        Reduction of d-lactate content in sauerkraut using starter cultures of recombinant Leuconostoc mesenteroides expressing the ldhL gene

        <P>The n-form of lactate, which causes metabolic stress upon excessive dietary intake, is mainly produced by Leuconostoc sp., the predominant species in sauerkraut. To shift the metabolic flux of D-lactate from pyruvate to L-lactate, we expressed the L-lactate dehydrogenase (ldhL) gene in Leuconostoc mesenteroides ATCC 8293. The IdhL gene from Lactobacillus plantarum was introduced into L. mesenteroides using the shuttle vectors pLeuCM and pLeuCM42. To elevate the expression level of IdhL in L. mesenteroides, the nucleotides for pyruvate kinase promoter were fused to IdhL and cloned into above vectors to construct pLC18pkL and pLC42pkL. As results, introduction of pLC42pkL in L. mesenteroides significantly improved both L-LDH activity and L-lactate productivity during fermentation, decreasing the D-/L-lactate ratio. When used as a starter culture for sauerkraut fermentation, recombinant L. mesenteroides harboring pLC42pkL increased L-lactate concentration and decreased D-lactate concentration compared to the wild type strain. We newly developed a recombinant L. mesenteroides which has high L-lactate dehydrogenase activity and applied this strain to minimize the harmful effect of D-lactate during the sauerkraut fermentation. To the best of our knowledge, we demonstrate for the first time the effective use of recombinant Leuconostoc sp. for quality improvement of fermented foods.         Jacobi–Stirling numbers, Jacobi polynomials, and the left-definite analysis of the classical Jacobi differential expression

        <P><B>Abstract</B></P><P>We develop the left-definite analysis associated with the self-adjoint Jacobi operator Ak(α,β), generated from the classical second-order Jacobi differential expression<SUB>ℓα,β,k</SUB>[y](t)=1<SUB>wα,β</SUB>(t)((-(1-t<SUP>)α+1</SUP>(1+t<SUP>)β+1</SUP><SUP>y′</SUP>(t)<SUP>)′</SUP>+k(1-t<SUP>)α</SUP>(1+t<SUP>)β</SUP>y(t))(t∈(-1,1)),in the Hilbert space Lα,β2(-1,1)≔<SUP>L2</SUP>((-1,1);<SUB>wα,β</SUB>(t)), where <SUB>wα,β</SUB>(t)=(1-t<SUP>)α</SUP>(1+t<SUP>)β</SUP>, that has the Jacobi polynomials {Pm(α,β)}m=0∞ as eigenfunctions; here, α,β>-1 and <I>k</I> is a fixed, non-negative constant. More specifically, for each n∈N, we explicitly determine the unique left-definite Hilbert–Sobolev space Wn,k(α,β)(-1,1) and the corresponding unique left-definite self-adjoint operator Bn,k(α,β) in Wn,k(α,β)(-1,1) associated with the pair (Lα,β2(-1,1),Ak(α,β)). The Jacobi polynomials {Pm(α,β)}m=0∞ form a complete orthogonal set in each left-definite space Wn,k(α,β)(-1,1) and are the eigenfunctions of each Bn,k(α,β). Moreover, in this paper, we explicitly determine the domain of each Bn,k(α,β) as well as each integral power of Ak(α,β). The key to determining these spaces and operators is in finding the explicit Lagrangian symmetric form of the integral composite powers of <SUB>ℓα,β,k</SUB>[·]. In turn, the key to determining these powers is a double sequence of numbers which we introduce in this paper as the <I>Jacobi–Stirling numbers</I>. Some properties of these numbers, which in some ways behave like the classical Stirling numbers of the second kind, are established including a remarkable, and yet somewhat mysterious, identity involving these numbers and the eigenvalues of Ak(α,β).</P>

