        LINC01272 Suppressed Cell Multiplication and Induced Apoptosis Via Regulating MiR-7-5p/CRLS1 Axis in Lung Cancer

        LINC01272 is a long non-coding RNA (lncRNA) that has been considered as a biomarker for many diseases including lung squamous cell carcinoma. Here, we investigated the function and mechanism of LINC01272 on lung cancer (LC). The differential expression of LINC01272 in LC and normal samples was analyzed by GEPIA based on the data from TCGA-LUAD database, as survival prognosis was analyzed through Kaplan-Meier Plotter. LINC01272 overexpression plasmid and miR-7-5p mimic were transfected into A549 and PC-9 cells. LINC01272, miR-7-5p and cardiolipin synthase 1 (CRLS1) mRNA expression was measured by quantitative reverse transcription-polymerase chain reaction. Cell viability was detected through MTT assay. Cell multiplication was evaluated by cell formation assay. Cell apoptosis was assessed through flow cytometry assay. Through bioinformatics, the target miRNA of LINC01272 and downstream genes of miR-7-5p were predicted. The targeting relationship was tested by dual luciferase reporter analysis. CRLS1, B-cell lymphoma-2 (Bcl-2), BCL2-associated X (Bax) and cleaved caspase-3 protein levels were detected through western blot. LINC01272 was downregulated in LC and low LINC01272 expression had poor prognosis. In A549 and PC-9 cells, LINC01272 inhibited cell viability and multiplication and induced apoptosis. LINC01272 negatively regulated miR-7-5p and CRLS1 was a target of miR-7-5p. MiR-7-5p reversed the effect of LINC01272 on viability, multiplication, apoptosis and expression of miR-7-5p and CRLS1 as well as apoptosis-related factors (Bcl-2, Bax and cleaved caspase-3). LINC01272 suppressed cell multiplication and induced apoptosis via regulating the miR-7-5p/CRLS1 axis in LC.

        주세 체계 개편으로 인한 주류 산업의 변화와 주세 부과 방안에 관한 연구

        On January 1, 2020, the liquor tax for beer and takju was reorganized from the ad valorem tax to the specific tax. The purpose of the reorganization of the liquor tax announced by the National Tax Service is to improve the quality of the liquor and to resolve unreasonable discrimination between domestic and imported liquor. However, it is necessary to determine whether the National Tax Service’s standard for levying the liquor tax is appropriate for the purpose. In this study, the change in the liquor industry is estimated due to the reorganization of the liquor tax using Hicks net price elasticity. In addition, the specific tax for each of the liquors and the alcohol content derived from the social cost minimization model is compared. The main findings are as follows. First, when the liquor tax of beer and takju is converted to the specific tax, social costs increase, and social welfare decrease. Second, if all the liquors are converted to the specific tax, social costs decrease. Third, when comparing specific tax by each of the liquors and the alcohol content according to the social cost minimization model, The specific tax by alcohol content can be considered more appropriate in terms of social cost and the stakeholders in the liquor industry.

        Smart farm (plant factory) technology is evolving into various farms of convergence technology through linkage with bio & IT industry, and urban agriculture. Particularly, it can serve as a key solution to societal issues, such as rural aging, rural migration settlement and rural job creation for young people. However, the level of technology and industrialization of the plant factories in domestic smart farms is still growing. For example, plant factories are centered on production of leaf vegetables due to limited technology to maintain light quality and intensity of artificial light sources. Therefore, it is important to acquire core technology necessary to design an automated system that enhances modeling and industrializing Korean smart farms. In this study, a natural light condensing and transmission device was designed to develop a next-generation light supply system that uses both natural and artificial light. The prototype of the natural light condensing and transmission device consisted of a condenser lens, an optical sensor, a light source tracking device, and an optical fiber. The condenser lens has increased the efficiency of the optical fiber by focusing more natural light on the optical fiber in the form of an aspherical convex lens. The optical sensor maximizes the efficiency of light collection by sensing from the sunrise. The light source tracking device enables more precise tracking of light sources as it uses an optical sensor system. The optical fiber has the highest degree of freedom among optical transmission technologies and it can transmit more than 200 m light sources with a low attenuation rate (0.06%/m). In the future, The spectroscopic measurement of light source and light efficiency (loss rate) will test using a spectrophotometer (FieldSpec 4, ASD Inc., USA) having a measurement range of 350 ~ 2,500 nm.

