        A toxicogenomics study of two chemicals in coffee roasting process

        Background Roasted coffee beans are broadly consumed in nearly all classes of the population. 2,3-Butanedione is a food flavor that causes obstructive bronchiolitis in microwave popcorn manufacturing workers. It can be naturally produced when coffee is roasted. Objective To determine effects of 2,3-butanedione (diacetyl) and 2,3-pentanedione on workers’ health during the coffee roasting process, we performed a toxicogenomics study for differentially expressed genes in lung cells after exposure to the two chemicals. Results Gene selection was performed by clustering, gene ontology/pathway, and data mining using microarray data. Target microRNAs and genes were selected based on the expression of microRNAs and correlation analysis of genes and microRNAs. As a result of expression analysis of target gene miRNAs affected by the two chemicals, it was evaluated that Fosl1, Rb1, Aspn, Dusp1, Rnf19b, Jun, and Hbegf were over-regulated by targeting down-regulated miRNAs mutated by two chemicals. Using OMIM database and KEGG pathway, we found that Terc and Bmpr2 were two changed genes by matching with the KEGG disease pathway database. Conclusion Dosage, duration, exposure, and extrapolations are necessary to reflect effects of diacetyl and 2,3-pentanedione on workers in the coffee roasting process. This study focuses on early biochemical changes, mechanisms, and early biomarker discovery when normal lung cells are exposed to these two chemicals.

        제주도 지하수 질산염 농도의 시·공간적 변화 특성: 장기(1993-2015) 모니터링 자료의 평가

        1993년부터 2015년까지 관측된 제주도 지하수 장기모니터링 관측정(N = 4,835)에서 수집된 지하수 수질자료(N = 21,568)를 기반으로 질산성질소의 시공간적 변동 특성을 평가하였다. 제주도 지하수의 질산성질소 농도의 중앙값은 2.5 mg/L로서 다른 국가나 대륙의 조사 결과에 비해 다소 높거나 유사한 것으로 나타났다. 또한 지하수 용도, 행정구역 및 고도 별로 유의한 차이를 보였다. 특히, 산간 지역에 비해 저지대 해안가에 위치한 농업 및 주거지역에서 농도가 높음을 확인하였다. Mann-Kendall 및 Sen’s slope 분석을 활용한 질산성질소 농도의 추세 분석 결과, 하류 저지 대에 비해 중산간지역에서의 질산성질소 농도 증가 경향이 뚜렷하였다. 제주도 내 토지 피복의 시계열 변화 특성과 결부 지어 보면, 중산간지역의 오염 증가 추세는 농업지역의 확장 등 인위적 활동 증가에 기인한 결과로 판단된다. 반면,기지정된 지하수자원특별관리구역에서는 전반적으로 질산성질소 농도의 감소 경향이 나타났는데, 이는 지하수 관리 측면에서 수질관리를 위한 적극적인 정책이 유효함을 시사한다. 본 연구에서는 제주도 지하수의 질산성질소 오염관리를 위한 적정 방안을 제안한다. The spatio-temporal variations of nitrate concentrations in groundwater of Jeju Island were evaluated by an analysis of time series groundwater quality data (N = 21,568) that were collected from regional groundwater monitoring (number of wells = 4,835) for up to 20 years between 1993 and 2015. The median concentration of NO 3 -N is 2.5 mg/L, which is slightly higher than those reported from regional surveys in other countries. Nitrate concentrations of groundwater in wells tend to significantly vary according to different water usage (of the well), administrative districts, and topographic elevations: nitrate level is higher in low-lying agricultural and residential areas than those in high mountainous areas. The Mann-Kendall trend test and Sen’s slope analysis show that nitrate concentration in mid-mountainous areas tends to increase, possibly due to the expansion of agricultural areas toward highland. On the other hand, nitrate concentrations in the Specially Designated Groundwater Quality Protection Zones show the temporally decreasing trend, which implies the efficiency of groundwater management actions in Jeju. Proper measures for sustainable groundwater quality management are suggested in this study.

