      • Evaluation of Several Screening Approaches for Detection of Cervical Lesions in Rural Shandong, China

        Purpose: The study was designed to: (1) investigate the prevalence of high-risk human papillomavirus (HR-HPV) infection and cervical neoplasia; and (2) evaluate clinical performance of visual inspection with acetic acid/ Lugol's iodine (VIA /VILI), Pap smear, high-risk human papillomavirus (HR-HPV) DNA test for detecting cervical intraepithelial neoplasia grade 2 or worse (CIN2+) and (3) explore appropriate screening approach in rural areas of Shandong Province. Materials and Methods: A total of 3,763 eligible women from Yiyuan County in Yimeng mountainous areas of rural Shandong, China, were enrolled and underwent Pap smear, HR-HPV DNA testing by Hybrid Capture 2 (HC2), and VIA /VILI tests. Women positive in any test were referred to colposcopy and biopsy as indicated. Results: The prevalence of HR-HPV infection among all enrolled women was 11.1% and that in healthy women was 9.9%. In total 33 cases of CIN1, 16 cases of CIN2, 6 cases of CIN3 but none of cervical cancer were detected and the crude prevalence of CIN2+ was 0.58%. For detecting CIN2+, the sensitivity of HR-HPV DNA testing, VIA/VILI, Pap smear was 90.9%, 77.3%, 81.8%, respectively. Pap smear had the best specificity of 98.2%, followed by HR-HPV DNA testing with specificity of 89.4%, VIA/VILI had the lowest specificity of 81.2%. Colposcopy referral rate of HR-HPV DNA testing, VIA/VILI, Pap smear was 11.1%, 18.5%, 2.3%, respectively. Conclusions: Our results suggest that HR-HPV DNA testing alone might be appropriate for primary cervical cancer screening in rural low-resource areas of Shandong Province, China.

        Knockout of the EgriBLOS2 gene results in the transparent integuments of Ectropis grisescens larvae

        The body colors of insects have evolved specialized roles in body protection, signaling and physiology. In some case, the larvae can camouflage their body colors to adapt the habitants and protect themselves. However, the genetic and molecular basis on larval body colors of the tea geometrid, Ectropis grisescens, remains poorly known. Here, we reported an effect of the lysosome-related organelles complex-1, subunit 2 (EgriBLOS2) gene knockout on larval integuments of E. grisescens, by using a clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein 9 (Cas9) system. Combining transcriptomic data and PCR approaches, we first identified the EgriBLOS2 gene from E. grisescens, which encoded 137 amino acids and comprised 3 introns. In the phylogenetic analysis, EgriBLOS2 clustered in the Lepidoptera clade with high conservation to members in other lepidopteran species. Developmental expression profiles revealed that EgriBLOS2 was constantly tran scribed at each stage, in which its expression was significantly lower in 2nd instar larvae than that of other instars. After injecting EgriBLOS2-specific guide RNA and Cas9 messenger RNA into eggs, 70% of larvae showed the translucent integuments in G0 generation, with an emphasis on black splayed patterns in the 2nd and 8th segments of abdomens. However, some typical characteristics of larvae were not obviously changed in mutant instars, such as ocelli, mouthparts and other appendants. This study has unraveled the roles of EgriBLOS2 in the formation of larval integument colors, and provides an alternative strategy for pest management based on the colors in this species.

        Glucose regulated protein 78 promotes cell invasion via regulation of uPA production and secretion in colon cancer cells

        Glucose regulated protein 78 (GRP78) is frequently highlyexpressed in tumor cells, contributing to the acquisition of severalphenotypic cancer hallmarks. GRP78 expression is also positivelycorrelated with tumor metastasis, and promotes hepatocellularcarcinoma cell invasion via increasing cell motility, however, other mechanisms involving the prometastatic roles of GRP78remain to be elucidated. Here we report that forced GRP78expression promotes colon cancer cell migration and invasionthrough upregulating MMP-2, MMP-9 and especially uPAproduction. These effects of GRP78 are mediated by enhancingthe activation of β-catenin signaling. Interestingly, we identify thatGRP78 interacts with uPA both in the cells and in the culturemedium, suggesting that GRP78 protein is likely to directlyfacilitate uPA secretion via protein-protein interaction. Takentogether, our findings demonstrate for the first time that besidesstimulation of cell motility, GRP78 can act by increasingproteases production to promote tumor cell invasion.

