        Heme oxygenase-1 (HO-1)/carbon monoxide (CO) axis suppresses RANKL-induced osteoclastic differentiation by inhibiting redox-sensitive NF-κB activation

        Heme oxygenase (HO-1) catalyzes heme to carbon monoxide (CO), biliverdin/bilirubin, and iron and is known to prevent the pathogenesis of several human diseases. We assessed the beneficial effect of heme degradation products on osteoclastogenesis induced by receptor activator of NF-κB ligand (RANKL). Treatment of RAW264.7 cells with CORM-2 (a CO donor) and bilirubin, but not with iron, decreased RANKLinduced osteoclastogenesis, with CORM-2 having a more potent anti-osteogenic effect. CORM-2 also inhibited RANKLinduced osteoclastogenesis and osteoclastic resorption activity in marrow-derived macrophages. Treatment with hemin, a HO-1 inducer, strongly inhibited RANKL-induced osteoclastogenesis in wild-type macrophages, but was ineffective in HO-1<sup>+/- </sup>cells. CORM-2 reduced RANKL-induced NFATc1 expression by inhibiting IKK-dependent NF-κB activation and reactive oxygen species production. These results suggest that CO potently inhibits RANKL-induced osteoclastogenesis by inhibiting redox-sensitive NF-κB-mediated NFATc1 expression. Our findings indicate that HO-1/CO can act as an antiresorption agent and reduce bone loss by blocking osteoclast differentiation. [BMB Reports 2017; 50(2): 103-108]

        성인에서 발생한 발진성 가성 혈관종증

        Background: Eruptive pseudoangiomatosis (EPA) is a rare, benign, spontaneously regressing childhood exanthem. It is characterized by the sudden onset of several bright red angioma-like papules surrounded by pale halos with a distinct histopathology from true angiomas. Objective: This study was performed to evaluate the clinical and histopathologic characteristics of EPA occuring in adults. Methods: Ten adult patients who visited Pusan National University Hospital and Mok Hye-SooㆍJang Ho-Sun Dermatology Clinic from March 2005 to September 2006 were evaluated. We prospectively evaluated the sex, age, onset season, past medical history including immunosuppressive abnormalities, systemic disorders and other diseases including allergies. We also investigated the relations of mosquito biting, patients` occupations and outdoor activities to occurrence of EPA. In addition, simultaneous occurrence in family members, the clinical, histopathologic, laboratory findings, disease courses and responses to treatment were evaluated. Based on medical records, photographs and pathologic slides, we retrospectively diagnosed another 20 EPA patients suspected as insect bite from October 2003 to March 2005. The same questions were inquired as for the 10 patients who prospectively underwent evaluation. Results: In the study, female predominance (76.7%) was observed and the average age of onset was 54.2 years. Interestingly, there was no child patient during the period of study. Multiple, 2∼5 mm sized, red angiomatous papules surrounded by pale halos occurred on exposed areas such as the arms (86.7%), legs (50%), and face (46.7%), although it could also occur to a non-exposed area. EPA occuring in adults usually appeared in summer (80%). The mean disease duration was 3.4 weeks. Although EPA spontaneously regressed, it had the potential of recurrence (46.7%). Histopathologic findings showed dilated dermal blood vessels without the evidence of increase in numbers, and perivascular lymphocytes infiltration. Inside the lumen of dermal blood vessels, plump endothelial cells were found. Conclusion: EPA occuring in adults usually happened to exposed sites in summer, so it can be misdiagnosed as insect bite. We suggest that dermatologists should be concerned about EPA in adults and conduct further investigation to have a better understanding of the disease. (Korean J Dermatol 2007;45(8):797∼803)

        Detection of benzalkonium chloride on glass surfaces using silver nanoparticles

        Raman reporter-based surface-enhanced Raman scattering (SERS) has been used to investigate the detection of one member of a class of the cationic surfactant quaternary ammonium compounds, alkyl dimethyl benzyl ammonium chloride, also known as benzalkonium chloride (BAC). For this study, SERS was performed using silver nanoparticles (AgNPs). Density functional theory (DFT) of BAC on Ag6 clusters was introduced to estimate binding at the level of the B3LYP/LANL2DZ basis sets. As the chain length increased from n = 8 to 18, the binding energies were expected to increase by 2.1 kcal/mol. Methylene blue (MB) Raman reporter molecules were subsequently treated on BAC thin-film-covered glass surfaces. Atomic force microscopy (AFM) images were used to determine the coverage of BAC on the glass, and field-emission electron microscope (FE-SEM) images revealed the surface morphologies of the SERS substrates. A standard calibration curve was prepared to quantify BAC within a concentration range of 0.0036805 to 36?805?ppm (r2?≥?0.99), according to the BAC-correlated SERS intensities of MB Raman peaks at ~1620?cm?1 adsorbed on AgNPs. A molecular dynamics (MD) simulation supported strong adsorption of BAC on glass surfaces.

