      • Reduced Telomere Length in Colorectal Carcinomas

        Purpose: Telomeres play a key role in the maintenance of chromosome integrity and stability, and telomere shortening is involved in initiation and progression of malignancies. The aim of this study was to determine whether telomere length is associated with the colorectal carcinoma. Patients and methods: A total of 148 colorectal cancer (CRC) samples and corresponding adjacent non-cancerous tissues were evaluated for telomere length, P53 mutation, and cyclooxygenase-2 (COX-2) mutation detected by fluorescent immunohistochemistry. Telomere length was estimated by real-time PCR. Samples with a T/S>1.0 have an average telomere length greater than that of the standard DNA; samples with a T/S<1.0 have an average telomere length shorter than that of the standard DNA. Results: Telomeres were shorter in CRCs than in adjacent tissues, regardless of tumor stage and grade, site, or genetic alterations (P=0.004). Telomere length in CRCs also had differences with COX-2 status (P=0.004), but did not differ with P53 status (P=0.101), tumor progression (P=0.244), gender (P=0.542), and metastasis (P=0.488). There was no clear trend between T/S optimal cut-off values (<1 or > 1) and colorectal tumor progression, metastasis, gender, P53 and COX-2 status. Conclusion: These findings suggesting that telomere shortening is associated with colorectal carcinogenesis but does not differ with tumor progression, gender, and metastasis.

        Hydraulic pumps are used to control the landing wheels of aircrafts, and their proper operation is vital to plane safety. It is well known that adhesive wear failure is a major cause of pump failure. A dual nitrogen plasma immersion ion implantation process calling for the implantation of nitrogen at two different energies and doses has been developed to enhance the surface properties of the disks in the pumps. The procedures meet the strict temperature requirement of <200℃, and after the treatment, the working lifetime of the pumps increases by more than a factor of two. This experimental protocol has been adopted by the hydraulic pump factory as a standard manufacturing procedure.

        Neural Network Based Visual Servo Control Under the Condition of Heavy Loading

        This paper proposes an image-based visual servo (IBVS) control system for hoist positioning under the condition of heavy loading. Various advanced visual servo methods for positioning have been proposed. However, the visual servo scheme under heavy load condition remains the following problems: 1) The deformation and oscillation of the end-effector under the condition of heavy loading will lead to coordinate deviation of end-effector and visual frame, which makes the system unstable and brings more errors. 2) The vibration velocity and offset caused by deformation and oscillation are difficult to measure and model. To address these concerns, we first model the system error based on visual error and differentiate it. We obtain that the basic vector of the system error derivative contains the vector of vibration velocity, then we use the basic vector of derivative of the system error to build up the relationship between system error and vibration velocity. Moreover, we construct a double parallel feed forward neural network to estimate the basic vectors which contain vibration velocity and offset, and compensate for them. Also, we give the proof of the boundedness of the parameters based on Lyapunov theory. Simulation results depict the superior performance of the proposed method against the other advanced visual servo methods under the condition of heavy loading. Furthermore, experiments are conducted on the hoisting platform in our laboratory, which verifies the effectiveness of the proposed method.

        Silence of LncRNA GAS5 Protects Cardiomyocytes H9c2 against Hypoxic Injury via Sponging miR-142-5p

        The regulatory role of long noncoding RNA (lncRNA) growth arrest-specific transcript 5 (GAS5) in both cancerous and noncancerous cells have been widely reported. This study aimed to evaluate the role of lncRNA GAS5 in heart failure caused by myocardial infarction. We reported that silence of lncRNA GAS5 attenuated hypoxia-triggered cell death, as cell viability was increased and apoptosis rate was decreased. This phenomenon was coupled with the down-regulated expression of p53, Bax and cleaved caspase-3, as well as the up-regulated expression of CyclinD1, CDK4 and Bcl-2. At the meantime, the expression of four heart failure-related miR-NAs was altered when lncRNA GAS5 was silenced (miR-21 and miR-142-5p were up-regulated; miR-30b and miR-93 were down-regulated). RNA immunoprecipitation assay results showed that lncRNA GAS5 worked as a molecular sponge for miR-142-5p. More interestingly, the protective actions of lncRNA GAS5 silence on hypoxia-stimulated cells were attenuated by miR-142-5p suppression. Besides, TP53INP1 was a target gene for miR-142-5p. Silence of lncRNA GAS5 promoted the activation of PI3K/AKT and MEK/ERK signaling pathways in a miR-142-5p-dependent manner. Collectively, this study demonstrated that silence of lncRNA GAS5 protected H9c2 cells against hypoxia-induced injury possibly via sponging miR-142-5p, functionally releasing TP53INP1 mRNA transcripts that are normally targeted by miR-142-5p.

