      • Curdione Inhibits Proliferation of MCF-7 Cells by Inducing Apoptosis

        Background: Curdione, one of the major components of Curcuma zedoaria, has been reported to possess various biological activities. It thus might be a candidate anti-flammatory and cancer chemopreventive agent. However, the precise molecular mechanisms of action of curdione on cancer cells are still unclear. In this study, we investigated the effect of curdione on breast cancer. Materials and Methods: Xenograft nude mice were used to detect the effect of curdione on breast cancer in vivo; we also tested the effect of curdione on breast cancer in vitro by MTT, Flow cytometry, JC-I assay, and western blot. Results: Firstly, we found that curdione significantly suppressed tumor growth in a xenograft nude mouse breast tumor model in a dose-dependent manner. In addition, curdione treatment inhibited cell proliferation and induced cell apoptosis. Moreover, after curdione treatment, increase of impaired mitochondrial membrane potential occurred in a concentration dependent manner. Furthermore, the expression of apoptosis-related proteins including cleaved caspase-3, caspase-9 and Bax was increased in curdione treatment groups, while the expression of the anti-apoptotic Bcl-2 was decreased. Inhibitors of caspase-3 were used to confirm that curdione induced apoptosis. Conclusions: Overall, our observations first suggested that curdione inhibited the proliferation of breast cancer cells by inducing apoptosis. These results might provide some molecular basis for the anti-cancer activity of curdione.

        Dammarane-type triterpene oligoglycosides from the leaves and stems of Panax notoginseng and their antiinflammatory activities

        Background: Inflammation is widespread in the clinical pathology and closely associated to the progress of many diseases. Triterpenoid saponins as a key group of active ingredients in Panax notoginseng (Burk.) F.H. Chen were demonstrated to show antiinflammatory effects. However, the chemical structures of saponins in the leaves and stems of Panax notoginseng (PNLS) are still not fully clear. Herein, the isolation, purification and further evaluation of the antiinflammatory activity of dammarane-type triterpenoid saponins from PNLS were conducted. Methods: Silica gel and reversed-phase C8 column chromatography were used. Furthermore, preparative HPLC was used as a final purification technique to obtain minor saponins with high purities. MS, NMR experiments, and chemical methods were used in the structural identifications. The antiinflammatory activities of the isolated saponins were assessed by measuring the nitric oxide production in RAW 264.7 cells stimulated by lipopolysaccharides. Real-time reverse transcription polymerase chain reaction was used to measure the gene expressions of inflammation-related gene. Results: Eight new minor dammarane-type triterpene oligoglycosides, namely notoginsenosides LK1-LK8 (1-8) were obtained from PNLS, along with seven known ones. Among the isolated saponins, gypenoside IX significantly suppressed the nitric oxide production and inflammatory cytokines including tumor necrosis $factor-{\alpha}$, interleukin 10, interferon-inducible protein 10 and $interleukin-1{\beta}$. Conclusion: The eight saponins may enrich and expand the chemical library of saponins in Panax genus. Moreover, it is reported for the first time that gypenoside IX showed moderate antiinflammatory activity.

        Comprehensive transcriptome and metabolome analysis reveals regulatory network for lignin biosynthesis in chicory sprouting

        Chicory is consumed worldwide and is an important commercial crop. However, excess lignin deposition may reduce its quality. The comprehension of the molecular mechanism underlying the biosynthesis of lignin in chicory is currently inadequate. To address this, an integrative analysis of the metabolome and transcriptome profiles was performed in chicory sprout at three different stages. A total of 706 metabolites were identified, with cinnamic acid, ferulic acid, coniferaldehyde, and sinapaldehyde enriched during the growth of chicory sprouts. This suggested that these four metabolites may affect the growth of chicory sprouts. Transcriptome analysis demonstrated that the expression of most of the differentially expressed genes (DEGs) involved in lignin biosynthesis was up-regulated during chicory growth. Importantly, the metabolite and gene expression profiles were closely correlated during sprout development, especially in association with lignin biosynthesis. The results will serve as a reference for lignin biosynthesis in chicory and may also assist biologists in improving chicory quality.

        Simultaneous analysis of bioactive metabolites from Ziziphus jujuba by HPLC–DAD–ELSD–MS/MS

        A high-performance liquid chromatography (HPLC) with diode array detector (DAD), evaporative light scattering detector (ELSD) and electrospray ionization mass spectrometry (ESI-MS) was established for the simultaneous determination of nine representative metabolites of the alkaloid, flavonoid and triterpenoid classes from Ziziphus jujuba var. spinosa. The optimal chromatographic conditions were obtained on an ODS column and the mobile phase was composed of water and methanol with 0.1% formic acid using a gradient elution system. Using the developed methods, all of the validation parameters were successfully obtained. In addition, effectiveness of diverse extraction methods was compared to each other for the development of standard analytic method. The verified method was successfully applied to the quantitative determination of representative metabolites in commercial samples of Z. jujuba and Z. mauritiana from different markets in Korea, China and Myanmar. The analytical results showed that the contents of the nine analytes vary significantly with sources and species, thus demonstrating its potential for the detection of this plant.

        Maximum Likelihood Multi-innovation Stochastic Gradient Estimation for Multivariate Equation-error Systems

        This paper focuses on the parameter estimation problems of multivariate equation-error systems. A multiinnovation generalized extended stochastic gradient algorithm is presented as a comparison. Based on the maximum likelihood principle and the coupling identification concept, the multivariate equation-error system is decomposed into several regressive identification subsystems, each of which has only a parameter vector, and a coupled subsystem maximum likelihood multi-innovation stochastic gradient identification algorithm is developed for estimating the parameter vectors of these subsystems. The simulation results show that the coupled subsystem maximum likelihood multi-innovation stochastic gradient algorithm can generate more accurate parameter estimates and has faster convergence rates compared with the multi-innovation generalized extended stochastic gradient algorithm.

