Comparision of doxorubicin-induced cardiotoxicity in the ICR mice of different sources

Doxorubicin is a widely used chemotherapeutic agents and is now part of standard therapeutic regimens for a variety of cancers (eg, hematopoietic malignancies and advanced solid tumors of the breast, ovary, thyroid, and bone). However, a potentially lethal and dose-dependent cardiotoxicity that appears within a short time after treatment limits the usage of doxorubicin in cancer patients. Although the mechanism of doxorubicin-induced cardiotoxicity is not completely understood, it is thought that free radical-induced oxidative stress and excessive production of reactive oxygen species are primary drivers of its toxicity. In this study, we compared the doxorubicin-induced cardiotoxicity of ICR mice obtained from three different sources and evaluated the utility of Korl:ICR stock established by the Korean FDA. Because doxorubicin-induced cardiotoxicity is thought to involve the excessive generation of ROS followed by oxidative stress, we determined the representative tissue index of oxidation, lipid peroxidation, and antioxidant, glutathione (GSH), as well as the parameters of heart injury. Doxorubicin treatment successfully induced cardiotoxicity as evidenced by histological examination and serum parameters (eg, levels of LDH and CK activities) in ICR mice. It was accompanied by increased lipid peroxidation and a decrease in both cysteine and GSH, further supporting previous reports that oxidative stress is a potential mechanism of doxorubicin-induced cardiotoxicity. Of interest, we did not observe a significant difference in doxorubicin-induced cardiotoxicity among mice of different origins. Collectively, our results suggest that Korl:ICR strain may be useful in the research of doxorubicin-induced cardiotoxicity.

Non-alcoholic fatty liver disease (NAFLD) is believed to be the most prevalent liver disease worldwide and a major cause of chronic liver injury. It is characterized by lipid accumulation in the absence of significant alcohol consumption and frequently progresses to steatohepatitis, liver fibrosis, and hepatocellular carcinoma. Although many studies have been conducted to better understand NAFLD since it was first recognized, there are still many gaps in knowledge of etiology, prognosis, prevention and treatment. Methionine-choline deficient (MCD) diet, a well-established experimental model of NAFLD in rodents, rapidly and efficiently produces the clinical pathologies including macrovesicular steatosis and leads to disease progression. In this study, we measured the response to MCD diet in C57BL/6N mice obtained from three different sources; Korea NIFDS, USA, and Japan. We evaluated changes in body weight, food consumption, and relative weights of tissues such as liver, kidney, gonadal white adipose tissue, inguinal white adipose tissue, and brown adipose tissue. These basic parameters of mice with an MCD diet were not significantly different among the sources of mice tested. After 3 weeks on an MCD diet, histopathological analyses showed that the MCD diet induced clear fat vacuoles involving most area of the acinus in the liver of all mice. It was accompanied by increased serum activities of alanine aminotransferase and aspartate aminotransferase, and decreased levels of serum triglyceride and cholesterol. In conclusion, the response of C57BL6N mice originating from different sources to the MCD diet showed no significant differences as measured by physiological, biochemical, and histopathological parameters.

Malignant fibrous histiocytoma was introduced in the early 1960s to describe a well known sarcoma of soft tissue, the tumors arise on the extremities or in the retroperitoneum and are located adjacent to deep fascia or within skeletal muscle. Local recurrence and distant metastasis are very common. Intraabdominal Malignant fibrous histiocytoma are very rare and only several cases were reported till now. We report one case of intraabdominal primary malignant fibrous histiocytoma in 21-year old female patient who had growing nontender mass of left lower abdomen. The presenting site was the mesenteric border of the proximal jejunum about 60 cm below the ligament of Treitz.