        Analysis of the Relationship between MHC-DRB1 Gene Polymorphism and Hydatidosis in Kazakh Sheep

        The objective of this work was to analyze the relationship between ovine major histocompatibility complex (MHC) DRB1 gene polymorphism and genetic resistance to hydatidosis in Kazakh sheep. The Ovar (ovine MHC) class II DRB1 second exon was amplified by polymerase chain reaction (PCR) from DNA samples of 702 Kazakh sheep, including 302 sheep with hydatidosis and 400 health controls. PCR products were characterized by the restriction fragment length polymorphism (RFLP) technique using five restriction enzymes, i.e., MvaI, HaeIII, SacI, SacII and Hin1I, yielding 14 alleles and 28 genotypes. Comparing the frequency of genotypes in hydatidosis sheep with the control group, it was found that the genotype frequencies of MvaIbc, Hin1Iab, SacIIab, HaeIIIde, HaeIIIdf and HaeIIIdd in control sheep were significantly (p<0.01) higher than in hydatidosis sheep, indicating that a significant correlation existed between these genotypes and resistance to hydatidosis. Genotype frequencies of MvaIbb, SacIIaa, Hin1Ibb and HaeIIIef in sheep with hydatidosis were extremely significantly (p<0.01) higher than in the control group, and the genotype frequency of HaeIIIab was significantly higher (p<0.05), indicating that a marked correlation existed between these genotypes and susceptibility to hydatidosis. By way of analyzing haplotype with these resistant genotypes, the hydatidosis resistant haplotype MvaIbc-SacIIab-Hin1Iab of Kazakh sheep was screened out, and then verified through artificial hydatid infection in sheep. The results indicated that the infection rate of sheep with the resistant haplotype of hydatidosis was significantly lower (p<0.01) than without this resistant haplotype. It showed that the genic haplotype MvaIbc-SacIIab-Hin1Iab of Ovar-DRB1 exon 2 was the resistant haplotype of hydatidosis in Kazakh sheep.

        Sealing design optimization of nuclear pressure relief valves based on the polynomial chaos expansion surrogate model

        Pressure relief valve (PRV) is one of the important control valves used in nuclear power plants, and its sealing performance is crucial to ensure the safety and function of the entire pressure system. For the sealing performance improving purpose, an explicit function that accounts for all design parameters and can accurately describe the relationship between the multi-design parameters and the seal performance is essential, which is also the challenge of the valve seal design and/or optimization work. On this basis, a surrogate model-based design optimization is carried out in this paper. To obtain the basic data required by the surrogate model, both the Finite Element Model (FEM) and the Computational Fluid Dynamics (CFD) based numerical models were successively established, and thereby both the contact stresses of valve static sealing and dynamic impact (between valve disk and nozzle) could be predicted. With these basic data, the polynomial chaos expansion (PCE) surrogate model which can not only be used for inputs-outputs relationship construction, but also produce the sensitivity of different design parameters were developed. Based on the PCE surrogate model, a new design scheme was obtained after optimization, in which the valve sealing stress is increased by 24.42% while keeping the maximum impact stress lower than 90% of the material allowable stress. The result confirms the ability and feasibility of the method proposed in this paper, and should also be suitable for performance design optimizations of control valves with similar structures.

      • Expression and Significance of the Wip1 Proto-oncogene in Colorectal Cancer

        Aim: To investigate the level of expression of proto-oncogene Wip1 and its physiological significance in colorectal cancer. Methods: Immunohistochemistry, semi-quantitative RT-PCR, and Western blotting were used to analyze Wip1 mRNA and protein expression in 120 cases of colorectal cancer and normal tissues to study relationships with clinical symptoms and disease prognosis. Results: The level of Wip1 protein expression was found to be significantly higher in colorectal cancer tissues (85% (102/120)) than in normal tissues (30% (36/120)) (P<0.05). The relative amount of Wip1 protein in colorectal cancer tissue was also found to be significantly higher (P<0.05) than in normal tissues ($1.060{\pm}0.02$ and $0.640{\pm}0.023$, respectively). Semi-quantitative RT-PCR showed average Wip1 mRNA expression levels to be $1.113{\pm}0.018$ and $0.658{\pm}0.036$ for colorectal cancer tissue and adjacent normal tissue (P<0.05). The level of Wip1 protein expression was not correlated with age, gender, or tumor site, but appeared linked with lymph node metastasis, Dukes stage, histological grade, and liver metastasis. Individuals with high and low levels of Wip1 expression showed statistically significant differences in the five-year overall survival and recurrence-free survival rates (P<0.05). Conclusion: Wip1 mRNA and protein are highly expressed in colorectal cancers and may be associated with colorectal cancer development and progression.

      • The Domain Ontology and Domain Rules Based Requirements Model Checking

        Many ontology-based methods have been proposed and applied in order to elicit system requirements correctly and unambiguously. However, most of ontologies in these methods are purely conceptual models. Furthermore, the domain knowledge base only captures domain concepts and neglects domain-restricted rules. If the requirements model violate these rules or contradict the usual business behavior, they become unreasonable. This paper suggests a formal approach to precisely describe ontology using description logic at first, and then model the integrity rules and derivation rules which restrict the business behavior. All the rules are represented in three aspects: syntax, semantics and visualization. Finally, the requirements model checking framework is provided combining domain ontology and domain rules, which makes the requirements elicitation process both guided by domain ontology and restricted by domain rules. Therefore, the acquired requirements would comply with both business needs and domain knowledge.

