        A Potent Brucella abortus 2308 Δery Live Vaccine Allows for the Differentiation between Natural and Vaccinated Infection

        Brucellosis is a globally distributed zoonotic disease that causes animal and human diseases. However, the current Brucella abortus vaccines (S19 and RB51) are deficient; they can cause abortion in pregnant animals. Moreover, when the vaccine S19 is used, tests cannot differentiate natural from vaccinated infection. Therefore, a safer and more potent vaccine is needed. A Brucella abortus 2308 ery promoter mutant (Δery) was constructed to overcome these drawbacks. The growth of the Δery mutant was significantly attenuated in macrophages and mice and induced high protective immunity in mice. Moreover, Δery induced an anti-Brucellaspecific IgG (immunoglobulin G) response and stimulated the expression of interferon-gamma (INF-γ) and interleukin-4 (IL-4). Furthermore, the expression of EryA antigen allowed for the serological differentiation between natural and vaccinated infection in mice. These results indicate that the Δery mutant is a potential attenuated live vaccine candidate against virulent Brucella abortus 2308 (S2308) infection.

        Improvement in Active Power Control Methods for a Wind Farm Based on Modified Wind Turbine Control in Danish Grid Codes

        The importance of power system stability has been emphasized with an increase of wind energy penetration in the power system. Accordingly, the guarantee on various control capabilities, including active and reactive power control of wind farms, was regarded as the most important aspect for the connection to the grid. To control the wind farm active power, the wind farm controller was introduced. The wind farm controller decides the power set points for each wind turbine generating unit and each wind turbine generating unit controls its power according to the set points from the wind farm controller. Therefore, co-relationship between wind farm controller and wind turbine controllers are significantly important. This paper proposes some control methods of wind farm active power control based on modified wind turbine control for power system stability and structures to connect wind turbine controllers to wind farm controller. Besides, this paper contributes to development of control algorithm considering not only electrical components but also mechanical components. The proposed contributions were verified by full simulation including power electronics and turbulent wind speed. The scenario refers to the active power control regulations of the Eltra and Elkraft system in Denmark.

      • L-carnitine Enhances Bovine Oocyte Quality and the Developmental Competence of Somatic Cell Nuclear Transfer Embryos

        Somatic cell nuclear transfer (SCNT) is the technique which generates embryos by transferring diploid nucleus into an enucleated oocyte, it has produced specific animals successfully in a variety of species. However, the developmental capacity of SCNT embryos is still relatively lower than that of embryos produced in vivo. Oocyte is a kind of lipid rich cells, its quality limits the efficiency of embryo production. L-carnitine is a co-enzyme facilitating the transportation of long chain fatty acids across the inner mitochondria membrane where fatty acids are used for generating adenosine triphosphate (ATP) via beta-oxidation. It also has antioxidant actions which may protect mitochondrial membranes and DNA against damage induced by reactive oxygen species (ROS). Whether L-carnitine is functional in bovine SCNT embryos are unknown. Therefore, the objective of this study was to examine the effects of L-carnitine on oocyte maturation and developmental competence of subsequent SCNT embryos. L-carnitine was supplemented during IVM, then intracellular ROS and GSH levels, mitochondrial activity, gene expression of COCs were analyzed at the end of IVM. SCNT embryos were produced subsequently, apoptosis detection and gene expression evaluation were performed in blastocysts. In the results, treatments with 1.5 mM and 3 mM L-carnitine significantly improved maturation rates (P<0.05). Treatments with 3 mM L-carnitine effectively induced improvement in nuclear maturation, intracellular GSH levels and mitochondrial activity, as well as a reduction in intracellular ROS levels (P<0.05). mRNA levels of CPT1A, ACAA1, ACAA2, AREG, EREG, SOD1, GPX4, GLUT1 and CDC2 transcripts were effectively up-regulated by 3 mM L-carnitine treatments (P < 0.05). Similarly, 3mM L-carnitine induced an increase in blastocyst developmental rates and an improvement in blastocyst quality (P<0.05). Our study indicates that L-carnitine treatment during IVM improves oocyte nuclear maturation and subsequent SCNT embryo development.

        Comparative proteomics of the mixotrophic dinoflagellate Prorocentrum micans growing in different trophic modes

        Protein profiles of a common mixotrophic dinoflagellate, Prorocentrum micans, growing autotrophically and mixotrophically (fed on the cryptophyte Rhodomonas salina) were compared using two-dimensional gel electrophoresis (2-DE) to determine if they vary in different trophic modes. Approximately 2.3% of the detected proteins were differentially expressed in the different trophic modes. Twelve proteins observed only in the mixotrophic condition had lower pI value (<5) than the fifteen proteins observed only in the autotrophic condition (>5). When the internal amino acid sequences of five selected proteins differentially expressed between autotrophic and mixotrophic conditions were analyzed using matrix-assisted laser desorption time-of-flight (MALDI-TOF) mass spectrometry, two proteins that were specifically expressed in the autotrophic condition showed homology to glyceraldehyde-3-phosphatase dehydrogenase (GAPDH) and a bacterial catalase. Three mixotrophy-specific proteins showed homology to certain hypothetical proteins from an insect and bacteria. These results suggested the presence of certain gene groups that are switched on and off according to the trophic mode of P. micans.

