      • Sensitive detection of human fucosidase by an activity-based probe

        Alpha-L-fucosidase catalyzes the removal of L-Fucose residues from the non-reducing end of glycoconjugates. There are two fucosidases in human, including  α-L-fucosidase-1 (FUCA1) and α-L-fucosidase-2 (FUCA2). Abnormal fucosidase activity has been associated with many diseases, such as fucosidosis (one of the lysosomal storage disorders), hepatocellular cancer and breast cancer. We previously demonstrated that gastric epithelial cells secrete FUCA2 extracellularly upon the infection of Helicobacter pylori, a strategy tomodify the cell surface glycoconjugates to prevent the bacterial adhesion. However the role and physiological function of FUCA2 still remains ambiguous, not mentioning the difficulty that the enzyme is highly unstable and the activity is often too small for detection. We developed an activity-based probe for sensitive detection of the enzyme activity in vitro and in vivo. First of all, the probe was utilized for dot-blot assay to measure the H. pylori-induced fucosidase activity in a quantitative manner. Secondly, the probe provided a real-time detection of the increased fucosidase activity by using confocal microscopy. The result also supported the idea that the bacterial infection not only leads to an increase in the extracellular activity of FUCA2, but also an intracellular increase of fucosidase activity. Western blotting analysis by FUCA1- and FUCA2-specific antibodies further corroborated the aforementioned increase levels of FUCA1 and FUCA2 activities, respectively. Our current emphasis is placed to search for the factors of H. pylori in association with the secretion of fucosidase during infection.

The main objective of paper is to help developers identify defects based on existing software metrics using data mining techniques and thereby improve the software quality.In this paper, variousclassification techniquesare revisitedwhich are employed for software defect prediction using software metrics in the literature.

        Comparative Study on Growth Patterns of 25 Commercial Strains of Korean Native Chicken

        Prediction of growth patterns of commercial chicken strains is important. It can provide visual assessment of growth as function of time and prediction body weight (BW) at a specific age. The aim of current study is to compare the three nonlinear functions (i.e., Logistic, Gompertz, and von Betalanffy) for modeling the growth of twenty five commercial Korean native chicken (KNC) strains reared under a battery cage system until 32 weeks of age and to evaluate the three models with regard to their ability to describe the relationship between BW and age. A clear difference in growth pattern among 25 strains were observed and classified in to the groups according to their growth patterns. The highest and lowest estimated values for asymptotic body weight (C) for 3H and 5W were given by von Bertalanffy and Logistic model 4629.7 g for 2197.8 g respectively. The highest estimated parameter for maturating rate (b) was given by Logistic model 0.249 corresponds to the 2F and lowest in von Bertalanffy model 0.094 for 4Y. According to the coefficient of determination ($R^2$) and mean square of error (MSE), Gompertz and von Bertalanffy models were suitable to describe the growth of Korean native chicken. Moreover, von Bertalannfy model was well described the most of KNC growth with biologically meaningful parameter compared to Gompertz model.

        Combined Application of Pseudomonas fluorescens and Trichoderma viride has an Improved Biocontrol Activity Against Stem Rot in Groundnut

        In an attempt to develop effective biocontrol system for management of stem rot disease in groundnut, 57 bacterial isolates and 13 isolates of Trichoderma spp. were evaluated for their antagonistic activity against Sclerotium rolfsii. The antagonists were selected based on their ability to inhibit the external growth of S. rolfsii from infected groundnut seeds. Four isolates of Pseudomonas fluorescens, GB 4, GB 8, GB 10 and GB 27, and T. viride pq 1 were identified as potent antagonists of S. rolfsii. T. viride pq 1 produced extracellular chitinase and parasitized the mycelium of S. rolfsii. Under controlled environment conditions, P. fluorescens GB 10, GB 27, T. viride pq 1 and the systemic fungicide Thiram(equation omitted) reduced the mortality of S. rolfsii inoculated to groundnut seedlings by 58.0%, 55.9%, 70.0% and 25.9%, respectively compared to control. In vitro growth of P. fluorescens GB 10 and GB 27 was compatible with T. viride pq 1 and Thiram(equation omitted). Integrated use of these two bacterial isolates with T. viride pq 1 or Thiram(equation omitted) improved their biocontrol efficacy. Combined application of either GB 10 or GB 27 with T. viride pq 1 was significantly effective than that with Thiram(equation omitted) in protecting groundnut seedlings from stem rot infection.

