        The purpose of this study was to investigate the component and quality characteristics of White Lotus Roots(LR), Leaves(LL), Seeds(LS) and Bunch(LB). Four kinds of LR, LL, LS, LB evaluated the component of moisture, ash, crude lipid and crude protein. The contents of thiamin, α-tocopherol, β-carotene and total flavonoid in LR, LL, LS, LB were evaluated. Also the contents Amino acids and mineral in LR, LL, LS, LB were evaluated. The contents of moisture were 2.87~11.85% which was the highest in LB and the lowest in LS. The contents of ash were 4.89~12.38% which was the highest in LB and the lowest in LR. Crude lipid contents of LR, LL, LS, LB were 0.13~1.82% which was the highest in LS and the lowest in LL. The contents of crude protein were 3.62~20.75% which was the highest in LL and the lowest in LB. The contents of thiamin were 3.23~5.86 mg/kg which was the highest in LL. α-Tocopherol contents were 3.91~272.15 mg/kg which was the highest in LL and the lowest in LS. The contents of β-carotene were 0.22~7.68 mg/kg which was the highest in LB and the lowest in LR. Total flavonoid contents were 43.21~6,404.05 mg/kg which was the highest in LL and the lowest in LR. Total ammo acid contents of LR, LL, LS, LB were 5758.61, 2328.71, 4075.36 ppm and 981.67 ppm. Amino acid of LR showed high content of aspartic acid, followed by glutamic acid and serine. Amino acid of LL and LS showed high content of glutamic acid. Amino acid of LB showed high content of histidine, followed by serine and glutamic acid. Potassium and sodium contents of which were better in LS than LL and LR. Magnesium and calcium contents of which were better in LR than LL, LR and LB.

        This study was performed to verify the craniofacial skeletal characteristics in anterior openbite group, in contrase to normal occlusion group, and also to find out, vertical factors which shows, correlate to the amount of anterior openbite. 21 individuals, including 10 males and 11 females, without orthodontic treatment history and anterior-posterior skeletal malrelationships, were selected and analyzed basic morphology and vertical factors, using standard lateral cephalogram. The obtained results as follows: 1. Comparison of anterior open-bite group with normal occlusion group using Moyers analysis. a) In basis morphologic analysis, Ba-SE-Me was lesser and Ba-SE-Mn. P. and Mn.P/A-B were greater in both sexes of anterior openbite group than in normal occlusion group. b) In angle measurements of vertical analysis, PMV/Pal. P. was lesser in male anterior openbite group and PMV/Occ. P. and PMV/Mn. P. were greater in both sexes of anterior openbite group than in normal occlusion group. c) In height ratio of vertical analysis, ATFH/PTFH and ALFH/ATFH were greater in both sexes of anterior openbite group than in normal occlusion group. 2. The amount of anterior openbite was correlated with PMV/Occ. P. and PMV/Mn. P.

      • 가토안의 Acid Phosphatase의 정제, 특성 및 아미노산 조성

        가토의 눈에서 분리한 망막과 맥락막으로부터 ACPase를 정제하여 다음과 같은 결론을 얻었다. 1. surfactant extraction, ammonium sulfate fractionation, pH4.0 acid extraction, DEAE-cellulose chromatography를 시행하여 ACPase를 정제하였다. Fraction Ⅲ의 비활성은 377.4로 정제율은 14.06배, 수득율은 5.7%이었다. 2. 정제한 효소액을 SDS-PAGE를 시행하여 분자량 48,000을 확인하였다. 3. SDS-PAGE의 band를 추출하여 아미노산 조성을 분석하였다. 4. 정제된 효소액을 이용하여 p-nitrophenylphosphate hexahydrate에 대한 km은 12.19mM, Vmax는 4.62×10^-2mM의 값을 얻었다. optimum pH는 4.7이었다. Acid Phosphatase(Orthophosphoric monoester Phosphohydrolase acid optimum, EC. 3.1.3.2, ACPase) was purified from Rentinal Pigment Epithelium of Rabbit eyes. The purification procedure includes surfactant extraction, ammonium sulfate fractionation, acid treatment, and ionexchange-chromatography on DEAE-cellulose. The purified enzyme migrates as a single band electrophoresis on SDS-Polyacrylamide gel. Acid phosphatase has been characterized with respect to its molecular weight, kinetic properties(Km Value determined with p-nitrophenylphophate were 1.2×10^-2nM.) Optimum pH were 4.7. The molecular weight of ACPase were 48,000 by SDS-PAGE. Amino acid composition of ACPase revealed a high content of Glumate

        The perceived result on 18 different family member concepts were measured on the psychological distance of the Osgood-type Semantic Differential. Subjects are 100 males and 100 females, ranging from the age of 16 to 19. All familymember concepts are perceived as positive and close. Comparing with the Kim's (1974) and Yoon's(1980) study, the young generation perceived more positive than the old on family member concepts and distance.

