        Silica-Polyaniline Based Bienzyme Cholesterol Biosensor: Fabrication and Characterization

        <P>In the present investigation, silica-polyaniline based bienzyme cholesterol biosensor is fabricated through a simple one-step electrochemical method. The one-step fabrication process involves electrochemical polymerization of N[3-(trimethoxysilyl)propyl]aniline to result poly(N[3-(trimethoxysilyl)propyl]aniline) (PTMSPA) and simultaneous immobilization of two enzymes, horseradish peroxidase (HRP) and cholesterol oxidase (ChOx) into PTMSPA matrix. The modified electrode is designated as PTMSPA-HRP/ChOx-ME. PTMSPA facilitates direct electron transfer between the electrode surface and the active redox centers of HRP. This enables the operation of a biosensor at a low working potential of about −150 mV (vs. Ag/AgCl) for the detection of hydrogen peroxide. The PTMSPA-HRP/ChOx-ME demonstrates excellent analytical performance for the detection of cholesterol between 1 and 25 mM with high sensitivity and selectivity. PTMSPA possesses features suited for the fabrication of third-generation biosensors.</P>

        Sperm and Seminal Plasma Proteomics: Molecular Changes Associated with Varicocele-Mediated Male Infertility

        Male infertility is a rising problem and the etiology at the molecular level is unclear. Use of omics has provided an insight into the underlying cellular changes in the spermatozoa of infertile men. Proteomics is one the promising omics techniques for biomarker screening that can provide complete information on molecular processes associated with male infertility. Varicocele is a pressing issue in the field of male infertility and the search for an appropriate diagnostic and therapeutic biomarker is still ongoing. In this review, we discuss the reports on proteomic profiles of sperm and seminal plasma in male infertility and provide an in-depth insight into varicocele studies associated with male infertility.

        Fabrication of Functional Nanofibrous Ammonia Sensor

        <P>A nanofibrous sensor for ammonia gas is fabricated by electrospinning the composite of poly(diphenylamine) (PDPA) with poly(methyl methacrylate) (PMMA) onto the patterned interdigit electrode. The composite electrospun membrane shows interconnected fibrous morphology. Functional groups in PDPA and the high active surface area of the fibrous membrane make the device detect a lower concentration of ammonia with a good reproducibility. The sensing capability of the device is studied by monitoring the changes in resistance of the membrane with different concentrations of ammonia. The changes in resistance of the membrane shows linearity with the concentration of ammonia in the limit of 10 and 300 ppm. UV-visible spectroscopy reveals the mechanism of sensing ammonia by the membrane.</P>

        Incidence and characteristics of major bleeding among rivaroxaban users with renal disease and nonvalvular atrial fibrillation

        Objective Patients with nonvalvular atrial fibrillation (AF) and renal disease (RD) who receive anticoagulation therapy appear to be at greater risk of major bleeding (MB) than AF patients without RD. As observed in past studies, anticoagulants are frequently withheld from AF patients with RD due to concerns regarding bleeding. The objective of this study was to evaluate the incidence and pattern of MB in those with RD, as compared to those without RD, in a population of rivaroxaban users with nonvalvular AF. Methods Electronic medical records of over 10 million patients from the Department of Defense Military Health System were queried to identify rivaroxaban users with nonvalvular AF. A validated algorithm was used to identify MB-related hospitalizations. RD was defined through diagnostic codes present within 6 months prior to the bleeding date for MB cases and end of study participation for non-MB patients. Data were collected on patient characteristics, comorbidities, MB management, and outcomes. Results Overall, 44,793 rivaroxaban users with nonvalvular AF were identified. RD was present among 6,921 patients (15.5%). Patients with RD had a higher rate of MB than those without RD, 4.52 per 100 person-years versus 2.54 per 100 person-years, respectively. The fatal bleeding outcome rate (0.09 per 100 person-years) was identical between those with and without RD. Conclusion In this post-marketing study of 44,793 rivaroxaban users with nonvalvular AF, RD patients experienced a higher MB rate than those without RD. The higher rate of MB among those with RD may be due to the confounding effects of comorbidities.

      • A Survey of EFL Instructors` Mediative Knowledge across Contexts of Teaching

        The current mixed-method study probed EFL instructors`` mediative knowledge across four different contexts of teaching including language institutes, universities, and senior and junior high schools in Iran. To this end, 181 EFL instructors, 80 males and 101 females, completed mediative closed-questionnaire which composed of twelve items to measure EFL instructors`` knowledge of mediation across context. Out of 181, 72 participants, 16 (22%) institute instructors, 20 (28%) university instructors, 19 (26.38%) senior instructors, and 17 (23.61%) junior instructors participated in oral interview. Results of descriptive statistics and Chi-square showed that there exist significant differences among EFL instructors in seven mediative items; furthermore, the difference shows that there exists relationship between EFL instructors`` mediative knowledge across the contexts; however, in five mediative items, no differences were found across contexts. Results of oral interview of EFL instructors in terms of assessment revealed that they mostly believed that learners through instructors`` guidance should self-assess their learning and behavior. To achieve such an end, they suggested instructors`` step by step scaffolding and strategic deliverance of assessment to the learners. In terms of cooperation and individualization, the results of oral interview revealed that instructors believed in unified improvement of both of them. To improve both, pair and group works were suggested to boost cooperation whereas competition and individual activities were suggested to foster individualization. Context variation due to its own community and unique characteristics, learners`` potential to self-assess their learning, and development of whole learner were suggested to be taken into consideration by instructors, educationalists, parents, and curriculum, and course designers.

