        Six new dammarane-type triterpene saponins from Panax ginseng flower buds and their cytotoxicity

        Background: Panax ginseng has been used for a variety of medical purposes in eastern countries formore than two thousand years. From the extensive experiences accumulated in its long medication usehistory and the substantial strong evidence in modern research studies, we know that ginseng hasvarious pharmacological activities, such as antitumor, antidiabetic, antioxidant, and cardiovascularsystemeprotective effects. The active chemical constituents of ginseng, ginsenosides, are rich instructural diversity and exhibit a wide range of biological activities. Methods: Ginsenoside constituents from P. ginseng flower buds were isolated and purified by variouschromatographic methods, and their structures were identified by spectroscopic analysis and comparisonwith the reported data. The 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H- tetrazolium bromidemethod was used to test their cytotoxic effects on three human cancer cell lines. Results: Six ginsenosides, namely 6’emalonyl formyl ginsenoside F1 (1), 3beacetoxyl ginsenoside F1 (2),ginsenoside Rh24 (6), ginsenoside Rh25 (7), 7behydroxyl ginsenoside Rd (8) and ginsenoside Rh26 (10)were isolated and elucidated as new compounds, together with four known compounds (3e5 and 9). Inaddition, the cytotoxicity of these isolated compounds was shown as half inhibitory concentration values,a tentative structureeactivity relationship was also discussed based on the results of our bioassay. Conclusion: The study of chemical constituents was useful for the quality control of P. ginseng flowerbuds. The study on antitumor activities showed that new Compound 1 exhibited moderate cytotoxicactivities against HL-60, MGC80-3 and Hep-G2 with half inhibitory concentration values of 16.74, 29.51and 20.48 mM, respectively.

        Recombinant-attenuated Salmonella Pullorum strain expressing the hemagglutinin-neuraminidase protein of Newcastle disease virus (NDV) protects chickens against NDV and Salmonella Pullorum challenge

        Newcastle disease virus (NDV) and Salmonella Pullorum have significant damaging effects on the poultry industry, but no previous vaccinecan protect poultry effectively. In this study, a recombinant-attenuated S. Pullorum strain secreting the NDV hemagglutinin-neuraminidase(HN) protein, C79-13ΔcrpΔasd (pYA-HN), was constructed by using the suicide plasmid pREasd-mediated bacteria homologousrecombination method to form a new bivalent vaccine candidate against Newcastle disease (ND) and S. Pullorum disease (PD). The effectof this vaccine candidate was compared with those of the NDV LaSota and C79-13ΔcrpΔasd (pYA) strains. The serum hemagglutinationinhibition antibody titers, serum immunoglobulin G (IgG) antibodies, secretory IgA, and stimulation index in lymphocyte proliferation wereincreased significantly more (p < 0.01) in chickens inoculated with C79-13ΔcrpΔasd (pYA-HN) than with C79-13ΔcrpΔasd (pYA) but werenot significantly increased compared with the chickens immunized with the LaSota live vaccine (p > 0.05). Moreover, the novel strain provides60% and 80% protective efficacy against the NDV virulent strain F48E9 and the S. Pullorum virulent strain C79-13. In summary, in this study,a recombinant-attenuated S. Pullorum strain secreting NDV HN protein was constructed. The generation of the S. Pullorum C79-13ΔcrpΔasd(pYA-HN) strain provides a foundation for the development of an effective living-vector double vaccine against ND and PD.

      • Expression of ERCC1, MSH2 and PARP1 in Non-small Cell Lung Cancer and Prognostic Value in Patients Treated with Platinum-based Chemotherapy

