The MAP Kinase Kinase Gene AbSte7 Regulates Multiple Aspects of Alternaria brassicicola Pathogenesis

        Kai Lu,Min Zhang,Ran Yang,Min Zhang,Qinjun Guo,Kwang-Hyun Baek,Hou-Juan Xu 한국식물병리학회 2019 Plant Pathology Journal Vol.35 No.2

Mitogen-activated protein kinase (MAPK) cascades in fungi are ubiquitously conserved signaling pathways that regulate stress responses, vegetative growth, pathogenicity, and many other developmental processes. Previously, we reported that the AbSte7 gene, which encodes a mitogen-activated protein kinase kinase (MAPKK) in Alternaria brassicicola, plays a central role in pathogenicity against host cabbage plants. In this research, we further characterized the role of AbSte7 in the pathogenicity of this fungus using ΔAbSte7 mutants. Disruption of the AbSte7 gene of A. brassicicola reduced accumulation of metabolites toxic to the host plant in liquid culture media. The ΔAbSte7 mutants could not efficiently detoxify cruciferous phytoalexin brassinin, possibly due to reduced expression of the brassinin hydrolase gene involved in detoxifying brassinin. Disruption of the AbSte7 gene also severely impaired fungal detoxification of reactive oxygen species. AbSte7 gene disruption reduced the enzymatic activity of cell walldegrading enzymes, including cellulase, β-glucosidase, pectin methylesterase, polymethyl-galacturonase, and polygalacturonic acid transeliminase, during host plant infection. Altogether, the data strongly suggest the MAPKK gene AbSte7 plays a pivotal role in A. brassicicola during host infection by regulating multiple steps, and thus increasing pathogenicity and inhibiting host defenses.

The MAP Kinase Kinase Gene AbSte7 Regulates Multiple Aspects of Alternaria brassicicola Pathogenesis

        Lu, Kai,Zhang, Min,Yang, Ran,Zhang, Min,Guo, Qinjun,Baek, Kwang-Hyun,Xu, Houjuan The Korean Society of Plant Pathology 2019 Plant Pathology Journal Vol.35 No.2

In this research, we further characterized the role of AbSte7 in the pathogenicity of this fungus using ${\Delta}AbSte7$ mutants. Disruption of the AbSte7 gene of A. brassicicola reduced accumulation of metabolites toxic to the host plant in liquid culture media. The ${\Delta}AbSte7$ mutants could not efficiently detoxify cruciferous phytoalexin brassinin, possibly due to reduced expression of the brassinin hydrolase gene involved in detoxifying brassinin. Disruption of the AbSte7 gene also severely impaired fungal detoxification of reactive oxygen species. AbSte7 gene disruption reduced the enzymatic activity of cell walldegrading enzymes, including cellulase, ${\beta}$-glucosidase, pectin methylesterase, polymethyl-galacturonase, and polygalacturonic acid transeliminase, during host plant infection. Altogether, the data strongly suggest the MAPKK gene AbSte7 plays a pivotal role in A. brassicicola during host infection by regulating multiple steps, and thus increasing pathogenicity and inhibiting host defenses.

Effects of dietary multi-strain probiotics supplementation in a low fishmeal diet on growth performance, nutrient utilization, proximate composition, immune parameters, and gut microbiota of juvenile olive flounder (Paralichthys olivaceus)

        Niu, Kai-Min,Khosravi, Sanaz,Kothari, Damini,Lee, Woo-Do,Lim, Jeong-Min,Lee, Bong-Joo,Kim, Kang-Woong,Lim, Sang-Gu,Lee, Sang-Min,Kim, Soo-Ki Elsevier 2019 FISH AND SHELLFISH IMMUNOLOGY Vol.93 No.-

<P><B>Abstract</B></P> <P>A 12-week feeding trial was conducted to evaluate the effects of multi-strain probiotics (MSP) in a low fish meal (FM) diet on overall performance, gut microbiota, selected non-specific immune responses and antioxidant enzyme activities of olive flounder (<I>Paralichthys olivaceus</I>) juveniles. A total of 225 healthy olive flounders (initial mean body weight, 13.5 ± 0.01 g) were randomly separated into 3 groups of 75 fish, each group having three replicates of 25 fish; first group was fed with a FM-based control diet (Con), 2nd group was fed with a low-FM diet containing a blend of plant and animal protein meals replacing 30% of the FM protein (FM30), and 3rd group was fed with the FM30 diet supplemented with 108–109 CFU kg−1 of the MSP (Pro). With the exception of lipid retention, which was significantly lower in fish fed the FM30 diet compared to the other two treatments, no other statistically significant differences were recorded with respect to any of the other growth and nutrient utilization parameters. Myeloperoxidase and lysozyme activities of fish fed the Pro diet were much higher and significantly different than those of fish fed the FM30 diet. Glutathione peroxidase activity was significantly higher in Pro- than in Con-fed fish, which, in turn, was significantly higher than FM30-fed fish. Expression of immune-related genes including IL-1β, IL-6, and TNF-α was markedly upregulated in livers of the fish fed Pro diet compared to those fed the Con and FM30 diets. Furthermore, supplementation of MSP in FM30 diet enriched the <I>Lactobacillus</I> abundance in the fish gut as well as predictive gene functions in relation to lipid and carbohydrate metabolisms. These data suggested that the MSP could reduce the potential adverse effects of the low-FM diet and might be used as a healthy immunostimulant for olive flounder.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Low fishmeal diet (FM30) significantly affected the lipid retention of olive flounder. </LI> <LI> The multi-strain probiotics (MSP) improved the activities of myeloperoxidase, lysozyme, and glutathione peroxidase. </LI> <LI> The MSP supplement markedly upregulated the immune-related gene expression of IL-1β, IL-6, and TNF-α in the liver. </LI> <LI> The MSP supplement enriched the predictive gene functions in relation to lipid and carbohydrate metabolisms. </LI> <LI> The MSP could be used as a healthy immunostimulant for olive flounder. </LI> </UL> </P>