<P>Endoplasmic reticulum (ER) stress is involved in non-alcoholic fatty liver disease (NAFLD), but the relationship between oxidative stress, another well-known risk factor of NAFLD, and ER stress has yet to be elucidated. In this study, we treated mice with tunicamycin (TM) (2 mg/kg body weight) for 48 h to induce ER stress in the liver and examined the metabolic pathway that synthesizes the endogenous antioxidant, glutathione (GSH). Tunicamycin (TM) treatment significantly increased mRNA levels of CHOP and GRP78, and induced lipid accumulation in the liver. Lipid peroxidation in the liver tissue also increased from TM treatment (CON vs. TM; 3.0 ± 1.8 vs. 11.1 ± 0.8 nmol MDA/g liver, <I>p</I> < 0.001), which reflects an imbalance between the generation of reactive substances and antioxidant capacity. To examine the involvement of GSH synthetic pathway, we determined the metabolomic changes of sulfur amino acids in the liver. TM significantly decreased hepatic S-adenosylmethionine concentration in the methionine cycle. The levels of cysteine in the liver were increased, while taurine concentration was maintained and GSH levels profoundly decreased (CON vs. TM; 8.7 ± 1.5 vs. 5.4 ± 0.9 µmol GSH/g liver, <I>p</I> < 0.001). These results suggest that abnormal cysteine metabolism by TM treatment resulted in a decrease in GSH, followed by an increase in oxidative stress in the liver. In HepG2 cells, decreased GSH levels were examined by TM treatment in a dose dependent manner. Furthermore, pretreatment with TM in HepG2 cells potentiated oxidative cell death, by exacerbating the effects of tert-butyl hydroperoxide. In conclusion, TM-induced ER stress was accompanied by oxidative stress by reducing the GSH synthesis, which made the liver more susceptible to oxidative stress.</P>

목적 : 양성 기형종에서 발견되는 난소 갑상선종의 다양한 임상적, 초음파적, 그리고 병리학적 특성을 살펴보고자 하였다. 연구 방법 : 본 저자들은 삼성제일 병원에서 최근 5년 (1997-2001년) 동안 경험한 33예의 난소갑상선종을 토대로 진단 당시의 연령 및 폐경유무, 비특이적 증상에서부터 갑상선 기능 항진증의 특이 증상을 포함한 다양한 임상양상, 초음파적 그리고 병리학적 특성을 비교 분석 하였다. 결과 : 난소갑상선종 환자의 평균 연령은 40.5세였고, 대부분 가임기 여성이였다. 14예만이 하복부 통증이나 복부종괴 촉지, 불규칙 자궁 출혈 등의 비특이적 증상을 호소하였고, 그 외의 다수 환자들은 정기 초음파 진단을 통해 난소 종양을 의심하게 되었다. 갑상선 조직이 포함된 종양임에도 불구하고 갑상선 기능 항진증을 보인 예는 없었으며, 11예에서 복수를 동반하였고, 3예에서는 CA-125 증가 소견을 보였다. 초음파상 11예에서는 악성 종양으로 진단 받았으나, 대부분의 예에서는 난소기형종으로 진단 받았다. 결론 : 난소 갑상선종은 임상양상, 검사실 소견, 영상 진단 소견 등에 있어 특징적 소견없이 매우 다양한 양상을 보이는 종양으로, 수술 후 조직검사 소견으로만 진단 가능하였다. 대부분의 난소갑상선종은 양성으로, 그 치료는 외과적 적출술만으로 충분한 것으로 사료되나, 악성 난소갑상선종의 치료에 있어서는 더 많은 경험이 축적되어야 할 것이다.
DNA Marker Traceability Techniques for Hanwoo Industry

The effectiveness of genetic markers on Hanwoo traceability systems was applied and evaluated from Korean 33 Hanwoo elite sire families. Five microsatellite markers were selected finally, which were located on chromosomes different chromosomes with the end sequencing of 100 HW-YUBAC that were recorded in the NCBI by Yeungnam University. Eleven major microsatellite markers were selected from allele amplified, their frequencies, H(Heterozygosity) and PIC(Polymorphism information content) with Hardy-Weinberg equilibrium. Next, in order to evaluate the power of the markers selected on the individual animal identification with experimental condition, the match probability(MP) and the relatedness coefficient(R) were computed.
Bootstrapping and DNA marker Mining of BMS941 microsatellite locus in Hanwoo chromosome 17

LOD scores and a permutation test for detecting and locating Quantitative trait loci(QTL) from the Hanwoo economic trait have been described and we selected a considerable major BMS941 locus. K -means clustering analysis of eight markers in BMS941 and four traits resulted in three cluster groups. Finally, we applied the bootstrap test method to calculate confidence intervals for finding major DNA markers. We conclude that the major markers of BMS941 locus in Hanwoo chromosome 17 are markers 85bp and 105bp.