        Proteomic Profiling of Seminal Plasma Proteins in Varicocele Patients

        Purpose: Seminal plasma provides a nutritive and protective milieu for spermatozoa. It contains factors/proteins required for sperm maturation, hyperactivation, capacitation and acrosome reaction. Alteration in the expression levels of seminal plasma proteins affect the fertilization process. The main objective of this study is to compare the seminal plasma proteome of healthy fertile men (control group) with varicocele patients in order to identify the differentially expressed seminal plasma proteins. Materials and Methods: Pooled seminal plasma samples from control (n=5) and varicocele (unilateral: n=5 and bilateral: n=5) subjects were used for proteomic profiling and functional bioinformatic analysis. Key differentially expressed proteins (DEPs) associated with binding of zona pellucida (acrosin; ACR), protein folding (heat shock related 70 kDa protein 2; HSPA2), oxidative stress (peroxiredoxin 2; PRDX2), lipid peroxidation and DNA fragmentation (apolipoprotein A2; APOA2) were validated by Western blot. Statistical analysis was conducted using Mann–Whitney test. Results: A total of 412 and 486 proteins were detected in seminal plasma of control group and varicocele patients respectively. Twenty-eight proteins were identified as DEPs between varicocele and control group. Validation of DEPs revealed downregulation of HSPA2 (p=0.0037) as well as APOA2 (p=0.0373), and upregulation of PRDX2 (p=0.0474). Conclusions: The seminal plasma protein profile of varicocele patients differ from healthy fertile men. Aberrant expression of seminal plasma proteins serve as an indicator of sperm pathology in varicocele patients.

        Protein Fingerprinting of Seminal Plasma Reveals Dysregulation of Exosome-Associated Proteins in Infertile Men with Unilateral Varicocele

        Purpose: Aberrant expression of seminal plasma proteins are associated with altered homeostasis that may affect the fertilizing ability of spermatozoa. However, the precise roles of seminal exosomes on sperm function remain unclear. The objective of this study was to identify the differentially expressed proteins (DEPs) associated with varicocele-mediated infertility by comparing seminal plasma protein profile of unilateral varicocele patients with proven fertile donors. Materials and Methods: Semen samples were obtained from 10 proven fertile donors with normal semen parameters and 33 infertile patients with unilateral varicocele. For proteomic analysis, 5 samples from each group were pooled and run in triplicate. Key DEPs (ANXA2, TF, CD63, KIF5B, SEMG1) associated with the exosome function were selected by bioinformatic tools and validated using Western blotting. Results: A total of 47 seminal plasma proteins were differentially expressed in unilateral varicocele patients compared to fertile donors. Validation of exosome-associated DEPs in unilateral varicocele patients (n=7) and fertile donors (n=7) revealed significant upregulation of ANXA2 (p=0.0016) and downregulation of KIF5B (p=0.009). The main upstream regulators of the DEPs in seminal plasma of unilateral varicocele group were androgen receptor, YB1 and NRF2. Conclusions: This is the first report to identify DEPs in seminal plasma of unilateral varicocele patients compared to fertile donors. Based on the detection of DEPs associated with exosomal function, Western blotting was used to validate the presence of defective exosome machinery in seminal plasma of unilateral varicocele patients. KIF5B and ANXA2 can be utilized as potential biomarkers of infertility in unilateral varicocele patients.

      • Enhanced Electrocatalysis for the Reduction of Hydrogen Peroxide at New Multiwall Carbon Nanotube Grafted Polydiphenylamine Modified Electrode

        <P>A new modified electrode having multiwall carbon nanotube (MWNT) grafted with polydiphenylamine (PDPA) as electrocatalytic layer is fabricated. FESEM image of the modified electrode shows a different morphology indicating the grafting of PDPA over MWNT. This morphology is in quite contrast from the conventional bilayer MWNT/conducting polymer modified electrode. The multiwall carbon nanotube grafted polydiphenylamine (MWNT-g-PDPA-ME) shows excellent electrocatalytic activity towards the reduction of hydrogen peroxide. The combined presence of MWNT and PDPA as a single unit provides better sensitivity than the bilayer configuration (MWNT/PDPA-ME). This modified electrode shows accelerated electron transfer at the interface with minimized surface fouling and surface renewability. The advantages of MWNT-g-PDPA-ME over the bilayer electrode are demonstrated with chronoamperometric studies. The amperometric response to H<SUB>2</SUB>O<SUB>2</SUB> obtained at −300 mV (vs. SCE) is rapid and highly sensitive as evident from the higher (2.83×10<SUP>−3</SUP> cm<SUP>3</SUP> mol<SUP>−1</SUP> s<SUP>−1</SUP>) rate constant for the diffusion reduction process.</P>

        Sensitive electrochemical detection of superoxide anion using gold nanoparticles distributed poly(methyl methacrylate)–polyaniline core–shell electrospun composite electrode

        <P>In the present communication, a novel composite nanofibrous electrode is developed for the detection of superoxide anion (O<SUB>2</SUB>˙<SUP>−</SUP>) in phosphate buffered saline (PBS). The composite fiber electrode is fabricated by dispersing gold nanoparticles onto poly(methyl methacrylate) (PMMA)–polyaniline (PANI) core–shell electrospun nanofibers. The constructed architecture is proven to be a favorable environment for the immobilization of the enzyme, superoxide dismutase (SOD). Direct electron transfer is achieved between SOD and the electrode with an electron transfer rate constant of 8.93 s<SUP>−1</SUP>. At an applied potential of +300 mV, PMMA/PANI–Au<SUB>nano</SUB>/SOD–ESCFM shows highly sensitive detection of O<SUB>2</SUB>˙<SUP>−</SUP>. In addition to this, quantification of different activities of SOD is realized at PMMA/PANI–Au<SUB>nano</SUB>/SOD–ESCFM.