        Purpose: To evaluate the prognostic value of the expression of excision repair cross-complementation group l (ERCC1), MutS protein homolog 2 (MSH2) and poly ADP-ribose polymerase 1 (PARP1) in non-small-cell lung cancer patients receiving platinum-based postoperative adjuvant chemotherapy. Methods: Immunohistochemistry was applied to detect the expression of ERCC1, MSH2 and PARP1 in 111 cases of non-small cell lung cancer paraffin embedded surgical specimens. Through og-rank survival analysis, we evaluated the prognostic value of the ERCC1, MSH2, PARP1 and the related clinicopathological factors. COX regression analysis was used to determine whether ERCC1, MSH2 and PARP1 were independent prognostic factors. Results: In the enrolled 111 non-small cell lung cancer patients, the positive expression rate of ERCC1, MSH2 and RARP1 was 33.3%, 36.9% and 55.9%, respectively. ERCC1 (P<0.001) and PARP1 (P=0.033) were found to be correlated with the survival time while there was no correlation for MSH2 (P=0.298). Patients with both ERCC1 and PARP1 negative cancer had significantly longer survival time than those with ERCC1 (P=0.042) or PARP1 (P=0.027) positive alone. Similalry, the survival time of patients with both ERCC1 and PARP1 positive cancer was shorter than those with ERCC1 (P=0.048) or PARP1 (P=0.01) positive alone. Conclusion: Patients with ERCC1 or PARP1 negative non-small cell lung cancer appear to benefit from platinum-based postoperative adjuvant chemotherapy.

        Improved Fiber Uniformity and Jet Number in Multi-spinneret Electrospinning via Auxiliary Electrode

        Auxiliary-electrode electrospinning process is a novel technique to improve nanofiber production rate Due to theformation of multiple jets. In the present work, dual-spinneret (DSE) and tri-spinneret electrospinning (TSE) with anauxiliary electrode were employed to evaluate the number of jets and fiber uniformity in this electrospinning process. Theresults showed that all the spinnerets in DSE and TSE with an auxiliary electrode could produce 2-12 jets, which could beadjusted by controlling the process parameters such as applied voltage and injection speed. The number of jets initiallyincreased and then decreased with a rise in applied voltage in these two systems. Moreover, the produced nanofibers weresmaller in size and possessed a more uniform fiber diameter per distribution in comparison to those without the auxiliaryelectrode. This work provides a new insight on mass production of nanofibers and designing electrospinning apparatus.

        Buckling Analysis of Columns Ended by Rotation-stiffness Spring Hinges

        Stabilities of compressed columns ended by spring hinges with particular rotation stiffness was a basic theory for more complex structures’ researches and engineering, however investigations on calculation approaches for these columns were rarely reported. This paper focus on buckling analysis on these spring hinge ended columns, and the derivation of the columns’ effective length factor equation were presented. Meanwhile a practical design formula to predicate the effective length factors was proposed and further validated to be highly accurate. Furthermore impacts of ideal-ends column method, which ignoring real ends’ rotation stiffness, were investigated. Finally conditions making ideal-ends column method accurate enough in practice were put forward and the practical condition equations were proposed.

        The Antifungal Mechanism of Electrolyzed Oxidizing Water against Aspergillus flavus

        Fungicidal electrolyzed oxidizing waters (EOW)include neutralized electrolyzed oxidizing water (NEW)and acidic electrolyzed oxidizing water (AcEW). Theseforms were evaluated against Aspergillus flavus conidia(spores) and mycelia. EOW possessed strong antifungalactivities against A. flavus conidia and mycelia due todestruction of the cellular structures of the A. flavusconidium and mycelium. K+ and Mg2+ leakage results fromdamage to the normal cellular functions of A. flavus conidiaand mycelia. Among the physicochemical parameters ofEOW, the available chlorine concentration (ACC) isprimarily responsible for the EOW antifungal activity. A. flavus can be eliminated by use of AcEW and NEW as anew, alternative method of fungal control that is superior tosome current physical methods and to synthetic chemicalfungicides.

        Novel Phage Display-Derived H5N1-Specific scFvs with Potential Use in Rapid Avian Flu Diagnosis

        The highly pathogenic avian influenza A (HPAI) viruses of the H5N1 subtype infect poultry and have also been spreading to humans. Although new antiviral drugs and vaccinations can be effective, rapid detection would be more efficient to control the outbreak of infections. In this study, a phage-display library was applied to select antibody fragments for HPAI strain A/Hubei/1/2010. As a result, three clones were selected and sequenced. A hemagglutinin inhibition assay of the three scFvs revealed that none exhibited hemagglutination inhibition activity towards the H5N1 virus, yet they showed a higher binding affinity for several HPAI H5N1 strains compared with other influenza viruses. An ELISA confirmed that the HA protein was the target of the scFvs, and the results of a protein structure simulation showed that all the selected scFvs bound to the HA2 subunit of the HA protein. In conclusion, the three selected scFVs could be useful for developing a specific detection tool for the surveillance of HPAI epidemic strains.
