        Is Human Kallikrein 11 in Non-small Cell Lung Cancer Treated Chemoradiotherapy Associated with Survival?

        Purpose Involvement of human kallikreins (hKs) in human cancers has been reported and several hKs are promising biomarkers of various cancers. The aim of this study was to evaluate the prognostic significance of hK11 expression in patients with non-metastatic non-small cell lung cancer (NSCLC). Materials and Methods The study included 44 patients with NSCLC. hK11 expression was determined by immuno- histochemical staining. Results The estimation of disease-free and overall survival by Kaplan-Meier was 11 months and 17 months, respectively. The estimation of overall survival by Kaplan-Meier was significantly higher in patients with hK11 strongly positive (2+) than in those with hK11 weakly positive (1+) (20 months vs. 11 months, p=0.032). Although not statistically different, the estimation of disease-free survival by Kaplan-Meier was higher in patients with hK11 strongly positive (2+) than in those with hK11 weakly positive (1+) (12 months vs. 9 months, p=0.113). Multivariate Cox regression analysis showed that the overall survival rates were significantly associated with response to chemoradiotherapy and the degree of staining with hK11. Conclusion The stronger hK11 expression in NSCLC appears to be associated with better survival rates. hK11 may be a prognostic biomarker of NSCLC.

      • Fibulin-3 as a Diagnostic Biomarker in Patients with Malignant Mesothelioma

        Background: New tumour biomarkers are being intensely investigated for malignant mesothelioma (MM). Fibulin-3 is produced in MM but its role remains uncertain. The aim of this study was to evaluate the validity of measuring serum fibulin-3 in the diagnosis and prognosis of MM. Materials and Methods: This prospective study was performed on 43 patients and 40 healthy controls who were admitted to our hospital between January 2012 and January 2014. Data from MM patients, including demographic and clinical features, routine laboratory data, levels of serum fibulin-3, and treatment outcomes were defined as potential prognostic factors. The receiver operating characteristic (ROC) curve for fibulin-3 was used to detect the cut-off value with highest sensitivity and specificity. Univariate survival analysis was performed using the Kaplan-Meier method in patients with MM. Afterwards, the possible factors identified with univariate analyses were entered into the cox regression analysis. Results: Our results revealed that patients with MM had significantly higher serum levels of fibulin-3 than controls. The results showed that the best cut-off point was 36.6 ng/ml with an AUC (area under the curve)=0.976, sensitivity=93.0% and specificity=90.0. In our study, the initial significant poor prognostic factors were advanced stage, high white blood cell count, high platelet count, high C-reactive protein (p<0.05 for each variable). Later, according to multivariate analysis the results showed only advanced stage as significant parameter (p=0.040). Conclusions: We determined that real use for serum fibulin-3 was not for prognosis but for diagnosis in MM. Also advanced stage was associated with poor MM prognosis.

        The letter recognition using BCI system

        In this paper, we show how such enhancement of Farwell-Donchin BCI enables a fresh, inexperienced user to achieve quickly an accurate BCI control with a high information transfer rate. This paper presents the results of a BCI experiment where the participant, who had no previous BCI experience, obtained, in about 20 min, a highly reliable and fast control over the BCI spelling device based on the Farwell-Donchin paradigm. Offline analysis showed that the high performance of the BCI was, to a high extent, due to the use of the ERP component N1, in addition to component P300, which has been considered the only ERP component important for the prediction of user's choice in the Farwell-Donchin paradigm in many publications.

      • Production and properties of tooth-colored yttria stabilized zirconia ceramics for dental applications

        <P><B>Abstract</B></P> <P>Dense zirconia stabilized with 3mol% yttria ceramics were produced in disc shape by first cold isostatically pressing at 100MPa and then sintering at 1450°C at ambient laboratory conditions. Coloring was accomplished by immersion the discs in NiCl<SUB>2</SUB>, MoCl<SUB>3</SUB>, and NiCl<SUB>2</SUB> + MoCl<SUB>3</SUB> solutions for 5, 30, and 60s. Different concentrations (0.1, 0.25, and 0.5wt%) were applied to get the color of natural tooth. The density, color, microhardness, fracture toughness, compressive strength, and wear rate of the discs were measured to evaluate the suitability of the colored discs for dental applications. Color assessments were made by measuring CIE Lab <I>L</I> <SUP> <I>*</I> </SUP>, <I>a</I> <SUP> <I>*</I> </SUP>, <I>b</I> <SUP> <I>*</I> </SUP>, and Δ<I>E</I> <SUP> <I>*</I> </SUP> values. Low temperature degradation of the samples was evaluated by aging sensitivity tests in autoclave for 2, 4, and 6h. Results have shown that color produced depends on the kind and concentration of the colorant solution while time of immersion has no significant effect on coloring process. Coloring solutions containing 0.1 and 0.25wt% MoCl<SUB>3</SUB> provided clinically acceptable color with the ΔE* value ranging from 5.16 to 6.42 for dental applications.</P>