        Effect of Lithium in Transition Metal Layers of Ni-Rich Cathode Materials on Electrochemical Properties

        <P>Li was incorporated into transition metal layers of Ni-rich Li[Ni<SUB>0.95</SUB>Co<SUB>0.05</SUB>]O<SUB>2</SUB> by formation of a solid solution with Li<SUB>2</SUB>MnO<SUB>3</SUB> (layer notation: Li[Li<SUB>0.33</SUB>Mn<SUB>0.67</SUB>]O<SUB>2</SUB>), which can be denoted as (1-<I>x</I>)Li[Ni<SUB>0.95</SUB>Co<SUB>0.05</SUB>]O<SUB>2</SUB> – <I>x</I>Li[Li<SUB>0.33</SUB>Mn<SUB>0.67</SUB>]O<SUB>2</SUB>, to understand the effect of Li on the structure, electrochemistry and thermal characteristic of the cathode. Structural analysis data obtained by Rietveld refinement of X-ray diffraction data indicate that the additional Li can be found in the transition metal layers of (1-<I>x</I>)Li[Ni<SUB>0.95</SUB>Co<SUB>0.05</SUB>]O<SUB>2</SUB> – <I>x</I>Li[Li<SUB>0.33</SUB>Mn<SUB>0.67</SUB>]O<SUB>2</SUB>. An interesting feature is that the average oxidation states of Ni and Mn are 3+ and 4+, respectively, for (1-<I>x</I>)Li[Ni<SUB>0.95</SUB>Co<SUB>0.05</SUB>]O<SUB>2</SUB> – <I>x</I>Li[Li<SUB>0.33</SUB>Mn<SUB>0.67</SUB>]O<SUB>2</SUB> as demonstrated by X-ray absorption study. The first discharge capacity was approximately 217 mAh g<SUP>−1</SUP>, and the resulting retention was above 90% for 0.9Li[Ni<SUB>0.95</SUB>Co<SUB>0.05</SUB>]O<SUB>2</SUB> – 0.1Li[Li<SUB>0.33</SUB>Mn<SUB>0.67</SUB>]O<SUB>2</SUB> (Li[Li<SUB>0.033</SUB>Ni<SUB>0.855</SUB>Co<SUB>0.045</SUB>Mn<SUB>0.067</SUB>]O<SUB>2</SUB>) in the range of 2.7 to 4.5 V for 100 cycles, which is a surprising result for Ni-rich compounds. Moreover, the thermal stability of (1-<I>x</I>)Li[Ni<SUB>0.95</SUB>Co<SUB>0.05</SUB>]O<SUB>2</SUB> – <I>x</I>Li[Li<SUB>0.33</SUB>Mn<SUB>0.67</SUB>]O<SUB>2</SUB> was significantly improved over the cathodes with identical Ni fraction. These results highlight the role of tetravalent Mn ions, even in small amounts, in stabilizing the electrochemical performances and thermal properties in the Ni-rich layer cathodes.</P>

        Glucose and Insulin Stimulate Lipogenesis in Porcine Adipocytes: Dissimilar and Identical Regulation Pathway for Key Transcription Factors

        Lipogenesis is under the concerted action of ChREBP, SREBP-1c and other transcription factors in response to glucose and insulin. The isolated porcine preadipocytes were differentiated into mature adipocytes to investigate the roles and interrelation of these transcription factors in the context of glucose- and insulin-induced lipogenesis in pigs. In ChREBP-silenced adipocytes, glucose-induced lipogenesis decreased by ~70%, however insulin-induced lipogenesis was unaffected. Moreover, insulin had no effect on ChREBP expression of unperturbed adipocytes irrespective of glucose concentration, suggesting ChREBP mediate glucose-induced lipogenesis. Insulin stimulated SREBP-1c expression and when SREBP-1c activation was blocked, and the insulin-induced lipogenesis decreased by ~55%, suggesting SREBP-1c is a key transcription factor mediating insulin-induced lipogenesis. $LXR{\alpha}$ activation promoted lipogenesis and lipogenic genes expression. In ChREBP-silenced or SREBP-1c activation blocked adipocytes, $LXR{\alpha}$ activation facilitated lipogenesis and SREBP-1c expression, but had no effect on ChREBP expression. Therefore, $LXR{\alpha}$ might mediate lipogenesis via SREBP-1c rather than ChREBP. When ChREBP expression was silenced and SREBP-1c activation blocked simultaneously, glucose and insulin were still able to stimulated lipogenesis and lipogenic genes expression, and $LXR{\alpha}$ activation enhanced these effects, suggesting $LXR{\alpha}$ mediated directly glucose- and insulin-induced lipogenesis. In summary, glucose and insulin stimulated lipogenesis through both dissimilar and identical regulation pathway in porcine adipocytes.