      • Antioxidant and Anti-Inflammatory Effects of Rhei Rhizoma and Coptidis Rhizoma Mixture on Reflux Esophagitis in Rats

        <P>The purpose of this study was to investigate the antioxidant and anti-inflammatory effects of the combined extract of Rhei rhizoma and Coptidis rhizoma (RC-mix) in experimental model of acute reflux esophagitis. The antioxidant activity was assessed by<I> in vitro</I> 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays. RC-mix was given at 100, 200, and 400 mg/kg body weight 2 h prior to induction of reflux esophagitis (RE). After 5 h, the effects of RC-mix treated rats were compared with those of normal and control rats. The representative flavonoid contents of RC-mix, such as sennoside A, epiberberine, coptisine, palmatine, and berberine, were detected using HPLC. The elevated esophageal mucosa damage was markedly ameliorated by RC-mix treatment in a dose-dependent manner. Furthermore, the administration of RC-mix reduced the increase of serum reactive oxygen species (ROS) and peroxynitrite (ONOO<SUP>−</SUP>). The improvement of superoxide dismutase (SOD) and heme oxygenase-1 (HO-1) levels were marked in the group given RC-mix. Moreover, the elevation of inflammatory mediators and cytokines by nuclear factor-kappa B (NF-<I>κ</I>B) activation in control rats decreased by RC-mix pretreatment. These results indicate that RC-mix treatment reduces the pathological states of esophagitis<I> via</I> regulating NF-<I>κ</I>B mediated inflammation related to oxidative stress. </P>

        경기도의 연합정치 성과에 대한 분석을 실시함에 있어 정책연합, 정부연합이라는 두 가 지 분류기준을 주요변수로 규정하였다. 아울러, 경기도 연합정치를 평가함에 있어 한국과 같은 지방자치단체의 독임제 기관구성 하에 연합정치의 성과를 분석하여 연합정치의 활성 화를 위한 방안으로서 지방자치제도 개선방안을 제시하였다. 경기도 연합정치의 한계로 행정업무 지연의 문제 등 연합정치로 인한 다양한 사업으로 인해 많은 예산이 반영되고 해당 부서의 중요사업에 대한 검토를 신속하게 이루어져야 함 에도 불구하고, 보고 및 결제단계 증가로 시간소요 또한 증가하는 등의 문제점이 나타났다. 향후 경기도 연합정치를 활성화시키고 나아가 이를 실천하기 위한 지방자치제도의 개선 방안으로 첫째, 기관구성의 다양화이다. 둘째, 지방장관제 도입이다. 셋째, 광역시도 의회의 역량을 강화하는 것이다. 넷째, 지방선거제도의 개편이다. On the other hand, comparing the first and second periods in terms of the governmental coalition, the allocation of power and the improvement of the coalition process can be mentioned. First, from the aspect of power allocation, the first stage handed over to the opposition party the vice governor of social integration, which is responsible for the three countries of health, welfare, environment, and women's family. In the second stage(Cho Sung-Ho et al., 2017), In this way, the opposition party is able to share power with the opposition party. At the same time, it introduced the coalition chairperson system, which further strengthened the power of political parties participating in the coalition. In addition, by establishing a unified political arbitration committee to facilitate the conflict situation that occurred under the coalition government, the coalition government made soft landing. In the future, we will suggest ways to improve the local autonomy system to revitalize the Gyeonggido coalition politics and to practice it. First, diversification of organization. Second, the introduction of local ministers. Third, the metropolitan city should strengthen its capacity. Fourth, it is a reorganization of local electoral system.

        코아-셀 구조를 가지는 전도성 폴리피롤 나노섬유를 이용한 메탄올 센서 제작

        Electrically conductive polypyrrole-polyvinylpyrrolidone (PPy-PVP) nanofiber mats with a core-shell structure have been successfully fabricated by a two-step process: the formation of FeCl3-containing PVP nanofiber mat by electrospinning, and the vapor-phase polymerization (VPP) of pyrrole monomer on the mat in a sealed chamber at room temperature. Surface morphology and chemical composition of the PPy-PVP mat were characterized by SEM, EDX and FTIR analyses. The as-prepared nonwoven mat was composed of PPy-PVP nanofibers with an average diameter of 300 nm. The sheet conductivity of the nanofiber mat was measured to be approximately 0.01 S/cm by a four-point probe. We have also investigated gas-sensing properties of PPy-PVP nanofiber mat upon exposure to methanol vapor. The PPy-PVP nanofiber sensors were observed to have excellent methanol-sensing performance. The nanofiberbased core-shell nanostructure could give an opportunity to fabricate a highly sensitive and fast response sensor due to its high surfaceto-volume ratio.