        Excitation of Molecular Hydrogen in the Orion Bar PhotodissociationRegion from a Deep Near-infrared IGRINS Spectrum

        <P>We present a deep near-infrared spectrum of the Orion Bar Photodissociation Region (PDR) taken with the Immersion Grating INfrared Spectrometer (IGRINS) on the 2.7 m telescope at the McDonald Observatory. IGRINS has high spectral resolution (R similar to 45,000) and instantaneous broad wavelength coverage (1.45-2.45 mu m), enabling us to detect 87 emission lines from rovibrationally excited molecular hydrogen (H-2) that arise from transitions out of 69 upper rovibration levels of the electronic ground state. These levels cover a large range of rotational and vibrational quantum numbers and excitation energies, making them excellent probes of the excitation mechanisms of H2 and physical conditions within the PDR. The Orion Bar PDR is thought to consist of cooler high density clumps or filaments (T = 50-250 K, n(H) = 10(5)-10(7) cm(-3)) embedded in a warmer lower density medium (T = 250-1000 K, n(H) = 10(4)-10(5) cm(-3)). We fit a grid of constant temperature and density Cloudy models, which recreate the observed H2 level populations well, to constrain the temperature to a range of 600-650. K and the density to n(H) = 2.5 x 10(3) -10(4) cm(-3). The best-fit model gives T = 625 K and n(H) = 5 x 10(3) cm(-3). This well-constrained warm temperature is consistent with kinetic temperatures found by other studies for the Orion Bar's lower density medium. However, the range of densities well fit by the model grid is marginally lower than those reported by other studies. We could be observing lower density gas than the surrounding medium, or perhaps a density-sensitive parameter in our models is not properly estimated.</P>

        Comparison of Cell Lysis Techniques via Q-TOF LC/MS

        Untargeted metabolomics is a useful tool for drug development focusing on novel chemotherapeutic and chemopreventative agents against cancer cells. In recent years, quadrupole time of flight liquid chromatography-mass spectrometry (Q-TOF LC/MS)-based untargeted metabolomic approaches have gained importance to evaluate the effect of these agents at the molecular level. The researchers working on cell culture studies still do not apply standardized methodologies on sample preparation for untargeted metabolomics approaches. In this study, the rough and wet lysis techniques performed on MCF-7 breast cancer cells were compared with each other via the Q-TOF LC/MS-based metabolomic approach. The C18 and hydrophilic interaction liquid chromatography (HILIC) columns were used for the separation of the metabolites in MCF-7 cell lysates. 505 peaks were detected through the HILIC column and 551 peaks were found through the C18 column for the wet lysis technique. This situation supported by the base peak chromatograms showed that the wet lysis technique allowed us to extract higher number of non-polar metabolites. Almost equal number of metabolites was found for the C18 and HILIC columns (697 peaks for the HILIC column and 695 peaks for the C18 column) when the rough lysis technique was used. However, the intensities of polar metabolites were higher for the rough lysis technique on base peak chromatograms for both the HILIC and C18 columns. Although cell lysis technique, which is the first step in the sample preparation for cell culture studies, does not cause dramatic differences in the number of the detected metabolite peaks, it affects the polar and non-polar metabolite ratio significantly. Therefore, it must be considered carefully especially for in vitro drug development studies.

        Evaluation of YouTube Videos on Hepatocellular Carcinoma

        Background: As in every field, online information has become increasingly used in health. However, it is well known that some information online containing health advice is incorrect and may even include false statements. For this reason, it is crucial for public health that individuals reach reliable, high-quality resources when they are trying to obtain health information. Studies have been conducted on the quality and reliability of online information about many diseases, but no similar study has been found in the literature on hepatocellular carcinoma (HCC). Methods: In this descriptive study, videos on YouTube (www.youtube.com) about HCC were evaluated using the Global Quality Scale (GQS) and the modified DISCERN tool. Results: While 129 (89.58%) of the videos examined within the study were considered useful, 15 (10.42%) were misleading. The GQS scores of the videos considered useful were significantly higher than the scores of the misleading videos, with a median (min–max) score of 4 (2–5) (P < 0.001). When we compared the DISCERN scores, the scores of the useful videos were significantly higher (P < 0.001) than the scores of the misleading videos. Conclusion: YouTube should be considered a complex structure where accurate and reliable health information can be presented, as well as erroneous and misleading information. Users should understand the importance of video sources and focus their research on videos from doctors, academics, and universities.