      • 골형성 종양의 골조직에서 비교원성 골단백질의 분포

        목적 : 골조직내에는 여러 가지 비교원성 단백질이 포함되어 있다. 이들 단백질은 골 구성 세포들의 분화, 성장 및 골조직 형성을 조절하는 것으로 알려져 있다. 본 연구는 골 조직에서 발생하는 각종 종양에서 비교원성 단백질의 분포를 관찰하여 이를 골종양 감별 진단적 표지자로서의 가치를 규명하고자 한다. 대상 및 방법 : 1990년부터 1995년 4월까지 경북대학교병원에서 각 종 골종양을 수술하여 얻은 조직을 osteocalcin, osteonectin, 및 단구-대식구 항원에 대한 면역조직화학적 방법으로 염색을 하여 이들 단백의 분포를 관찰하였다. 결과 : osteocalcin 및 osteonectin은 골육종 및 골모 세포종에서 강한 양성 반응을 보였다. 그리고 osteonectin은 일부 연골 형성 종양 및 거대세포 종양에도 부분적인 양성 반응을 보였고 osteocalcin은 거대세포 종양에도 양성 반응을 보였다. Ewing 육종, 악성 림프종, 및 골수종 등은 osteonectin 및 osteocalcin 모두 음성 반응을 나타내었다. 그리고 단구-대식구 항원에 대한 반응은 거대세포 종양에서 강하게 발현하였다. 결과 및 결론 : 이상의 성적으로 보아 osteocalcin 및 osteonectin은 골형성 종양의 감별 진단에 유효하며 단구-대식구 항원 검사는 거대세포종 진단에 도움이 될 것이라고 생각된다. To ascertain the effective value of non-collagenous bone proteins as a diagnostic marker of osteogenic neoplasm, the expressions of osteonectin and osteocalcin, together with monocyte-macrophage antigen were evaluated in tissue sections obtained from primary bone tumors. Osteonectin and osteocalcin were detected in entire neoplastic component of osteogenic bone tumors and giant cell tumors. Osteonectin was also demonstrated in neoplastic component of some cartilage-forming tumors, whereas osteocalcin was specifically detected in bone forming tumor with osteoblastic differentiation. osteonectin and osteocalcin were not detected in Ewing's sarcoma, malignant lymphoma, and multiple myeloma. On the other hand, monocyte-macrophage antibody positive stains in the mononuclear and multinucleated giant cell components of giant cell tumor and the reactive multinucleated giant cells of entire primary bone tumors were observed. The results suggested that osteonectin and oiteocalcin as well as monocyte-macrophage antigen may be helpful in the histologic diagnosis of bone tumors, particularly osteocalcin is expected to be more useful for the diagnosis of osteosarcomas.

      • 타우로우루소데옥시콜린산이 흰쥐의 적출심장에서 허혈 및 재관류 손상에 미치는 영향

        Abstract - In this study, the effects of tauroursodeoxycholic acid (TUDCA) on ischemia/ reperfusion injury were investigated on isolated heart perfusion models. Hearts were perfused with oxygenated Krebs-Henseleit solution (pH 7.4, 37℃) on a Langendorff apparatus. After equilibration, isolated hearts were treated with TUDCA 100 and 200μM or vehicle (0.02% DMSO) for 10 min before the onset of ischemia in single treatment group. In 7 day pretreatment group, TUDCA 50, 100 and 200 mg/kg body weight were given orally for 7days before operation. After global ischemia (30 min), ischemic hearts were reperfused for 30 min. The physiological (i.e. heart rate, left ventricular developed pressure, coronary flow, double product, time to contracture formation) and biochemical (lactate dehydrogenase; LDH) parameters were evaluated. In vehicle-treated group, time to contracture formation was 810 sec during ischemia, LVDP was 34.0 mmHg at the endpoint of reperfusion and LDH activity in total reperfusion effluent was 34.3 U/L. Single treatment with TUDCA did not change the postischemic recovery of cardiac function, LDH and time to contracture compared with ischemic control group. TUDCA pretreatment showed the tendency to decrease LDH release and to increase time to contracture and coronary flow. Our findings suggest that TUDCA does not ameliorate ischemia/reperfusion-induced myocardial damage.

