      • 염색체 위치 특이적 삽입과 안정적인 유전자 발현을 유도하는 플라스미드 백터의 제작

        Insertion of reporter constructs into the mammalian genome leads to variable gene expression due to position effects at the site of integration. This random integration has limited the gene therapy of human genetic disorders by its undesirable effects. We report here the newly constructed plasmid vector(pIRES-neo-YJ) based on the concepts of homologous recombination and position-independent promoter enhancing of beta-globin matrix attachment region(Glb-MAR). chromosome 7 centromere-specific alpha satellite(alphoid) DNA sequence was cloned into pIRES-neo-YJ for homologous recombination of the cloned gene with the centromeric region of chromosome 7, which is genetically silent. Beta Glb-MAR sequence that allows high levels of transcription independent of the chromosomal site of integration was also inserted into pIRES-neo-YJ to ensure the stable and higher expression of the cloned genes. We expect that pIRES-neo-YJ would provide a valuable tool to eliminate random integration of cloned genes into the undesirable chromosomal region and their short-lived expression which often encounters during the construction of transgenic animals and human gene therapy.

      • 부산시 공장지역·주거지역 하천의 수질특성분석

        This study is carried out to investigate the water quality changes in Busan Metropolitan streams. The results are as follows. In Busan Metropolitan streams, the loading rates of contaminants are greater in industrial waste water streams than domestic waste water streams. Some sampling points show a little high loading in autumn, but this needs more investigation to make clear the causes. The level of organic contaminants(BOD_(5) and COD_(MN)) is found to be low and does not show seasonal variation in domestic waste water streams. But, the level of organic contaminants in industrial waste water streams is relatively high and seems to be seasonally variable, which is affected by other factors. The nutrient materials, such as nitrogen(as T-N, about 20mg/L) and phosphorus(as T-P, about 2.0mg/L), are abundant than Nak Dong River and the general trends of contaminants level are similar to those of organic contaminants.

        In this paper, the T-junction branch network of the orthomode transducer is analyzed. The T-junction blanch network consists of the cylindrical common waveguide and the rectangular sub-waveguide. Using the vector potential of each region, the hybrid mode is considered. Based on the derived electromagnetic components, scattering parameters of the T-junction branch network are calculated. The cylindrical waveguide size and the rectangular waveguide size are WR62 and WC55. In order to analyze the discontinuity of the T-junction branch network, high order mode of the branch region is considered the 15th. From the results, the transmission coefficient. S31 and S21 show -3dB, the reflection coefficient, S11 and S33 are -10dB, -8dB in the range of 13.3GMz∼16GHz.

      • 디-젤 批塵用 移動벨트型 電氣集塵裝置의 小型化에 관한 硏究

        A Moving-Belt Type Electrostatic Precipitator (MBEP) proposed by one of authers has been improved by introducing systematic control concept for the diesel soot particulates from diesel engine exhaust. The proto type MBEP was designed without helping of any precharger or precoagulator of the soot particles, and so it is too large for equipping to the vehicles. The improved one is designed as a systematic control device consisted of a precharger and a MBEP. This systematization makes it possible miniaturizing the size of MBEP without any drop of its collection performance by enhancing the corona field intensity and current density in the collection space concurrently obtained by reducing the interelectrode spacing of corona electrode to collection electrode. As a result, the collection efficiency of present MBEP system, which is reduced by ca. 1/2 in volume to the proto type MBEP, amounts to 82% at the gas velocity of 1.0m/s (this nearly equals to the gas exhaust of a 2000 cc diesel powered vehicle) and gas temperature of ca. 50℃ for the simulated soot particles ranged in size from 0.005μm (mass median diam is 1.0 μm). The collection efficiency of the improved MBEP system of 82% is higher enough to the US EPA regulation limit of 70% of the year of 1985 for the diesel powered light-duty trucks and passenger cars.

      • 우리나라 中部 農村地域住民 死亡樣相에 關한 調査硏究

        For the purpose of analizings the causes and the trends of deaths, and to know the actual states of death certification, we analyzed the records of total 10,415 deaths from 15 Myons of Chung cheong Province during past 10 years, from the January 1, 1975 to the December 31, 1984. Followings are the conclusions therfrom: 1. Of the total deaths, the rate of notification of death with medical certificate was 16.0%, and that with other certificate occupied 76.0%. The rate of notification of death with medical certificate was the highest in the plain area, but that with other's certificate was the highest in the seaside area. 2. 90.6% of deaths in hospital of clinic were reported with medical certificate, but 82.1% of home death was reported with other certificate. The rate of that reported with Medical Certificate become gradual increased in incidence with years. 3. 82.0% of total deaths were reported within one month, the rate become gradual increased in incidence with years. 4. Of the total deaths, below 10 years of age group occupied 8.1%, in the twenties the rates were 5 to 6%. After the forties the rate was increased by ageing, that is 10.2% in the forties, 18.5% to 13.1% in the fifties, 18.5% in the sixties and 32.5% in after the seventies age group, respectively. 5. The proportional mortality indicator was below 60 in time during 1975-1979, 62-63 in 1980-1982, but above 70 in 1983 and 1984. 6. The proportion of hospital or clinical deaths was slightly increased with 3.3% in 1975, 5.0% in 1980 and 6.6% in 1984. 7. Sex ratio of the deaths was higher in male children up to the sixties. Home deaths occupied 90.1% and hospital or clinic deaths occupied only 5.6%. 8. The proportion of deaths was higher, but lower in Summer in seasonal difference. The rate of below thirties was high in Spring and Summer, but the rate of over fifties was high in Autumn and Winter. 9. In the past 10 years from 1975 to 1984, the main causes of death were of Unknown cause, Disease of circulatory system, Symptoms·signs and ill-difind conditions, Neoplasms, and Injuly and poisoning in that order. But those that were of Unknown cause, Disease of circulatory system, Symptoms·signs and ill-defined conditions, Neoplasms, Injury and poisoning, Disease of digestive system, Infection and parasitic disease in that order in 1975-1979, and Unknown cause, Disease of circulatory systems, Symptoms·signs and ill-defined conditions, Injury and poisoning, Neoplasms, Disease of circulatory system and Infection and parasitic disease, in that order in the year 1980-1984. 10. The rate that we could have analized the cause of death was 87.1% only by the Medical Certificate, but the rate of that was 55.3% by the Medical Certificate and Neighbour's Certificate. 11. The cause of death in male were Unknown and others, Disease of the circulatory system, Symptoms·signs and ill-defined conditions, Injury and poisoning, in that order, but those in female were Unknown and others, symptoms·signs and ill-difined conditions, Disease of circulatory system, Neoplasms, Injury and poisoning, in that order. 12. For age specific causes of death, the highest rate was Unknown and others in all age group, without that the main causes were Disease of respiratory system, Infection and parasites, Injury and poisoning, Disease of nervous system and sense organ in the below ten age group; Injury and poisoning, Infection and parasites, Neoplasms, and Disease of the respiratory system in tens and thirties age group; Disease of the respiratory system, Disease of the digestive system, and Infection and poisoning fifties group, but Disease of the respiratory system, Symptoms·signs and ill-defined conditions, Neoplasms, and Disease of the digestive system in after sixties age group, respectively. 13. The frequent causes of death were different by the season, mainly, Unknown and others, Disease of the circulatory system, Symptoms·signs and ill-defined conditions, Injury and poisoning, and Neoplasems in that order in Autumn; but Unknown and others, Disease of the circulatory systems, Symptoms·sign and ill-defined conditions, Neoplasms and Injury and poisoning in that order in other seasons.
