부 움직임 표적으로 사용된 분자체는 약 37 kBq의 Cu-64를 흡착시켜 폐종양을 모사하였다. 소동물 PET 영상은 Siemens Inveon 스캐너를 이용하여 획득하였으며 외부 움직임 데이터는 트리거 생성 장치인 BioVet을 이용하였다. SD-Rat PET 영상은 $^{18}F$-FDG 37 MBq/0.2 mL을 미정맥으로 주사하고 60분 후 20분간 데이터를 획득하였다. 리스트모드 데이터의 각 선응답은 외부 트리거 장치에 의해 획득된 트리거신호를 이용하여 2 bin에서 16 bin으로 사이노그램을 획득하였다. 획득된 사이노그램 데이터는 OSEM 2D 알고리즘을 이용하여 4회의 반복으로 재구성하였다. 종양의 정량적 분석을 위한 PET 영상은 종양을 묘사한 분자체 영역에 관심영역을 설정하고 계수와 SNR 그리고 FWHM을 이용하여 평가하였다. 움직임 표적으로 사용된 분자체의 크기는 $1.59{\times}2.50mm$이었으며, 기준 영상으로 획득한 체외 분자체 수직 및 수평 FWHM은 $2.91{\times}1.43mm$이었다. 정적영상과 4 bin 그리고 8 bin 영상에서의 수직 FWHM은 각각 3.90 mm, 3.74 mm, 3.16 mm이었으며 수평 FWHM은 각각 2.21 mm, 2.06 mm, 1.60 mm이었다. 정적영상, 4 bin, 8 bin, 12 bin 그리고 16 bin의 계수 값은 각각 4.10, 4.83, 5.59, 5.38, 5.31이었다. 정적영상, 4 bin, 8 bin, 12 bin 그리고 16 bin의 SNR은 4.18, 4.05, 4.22, 3.89, 3.58이었다. FWHM은 게이트 수의 증가에 따라 계속 향상됨을 확인하였다. 그러나 계수 값과 SNR은 게이트 수의 증가에 따라 계속 향상되지 않고 특정 bin 수에서 가장 높은 값을 보여 소동물 폐 영역에서의 종양 영상화시 SNR의 손실을 최소화하면서 향상된 계수 값을 얻을 수 있는 게이트 수를 획득하였다. 내부 움직임 측정은 최적화된 종양 국소화 영상을 획득할 수 있으며 외부 움직임 모니터링 시스템을 사용하지 않고 장기별 움직임 예측 모델링을 위한 유용한 방법이 될 것으로 기대된다. The purpose of this study was to estimate internal motion using molecular sieve for quantitative improvement of lung tumor and to localize lung tumor in the small animal PET image by evaluated data. Internal motion has been demonstrated in small animal lung region by molecular sieve contained radioactive substance. Molecular sieve for internal lung motion target was contained approximately 37 kBq Cu-64. The small animal PET images were obtained from Siemens Inveon scanner using external trigger system (BioVet). SD-Rat PET images were obtained at 60 min post injection of FDG 37 MBq/0.2 mL via tail vein for 20 min. Each line of response in the list-mode data was converted to sinogram gated frames (2~16 bin) by trigger signal obtained from BioVet. The sinogram data was reconstructed using OSEM 2D with 4 iterations. PET images were evaluated with count, SNR, FWHM from ROI drawn in the target region for quantitative tumor analysis. The size of molecular sieve motion target was $1.59{\times}2.50mm$. The reference motion target FWHM of vertical and horizontal was 2.91 mm and 1.43 mm, respectively. The vertical FWHM of static, 4 bin and 8 bin was 3.90 mm, 3.74 mm, and 3.16 mm, respectively. The horizontal FWHM of static, 4 bin and 8 bin was 2.21 mm, 2.06 mm, and 1.60 mm, respectively. Count of static, 4 bin, 8 bin, 12 bin and 16 bin was 4.10, 4.83, 5.59, 5.38, and 5.31, respectively. The SNR of static, 4 bin, 8 bin, 12 bin and 16 bin was 4.18, 4.05, 4.22, 3.89, and 3.58, respectively. The FWHM were improved in accordance with gate number increase. The count and SNR were not proportionately improve with gate number, but shown the highest value in specific bin number. We measured the optimal gate number what minimize the SNR loss and gain improved count when imaging lung tumor in small animal. The internal motion estimation provide localized tumor image and will be a useful method for organ motion prediction modeling without external motion monitoring system.