        CRISPR/Cas9‑mediated editing of CsWRKY22 reduces susceptibility to Xanthomonas citri subsp. citri in Wanjincheng orange (Citrus sinensis (L.) Osbeck)

        Key message CRISPR/Cas9-mediated editing of CsWRKY22 repressed canker development in Wanjincheng orange. Abstract Citrus canker, a destructive disease of citrus, is threatening the citrus industry worldwide. Breeding resistant cultivars is the most economical and effective approach to control citrus canker. Recently, CRISPR/Cas9-mediated genome editing was demonstrated to be a powerful tool for the improvement of citrus resistance to the disease. In our previous works, we confirmed that CsWRKY22 is involved in the plant immunity response to citrus canker in Wanjincheng orange (Citrus sinensis (L.) Osbeck). In this study, we targeted this gene to improve the resistance of Wanjincheng orange against citrus canker by CRISPR/Cas9-mediated editing. Sanger sequencing confirmed that CsWRKY22 in Wanjincheng orange contains CsWRKY22G and CsWRKY22C alleles, and the ratio of CsWRKY22G to CsWRKY22C is approximately 2:1. Four sgRNAs, which targeted the first exon of CsWRKY22, were selected for testing. In vitro cleavage activity analysis showed that two (W1 and W2) of the four sgRNAs displayed robust cleavage activities using PCR amplicons from the Wanjincheng orange genome as template. Subsequently, two constructs, pCas9/WRKY22sgRNA-W1 and pCas9/WRKY22sgRNA-W2, were used to modify the CsWRKY22. Three mutant plants were identified from seven independent transgenic plants. Based on Sanger sequencing, the W1-1, W2-2, and W2-3 mutant lines displayed 85.7%, 79.2%, and 68.2% mutation rates, respectively. Resistance evaluation indicated that the mutant plants showed decreased susceptibility to citrus canker. These results indicate that CRISPR/Cas9-targeted gene modification is an efficient approach for enhancing disease resistance in citrus.

        An iterative method for solving equilibrium problem fixed point problem and generalized variational inequalities problem

        In this paper, we introduce a new iterative scheme for nding a common element of the set of an equilibrium problem, the set of xed points of nonexpansive mapping and the set of solutions of the generalized variational inequality for α-inverse strongly g-monotone mapping in a Hilbert space. Under suitable conditions, strong convergence theorems for approximating a common element of the above three sets are obtained.

        Bond Behaviors between Ribbed BFRP Bars and Ecological High Ductility Cementitious Composites

        Direct pull-out method was conducted to explore the bond stress-slip response of ribbed basalt fiber reinforced polymer (BFRP) bars embedded in ecological high ductility cementitious composites (Eco-HDCCs). The effects of the embedment length and cover thickness on the bond properties between Eco-HDCCs and the bars were described. Results indicate that the failure characteristics of the specimens depend on the embedment length and cover thickness of the BFRP bars. As the embedment lengths of the bars increase, the ultimate bond stress decreases while the ultimate slip increases. Besides, both the ultimate bond stress and ultimate slip show an increasing trend if the specimen has a cover thickness of less than 45 mm, while the increasing trend is slower if the specimen has a cover thickness of more than 45 mm. In addition, the strain of the specimen surface depends on the cover thickness, and the strain increases when the cover thickness is in a range of 15 mm − 35 mm. Moreover, certain embedment lengths and cover thicknesses of the BFRP bars are recommended in the structural design of bridge deck link slabs. The bond stress-slip relationship models of the BFRP bars in Eco-HDCCs are proposed based on the test results.

        Viscosity approximation methods for nonexpansive semingroups and monotone mapppings

        Let C be a nonempty closed convex subset of real Hilbert space H and F = {S(t) : t ≥ 0} a nonexpansive self-mapping semigroup of C, and f : C → C is a fixed contractive mapping. Consider the process {xn} :x_(n+1) =β_nx_n+(1−β_n)z_n z_n = α_nf(x_n) + (1 − α_n)S(t_n)P_C(x_n − r_nAx_n). It is shown that {xn} converges strongly to a common element of the set of fixed points of nonexpansive semigroups and the set of solutions of the variational inequality for an inverse strongly-monotone mapping which solves some variational inequality.

        Preparation and photoluminescence properties of the Eu2+, Sm3+ co-doped Li2SrSiO4 phosphors

        A series of Eu2+ and Sm3+ co-doped Li2SrSiO4 phosphors are prepared by the high temperature solidstate reaction. The morphology, structure and spectroscopic properties of the prepared samples are characterized by scanning electron microscopy, X-ray diffraction, diffuse reflection spectra, photoluminescence spectra and electron paramagnetic resonance spectra, respectively. The effect of Sm3þdoping concentration on the photoluminescence intensity of the prepared samples is also investigated. The results indicate that the crystal structure of Li2SrSiO4 is not changed with the Eu2+, Sm3+ co-doping. The spherical-like particle size of the obtained product is about 20-30 nm in diameter. When the Sm3+ concentration is 0.3 mol% and the Eu2+ concentration is 0.7 mol%, the phosphors show the maximum emission intensity, which is 50% higher than that of Eu2+ doped Li2SrSiO4. Excited at 420 nm, the phosphor presents a single broad emission band peaking at 558 nm, which is ascribed to the 4f65d1 / 4f7 transitions of Eu2+ and 4G5/2 / 6H5/2 and 4G5/2 / 6H7/2 transitions of Sm3+. The Commission International de I0Eclairage chromaticity coordinates of Li2SrSiO4:0.7 mol% Eu2+, 0.3 mol%Sm3+ are x = 0.28, y = 0.28.