        Synthesis of Chitosan-g-Poly(ethylene glycol)-g-Polyethyleneimine Copolymer and Its Research as Drug Carrier

        Tumors have emerged as the number one cause of death with the incidence rate rising rapidly due to various factors. Nanoparticle-mediated PolyI:C (PIC) delivery is a promising approach in the treatment of various tumors. However, the delivery of PIC to target cells is very difficult. Therefore, it is necessary to develop carriers which can safely deliver PIC to the target cells. In this study, chitosan-g-poly (ethylene glycol)-g-polyethyleneimine copolymer (CS-g-PEG-g-PEI) which delivers PIC as a carrier was synthesized by carbonyldiimidazole (CDI) coupling method using ionic liquid as the reaction medium. The results of Fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance spectroscopy (1H NMR and 13C NMR), elemental analysis and gel permeation chromatography (GPC) showed that PEG and PEI were grafted onto CS successfully. The result of cytotoxicity assay indicated low levels of cytotoxicity in CS-g-PEG-g-PEI. The positively charged CS-g-PEG-g-PEI self-assembled with the negatively charged PIC to form composite nanoparticles whose physicochemical properties were studied. The results of stability experiments and subsequent gel electrophoresis indicated that the stability of nanoparticles solution was good with water as a solvent, CS-g-PEG-g- PEI as a carrier, the ratio of nitrogen/phosphorus (N/P) is 8, and the concentration of PIC as 0.5 mg/mL. CS-g-PEG-g-PEI could bind to PIC almost completely and effectively protect it from degradation of Rnase A at N/P=8, and the PIC encapsulation efficiency was over 87%. The results of morphological characterization showed that nano-sized self-assembled aggregates with core-shell structure could be prepared successfully. This provides a new carrier for long-circulation and research of solid tumor drug delivery system.

        ZNF488 Enhances the Invasion and Tumorigenesis in Nasopharyngeal Carcinoma Via the Wnt Signaling Pathway Involving Epithelial Mesenchymal Transition

        Purpose The purpose of this study was to investigate the function of Zinc finger protein 488 (ZNF488) in nasopharyngeal carcinoma (NPC). Materials and Methods The endogenous expression of ZNF488 in NPC tissues, normal nasopharyngeal epithelium tissues and NPC cell lines were detected by quantitative reverse transcription polymerase chain reaction. ZNF488 over-expressing and knock-down NPC cell line models were estab- lished through retroviral vector pMSCV mediated over-expression and small interfering RNA (siRNA) mediated knock-down. The invasion and migration capacities were evaluated by wound healing and transwell invasion assays in ZNF488 over-expressing and control cell lines. Soft-agar colony formation and a xenograft experiment were performed to study tumorigenic ability in vitro and in vivo. Immunofluorescence and western blotting analysis were used to examine protein changes followed by ZNF488 over-expression. Microarray analysis was performed to explore gene expression profilings, while luciferase reporter assay to evaluate the transcriptive activity of Tcf/Lef. Results ZNF488 was over-expressed in NPC tissues compared with normal tissues, especially higher in 5-8F and S18, which are well-established high metastatic NPC clones. Functional studies indicate that over-expression of ZNF488 provokes invasion, whereas knock-down of ZNF488 alleviates invasive capability. Moreover, over-expression of ZNF488 promotes NPC tumor growth both in vitro and in vivo. Our data further show that over-expression of ZNF488 induces epithelial mesenchymal transition (EMT) by activating the WNT/β -catenin signaling pathway. Conclusion Our data strongly suggest that ZNF488 acts as an oncogene, promoting invasion and tumorigenesis by activating the Wnt/β -catenin pathway to induce EMT in NPC.

        Cytogenetic Analysis of Polygala tenuifolia Wild by Different Staining Techniques and Fluorescent In Situ Hybridization

        Karyotype of P. tenuifolia was characterized with emphasis on heterochromatin distribution using Giemsa C-banding, Chromomycin A3 (CMA3), DAPI, silver impregnation and localization of ribosomal (18S-5.8S-26S rDNA) by fluorescence in situ hybridization (FISH). Diploid chromosome complement, 2n = 2x = 38, consisted of 13 pairs of submetacentric and 6 pairs of metacentric chromosomes. C-banding and silver staining showed a conspicuous bands on the short arms of pair 13, where the secondary constriction (SC) was located. The only GC rich heterochromatin, as revealed by fluorochrome Chromomycin A3 (CMA) staining, was that associated with nucleolar organizer regions (NORs), where 4, 6-diamino-2-phenylindole (DAPI) apparently stained pale. AT rich heterochromatin stained with DAPI was distributed uniformly on all chromosomes. FISH with 45S rDNA probe revealed one 18S-5.8S-26S rDNA loci on secondary constriction of chromosome pair 13, where they corresponded to nucleolar organizer regions. The ribosomal DNA behaviors during the cell cycle were analyzed on interphase nuclei, prophases, metaphases, anaphase and telophase; indicate that the activity of rDNA at individual loci may also vary through different phases.