      • 원심력을 이용한 광통신용 광섬유 모재제조 : 미세입자 크기 및 이동식 injection tube의 영향

        In this paper, the technique to fabricate the optical fiber preforms by centrifuge was investigated, using silica particles of different sizes. The injection tube was designed to traverse axially so that uniform coating of tiny silica particles onto the substrate tube can be certified. The deposition efficiencies and deposition rates of SiO2 Particles were measured to elucidate the effects of process variables such as rotation speed of rotor, aqueous flow rate, suspension concentration, binder concentration and overflow weir diameter. This study shows clearly the merit of this technique by enhancing abruptly the deposition rates and deposition efficiencies, comparing to the conventional processes for optical fiber preforms.

      • 펄스 코로나 방전 공정에서 탈질 및 탈황 효율 분석

        In this study, we analyzed the removal efficiencies of SOx and NOx by the pulsed corona discharge process and investigated the effect of several process variables. The NO removal efficiencies and the SO_2 removal efficiencies were measured and analyzed as the function of initial concentrations of NO, H_2O, and NH_3, SO_2 applied voltage, pulse frequency and residence time.As the frequency of applied voltage increases, or as the applied voltage increases or as the residence of applied voltage increases, or as the applied voltage increases or as the residence time increases, the NO, SO_2 removal efficiency increases. The NO, SO_2 removal efficiency increases by the addition of O_2 or H_2O, or by using the large diameter of the discharge. The experimental results can be used as a basis to design the pulsed corona discharge to remove NOx, SOx and VOCs.

        수용액에서 γ형 알루미나 표면에 대한 니켈이온(Ⅱ)의 흡착특성

        γ형 알루미나의 흡착특성을 구명하기 위하여 본 실험에서는 수용액에서 γ형 알루미나를 흡착제로 이용하여 온도와 pH가 니켈이온(Ⅱ)의 흡착능에 미치는 영향을 관찰하고 알루미나 표면과 니켈이온(Ⅱ) 간의 Langmuir와 Freundlich 흡착등온식과 Lagergren 흡착속도식을 결정했다. 5 g Al_2O_3/L의 조건에서 니켈이온은 10 h에 흡착평형상태에 도달했다. PH 2~6의 범위에서 pH가 증가할 때 흡착량은 급격히 증가하다가 PH 6에서 최대 흡착량을 나타냈으며 pH 6~9의 범위에서는 pH가 증가하여도 흡착량은 일정했다. 온도 20℃에서는 흡착량이 적었으나 30℃에서 급격히 흡착량이 증가하였으며 30~50 ℃의 범위에서는 온도 증가에 따라 흡착량이 완만하게 증가했다. Freundlich 흡착등온식의 결정계수는 0.9489로 Langmuir식의 0.9265보다 높았으며 Lagersen 흡착속도식에서의 k_ad값은 0.0023 h^-1으로 결정계수는 0.9497이였다. In this paper, Langmuir and Freundlich adsorption isotherms, and Lagergen adsorption rate were determined and the effect of temperature and pH on the adsorptivity of Ni(Ⅱ) ions were investigated in order t() stud)'the adsorption characteristics of γ-type alumina. For 5 g Al_2O_3/L, the adsorption equilibrium was obtained in 10 h. The adsorption amount increased rapidly when pH incrteased from 2 to 6, and the maximum adsorption amount was observed at pH 6. For pH 6 to 9, however, the adsorption amount remained constant. The adsorption amount was low at 20℃. However, The adsorption amount increased with increased in temperature; the amount increased rapidly at 30℃ and increased linearly with temperature from 30 to 50℃. The correlation coefficient (R_2) for Freundlich adsorption isotherm was 039489%, and this was higher than that for Langmuir adsorption isotherm of 0.9268%. The adsorption coefficient (k_ad) of lagergen equation was 0.0023h^-1, and the correlation coefficient (R^2) was 0.9497.
