The low solubility and high-cost recovery of Propionibacterium acnes polyunsaturated fatty acid isomerase (PAI) are key problems in the bioproduction of high value-added conjugated linoleic acid (CLA). To improve the solubility of recombinant PAI, six chaperone proteins were coexpressed with PAI. Introduction of GroELS proteins dramatically improved the PAI solubility from 29% to 97%, with increased activity by 57.8%. Combined expression of DnaKJGrpE and GroELS proteins increased the activity by 11.9%. In contrast, coexpression of DnaKJGrpE proteins significantly reduced the activity by 57.4%. Plasmids pTf16 harboring the tig gene and pG-Tf2 containing the tig and groEL-groES genes had no visible impact on PAI expression. The lytic protein E was then introduced into the recombinant Escherichia coli to develop a cell autolysis system. A 35% activity of total intracellular PAI was released from the cytoplasm by suspending the lysed cells in distilled water. The PAI recovery was further improved to 81% by optimizing the release conditions. The lipase from Rhizopus oryzae was also expressed in E. coli, with an extracellular activity of 110.9 U/ml. By using the free PAI and lipase as catalysts, a joint system was established for producing CLA from sunflower oil. Under the optimized conditions, the maximum titer of t-10,c-12-CLA reached 9.4 g/l. This work provides an effective and low-cost strategy to improve the solubility and recovery of the recombinant intracellular PAI for further large-scale production of CLA.
Metabolic Syndrome with Hyperglycemia and the Risk of Ischemic Stroke

Purpose: The association of ischemic stroke and metabolic syndrome (MetSyn) with or without diabetes mellitus (DM) is not clear. The present study aimed to identify the impact of diabetes or hyperglycemia on the risk of MetSyn-associated ischemic stroke. Materials and Methods: This study comprised an Asian population of 576 patients with acute nonembolic cerebral infarction and 500 controls. MetSyn was defined according to the criteria of the International Diabetes Federation. MetSyn patients were further subgrouped according to their glucose levels: MetSyn with DM, MetSyn with impaired fasting glucose (IFG) and MetSyn with normal glucose tolerance (NGT). The impact of MetSyn on cerebral infarction was then evaluated. Results:At baseline, the prevalence of MetSyn in patients with cerebral infarction was higher than that of the controls (57.29% vs. 10.00%, p<0.01). In the stroke group, the prevalences of MetSyn with DM, IFG, and NGT were 25.69%, 8.85% and 22.74%, respectively, all of which were higher than that of the controls (all p-values <0.05). By multiple logistic regression analysis, we discovered that MetSyn was associated with an increased risk of cerebral infarction (odds ratio: 5.73, p<0.01). After adjustment for all the components of MetSyn, the odds ratios of MetSyn with DM, IFG, and NGT were 5.70, 2.24 and 2.19 (all p-values <0.05), respectively. Conclusion: In Asian population, patients with MetSyn accompanied by T2DM are at the greatest risk for acute non-embolic stroke. Additionally, IFG was not observed to be associated with an increased risk for MetSyn-related ischemic stroke.

Stimulus-responsive materials with controllable oil/water separation performance have prodigious potentialapplication. Here, a new thought for low-cost, time-saving, flexible approach has been developed to prepare a pH-responsivematerial with switchable superamphiphobicity and superoleophobicity-superhydrophilicity. The as-prepared material can beeasily applied onto multifarious substrates and presents stable superamphiphobicity. However, when the modified material istreated by alkaline solution, its surface wettability turns superhydrophilicity and superoleophobicity, thus water is allowed topenetrate through the material whereas the oil was blocked on the surface. Moreover, the surface wettability can be recoveredto superamphiphobicity quickly via treating the material with acidic aqueous. Hence, such a controllable water wettabilityand stable oil repellency property endows the as-prepared material with excellent capability to separate water from oil/watermixture. In addition, the pH-responsive materials can maintain switchable wettability after being treated by acid and alkalinumerous cycles. Furthermore, the obtained materials also exhibit excellent recyclable, self-cleaning and flame-resistantperformance, which shows potential applications for smart water-oil separators and fire-shielding protectors.
중국 우한 코로나 사례를 통한 재난지역 긴급물류의 성과에 미치는 요인에 관한 연구 : 한국 긴급물류 운영에 대한 시사점 도출

Purpose - This study, against the backdrop of the city of Wuhan, China, where COVID-19 first appeared, examines the factors that affect the effectiveness and efficiency of emergency logistics through empirical research on relief organizations that support Wuhan, China, during the fight against COVID-19. This study also studies how the government’s control situation affects the effectiveness and efficiency of emergency logistics. Design/Methodology/Approach - We employed a quantitative research method using survey data collected in Wuhan, China. This paper tested five hypotheses which assumes the effect of transportation system, inter-organizational cooperation, emergency logistics center location and demand and inventory management on the emergency logistics performance. In addition, we also examined a moderating role of government control on those hypothesized relationships. Findings - We found that transportation system, inter-organizational cooperation, emergency logistics center location and demand and inventory management affected the emergency logisitcs performance positively, and government control have moderated those reloationships partially. Research Implications - This findings may give meaningful insights on successful management of emergency logistics in Korea.