        3,4,5-Trihydroxycinnamic Acid Inhibits Lipopolysaccharide-Induced Inflammatory Response through the Activation of Nrf2 Pathway in BV2 Microglial Cells

        3,4,5-Trihydroxycinnamic acid (THC) is a derivative of hydroxycinnamic acids, which have been reported to possess a variety of biological properties such as anti-inflammatory, anti-tumor, and neuroprotective activities. However, biological activity of THC has not been extensively examined. Recently, we reported that THC possesses anti-inflammatory activity in LPS-stimulated BV2 microglial cells. However, its precise mechanism by which THC exerts anti-inflammatory action has not been clearly identified. Therefore, the present study was carried out to understand the anti-inflammatory mechanism of THC in BV2 microglial cells. THC effectively suppressed the LPS-induced induction of pro-inflammatory mediators such as NO, TNF-a, and IL-1b. THC also suppressed expression of MCP-1, which plays a key role in the migration of activated microglia. To understand the underlying mechanism by which THC exerts these anti-inflammatory properties, involvement of Nrf2, which is a cytoprotective transcription factor, was examined. THC resulted in increased phosphorylation of Nrf2 with consequent expression of HO-1 in a concentration-dependent manner. THC-induced phosphorylation of Nrf2 was blocked with SB203580, a p38 MAPK inhibitor, indicating that p38 MAPK is the responsible kinase for the phosphorylation of Nrf2. Taken together, the present study for the first time demonstrates that THC exerts anti-inflammatory properties through the activation of Nrf2 in BV2 microglial cells, suggesting that THC might be a valuable therapeutic adjuvant for the treatment of inflammation-related disorders in the CNS.

      • <i>Toxoplasma gondii</i> protects against H<sub>2</sub>O<sub>2</sub>‐induced apoptosis in ARPE‐19 cells through the transcriptional regulation of apoptotic elements and downregulation of the p38 MAPK pathway

        <P><B>Abstract.</B></P><P><B>Purpose: </B> Toxoplasmosis, which is caused by the protozoan parasite <I>Toxoplasma gondii</I>, can lead to severe visual impairment. <I>T.?gondii</I> inhibits or delays programmed cell death caused by various apoptotic triggers; however, the mechanisms involved in the <I>T</I>.?<I>gondii</I>‐induced suppression of apoptosis in retinal cells have not been analysed in detail.</P><P><B>Methods: </B> We investigated the role of <I>T</I>.<I>?gondii</I> infection in H<SUB>2</SUB>O<SUB>2</SUB>‐induced apoptosis in human retinal pigment epithelial cells (ARPE‐19) by monitoring the activities of apoptosis‐regulating molecules and mitogen‐activated protein kinases (MAPKs), including p38 MAPK. We also examined the gene downstream from p38 MAPK.</P><P><B>Results: </B> <I>T.?gondii</I> infection significantly inhibited the cellular toxicity of H<SUB>2</SUB>O<SUB>2</SUB> (500 μ<SMALL>m</SMALL>) and increased cell viability in a multiplicity of infection (MOI)‐dependent manner by reducing DNA fragmentation and reactive oxygen species (ROS) generation in ARPE‐19 cells. Western blot analysis also showed that <I>T</I>.?<I>gondii</I> infection prevented the host cell expression of pro‐apoptotic factors, such as Bad and Bax, and the activation of caspase‐3. Infection with <I>T</I>.?<I>gondii</I> increased the expression of the anti‐apoptotic factor Bcl‐2 in ARPE‐19 cells under oxidative stress. In accordance with these findings, <I>Toxoplasma</I> infection was protective enough to suppress the phosphorylation of p38 MAPK following H<SUB>2</SUB>O<SUB>2</SUB> treatment. Exposure to H<SUB>2</SUB>O<SUB>2</SUB> increased the expression of heme oxygenase‐1 (HO‐1) in ARPE‐19 cells, and its expression was significantly inhibited in H<SUB>2</SUB>O<SUB>2</SUB>‐treated infected cells.</P><P><B>Conclusion: </B> The protective function of <I>T</I>.?<I>gondii</I> infection against ROS‐induced apoptosis results from changes in the expression of apoptotic molecules and the downregulation of stress‐induced intracellular signalling.</P>