        내부 응력이 .06 taper Profile의 피로 파절에 미치는 영향

        Objectives: The purpose of this study was to evaluatie the reation between intentionally induced internal stress and cyclic fatigue failure of .06 taper ProFile. Materials and Methods: Length 25 mm, .06 taper ProFile (Dentsply Maillefer), and size 20, 25, 30, 35, and 40 were used in this study. to give the internal stress, the rotary NiTi files were put into the .02 taper, Endo-Training Bloc (Dentsply Maillefer) until auto-stop by torque controlled motor. Rotary NiTi files were grouped by the number of induced internal stress and randomly distributed among one control group and three experimental groups (n = 10, Stress 0 [control], Stress 1, Stress 2 and Stress 3). For cyclic fatigue measurement, time for separation of the rotary NiTi files was recorded. the fractured surfaces were observed by field emission scanning electron microscope (FE-SEM, SU-70, Hitachi). The tiem for separation was statistically analyzed using two-way ANOVA and post-hoc Scheffe test at 95% level. Results: In .06 taper ProFile size 20, 25, 30, 35, and 40, there were statistically significant difference on time for separation between control group and the other groups (p < 0.05). Conclustion: In the limitation of this study, cyclic fatigue failure of .06 taper ProFile is influenced by internal stress accumulated in the files. (Restor Dent Endod 2012;37(2):79-83)

        Performance Evaluation of end mills for high speed cutting has been performed in a view of dynamic characteristics and noise-vibration under operation. The tools tested in this research consist of three foreign country made and one korean made. In addition, numerical models using finite element method are established, which are confirmed by experimental results. The evaluation results has been feedback for developing high performance end mills for high speed cutting tools.

        In the tandem pencil slat saw lines, feeding of cedar blocks is ofter stopped because excessive motor current is required in a saw motor. These events are called "kick-offs" in technical reports. Kick-offs decrease productivity due to machine down-time and damage to saw blades often accompanies them. Research on saw behavior at kickoff is required to understand and reduce the frequency and severity of kickoff events. This research aims at understanding the fundamental mechanisms of kickoffs during cutting, predicting the impending kickoff and evolving design improvements for high cutting performance with fewer and less severe kickoffs.

      • 림프구에 대한 clozapine의 생체내 유전독성

        최근 난치성 정신분열증 환자에서 가장 효과적인 약으로 알려진 clozapine은 골수에 작용하여 심각한 부작용인 무과립 구증을 일으킬 수 있다. 무과립구증의 원인 및 기전에 대한 연구는 매우 많으나 현재까지 확실한 결론이 나지 않은 상태이다. 저자들은 clozapine의 이러한 골수 독성에 근거하여 clozapine이 인체내에서 염색체에 어떤 변화를 주는지 아닌지를 알아보기 위해 clozapine 단독치료를 시행한 환자에게서 치료전과 치료중에 얻어진 말초혈액 림프구를 배양하여 염색체에 어떤 변화가 생기는지를 조사하였으나, clozapine에 의한 염색체의 자발적인 변이는 관찰되지 않았다. Objective : Many drugs and environmental chemicals can induce aneuploidy or chromosome instability in vivo or in vitro. Modification of the fidelity of cellular division may occur via a variety of mechanisms and chemical interactions. The clozapine is new antipsychotic drug with superior effect than classic antipsychotics. But, it may produce agranulocytosis and seizure in some patients. Agranulocytosis is a serious side effect of this drug but the cause and mechanism of this side effect is not clearly explained. Some study suggested that oxidative stress can induce chromosome aberration and the other study suggested that clozapine produce free radical and it can induce oxidative stress on some kind of cells. Therefore we decided to investigate whether clozapine and its metabolites can induce genotoxic effect in vivo. Method : The patients were chronic schizophrenic patients without a treatment at least one year. Diagnosis was made by DSM-IV criteria and agreed by both of two psychiatrists. Blood sampling was done before the treatment and during the treatment with considerable time interval. We used peripheral blood lymphocyte culture method and we counted the numerical aberrations, sister chromatid exchanges. Results : Chromosome aberration was not detected in all samples. Conclusion : Clozapine did not induce spontaneous chromosome aberration to human lymphocytes in vivo.