        LincR-PPP2R5C Promotes Th2 Cell Differentiation Through PPP2R5C/PP2A by Forming an RNA–DNA Triplex in Allergic Asthma

        Purpose: The roles and mechanisms of long noncoding RNAs (lncRNAs) in T helper 2 (Th2) differentiation from allergic asthma are poorly understood. We aimed to explore a novel lncRNA, LincR-protein phosphatase 2 regulatory subunit B' gamma (PPP2R5C), in Th2 differentiation in a mouse model of asthma. Methods: LincR-PPP2R5C from RNA-seq data of CD4+ T cells of asthma-like mice were validated and confirmed by quantitative reverse transcription polymerase chain reaction, northern blotting, nuclear and cytoplasmic separation, and fluorescence in situ hybridization (FISH). Lentiviruses encoding LincR-PPP2R5C or shRNA were used to overexpress or silence LincR-PPP2R5C in CD4+ T cells. The interactions between LincR-PPP2R5C and PPP2R5C were explored with western blotting, chromatin isolation by RNA purification assay, and fluorescence resonance energy transfer. An ovalbumin-induced acute asthma model in knockout (KO) mice (LincR-PPP2R5C KO, CD4 conditional LincR-PPP2R5C KO) was established to explore the roles of LincR-PPP2R5C in Th2 differentiation. Results: LncR-PPP2R5C was significantly higher in CD4+ T cells from asthmatic mice ex vivo and Th2 cells in vitro. The lentivirus encoding LincR-PPP2R5C suppressed Th1 differentiation; in contrast, the short hairpin RNA (shRNA) lentivirus decreased LincR-PPP2R5C and Th2 differentiation. Mechanistically, LincR-PPP2R5C deficiency suppressed the phosphatase activity of the protein phosphatase 2A (PP2A) holocomplex, resulting in a decline in Th2 differentiation. The formation of an RNA-DNA triplex between LincR-PPP2R5C and the PPP2R5C promoter enhanced PPP2R5C expression and activated PP2A. LincR-PPP2R5C KO and CD4 conditional KO decreased Th2 differentiation, airway hyperresponsiveness and inflammatory responses. Conclusions: LincR-PPP2R5C regulated PPP2R5C expression and PP2A activity by forming an RNA-DNA triplex with the PPP2R5C promoter, leading to Th2 polarization in a mouse model of acute asthma. Our data presented the first definitive evidence of lncRNAs in the regulation of Th2 cells in asthma.

        A successive ‘‘conversion-deposition” mechanism achieved by micro-crystalline Cu2O modified current collector for composite lithium anode

        Lithium (Li) metal is a promising material for high-energy–density batteries, but it is still plagued byobvious capacity degradation and low average Coulombic efficiency resulting from dendrite Li propagation. One main reason is the electro-mechanic coupled failure of plated Li on the current collector, whichcontributes to non-dense Li deposition on the anode. Transition metal oxides (TMOs) with a conversiontypemechanism have been used directly as the anode materials for lithium ion batteries, which demonstratedbetter electro-mechanical stability than metal Li. Herein, a successive ’’conversion-deposition’’mechanism is ingeniously developed to restrain the generation of dendritic Li. Specifically, a microcrystallineCu2O modified current collector was prepared, in which Li+ are sequentially inserted intoCu2O and deposited in the form of Li metal at successive low potential. A Li-Cu half-cell based on thehybrid mechanism sustains a high Coulombic efficiency of over 99.3 % in up to 800 cycles. This work ingeniouslyinhibits the generation of dendrite Li by incorporating conversion-type materials withdeposition-dissolution type metal Li, which contributes to a novel concept for the design of functionalcurrent collectors for composite Li anodes.

        북한 교과서 개편에 따른 문학교육 양상 -김정일 시대를 중심으로

        The North Korean education has been changed into by the place that the situation of the times and a social system intended. This study reach it in Kim Jong-il era, and, on the other hand, an aspect of North Korea education shows a change in contents and a system while inheriting former quality This study considered a literature education aspect of Kim Jong-il era through a change process of national language literature textbook. The education was able to confirm that I was in charge of a function to maintain the North Korea system through this in North Korea. Because education has an important duty to build socialist society in North Korea, it is put with guidance and influence of the bonbon receipt, and literature education performs the reflection that is substantiality for this. 1996 when textbook reorganization made ends meet in things of the times that were Kim Jong-il whom this report paid attention to was the time when North Korea experienced a thorny march. It appears that the revolution person ability training of the principle of good chance is a major object and educates independent literary arts theory, Historic understanding for basics theory for culture word and literature and the acquisition for the trend of thought of world literature formed main contents. Generally, National language literature textbook is inclined to a duty of politics thought education and communism revolution person ability training education than a peculiar function of literature education. This study understands that North Korea literature education establishes an important point for ideology education to get over a North Korean crisis after 1996.