      • Microbiological Quality of Alternative Water Sources in the Urban Environment

        In Australia, decreasing rainfall, frequent drought and population growth in urban environments, along with an overall desire to achieve greater water sustainability, has increased the demand for alternative water sources such as stormwater, roof-harvested rainwater, and treated wastewater effluent. The potential public health risks associated with alternative water sources predominantly originate from the presence of enteric viruses, pathogenic bacteria, and protozoan parasites. Empirical data on the presence of pathogens is a prerequisite to accurately determine human health risks and the extent of treatment required prior to use for potable and nonpotable purposes. We have carried out a number of targeted studies on the microbiological quality of the roof-harvested rainwater, stormwater, and treated wastewater effluent in the Southeast Queensland (Australia). The main of aim these studies was to collect quantitative data on the presence of pathogens and make an assessment of the potential health risk associated with water reuse. A dedicated survey of the roof-harvested rainwater tanks in Brisbane has shown the presence of Campylobacter spp. (21%), Salmonella spp., (4%), and Giardia lamblia (13%). In addition, opportunistic pathogens Legionella spp. (99%) and Mycobacterium intracellulare (78%) were also detected\. The numbers of bacteria ranged up to 3.1 x 10<sup>6</sup> PCR detectable units (pdu) L<sup>-1</sup> for Legionella spp., 9.6 X 10<sup>5</sup> pdu L<sup>-1</sup> for P. aeruginosa, 6.8 X 10<sup>5</sup> pdu L<sup>-1</sup> for M. intracellulare, 6.6 x 10<sup>5</sup> pdu L<sup>-1</sup> for Acanthamoeba spp., 1.1 x 10<sup>5</sup> pdu L<sup>-1</sup> for M. avium, and 9.8 X 10<sup>3</sup> pdu L<sup>-1</sup> for L. pneumophila. The presence of potential opportunistic pathogens in tank water may present health risks if un-treated water is for potable and non-potable use in the house. Stormwater runoff from two urban catchments with different drainage area, impervious area, and land use, a potential source of pollution and population density was collected and analyzed for the presence of pathogens and fecal indicator bacteria. The mean FIB numbers in water samples collected after the storm event ranged from 10 <sup>3</sup> to 10<sup>4</sup> L<sup>-1</sup> for E. coli and 10 <sup>4</sup> to 10<sup>5</sup> L<sup>-1</sup> for Enterococcus spp. Salmonella enterica numbers varied between 10 to 2400 MPN L<sup>-1</sup>. Human adenovirus (HAdV) numbers in the stormwater varied between 10 to 10<sup>3</sup> pdu L<sup>-1</sup>, human polyomavirus (HPyV) numbers varied between 1 to 10<sup>3</sup> pdu L<sup>-1</sup>. Torque teno virus (HTtV) numbers varied between 10 to 7.45 x 10<sup>3</sup> pdu L<sup>-1</sup>. Somatic coliphages (Microviridae) was also widely prevalent with numbers varying between 10 to 10<sup>3</sup> pdu L<sup>-1</sup>. The results of this study suggest that enteric virus and bacterial pathogens frequently occur in the stormwater runoff in significant numbers which could lead to increased human health risks. A quantitative survey HAdV, HPyV, HTtV and somatic coliphage (Microviridae) numbers in influent and effluent samples was carried out from three wastewater treatment plants (WWTP) employing an activated sludge process to produce secondary treated effluent in Brisbane. HPyV, HAdV, HTtV and Microviridae were consistently detected in the primary influent in high numbers (10<sup>5</sup> to 10<sup>6</sup> pdu L<sup>-1</sup>) and secondary treated effluent (10<sup>2</sup> to 10<sup>3</sup> pdu L<sup>-1</sup>) across all three treatment plants. Under appropriate conditions, WWTPs with activated sludge process could be an effective treatment barrier with > 3 log10 removal of enteric. The results confirm the occurrence of enteric viruses in significant numbers in secondary treated effluent, tertiary treatment may be required prior to effluent reuse or discharged into the environmental to prevent exposure of people to health hazards.

        Characterization of Lactobacillus acidophilus Isolated from Piglets and Chicken

        Lactic acid bacteria were isolated from piglets and chicken and characterized. Lactic acid bacteria showing resistance to low pH and bile, adhesion to intestinal epithelium cells, and the inhibition of Escherichia coli and Salmonella spp. were identified as Lactobacillus acidophilus. L. acidophilus PF01 survived for 2 h in MRS broth adjusted to pH 2. L. acidophilus CF07 was less resistant than L. acidophilus PF01 to pH 2, but survived at pH 2.5 for 2 h. Both of isolates were able to grow in MRS broth containing 0.3% (w/v) bile, with L. acidophilus CF07 being more tolerant to bile than L. acidophilus PF01. L. acidophilus PF01 and CF07 adhered specifically to the duodenal and jejunal epithelium cells of piglet, and the cecal and duodenal epithelium cells of chicken, respectively. Both of isolates did not adhere to the epithelium cells of the various animal intestines from which they were isolated. When L. acidophilus was cultured with E. coli and Salmonella spp. in MRS broth, MRS broth containing 2% skim milk powder or modified tryptic soy broth at $37^{\circ}C$, L. acidophilus PF01 and CF07 inhibited the growths of E. coli K88 and K99, and S. enteritidis and S. typhimurium, respectively. Both of isolates were found to possess the essential characteristics of probiotic lactic acid bacteria for piglet and chicken.

      • DHS-21, a dicarbonyl/l-xylulose reductase (DCXR) ortholog, regulates longevity and reproduction in Caenorhabditis elegans

        Dicarbonyl/l-xylulose reductase (DCXR) converts l-xylulose into xylitol, and reduces various α-dicarbonyl compounds, thus performing a dual role in carbohydrate metabolism and detoxification. In this study, we identified DHS-21 as the only DCXR ortholog in Caenorhabditis elegans. The dhs-21 gene is expressed in various tissues including the intestine, gonadal sheath cells, uterine seam (utse) cells, the spermathecal-uterus (sp-ut) valve and on the plasma membrane of spermatids. Recombinant DHS-21 was shown to convert l-xylulose to xylitol using NADPH as a cofactor. Dhs-21 null mutants of C. elegans show defects in longevity, reproduction and egg-laying. Knock-down of daf-16 and elt-2 transcription factors affected dhs-21 expression. These results suggest that DHS-21 is a bona fide DCXR of C. elegans, essential for normal life span and reproduction.

