        Osteogenic Activities of Genistein Derivatives were Influenced by the Presence of Prenyl Group at Ring A

        Our recent report indicated that the crude extract from stem bark of Erythrina variegata L. (Leguminosae) (EV) exerted beneficial effects against osteoporosis induced by estrogen deficiency in vivo. Followup phytochemical study has isolated genistein-derivatives mainly in the form of prenylgenistein from this extract, including 6-prenylgenistein, 8-prenylgenistein, and 6, 8-diprenylgenistein. The present study was performed to investigate the structure-function relationship of these compounds on osteoblastic proliferation, differentiation and mineralization in UMR 106 cells. Our results showed that genistein did not stimulate cell growth while 8-prenylgenistein promoted cell growth significantly by 10~23%. In contrast, the treatment by 6-prenylgenistein for 48 h reduced UMR 106 cell proliferation when compared to cells treated with genistein. The proliferation of 6,8-diprenylgenistein-treated cells was greater than those treated by 6-prenylgenistein at all testing concentrations. For ALP activity, significant increase was found in cells treated by either 8-prenylgenistein or 6,8-diprenylgenistein for 48 h at the concentration of 10-10 M. In mineralization study, the content of Ca and P in extracellular matrix were significantly increased in 8-prenylgenistein treated cells. The results showed that genistein derivativesisolated from EV demonstrated stimulatory effects on osteogenesis in UMR 106 cells. Based on thestudy of structure-activity relationship, it appears that prenylation at C-8, but not at C-6, could increasethe bone-protective effect of genistein.

        <P>One-dimensional smart probes based on nanowires and nanotubes that can safely penetrate the plasma membrane and enter biological cells are potentially useful in high-resolution and high-throughput gene and drug delivery, biosensing and single-cell electrophysiology. However, using such probes for optical communication across the cellular membrane at the subwavelength level remains limited. Here, we show that a nanowire waveguide attached to the tapered tip of an optical fibre can guide visible light into intracellular compartments of a living mammalian cell, and can also detect optical signals from subcellular regions with high spatial resolution. Furthermore, we show that through light-activated mechanisms the endoscope can deliver payloads into cells with spatial and temporal specificity. Moreover, insertion of the endoscope into cells and illumination of the guided laser did not induce any significant toxicity in the cells.</P>

        Low-Frequency Intermittent Hypoxia Suppresses Subcutaneous Adipogenesis and Induces Macrophage Polarization in Lean Mice

        Background: The relationship between obstructive sleep apnoea (OSA) and metabolic disorders is complex and highly associated. The impairment of adipogenic capacity in pre-adipocytes may promote adipocyte hypertrophy and increase the risk of furthermetabolic dysfunction. We hypothesize that intermittent hypoxia (IH), as a pathophysiologic feature of OSA, may regulate adipogenesisby promoting macrophage polarization. Methods: Male C57BL/6N mice were exposed to either IH (240 seconds of 10% O2 followed by 120 seconds of 21% O2, i.e., 10 cycles/hour) or intermittent normoxia (IN) for 6 weeks. Stromal-vascular fractions derived from subcutaneous (SUB-SVF) and visceral(VIS-SVF) adipose tissues were cultured and differentiated. Conditioned media from cultured RAW 264.7 macrophages afterair (Raw) or IH exposure (Raw-IH) were incubated with SUB-SVF during adipogenic differentiation. Results: Adipogenic differentiation of SUB-SVF but not VIS-SVF from IH-exposed mice was significantly downregulated incomparison with that derived from IN-exposed mice. IH-exposed mice compared to IN-exposed mice showed induction of hypertrophicadipocytes and increased preferential infiltration of M1 macrophages in subcutaneous adipose tissue (SAT) comparedto visceral adipose tissue. Complementary in vitro analysis demonstrated that Raw-IH media significantly enhanced inhibition ofadipogenesis of SUB-SVF compared to Raw media, in agreement with corresponding gene expression levels of differentiation-associatedmarkers and adipogenic transcription factors. Conclusion: Low frequency IH exposure impaired adipogenesis of SAT in lean mice, and macrophage polarization may be a potentialmechanism for the impaired adipogenesis.

