In this work, nanocrystalline hexagonal tungsten oxide was prepared by acidic precipitation from sodium tungstate solution. TEM studies of nanopowders showed that the average size of the hexagonal nanoparticles is 30–50 nm. Novel nanocomposites were prepared by embedding a low amount of gold-decorated carbon nanotubes into the hex-WO3 matrix. The addition of MWCNTs lowered the temperature range of sensitivity of hex-WO3 nanocomposites to NO2 hazardous gas. The sensitivity of hex-WO3 with Au-decorated MWCNTs to NO2 is at the temperature range between 25°C and 250°C.

Objective Purpose of the study was to identify the relationship among actual plasmatic levels of steroid hormones and behavioral manifestations in boys with autism and to assess the genetic contribution to these manifestations.Methods 172 boys with autism under 10 years of age and 135 neurotypical boys attended the study. ADI-R and ADOS-2 were used to evaluate the core symptom severities. Problem behavior was assessed using BPI-01 questionnaire. Levels of testosterone, estradiol, dehydroepiandrosterone, dehydroepiandrosterone-sulfate and sex hormone binding globulin (SHBG) were measured in plasma of autistic boys. Three SNPs (in ESR1, SHBG, SRD5A2 genes) and one STR in AR gene (number of CAG repeats in first exon) were assessed. Hormonal levels and number of CAG repeats in AR gene were used for correlation analysis with behavioral measures. Genotype and allelic frequencies were compared among autistic and neurotypical boys.Results We found negative relationship among SHBG levels and restricted, repetitive behaviors (measured by ADOS-2) and positive relationship among actual testosterone levels and frequency of stereotyped behavior (measured by BPI-01).Conclusion Actual levels of SHBG and testosterone are related to severities of restricted and repetitive behaviors in boys with autism. Mechanisms of action of these hormones in brain require further investigation.

Historical and local varieties are an important source of genetic diversity in connection with the wheat baking quality. By analysis of DNA from the coding part of the genome of the Slovak regional wheat (Triticum aestivum L.) variety ‘Trebišovská 76’, differences in the Dy subunit composition coded by locus Glu-1D were found. Comparing the newly found sequence with a generally used 1Dy12 (GenBank accession no. X03041)sequence, existence of a new high-molecular-weight glutenin subunit, which was marked as 1Dy12* (GenBank accession no. EU266533) was confirmed. The molecular weight of probable protein coded by 1Dy12* subunit reaches 72.35kDa comparing to the 1Dy12, which reaches 68.71 kDa. This difference is not likely to distinguish by SDS-PAGE. The easiest way for cereal genotyping according to baking quality is proteomic analysis. More exact and suitable method for genotype identification appears to be marker assisted selection using PCR.

Grapevine rupestris stem pitting-associated virus(GRSPaV) is a worldwide-distributed pathogen ingrapevines with a high genetic variability. Our studyrevealed differences in the complexity of GRSPaVpopulation in a single host. A single-variant GRSPaVinfection was detected from the SK30 grapevine plant. On the contrary, SK704 grapevine was infected bythree different GRSPaV variants. Variant-specific RTPCRdetection protocols have been developed in thiswork to study distribution of the three different variantsin the same plant during the season. This studyshowed their randomized distribution in the infectedSK704 grapevine plant. Comparative analysis of fulllengthgenome sequences of four Slovak GRSPaVisolates determined in this work and 14 database sequencesshowed that population of the virus clusterinto four major phylogenetic lineages. Moreover, ouranalyses suggest that genetic recombination along withpoint mutations could play a significant role in shapingevolutionary history of GRSPaV and contributedto its extant genetic diversification.

Salt effect on the liquid phase equilibrium of the water-furfuryl alcohol-furfural system at 298.15 K was studied. The effects of salt concentration (1, 3 and 10% w/wNaCl) and type of salt (LiCl, KCl and RbCl) were investigated. The solubility and tie-line data of these systems were determined by experimental analysis. The results showed that addition of salt significantly affected the two-phase region of ternary system; the enlargement of two-phase region occurred with an increase of salt concentration in initial aqueous phase and the type of the salt in the following order: LiCl>NaCl>KCl>RbCl. Distribution coefficients and separation factors were also calculated. The reliability of the LLE experimental results was validated by the Othmer-Tobias equation, Hand equation and Bachman equation. NRTL was employed to correlate the results obtained by the modeling with the experimental data. Good correlation result was confirmed by the rmsd values of less than 0.3%.