      • MiR-133b Acts as a Tumor Suppressor and Negatively Regulates TBPL1 in Colorectal Cancer Cells

Xiang, Kai-Min,Li, Xiao-Rong Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.8

Introduction: MicroRNAs have emerged as post-transcriptional regulators that are critically involved in tumorigenesis. This study was designed to explore the effect of miRNA 133b on the proliferation and expression of TBPL1 in colon cancer cells. Methods: Human colon cancer SW-620 cells and human colon adenocarcinoma HT-29 cells were cultured. MiRNA 133b mimcs, miRNA 133b inhibitors, siRNA for TBPL1 and scrambled control were synthesized and transfected into cells. MiR-133b levels in cells and CRC tumor tissue was measured by real-time PCR. TBPL1 mRNA was detected by RT-PCR. Cell proliferation was studied with MTT assay. Western blotting was applied to detect TBPL1 protein levels. Luciferase assays were conducted using a pGL3-promoter vector cloned with full length of 3'UTR of human TBPL1 or 3'UTR with mutant sequence of miR-133b target site in order to confirm if the putative binding site is responsible for the negative regulation of TBPL1 by miR-133b. Results: Real time PCR results showed that miRNA 133b was lower in CRC tissue than that in adjacent tissue. After miR-133b transfection, its level was elevated till 48h, accompanied by lower proliferation in both SW-620 and HT-29 cells. According to that listed in http://www.targetscan.org, the 3'-UTR of TBPL1 mRNA (NM_004865) contains one putative binding site of miR-133b. This site was confirmed to be responsible for the negative regulation by miR-133b with luciferase assay. Further, Western blotting and immunohistochemistry both indicated a higher TBPL1 protein expression level in CRC tissue. Finally, a siRNA for TBPL1 transfection obviously slowed down the cell proliferation in both SW-620 and HT-29 cells. Conclusion: MiR-133b might act as a tumor suppressor and negatively regulate TBPL1 in CRC.

Developed a high-performance sensor based on cumarin derivative for rapid and sensitive detection of palladium ion in organic wastewater

        Ai-min Ren,Yu Zhang,Weiwei Yu,Kai Zhao,Zhiru Hu,Zhiquan Zhang,Guodong Feng,Zhi-Guang Song 한국공업화학회 2021 Journal of Industrial and Engineering Chemistry Vol.99 No.-

In this article, a series of coumarin derivativefluorescent probes have been synthesized, which can realizesimple, rapid and sensitive detection of Pd2+. We tested thefluorescence properties of threefluorescentprobes, (E)-N0-(((4-chloropyridin-2-yl) methylene)-7-(diethylamino)-2-oxo-2H-chromene-3-carbohy(CMDCC) has the betterfluorescence properties and anti-interference ability. Then, CMDCC as thefluorescence probe for detection of Pd2+ was systematic studied. Under optimized conditions, this probehas a wide pH range, a lower detection limit (4.4510 8 mol L-1), and a faster response speed (3 min). Inthe Pd2+ concentration range of 0.1 5 mmol L-1, thefluorescence intensity of this probe and concentrationof Pd2+ show a good linear response. In this work, Pd2+ content in organic waste liquid was successfullydetected and a standard addition test was also performed. The satisfactory recovery rate was obtained. Moreover, it can be observed with naked eyes that the palladium ion changes the solution color of CMDCCfrom green to red. This color change can easily be compared with other metal salts. It indicates thatCMDCC can also detect Pd2+ by colorimetric method. Therefore, CMDCC should be used to rapidly, conveniently detect Pd2+ in environmental samples.