        Study on Information Ethics Education and Production of Tools in Domestic Early Childhood Education Institutions

        4차 산업혁명으로 인하여 날이 갈수록 연령과 상관없이 과도하게 스마트기기에 많이 노출되고 있으며, 이에 대한 의존도가 급증하고 있는 실정이다. 최근 유아교사 및 유아 대상으로 무분별한 스마트폰 기기 노출 등으로 인한 스마트폰과의존 등 다양한 정보윤리의 문제점이 등장하고 있다. 이에 따라 유아교육학과에 재학 중인 예비유아교사들을 대상으로 국내 유아대상 정보윤리교육의 현황에 대해 설문조사를 하고, 이를 바탕으로 정보윤리교육 활성화 방안을 제시, 다양한 교재교구 개발 사례를 제안하고자 한다. 본 연구의 가장 큰 의의는 유아들에게 정보윤리교육을 균형있게 실시하며, 상호작용이 활발한 다양한 교재교구 개발하고자 한다. 또한 유아 뿐만 아니라 유아교사들에게 정보윤리교육의 필요성을 강조하고 바람직한 유아 미디어 교육을 실시하도록 하는 것이다. Due to the fourth industrial revolution, people are excessively exposed to smart devices regardless of age, and the dependence is increasing rapidly. In the same context, various problems on information ethics have emerged to infant teachers and children. Therefore, there is a need to present guidelines for infants on the correct use of smart devices. This study proposes means to promote information ethics education in infant education based on a survey on future teachers attending Department of Early Childhood Education. Also, it suggests development of various teaching materials. The main significance of the research is to provide a balanced information ethics education and to develop interactive teaching materials. It also calls for infant teachers, as wells as infants, to understand the need for information ethics education and to provide desirable media education for infants.

        Incidence of the Rice Root-Knot Nematode, Meloidogyne graminicola, in Guangxi, China

        Root-knot nematodes (Meloidogyne spp.) are the most destructive group of plant-parasitic nematodes. Plants infected by Meloidogyne spp. develop above-ground symptoms, stunting, yellowing, nutrient deficiencies, and gall formations with typical hook-shaped root tips. Infected plants experience yield losses. During 2018-2019 survey, leaf chlorosis rice plants were found in 206 fields of 67 counties in Guangxi, China, around 30 days after transplanting. Galls and hooked tips on the roots and pear-shaped females were observed. About 32.04% of fields were infested with the nematode. The nematodes were identified as Meloidogyne graminicola base on morphological and molecular analysis. To the best of our knowledge, this is the first report of M. graminicola on rice plants in Guangxi, China. The results of this study urge the discovery of resistant cultivars and the development of management strategies.

        Experimental study on particle circulation characteristics of external circulating fluidized bed evaporator

        In order to solve the particle circulation problem of external circulation fluidized bed evaporator, a new particle circulation device was developed, and a cold experiment device was set up for reference to the principle of gassolid circulating fluidized bed U-loop seal valve. The adjustment characteristics of the particle circulation device were studied experimentally. The effects of different flow combinations on pressure drop, particle circulation rate and particle volume fraction of heat exchange tube were investigated when the main flow and any auxiliary flow of the sealing valve were combined. It was found that when the main flow rate is constant, the pressure drop of the heat exchange tube increases with the increase of the auxiliary flow rate, and the pressure drop trend is stable when the main flow and loose flow are combined. The flow rate of particle circulation can be adjusted by adjusting the flow rate of any auxiliary flow alone, and fluidized flow has the strongest ability to regulate the particle circulation rate. The amplitude of the power spectral density of the pressure differential fluctuation signal is related to the particle circulation rate and volume fraction, so the flow state in the pipe can be studied by this method.

        WEIGHTED EVIDENTIAL FUSION METHOD FOR FAULT DIAGNOSIS OF MECHANICAL TRANSMISSION BASED ON OIL ANALYSIS DATA

        Condition monitoring (CM) and fault diagnosis are critical for the stable and reliable operation of mechanical transmissions. Mechanical transmission wear, which leads to changes in the physicochemical properties of the lubrication oil and thus severe wear, is a slow degradation process that can be monitored by oil analysis, but the actual degradation degree is difficult to evaluate. To solve this problem, we propose a new weighted evidential data fusion method to better characterize the degradation degree of the mechanical transmission through the fusion of multiple CM datasets from oil analysis. This method includes weight allocation and data fusion steps that lead to a more accurate data-based fault diagnostic result for CM. First, the weight of each evidence is modeled with a weighted average function by measuring the relative scale of the permutation entropy from each CM dataset. Then, the multiple CM datasets are fused by the Dempster combination rule. Compared with other evidential data fusion methods, the proposed method using the new weight allocation function seems more reasonable. The rationality and superiority of the proposed method were evaluated through a case study involving an oilbased CM dataset from a power-shift steering transmission.