Aliphatic polycarbonate-based block copolymers have received considerable attention as carriers for targeted drug and gene delivery because of their biocompatibility and biodegradability. However, there is little understanding of their phase behaviour and physicochemical characterization of the particles made from them. Here, we prepared a series of well-defined poly(trimethylene carbonate) (PTMC)- based copolymers with molar masses of 3-9 kg·mol-1 by metal-free ring-opening polymerization using dihydroxy-terminated poly(ethylene oxide) as a macroinitiator. Micellar nanoparticles self-assembled from copolymers had a size of less than 130 nm. They were degraded by the action of a model lipase from Mucor Miehei at 37 °C, which is of high importance for biodegradability in the living organism. X-ray diffraction and differential scanning calorimetry proved that amorphous copolymers with more than 39 mol% of carbonate units and representative particles were prone to the rearrangement of PTMC chains during storage and to thus undergo post-crystallization. Our findings can contribute to the comprehensive characterization of polycarbonate biomaterials for medical applications.

The complete genome sequence of a Slovak SL-1 isolate of Tomato mosaic virus (ToMV) was determined from the next generation sequencing (NGS) data, further confirming a limited sequence divergence in this tobamovirus species. Tomato genotypes Monalbo, Mobaci and Moperou, respectively carrying the susceptible tm-2 allele or the Tm-1 and Tm-2 resistant alleles, were tested for their susceptibility to ToMV SL- 1. Although the three tomato genotypes accumulated ToMV SL-1 to similar amounts as judged by semiquantitative DAS-ELISA, they showed variations in the rate of infection and symptomatology. Possible differences in the intra-isolate variability and polymorphism between viral populations propagating in these tomato genotypes were evaluated by analysis of the capsid protein (CP) encoding region. Irrespective of genotype infected, the intra-isolate haplotype structure showed the presence of the same highly dominant CP sequence and the low level of population diversity (0.08-0.19%). Our results suggest that ToMV CP encoding sequence is relatively stable in the viral population during its replication in vivo and provides further demonstration that RNA viruses may show high sequence stability, probably as a result of purifying selection.
The efficiency of different types of extenders for semen cooling in stallions

Objective: This study was conducted to examine influence of skimmed milk-based extender (SM), INRA 96 extender and BotuSemen Gold extender on parameters of stallions' ejaculate during storage. Methods: In this study, 14 stallions between 4 and 20 years of age were monitored. Total and progressive motility, viability and morphology of sperm were evaluated at time intervals of 24, 48, and 72 hours after collection. Results: The total motility, progressive motility, and values of sperm with normal morphology were significantly higher in the INRA 96 and BotuSemen Gold extenders than in the SM (p<0.01). The sperm viability differed significantly in all extenders (p<0.01). The highest value of sperm viability was in INRA 96 (64.69%±0.67%) and lowest in SM (59.70%±0.81%). The highest differences occurred at 72 hours of storage. Values of total motility, progressive motility and sperm viability decreased over time (p<0.01). In case of sperm morphology there was no statistically significant decrease between 48- and 72-hour time intervals. Conclusion: It can be concluded that the extenders with a chemically defined composition have shown better indicators of insemination capabilities in ejaculates than the SM. The BotuSemen Gold extender is a suitable alternative to the INRA 96, when used within 48 hours; after 72 hours of storage, however, the INRA 96 showed a higher share of viable spermatozoa.

How fullerenes inhibit the amyloid fibril formation of hen lysozyme

The inhibition of protein unfolding and self-assembly into fibrils using nanoparticles is a potential medicinalstrategy to impede amyloid-related diseases. Among various nanoparticles, fullerenes have attractedgreat interest because of their inhibition effect on protein fibrillization, but the mechanism of the inhibitionprocess is not properly understood. To explore the inhibition mechanisms, molecular dynamics simulationsof hen egg white lysozyme (HEWL), a model of an amyloid-prone protein, have been performedin the presence of varied C60 concentrations. It is found that the structural fluctuation of HEWL decreasesas the concentration of C60 increases. As the concentration of C60 increases, clustered fullerene aggregatesare formed via hydrophobic interaction rather than interacting with HEWL, and HEWL was adsorbed onthe surface of the fullerene aggregates. The maintenance of the structure and the inhibition of HEWL fibrillizationby C60 is also experimentally confirmed by thioflavin T fluorescence assays and atomic forcemicroscopy. Thus, two independent methods confirmed that C60 is able to inhibit the formation of amyloidfibrils in a dose-dependent manner. This study is an initial step in understanding by which mechanismC60 inhibits HEWL fibrillization, allowing to further design strategies for amyloid inhibition basedon fullerenes and other nanoparticles.