      • 감과실의 연화에 따른 물성 변화와 헤미셀루로즈의 변화

        감의 연화에 따른 물성 변화와 이에 따른 세포벽 다당류의 조성과 함량 및 column chromatography를 통한 계략적인 분자량의 변화를 조사하였다. 조세포벽의 함량은 연화된 감에서 현저히 감소되었다. 또한 온도변화에 따른 감의 물성변화와 세포벽 다당류의 변화를 조사하였다. 그 결과 25℃에서 가장 쉽게 연화 되었으며 45℃에서는 연화가 억제되었다. 연화가 진행되는 동안에 부착성은 증가하였으나 경도는 감소되었다. 이러한 현상은 25℃에서 가장 현저하게 나타났다. 조세포벽의 함량은 연화된 감에서 현저히 감소되었은데 구체적으로 ionically associated pectin(IAP)은 59%, covalently bounded pectin(CBP)은 60%, hemicellulose fraction 2(HF2)는 74%가 감소된 반면 hemicellulose fraction 1(HF1)과 cellulose fraction(CF)은 큰 변화가 없었다. 연화된 감의 IAP, CBP에서는 pentose와 hexose의 감소율이 높았다. HF1과 HF2역시 uronic acid의 함유율이 높았는데 HF2가 현저하였다. 또 연화에 따라 HF1은 pentose와 HF2는 uronic acid와 hexose의 손실율이 높았다. 특히 HF2는 수백만 단위의 거대분자가 만단위까지 저분자화 하였다. 이상의 결과에서의 감의 연화는 CBP에 연결된 hemicellulose중 특히 HF2의 저분자화와 밀접한 관련이 있는 것으로 판단 되었다. The changes of cell wall polysaccharides were studies by gel filteration chromatography. Various cell wall polysaccharides such as ionically associated pectin(IAP), covalently bounded pectin(CBP), 2N potasium hydroxide soluble hemicellulosic fraction(HF1) and 0-3N potasium hydroxide soluble hemicellulosic fraction(HF2) were fractionated by chemical method. The content of crude cell wall remarkably decreased in the soft persimmon. The changes of texture, composition of cell wall polysaccharides during preserving of persimmon at various temperature were estimated to investigate the softening characteristics of persimmon fruit. The softening of persimmon was the most promoted at 25℃, Whereas it was inhibited at 45℃. During softening adhesiveness increased and hardness decreased. This phenomenon was obvious at 25℃. The decreasing rates of IAP, CBP and HF2 were 59, 60 and 74%, respectively, while HF1 and cellulose changed only a little during softening. The loss rate of pentose in IAP, of hexose in CBP, of hexose and uronic acid in HF2, of pentose in HF1 increased during softening. Though apparent molecular weight of all polysaccharides shifted from high molecular weight to low molecular weight polymer, the shifting degree of CBP and HF2 was especially remarkable during softening. It is suggested that the servere softening phenomenon of persimmon involved the degradation and dissolution of wall bounded-CBP and HF2 which were associated with each other.

      • 가토안의 Acid Phosphatase의 활성 및 분포

        Lysosomal enzyme activity of choroid, newrosensory retina, iris and ciliarybody, cornea and sclea of the rabbit eye was measured by acid phosphatase(ASPase)as enzyme maker. We reported lysosomal enzyme activity on retinal pigment epitherium in 1998. Distribution of ACPase in the tissues was carried out using Fukuzawa method and Kind-king method. Of the tissues of the rabbit eye the ACPase activity was highest in the choroid(28.6583±12.6920)by Kind-king method. The meurosensory retina(4.5433±2.2091), Iris & ciliary body(2.4138±0.8307), cornea(1.7223±0.6492)and sclea(0.9550±0.0646)also showed the specipic activity of enzyme.

        자동혼합 부가중합형 실리콘고무인상재의 인장강도 및 유연성에 관한 연구

        The purpose of this study was to investigate the tensile strength and flexibility of four automixing additional silicone rubber impression materials. To evaluate the tear resistance of impression material, the automixed impression material was cured in a 32℃ water bath with the necked specimen preparation mold for 5 minutes. Three minutes after the withdrawl from the water bath the specimens were tested for their maximum tensile strength with universal testing machine(Instron 4302, England). To evaluate the flexibility of the impression materials, the cylindrical specimen was compress 10% of the original length, and the stress produced by that compression was evaluated. The flexibility of the cylindrical specimen was evaluated at 2 minutes-, 1 hour-, and 24 hours- after being withdrawn from the curing water bath by checking the stress at 10% compression. The results were as follows; 1. Examixfine showed the highest tensile strength value at 18.11 ㎏/㎠ followed by Provil L C.D. at 16.77 ㎏/㎠. The next was Express at 12.40 ㎏/㎠ and Correct VPS showed a significantly low value at 9.62 ㎏/㎠ when compared with other groups(P<0.01). 2. Under the 10% compression, Correct VPS showed significantly low stress at 1.511 ㎏/㎠ when compared with other groups(P<0.01). The other groups were Examixfine at 1.879 ㎏/㎠, Provil at 2.025 ㎏/㎠, and Express showed significantly higher stress at 3.689 ㎏/㎠(P<0.01). 3. The order of stress from lowest to highest for the 10% compression at 1 hour- and 24 hours- after being withdrawn from the curing water bath was Correct VPS, Examixfine, Provil L C.D., and Express. In all experimental groups the stress for dimensional shortening increased as the elapsed time increased.