        Dimer-type heterostructure of Ag/Zn0.995Ni0.005O microspheres: Microwave-assisted synthesis and synergistic antibacterial activity

        The dimer-type heterostructured Ag/Zn0.995Ni0.005O microspheres were prepared via two-stepmicrowave-assisted process, which exhibited stronger visible-light-activated antibacterial performancethan visible-light only, ZnO/Vis, Zn0.995Ni0.005O/Vis, or AgNPs/Vis. It is attributed to not only theinteraction of released metal ions with functional groups of vital enzymes and proteins, but also thecollapsing force of reactive oxygen species to the cells. Photoelectrochemical measurements show thatthe photogenerated charges of Ag/Zn0.995Ni0.005O possess longer lifetime and higher separation resultingfrom the synergistic effect between Ni and Ag, which leads to its superior visible-light-drivenantibacterial performance. Gram-negative bacteria are more effectively restrained by Ag/Zn0.995Ni0.005Othan Gram-positive bacteria because of structural difference between their cell walls.

      • hARIP2 is a Putative Growth-promoting Factor Involved in Human Colon Tumorigenesis

        Activin is a multifunctional growth and differentiation factor of the growth factor-beta (TGF-${\beta}$) superfamily, which inhibits the proliferation of colon cancer cells. It induces phosphorylation of intracellular signaling molecules (Smads) by interacting with its type I and type II receptors. Previous studies showed that human activin receptor-interacting protein 2 (hARIP2) can reduce activin signaling by interacting with activin type II receptors; however, the activity of hARIP2 in colon cancer has yet to be detailed. In vitro, overexpression of hARIP2 reduced activin-induced transcriptional activity and enhanced cell proliferation and colony formation in human colon cancer HCT8 cells and SW620 cells. Also, hARIP2 promoted colon cancer cell apoptosis, suggesting that a vital role in the initial stage of colon carcinogenesis. In vivo, immunohistochemistry revealed that hARIP2 was expressed more frequently and much more intensely in malignant colon tissues than in controls. These results indicate that hARIP2 is involved in human colon tumorigenesis and could be a predictive maker for colon carcinoma aggressiveness.

        A hemoglobin-based oxygen-carrying biomimetic nanosystem for enhanced chemo-phototherapy and hypoxia alleviation of hepatocellular carcinoma

        Tumor microenvironment is characterized by low pH, high reactive oxygen species and hypoxia, whichprovides a suitable environment for cancer growth. The hypoxia not only elevates tumor angiogenesisand metastasis, but also is responsible for the development of treatment resistance, which graduallybecomes a significant impediment for cancer therapy. Therefore, we developed a biomimetic nanosystemcontaining hemoglobin extracted from red blood cells, chemotherapy drug sorafenib, sensitizer ursolicacid and photosensitizer indocyanine green for enhanced chemo-photo combination therapy of hepatocellularcarcinoma, which could not only enhance the chemotherapy effect of sorafenib bowing to thesensitizing effect of ursolic acid, but also achieved synergetic phototherapy in virtue of indocyaninegreen. Besides, the nanoparticles could effectively delivery exogenous oxygen to tumor site and amelioratethe tumor hypoxic environment with the assistance of hemoglobin. The dual-sensitization drugdelivery system was expected to effectively reduce the resistance of traditional treatment methodsagainst tumor hypoxia, providing a novel prospect for the synergistic hepatocellular carcinomatreatment.

        The purified extract of steamed Panax ginseng protects cardiomyocyte from ischemic injury via caveolin-1 phosphorylation-mediating calcium influx

        Background: Caveolin-1, the scaffolding protein of cholesterol-rich invaginations, plays an important rolein store-operated Ca2þ influx and its phosphorylation at Tyr14 (p-caveolin-1) is vital to mobilize protectionagainst myocardial ischemia (MI) injury. SOCE, comprising STIM1, ORAI1 and TRPC1, contributesto intracellular Ca2þ ([Ca2þ]i) accumulation in cardiomyocytes. The purified extract of steamed Panaxginseng (EPG) attenuated [Ca2þ]i overload against MI injury. Thus, the aim of this study was to investigatethe possibility of EPG affecting p-caveolin-1 to further mediate SOCE/[Ca2þ]i against MI injury in neonatalrat cardiomyocytes and a rat model. Methods: PP2, an inhibitor of p-caveolin-1, was used. Cell viability, [Ca2þ]i concentration were analyzedin cardiomyocytes. In rats, myocardial infarct size, pathological damages, apoptosis and cardiac fibrosiswere evaluated, p-caveolin-1 and STIM1 were detected by immunofluorescence, and the levels ofcaveolin-1, STIM1, ORAI1 and TRPC1 were determined by RT-PCR and Western blot. And, release of LDH,cTnI and BNP was measured. Results: EPG, ginsenosides accounting for 57.96%, suppressed release of LDH, cTnI and BNP, and protectedcardiomyocytes by inhibiting Ca2þ influx. And, EPG significantly relieved myocardial infarct size, cardiacapoptosis, fibrosis, and ultrastructure abnormality. Moreover, EPG negatively regulated SOCE viaincreasing p-caveolin-1 protein, decreasing ORAI1 mRNA and protein levels of ORAI1, TRPC1 and STIM1. More importantly, inhibition of the p-caveolin-1 significantly suppressed all of the above cardioprotectionof EPG. Conclusions: Caveolin-1 phosphorylation is involved in the protective effects of EPG against MI injury viaincreasing p-caveolin-1 to negatively regulate SOCE/[Ca2þ]i.