Electrophysiological and alarm behavioral responses of Solenopsis invicta Buren (Hymenoptera: Formicidae) to alkoxypyrazines

        Yan Sun,Kai-Min Shao,Yongyue Lu,Qun-Hui Shi,Wen-Kai Wang,Li Chen 한국응용곤충학회 2017 Journal of Asia-Pacific Entomology Vol.20 No.2

The red imported fire ant, Solenopsis invicta produces an alarm pheromone component, 2-ethyl-3,6- dimethylpyrazine, and responds to its pyrazine analogs in a similar manner but at varied detection thresholds. Herein, the responses of fire ant workers by electroantennogram (EAG) and behavior were tested with twelve structurally-related oxygen-containing pyrazines (alkoxypyrazines) and the synthetic alarm pheromone. All tested compounds elicited a dose-dependent EAG response, with S. invicta responding greatest to the synthetic alarmpheromone. Chemical structure of pyrazines influenced the EAG response but not always alarmbehavioral response. Among the 13 tested compounds, 7 compounds displayed significantly greater EAG response than 2- isopropyl-3-methoxypyrazine and 2-ethoxy-3-isopropylpyrazine at the dose of 1000 μg. Four of these 7 compounds, 2-ethyl-3,6-dimethylpyrazine, 2-methoxy-3-methylpyrazine, 2-ethoxy-3(5 or 6)-methylpyrazine, and 2-chloro-3-methoxypyrazinewith characteristic substituents on pyrazine ringwere further subjected to bait discovery bioassay.Hotdog bait containing pyrazines attracted significantlymore fire antworkers in the first 15-min period, resulting in quicker recruitment to food block than hexane control. The potential of using alkoxypyrazines in fire ant control is discussed.

Plant Virome Analysis by the Deep Sequencing of Small RNAs of Fritillaria thunbergii var. chekiangensis and the Rapid Identification of Viruses

        Lu-xi Chen,Hang-kai Pan,Yu-tian Tao,Dang Yang,Hui-min Deng,Kai-jie Xu,Wen-bin Chen,Jun-min Li 한국식물병리학회 2022 Plant Pathology Journal Vol.38 No.5

Thunberg fritillary (Fritillaria thunbergii), a perennial used in traditional Chinese herbal medicine, is a members of the family Liliaceae. The degeneration of germplasm is a severe problem in the production of Fritillaria thunbergii var. chekiangensis. However, no information about viral infections of F. thunbergii var. chekiangensis has been reported. In this study, we sequenced the small RNAs of F. thunbergii var. chekiangensis from leaves and bulbs, and viruses were identified using a phylogenetic analysis and BLAST search for sequence. In addition, multiplex reverse transcriptase-polymerase chain reaction (RT-PCR) was used to rapidly detect viruses in this variety. Our study first reported that five viruses infected F. thunbergii var. chekiangensis. Among them, fritillary virus Y (FVY), lily mottle virus (LMoV), Thunberg fritillary mosaic virus (TFMV), and hop yellow virus (HYV) had been reported in F. thunbergii, while apple stem grooving virus was first reported in the genus Fritillaria. A multiplex RT-PCR method was developed to rapidly test the four viruses FVY, LMoV, TFMV, and HYV in F. thunbergii var. chekiangensis. Our results provide a better understanding of the infection of F. thunbergii var. chekiangensis by viruses and a basic reference for the better design of suitable control measures.

      • The Role of Maternal Interpersonal Relation Satisfaction in the Relationship between Confl icted Teacher-Child Relationship and Negative Peer Interaction Quality in Young Children

( Kai Sook Chung ),( Min A Kim ) 대한가정학회 2014 International Journal of Human Ecology Vol.15 No.1

The effects of conflicted teacher-child relation on conflicted or passive peer interaction and a moderation effect of mothers’ interpersonal relation satisfaction on the associations were assessed. Children from 2- to 6-year-olds(184 girls, 185 boys) mostly from middle socioeconomic-status urban community in Korea and their teachers and mothers participated. Conflicted teacher-child relation predicted conflicted peer interaction but not passive peer interaction. Children, whose relationship with teachers were conflicted, engaged in conflicted play with peers more often than children who were in less conflicted relationship with the teachers. Teachers who were in conflicted relationship with the children, perceived the children having conflicted interaction with peers more often, if mothers of the children were less satisfying in relationship with significant others, especially boys. Children, whose mothers are in less satisfying interpersonal relation with others, were more passive in peer interaction than children whose mothers are in more satisfying interpersonal relationship.

Full Endoscopic Removal of Cervical Spinal Epidural Abscess: Case Report and Technical Note

        Kai-Sheng Chang,Li-Wei Sun,Chun-Yuan Cheng,Shang-Wen Chang,Chien-Min Chen 대한척추신경외과학회 2020 Neurospine Vol.17 No.sup.

Spinal epidural abscess is a rare disease that is less likely to occur in the cervical region. When it occurs here, cervical spondylodiscitis can develop. Surgical treatment is recommended because of possible life-threatening septic and neurological complications. We present a case of an 81-year-old man who suffered from right side paralysis and was subsequently diagnosed with a C4 to C7 epidural abscess. We utilized full endoscopic surgery for patient management. The traditional surgical methods for treating cervical epidural abscesses may cause spinal instability. There has only been one previous case report on the endoscopic-assisted method. Minimal invasive surgery by a full endoscopic method can be done with a small incision and is associated with minimal blood loss and muscle damage. This is the first report on cervical epidural abscess drainage utilization a full endoscopic method. We recommend this alternative minimally invasive method to manage cervical epidural abscess.