        POU class 1 homeobox 1 gene polymorphisms associated with growth traits in Korean native chicken

        Objective: POU class 1 homeobox 1 (POU1F1) mediates growth hormone expression and activity by altering transcription, eventually resulting in growth rate variations. Therefore, we aimed to identify chicken POU1F1 polymorphisms and evaluate the association between single nucleotide polymorphisms (SNPs) and growth-related traits, and logistic growth curve parameter traits (${\alpha}$, ${\beta}$, and ${\gamma}$). Methods: Three SNPs (M_1 to M_3) were used to genotype 585 $F_1$ and 88 $F_0$ birds from five Korean native chicken lines using a polymerase chain reaction-restriction fragment length polymorphism method. Results: Single marker analyses and traits association analyses showed that M_2 was significantly associated with body weight at two weeks, weight gain from hatch to 2 weeks, and weight gain from 16 to 18 weeks (p<0.05). M_3 was significantly associated with weight gain from 14 to 16 weeks and from 16 to 18 weeks, and asymptotic body weight (${\alpha}$) (p<0.05). No traits were associated with M_1. The POU1F1 haplogroups were significantly associated with weight gain from 14 to 16 weeks (p = 0.020). Linkage disequilibrium test and Haploview analysis shown one main haploblock between M_2 and M_3 SNP. Conclusion: Thus, POU1F1 significantly affects the growth of Korean native chickens and their growth curve traits.

        Temporal and spatial assemblages of invasive birds occupying the urban landscape and its gradient in a southern city of India

        Common birds play an important role in regulating the functioning of urban ecosystems. Typically, a few common species have become invasive species threatening biodiversity worldwide. Our understanding of the dynamics of invasive birds in an Indian context is still in its infancy. Hence, we studied the gradual adaptation of invasive birds to novel habitats and their dispersal dynamics in a southern city of India. We tested the prediction that urban matrix are increasingly composed of invasive generalist species. The results illustrate the dominance of invasive species such as Corvus splendens, Acridotheres tristis, Acridotheres livia, and Milvus migrans in the urban environment. The significant abundance of C. splendens exhibited urbanization-induced homogenization. The land-use pattern showed more inclination toward the urban structures than the vegetative attributes. Specialist groups from the frugivore guild were found to decline from the urban environment, which may shed light on the ecological factors that constrain their distribution.

        Kisspeptin regulates the development of caprine primordial follicles in vitro

        Kisspeptin, a neuropeptide and the master controller of reproductive axis upstream to GnRH neurons, and its receptor are also expressed in extrahypothalamic tissues, such as ovaries. As systemic kisspeptin has been shown to modulate follicular dynamics in cattle, we hypothesized that kisspeptin has direct actions on the ovarian follicular development. We also hypothesized that kisspeptin regulation of primordial follicle development is via modulation of VEGF expression. In order to test these hypotheses, we cultured caprine ovarian cortical strips in vitro for 7 days with supplementation of kisspeptin at 1, 10 and 100 μM concentration and observed the development of primordial follicles into intermediate, primary and secondary follicles. We also studied the alteration in the expression profile of VEGF and VEGF transcript variant 2 mRNA during follicular development in the presence of kisspeptin. We confirmed the presence of GPR54 in goat ovaries in our preliminary studies. Supplementation of kisspeptin at 1 and 10 μM concentration facilitated the development of primordial follicles into intermediate, primary and secondary follicles with less number of degenerated follicles while the same at 100 μM resulted in degeneration of follicles. We observed a drastic increase in the expression profile of VEGF and VEGF transcript variant 2 mRNA upon culture which was independent of kisspeptin treatment. In conclusion, our studies show that kisspeptin facilitates ovarian primordial development in vitro.

        Single Nucleotide Polymorphism of TBC1D1 Gene Association with Growth Traits and Serum Clinical-Chemical Traits in Chicken

        TBC1D1 gene has known functional effects on body energy homeostasis and glucose uptake pathway in skeletal muscle tissue. This biological function is reported to have significant effects on traits of growth and meat quality in chicken. In this study, we focused on two single nucleotide polymorphisms (SNPs) (g.70179137A>G and g.70175861T>C) identified through SNP annotation information of Korean native chicken and previous literature for TBC1D1 in chicken. Association of SNPs in TBC1D1 with growth and serum clinical-chemical traits were evaluated. A total of 584 male and female birds from five Korean native chicken lines were used in the study. The SNP1 (g.70179137A>G) is located in intron 11 and SNP2 (g.70175861T>C) is a non-synonymous missense mutation in exon 10, responsible for the amino acid change from Methionine to Valine. The A allele of SNP1 and T allele of SNP2 had the highest allele frequencies. Both SNPs indicated moderate polymorphism information content values (0.25<PIC<0.5). Association results shown that SNP1 was significantly associated with BW00, BW20, GR14-16 and carcass weight (P<0.05). The AA genotype had higher value for all these traits except BW00. Whereas, genotype GG have lower values for BW20, GR14-16, GR18-20 and carcass weight. The sGOT level was significant with SNP1 but not with the SNP2. The GG genotype had the highest sGOT value and AG reported the lowest. BW20 was significantly associated with SNP2 (P<0.05). The body weight and carcass weight values of CC and TC genotypes were higher than those of TT genotype.