        Levosimendan Reduces Lung Injury in a Canine Model of Cardiopulmonary Bypass

        Background and Objectives: To explore the lung-protective effect of levosimendan (LS) during cardiopulmonary bypass in a canine modelby determining the wet/dry weight (W/D) ratio of lung tissue, malonaldehyde (MDA) and superoxide dismutase (SOD) concentrations, andperforming a histological evaluation. Materials and Methods: Thirty-two canines were divided randomly into four groups and underwent a routine aortic cross-clampingcardiopulmonary bypass procedure for 1 h, followed by recovery for 2 h. Animals were handled as follows: group C (means controlgroup), no special treatment after aortic cross clamping; group P (means pulmonary artery perfusion group), pulmonary artery perfusionwith cold oxygenated blood after aortic cross clamping; group LSIV (means intravenous injection of LS group), intravenous injection of LS(65 μg/kg) before thoracotomy, and the rest of the procedure was identical to the control group; group LPS (means pulmonary perfusionwith LS group), pulmonary perfusion with cold oxygenated blood combined with LS (65 μg/kg) after aortic cross clamping. Lung tissueswere removed and subjected to evaluation of pathological alterations, W/D ratio and MDA and SOD concentrations. Results: In group C, the W/D ratio and MDA concentration were higher, while the SOD concentrations were lower (p<0.05). Comparedwith groups P and LSIV, the MDA concentration was lower in group LPS, while that of SOD was higher (p<0.05); Light and electronmicroscopy indicated that LS intervention reduced impairment of lung tissues. Conclusion: Our findings suggest that LS plays an important role in protecting lung tissues.

        MicroRNA-148a Can Regulate Runt-Related Transcription Factor 3 Gene Expression via Modulation of DNA Methyltransferase 1 in Gastric Cancer

        Underexpression of the gene runt-related transcription fac-tor 3 (RUNX3), an important tumor suppressor, is known to contribute to gastric cancer progression. However, the mechanism underlying aberrant RUNX3 expression has not been fully elucidated. We investigated the role of microRNA-148a (miR-148a) and DNA methyltransferases (DNMTs) in RUNX3 promoter methylation and gene expression. RUNX3 mRNA, RUNX3 protein, and methylation levels were assayed in human gastric cancer tissues and matched normal tissues, and AGS and BGC-823 cells by real-time reverse transcription PCR, Western blot, and methylation-specific PCR, respectively. A correlation between RUNX3 mRNA levels and that of miR-148a was also investigated in gastric cancer tissues. We found that RUNX3 mRNA levels were significantly downregulated in gastric cancer tissues compared with their matched normal tissues, and were closely associated with miR-148a expression. After treatment of human gastric cancer AGS and BGC-823 cells with the DNA methylation inhibitor 5-aza-2’-deoxycytidine, a significant increase in RUNX3 mRNA, RUNX3 protein, and the non-methylated form of the RUNX3 promoter were observed relative to untreated cells. Enforced expression of miR-148a, which can modulate DNMT1 and DNMT3B, also increased the expression of RUNX3 in gastric cancer cells. Knockdown of DNMT1 was associated with increased levels of RUNX3 mRNA and RUNX3 protein, while knockdown of DNMT3B did not have any effect on these in BGC-823 cells. Our results show that miR-148a may regulate RUNX3 expression through modulation of DNMT1-dependent DNA methylation in gastric cancer and highlight a miRNA-epigenetics regulation mechanism of gene expression.

      • Features Fusion with Adaptive Weights for Pedestrian Classification

        In this paper, we study the problem of pedestrian classification, which could lead to an improvement of performance of the Pedestrian Detection Systems. Since the traditional approaches merely focus on the recognition of pedestrian, the device would keep alerting the drivers even if the pedestrians are walking on a safe track. We attempt to classify pedestrians in order to make those devices, equipped in the cars, more intelligent and pragmatic. We propose a method to extract features including HOGs (Histogram of Oriented Gradient), LTPs (Local Ternary Pattern), Color Names and to fuse them efficiently. The three features are weighted fused depending on the size of patches as well as each patch’s gradient value which is computed via a 3<SUP>*</SUP>3 Sobel operator. Afterwards we will train a random forest with 50 discriminative decision trees, using the fused features. Our method is tested on the images of humans from INRIA dataset. The experimental results show that our method of features fusion, with adaptive weights assigned to the different features, yields a significant gain of 12.9% in mean AP (Average Precision) over the simple features concatenation framework. Accordingly, our method is practicable for classifying pedestrians.

        Trajectory Tracking Performance Analysis of Underwater Manipulator for Autonomous Manipulation

        In this study, the end-effector tracking performance of a manipulator installed on a remotely operated vehicle (ROV) for autonomous underwater intervention is verified. The underwater manipulator is an ARM 7E MINI model produced by the ECA group, which consists of six joints and one gripper. Of the six joints of the manipulator, two are revolute joints and the other four are prismatic joints. Velocity control is used to control the manipulator with forward and inverse kinematics. When the manipulator approaches a target object, it is difficult for the ROV to maintain its position and posture, owing to various disturbances, such as the variation in both the center of mass and the reaction force resulting from the manipulator motion. Therefore, it is necessary to compensate for the influences and ensure the relative distance to the object. Simulations and experiments are performed to track the trajectory of a virtual object, and the tracking performance is verified from the results.

        Although eliminating ring ghost has been industrially required, it is only removed from data biased to certain phenomena due to lack of learning data, and there are limitations to performance. To solve this problem, this paper proposes a method for removing ring-ghost harmful noise. The proposed method is performed by in-painting for detecting the ring ghost region and removing ring ghost noise, and it can provide quality improvement results by removing ring pattern noise from specific lenses and cameras.
