        The Association of Acquired T790M Mutation with Clinical Characteristics after Resistance to First-Line Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitor in Lung Adenocarcinoma

        Purpose The main objective of this study was to investigate the relationship among the clinical characteristics and the frequency of T790M mutation in advanced epidermal growth factor receptor (EGFR)mutant lung adenocarcinoma patients with acquired resistance after firstline EGFRtyrosine kinase inhibitor (TKI) treatment. Materials and Methods We enrolled EGFR-mutant stage IIIB-IV lung adenocarcinoma patients, who had progressed to prior EGFR-TKI therapy, and evaluated their rebiopsy EGFRmutation status. Results A total of 205 patients were enrolled for analysis. The overall T790M mutation rate of rebiopsy was 46.3%. The T790M mutation rates among patients with exon 19 deletion mutation, exon 21 L858R point mutation, and other mutations were 55.0%, 37.3%, and 27.3%, respectively. Baseline exon 19 deletion was associated with a significantly higher frequency of T790M mutation (adjusted odds ratio, 2.14; 95% confidence interval [CI], 1.20 to 3.83; p=0.010). In the exon 19 deletion subgroup, there was a greater prevalence of T790M mutation than other exon 19 deletion subtypes in patients with the Del E746-A750 mutation (61.6% vs. 40.6%; odds ratio, 2.35; 95% CI, 1.01 to 5.49; p=0.049). The progression- free survival (PFS) of first-line TKI treatment > 11 months was also associated with a higher T790M mutation rate (54.1% vs. 39.3%; adjusted odds ratio, 1.82; 95% CI, 1.02 to 3.25; p=0.044). Patients who underwent rebiopsy at metastatic sites had more chance to harbor T790M mutation (52.6% vs. 33.8%; adjusted odds ratio, 1.97; 95% CI, 1.06 to 3.67; p=0.032). Conclusion PFS of first-line EGFR-TKI, rebiopsy site, EGFR exon 19 deletion and its subtype Del E746- A750 mutation are associated with the frequency of T790M mutation.

        The Clinical Outcomes of Different First-Line EGFR-TKIs Plus Bevacizumab in Advanced EGFR-Mutant Lung Adenocarcinoma

        Purpose The aim of this study was to investigate the efficacy of various epidermal growth factor receptor (EGFR)–tyrosine kinase inhibitors (TKIs) plus bevacizumab in advanced EGFR-mutant lung adenocarcinoma patients. Materials and Methods From August 2016 to October 2020, we enrolled advanced lung adenocarcinoma patients harboring exon 19 deletion or L858R receiving gefitinib, erlotinib and afatinib plus bevacizumab as the first-line treatment for the purposes of analysis. Results A total of 36 patients were included in the final analysis. Three patients received gefitinib, 17 received erlotinib, and 16 received afatinib combined with bevacizumab as the first-line treatment. The objective response rate was 77.8%, and disease control rate was 94.4%. The overall median progression-free survival (PFS) was 16.4 months, while the median PFS was 17.1 months in patients with exon 19 deletion, and 16.2 months in patients with L858R mutation (p=0.311). Regarding the use of different EGFR-TKIs, the median PFS was 17.1 months in the erlotinib group and 21.6 months in the afatinib group (p=0.617). In patients with brain metastasis at baseline, the median PFS was 18.9 months in the erlotinib group and 16.4 months in the afatinib group (p=0.747). Amongst patients harboring exon 19 deletion, the median PFS was 16.2 months in the erlotinib group and not-reached in the afatinib group (p=0.141). In patients with L858R mutation, the median PFS was 18.9 months in the erlotinib group and 16.2 months in the afatinib group (p=0.481). Conclusion Our research demonstrates that not only erlotinib combined with bevacizumab, but also afatinib plus bevacizumab as first-line treatment, provides solid clinical efficacy in advanced EGFR-mutant lung adenocarcinoma patients.         Non-pharmacological Approach in the Management of Functional Dyspepsia

        Functional dyspepsia (FD) is a common functional gastrointestinal disease which bears a significant burden on society and individuals. Despite the high prevalence of FD, its pathophysiology remains poorly understood and the treatment options are limited and unsatisfactory. In the absence of effective pharmacological treatments for FD, non-pharmacological approaches, including: reassurance, lifestyle modification, psychotherapy, dietary interventions, medical food, acupuncture, and electrical stimulation and modulation are sought after by many physicians and FD patients. In this article, we review clinical studies which investigate non­pharmacological therapies for FD. We will also discuss potential mechanisms involved in the therapeutic effects of these non­pharmacological approaches. Though the evidences to support the routine use of the non-pharmacological management is still lacking, the non-invasive nature and potentially minimal side-effects of these therapies may be attractive in the FD management. In order to confirm the clinical effectiveness of these non-pharmacological approaches, more well-conducted, methodologically rigorous, and large-scaled clinical trials are required.

        Isoamylamine Induces B16-F1 Melanoma Cell Autophagy by Upregulating the 5′ Adenosine Monophosphate-Activated Protein Pathway

        Isoamylamine (IA) is an aliphatic monoamine molecule present in cheese, eggs, and wine. It belongs to the family of polyamines and also can be synthesized endogenously. It has been known that regulation of polyamines in cells is related to cell cycle and tumor formation. Malignant melanoma is difficult to treat and easily resistant to chemotherapy/radiotherapy through autophagy. In this study, we aim to clarify whether IA has a growth control effect on melanoma tumor cells and the regulatory mechanism. We treated B16-F1 melanoma cells with IA at concentrations of 0, 200, 400, and 600 ppm for 24 h. The 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay was checked for cell viability and results showed that IA has an inhibitory effect on B16-F1 melanoma cells. The signaling molecules, which included Raf/MEK/ERK, were activated, while MSK1 and protein kinase B (AKT) were suppressed. Autophagy was also confirmed to be induced by IA. The acridine orange stain-positive cells were increased and BECN-1/LC3 upregulated. The data also showed that the autophagy regulatory molecule, 5′-adenosine monophosphate-activated protein kinase (AMPK), was induced after IA treatment, so we used dorsomorphin to inhibit AMPK and found that it could suppress autophagy. In conclusion, IA has an effect of inducing autophagy in B16-F1 cells and it is regulated through AMPK.