      • Insertion Sequence 6110의 Mixed-Linker 중합효소 연쇄반응을 이용한 Mycobacterium tuberculosis의 Molecular Typing

        목적 : M. tuberculosis에서 mixed-linker를 이용한 IS6110의 PCR을 시행하여 그 RFLP를 분석하고, 이를 기존의 RFLP법에 의한 결과와 비교 분석하여 신속하고 효과적인 molecular typing법으로서의 유용성을 검토하였다. 방법 : Mixed-linker PCR법을 이용하여 다양한 약제 감수성을 보이는 13명의 환자에서 분리된 14결핵 균주들의 임상 분리균을 대상으로 IS6110과 이에 인접된 염기서열을 포함하는 다양한 절편들을 증폭시켜 얻어진 mixed-linker RFLP를 conventional RFLP와 비교하였다. 결과 : 13명의 환자에서 분리된 13균주들의 mixed-linker RFLP 결과 모든 다른 RFLP양상을 보였으며, 이는 conventional RFLP의 결과와 일치하였다. Mixed-linker PCR법으로 증폭된 절편들은 기본적으로 150-600 bp 사이에 존재하였으며, IS6110의 한절편인 소식자로 Southern hybridization하여 IS6110에 특이함을 증명하였다. Mixed-linker RFLP는 conventional RFLP보다 더 많은 수의 절편들을 보여 주었다. Mixed-linker와 conventional RFLP 양상들은 약제 내성과 무관하였다. 결론 : Mixed-linker PCR법은 기존의 conventional RFLP법보다 더욱 간편한 방법으로 결핵균의 신속한 동정 및 molecular typing을 가능하게 하여 결핵의 역학조사에 효과적인 방법임을 알 수 있다. Background : Various classic typing methods including phage typing, antibiotic resistance typing and multilocus enzyme electrophoresis have been proven inefficient because of the phenotypic and genotypic homogeneities of the Mycobacterium tuberculosis(M.tuberculosis) species. Thus, DNA restriction fragment length polymorphism(RFLP) analysis using the repetitive element insertion sequence 6110(IS6110) of the M. tuberculosis genome has been proposed to be a powerful epidemiologic tool. However, this method requires large amount of genomic DNA and Southern hybridization procedures. The aim of this study was to evaluate DNA RFLP analysis by mixed-linker polymerase chain reaction(mixed-linker PCR) for a rapid, simple typing method, which specifically amplified genomic RFLP fragments containing IS6110 with using one primer specific for IS6110 and a second primer complementary to a linker ligated to the restricted genomic DNA. Methods : Fourteen clinical isolates of M. tuberculosis with various drug susceptibilities including 2 strains isolated from a patient were used for the study. Their RFLP patterns by mixed linker-PCR(mixed liker RFLP) were analyzed and compard with those by the conventional RFLP method. Results : Mixed-linker RFLP obtained from 13 isolates of 13 patients showed all different reproducible patterns and 2 isolates from the same patient showed almost the same RFLP patterns which had different drug susceptibility. mixed-linker RFLP patterns were correlated to the results by the conventional RFLP. And mixed-linker RFLP showed a higher number of fragments containing IS6110 than those by the conventional RFLP. However, both mixed-linker and conventional RFLP were independent of drug resistance patterns. Conclusion : Mixed linker-RFLP is a rapid, simple typing method with high discrimination and reproducibility, and seems to be useful for epidemiologic study of M. tuberculosis.

        A New Comprehensive Short-form Health Literacy Survey Tool for Patients in General

        Purpose: To validate a conceptual short-form health literacy 12 items questionnaire (HL-SF12) in patient populations. Methods: A cross-sectional study was conducted via a convenient sample of 403 patients from three departments of a community general hospital in the northern Taiwan. Patients’ health literacy was assessed with a validated HL-SF12, derived from the full scale, the European Health Literacy Survey Questionnaire (HLS-EU-Q), as well as a single-item from Chew’s Set of Brief Health Literacy Question. A reference population in Northern Taiwan (n = 928) via the HLS-EU-Q in 2013e2014 was used as a reference to compare the health literacy between that of the general public and the patients. Data was analyzed by confirmatory factor analysis (CFA), internal consistency analysis, correlation analysis, and linear regression models. Results: Patients’ health literacy assessed with the HL-SF12 was shown with high internal consistency (Cronbach a = .87), and moderately correlated with the single-item from Chew's Set of Brief Health Literacy Question, with satisfactory item-scale convergent validity (item-scale correlation> .40), without floor/ceiling effect, and with satisfactory goodness of fit indices of the three-factor construct model for most of the patients. Their health literacy was significantly positively associated with female gender, higher income, and more often watching health-related TV programs. On the other hands, patients were reported with significantly higher healthcare health literacy than the general public, but not in general health literacy, disease prevention health literacy, or health promotion health literacy. Conclusion: The comprehensive HL-SF12 was a valid and easy to use tool for assessing patients’ health literacy in the hospitals to facilitate healthcare providers in enhancing patients’ health literacy and healthcare qualities.