        Current status of arsenic exposure and social implication in the Mekong River basin of Cambodia

        <P>To evaluate the current status of arsenic exposure in the Mekong River basin of Cambodia, field interview along with urine sample collection was conducted in the arsenic-affected area of Kandal Province, Cambodia. Urine samples were analyzed for total arsenic concentrations by inductively coupled plasma mass spectrometry. As a result, arsenicosis patients (n = 127) had As in urine (UAs) ranging from 3.76 to 373 A mu g L-1 (mean = 78.7 +/- A 69.8 A mu g L-1; median = 60.2 A mu g L-1). Asymptomatic villagers (n = 108) had UAs ranging from 5.93 to 312 A mu g L-1 (mean = 73.0 +/- A 52.2 A mu g L-1; median = 60.5 A mu g L-1). About 24.7 % of all participants had UAs greater than 100 A mu g L-1 which indicated a recent arsenic exposure. A survey found that females and adults were more likely to be diagnosed with skin sign of arsenicosis than males and children, respectively. Education level, age, gender, groundwater drinking period, residence time in the village and amount of water drunk per day may influence the incidence of skin signs of arsenicosis. This study suggests that residents in Kandal study area are currently at risk of arsenic although some mitigation has been implemented. More commitment should be made to address this public health concern in rural Cambodia.</P>

        Effects of L-tryptophan, Fructan, and Casein on Reducing Ammonia, Hydrogen Sulfide, and Skatole in Fermented Swine Manure

        The effects of daily dietary Bacillus subtilis (Bs), and adding L-tryptophan, fructan, or casein to fecal fermentation broths were investigated as means to reduce the production of noxious gas during manure fermentation caused by ammonia, hydrogen sulfide (H2S), and 3-methylindole (skatole). Eighty swine (50.0±0.5 kg) were equally apportioned to an experimental group given Bs in daily feed, or a control group without Bs. After 6 weeks, fresh manure was collected from both groups for fermentation studies using a 3×3 orthogonal array, in which tryptophan, casein, and fructan were added at various concentrations. After fermentation, the ammonia, H2S, L-tryptophan, skatole, and microflora were measured. In both groups, L-tryptophan was the principle additive increasing skatole production, with significant correlation (r = 0.9992). L-tryptophan had no effect on the production of ammonia, H2S, or skatole in animals fed Bs. In both groups, fructan was the principle additive that reduced H2S production (r = 0.9981). Fructan and Bs significantly interacted in H2S production (p = 0.014). Casein was the principle additive affecting the concentration of ammonia, only in the control group. Casein and Bs significantly interacted in ammonia production (p = 0.039). The predominant bacteria were Bacillus spp. CWBI B1434 (26%) in the control group, and Streptococcus alactolyticus AF201899 (36%) in the experimental group. In summary, daily dietary Bs reduced ammonia production during fecal fermentation. Lessening L-tryptophan and increasing fructan in the fermentation broth reduced skatole and H2S.

        S-1. Baculovirus 에 삽입한 Copia 와 같은 운반요소의 성질

        Baculoviruses possess large(∼100 Kilobase pair), double-stranded, circular, DNA genomes which are packaged in enveloped, rod-shaped capsids. A mutant of the baculovirus Autographa californica nuclear polyherodsis virus (AcNPV) was recently isolated following serial passage of the virus through a cell line of the lepidopteran insect Trichoplusia ni (1). The, mutant, known as FP-D, has a most unusual genotpe; restriction endonuclease analysis of the viral DNA reveals the presence of a genetically unstable sequence inserted in the mutant DNA. Further analysis has established that this sequence is a mobile genetic element of T. ni and is a member of the copia-like class of transposable elements (2). We call the element TE-D. TE-D is 7.3 Kilobases (Kb) in length and contains 0.27 Kb direct terminal repeats so that it's structure is similar to copia-like elements of Drophila as well as the evolutionarily related retroviruses of vertebrates. The TE-D sequence is genetically unstable; a second virus, referred to as FP-DS, arises spontaneously and at high frequency in FP-D infected cells. FP-DS contains only one of the two 0.27 Kb terminal repeats of TE-D and may arise from recombination between the direct terminal repeats of TE-D. We alse have evidence that the element exists in free linear and circular from in FP-D infected cells; transposition of the element may therefore involve a 7.05 Kb circular and/or linear interm ediate. TE-D is a mid-repetitive, dispersed, element of T. ni cells. It is found in 45 or 50 different locations in the T. ni genome. The element is mobile as determined by experiments in which the position of TE-D sequences in T. ni larvae and T. ni cell culture are compared. In T. ni cell culture, the TE-D sequences have amplified several fold and the internal sequences of TE-D have diverged slightly. We are currently investigating the effects of element insertion on viral transcription and have observed the enhancements of transcription of viral sequences next to terminal repeats. FP-D will be a very valuable tool for studying transposable element movement and expression. We have a permissive insect cell line, S. frugiperda, which lacks TE-D so that the element can be studied in a naive cell environment. We expect that an analysis of FP-D will help to define the relationship between copia-like transposable elements and retroviruses. (1) Potter, K.N. and Miller, L.K.