        Microwave dielectric properties of (1 - x)(Mg0.95Co0.05)TiO3–x(Na0.5La0.5)TiO3 ceramic system

        The microstructures and the microwave dielectric properties of the (1 - x)(Mg0.95Co0.05)TiO3– x(Na0.5La0.5)TiO3 ceramic system were investigated. Two-phase system was confirmed by the XRD patterns and the EDX analysis. A co-existed second phase (Mg0.95Co0.05)Ti2O5 was also detected. The microwave dielectric properties are strongly related to the density and the matrix of the specimen. A new microwave dielectric material 0.88(Mg0.95Co0.05)TiO3–0.12(Na0.5La0.5)TiO3, possessing an excellent combination of dielectric properties: εr ~22.36, Q × f ~ 110,000 GHz (at 9 GHz), τf ~ 2.9 ppm/℃), is proposed as a candidate dielectric for GPS patch antennas.         An approach to hybrid inorganic nanoparticles in reactive PS-b-PMSMA amphiphilic copolymers

        A series of well-defined amphiphilic poly(styrene)-block-poly 3-(trimethoxysilyl) propyl methacrylate (PS-b-PMSMA) copolymers with controlled molecular weight and block length were prepared by the atom transfer free radical polymerization. The cadmium sulfide (CdS) nanoparticles were fabricated in the spherical micelles self-assembled from these prepared PS-b-PMSMA copolymers. Then, the CdS/PSb-PMSMA films were obtained by spin coating the CdS/PS-b-PMSMA solution on silicon wafer. The experimental results showed the addition of Cu(II) could decrease the value of polydispersity index for the prepared copolymers. Nuclear magnetic resonance and Fourier transform infrared spectra showed the synthesis of PS-b-PMSMA copolymer. The average roughness and mean square roughness of the prepared CdS/PS-b-PMSMA films obtained from the atomic force microscopy analysis were 3.0e3.4 nm and 1.7e2.0 nm, respectively, indicating the excellent surface planarity. On the other hand, the ratio of block length between PS and PMSMA had a great influence on the micelle size. The larger ratio of PS to PMSMA block length resulted in the larger size of micelles and CdS nanoparticles that caused a red-shift of ultravioletevisible and photoluminescence spectra. The red-shift of spectra was explained by the quantum confinement effect associated with the tiny size of the CdS nanoparticles.

        Metal Chloride Precursor Synthesization of Cu2ZnSnS4 Solar Cell Materials

        Cu2ZnSnS4 (CZTS) thin films with kesterite structures were prepared by directly sol-gel synthesizingspin-coated precursors on soda-lime-glass (SLG) substrates. The CZTS precursors wereprepared by using solutions of copper (II) chloride, zinc (II) chloride, tin (IV) chloride, and thiourea. The ratio of SnCl4 in the precursors was found to play a critical role in the synthesization of CZTS. CZTS phases of SnS and SnS2 were observed in the synthesized films as prepared using precursorswith a close to stoichiometric ratio of CuCl2:ZnCl2:SnCl4:CH4N2S = 4:1:1:8, where SnCl4 was 1mol/l. The amounts of the educed SnS and SnS2 phases observed in the SEM images could bereadily reduced by decreasing the volume of SnCl4 in the mixed solution. With decreasing amountof SnCl4 from 1 mol/l, the as prepared CZTS reveals a significant improvement in its crystallineproperties. In this work, CZTS with an average absorption coefficient and an optical energy gapof over 104 cm−1 and 1.5 eV, respectively, was obtained using precursors of copper (II) chloride,zinc (II) chloride, tin (IV) chloride, and thiourea mixed in a ratio of 2:1:0.25:8, and it had goodcrystallinity revealing a Cu-poor composition.

        Improving discharge uniformity of industrial-scale very high frequency plasma sources by launching a traveling wave

        Very high frequency (VHF) PECVD has been demonstrated to be able to significantly increase the deposition rate without compromising the film quality for the manufacture of silicon heterojunction and silicon thin film solar cells. To further reduce the production costs by enhancing the throughput, larger electrode and higher frequency are often required at the same time. Nevertheless, raising frequency in large-area PECVD results in non-uniform discharge caused by the standing wave effect and deteriorates the processing uniformity. In this study, a technique that generates a traveling wave via superposing two specific standing waves launched simultaneously is proposed to resolve this issue. An industrial-scale linear plasma reactor with length and width of 125 and 10 cm, respectively, is adopted for experimental tests and two 80 MHz power supplies are utilized to separately control the standing waves. The experimental results show that the discharge gap is only partially covered by plasma discharge when only one standing wave is applied. However, as both standing waves are launched, the non-uniformity of plasma discharge can be effectively reduced to <±5%. In addition, numerical simulation is also conducted in this study to clarify whether the proposed technique can be applied to large-area rectangular PECVD (substrate size: 1.4 m 1.1 m). By arranging multiple feeding points on opposite sides of the powered electrode, the simulation results indicate the non-uniformity of electric field can be maintained within ±10%.
