호알칼리성 Bacillus sp.F204와 Bacillus sp.K17의 원형질체 융합

To develop cellulase and xylanase-producing strain by protoplast fusion, alkalophilic Bacillus sp. F204 and K17 were treated with NTG(N-methyl-N'-nitro-N-nitrosoguanidine) and isolated antibiotics resistant strains of S20 (Km', Cm') and G70(Str'). The frequency of protoplast formation was about 95% when cells of mid-log phase were treated with 200㎍/㎖ lysozyme at 37℃ for 30-45 minutes. Under addition of 0.4-0.5M sodium succinate, 0.5% casamino acid, 1.5% polyvinylpyrrolidone, 25mM MgCl_2 and 50mM CaCl_2 to the regeneration medium, the regeneration frequency of Bacillus sp. F204 and K17 was 24.9% and 26.2%, respectively. the fusion frequency was 6.6×10^-6 in the presence of 30% polyethylene glycol 6000 containing 50mM Ca^++ at 45℃ for 5 minutes. Cellulase complex and xylanase activities of fusant were compared with parental strains.

For the purpose of obtaining microorganisms which produced an extracellular pepsin inhibitor, screening was carried out. one strain of Actinomycetes(GF 155-2) isolated from soil samples showed a high inhibitory activity against porcine pepsin. The morphological, physiological and cultural characteristics of the strain GF 155-2 on various culture media were studied according to ISP methods and Bergey's manual of determinative bacteriology (8th ed). this actinomycetes GF 155-2 was found to be similar to the genus Microtetraspora.
Equisetum arvense L.이 고혈당 쥐의 혈중 포도당 농도에 미치는 영향

The effect of methanol extract of Equisetum arvense L.(EM) on blood glucose level in streptozotocin-induced diabetic rat and acute toxicity of EM were investigated. Male Sprague-Dawley rats(200∼250g), with free access to commercial rat chow and water, were divided into four groups: Group 1 served as control; Group 2 received STZ only: Group 3 received EM only; Group 4 received pretreatment with EM followed by STZ administration. EM did not affect the normal blood glucose level and LD50 was about 6,250㎎/㎏. EM pretreatment in STZ-induced diabetic rats slightly inhibited Weight loss and water consumption. EM pretreatment in STZ-induced diabetic rats slightly inhibited the rise of serum glucose concentration. Serum transaminase(AST, ALT) and sorbitol dehydrogenase(SDH) were slightly increased by STZ induced changes in the serum glucose level.

The present study was undertaken in order to elucidate the effects of pretreatment with Equisetum arvense L.(EM) on change in serum lipid compositions and pancreatic enzymes induced by streptozotocin(STZ) in rats with pancreatic histopathology. 1. Total serum lipid and triglyceride levels in the STZ-induced diabetic rats were significantly higher than those in the control group, and those in the group pretreated with EM but the phopholipid serum level was not significantly different in any of the group. 2. In the STZ-induced diabetic group, the total serum cholesterol, VLDL-and LDL-cholesterol levels, and the atheroscerotic index were higher, while the HDL-cholesterol level was lower when compared to the control group. HOwever, these changes were slightly prevented by EM. 3. Pancreatic lipase and trypsin activities were increased, but amylase activity was decreased by STZ, and pretreatment with EM slightly prevented these STZ-induced changes, in the histological changes of the pancreatic β-cell. These results suggest that EM pretreatment slightly supresses STZ induced changes in the serum lipid compositions and pancreatic enzyme via maintenance of prevent of β-cell destruction by STZ.
EDAP LT-01+ 체외충격파쇄석기를 이용한 요로결석 치험 1,816례 : A Report of 1,816 Cases

Between February 8. 1990 and December 31. 1994, 1.816 renal units in 1.611 patients with urinary stones were treated with extracorporeal shock wave lithotripsy(ESWL) using the EDAP LT-01+ lithotriptor. The patients' mean age was 44.3 years with the range of 16 months to 92 years. Sex ratio was 1.39:1 with 936(58.2%) male and 675(41.8%) female patients. Locations of the stones were kidney in 746(41.1%) including 39 cases of staghorn calculi, ureter in 1061(58.4%) and bladder in 9 cases(0.5%). And mean diameter of the stones was 13mm ranging from 2mm to 80mm. Average number of treatments, average treatment time per session and average total storage per patient were 4.0 sessions, 37.6 minutes and 134.4. respectively. Of the 1,816 cases. 141 cases(7.8%) complained of severe pain requiring intravenous or intramuscular analgesics. Success rates by stone size were 98.0%(1,069 cases/1,091 cases, storage/treatment number: 74.7/2.7) in less than 10㎜, 94.4%(453 cases/480 cases, 146.1/4.5) in 10㎜ to 19㎜. 73.4%(116 cases/158 cases, 242.4/7.1) in 20㎜ to 29㎜ and 60.9%(53 cases/87 cases. 402.6/11.2) in more than 30㎜. Success rates by stone location were 66.7%(6/9) in bladder stones. 66.7%(26/39) in staghorn calculi. 77.9%(60/77) in ureteropelvic junction(UPJ) stones. 93.1%(658/707) in renal stones, 94.8%(458/483) in upper ureteral stones and 96.4%(483/501) in lower ureteral stones. Success rate(when patients having stone particles less than 4mm in diameter without causing urinary tract obstrution, were considered to be treated successfully) was 93.l%( 1,691/1,816) and stone-free rates was 88.9% (1,615/1.816). Additional measures used were double-J stenting in 91 cases, percutaneous nephrostomy in 42 cases and push-up in 39 cases. Post-ESWL complications were persistent gross hematuria lasting more than three days in 29 cases, high fever(>38℃) in 38 cases and steinstrasse in 109 cases. We conclude that extracorporeal shock wave lithotripsy with EDAP LT-01 + lithotriptor is an effective treatment modality for urinary stone less than 2.0㎝ in diameter even though retreatment rate is high.

Protein kinase C (PKC) is a multigene family of at least nine serine/threonine kinases that are central to many signal transduction pathways. And the expression of PKC gene is strictly con-trolled by developmental stage. Mutation of the W (c kit) gene, which encodes a transmembrane tyrosine kinase receptor, affect the development and differentiation stem cells. Most homozygous W mutant mice are sterile, due to a lack of germ cells arising during embryonic development, but one of notable exception is W */Wh mice, which are fully fertile in both sexes. In order to elucidate the biological functions of PKC S and c-kit in the testes, we have examined the expression of PKC b mRNA and c-kit mRNA in While mouse testes by means of Northern blotting. North-em blot results was demonstrated that only PKC 8, 4, 8 mRNA can be detected in normal mouse testes. PKC, 8 mRNA was weakly expressed in 7 and 8-week-old mice and highly ex-pressed by 12 weeks. The PKC 8 mRNA and the c-kit mRNA was detectable in the testes of Whl Wh mouse. These results indicated that the function of the PKC a gene is necessary for the development and the W (c-kit) gene is important for the transmission of signal in testes.
