        The Interpretation of Ironic Utterances

        The purpose of this paper is to examine the process of understanding verbal irony in cognitive pragmatics (relevance theory). According to Sperber and Wilson (1995), an ironic utterance is a second degree of interpretation of someone's thought or utterance; the speaker echoes an attributed opinion to express her attitude of disapproval or ridicule, while dissociating herself from it. In this paper, we will show that perceiving a given utterance as ironic depends on the listener's ability to catch and understand the speaker's intention, and that an echoic interpretation of an attributed thought, in relevance-theoretic terms, can be applied to more ironic utterances than Grice's account.

        Modern Asian Poets under Western Gaze: Bei Dao and Yosano Akiko in World Literature

        Although anthologies from Norton, Longman and Bedford have begun to encompass literatures from other countries and cultures under the category of world literature, they are often criticized for their Eurocentric perspective in introducing, describing, and selecting the works in their anthologies. In this paper, I will examine the ongoing discourse on world literature, and attempt to place Asian literature, especially modern East Asian poetry in this context by looking into two anthologies. I am mainly focusing on the poetry in The Longman Anthology of World Literature (2009) and The Norton Anthology of World Literature (2012) in order to examine how they explain world history and represent East Asian poets, and finally to question the validity of that representation. Both of the anthologies guide the readers through the modern history of the world in a rather Eurocentric way, largely focusing on modernism and its movements along with two world wars. It is not surprising therefore, when modern Asian poetry is either missing or unfitting to the theme under which it is explained. The Norton especially does not include any East Asian poets in its modern poetry section, and its editors might be the reason why. Although The Longman includes two East Asian poets, Bei Dao from China and Yosano Akiko from Japan, they are squeezed into the context of modernism and war narrative not for their writing style but for their political and cultural positions. While the anthology is often acknowledged as a marketing product, it also plays significant role in guiding the students academically, reflecting and contributing to the discourse on world literature. Such characteristics of the anthology necessitate editors and anthologists to mediate and interact more with unfamiliar non-Western scholars and authors in order to avoid Eurocentrism and include literatures outside of Europe and America without the risk of misrepresentation or negligence over distinctive poetics.

        Aspects of Meaning in Interaction: Incomplete Interpretive Hypotheses in L2 Discourse

        From a relevance theory perspective, it is assumed that verbal communication processes are governed by a ‘less-than-perfect heuristic’, entailing failures in communication, and that what requires explanation is how successful communication may be achieved (Sperber and Wilson 1995). Accordingly, little attention has been paid to miscommunication or hypotheses about jointly-adjusted interpretive meaning in the relevance theory framework. Using the relevance theory perspective (Carston 2002; Sperber and Wilson 1995; Wilson and Sperber 2004) with a view to extending the understanding of meaning by language users in interaction (Culpeper and Haugh 2014), this paper is concerned with a nuanced understanding of the meaning of the utterances as part of the interpretation process, including miscommunication, in second language (L2) English conversational discourse. Drawing on the relevance-theoretical notions of the individual’s ‘expectations of relevance’ (Sperber and Wilson 1995), ‘parallel adjustment process’ (Wilson and Sperber 2004) and Culpeper and Haugh’s (2014) notion of ‘recipient meaning’, this paper highlights that the potential for miscommunication in interaction is not uncommon, particularly in terms of meaning construction.

        Use of Clarification Requests and Confirmation Checks

        Clarification requests and confirmation checks are regarded as negotiation of meaning strategies in second language acquisition, by which assistance is requested to resolve incomprehension in a conversational discourse (e.g. Long 1996; Gass 2003). The present study attempts to explore pragmatic aspects of the use of clarification requests and confirmation checks in casual conversations between advanced adult Korean learners of English. Adopting Spencer-Oatey’s (2008) rapport management and Arundale’s (2010) face interpretation approaches, this paper explores how some clarification requests and confirmation checks may be related to potential pragmatic concerns in communication within a particular context. By examining utterances through dialogue interactions, this study demonstrates that some clarification requests and confirmation checks may involve rapport consideration or induce face interpretation of the discourse participants at the moment of utterance in ongoing conversations. This may suggest that, in such cases, the speaker may achieve connectedness or separateness (Arundale 2010), rather than correct comprehension.

        Interaction of Face and Rapport in an American TV Talk Show

        This paper is concerned with some features of English spoken discourse which appear in an American TV talk show. It focuses on the conversational interactions between a host and guests in a particular context from a rapport management perspective. The paper discusses whether certain speech acts are related to face, and shows that managing rapport involving positive politeness also appears to be manifested. With particular reference to the speaker’s role, this paper highlights the speakers’ awareness of the role of the listener and the rapport with them by interpreting the use of deictic shift, repetition and acts which may, unintentionally, threaten face.

        Development and Optimization of a Rapid Colorimetric Membrane Immunoassay for Porphyromonas gingivalis

        Porphyromonas gingivalis (P. gingivalis) is a major bacterial pathogen that causes periodontitis, a chronic inflammatory disease of tissues around the teeth. Periodontitis is known to be related to other diseases, such as oral cancer, Alzheimer’s disease, and rheumatism. Thus, a precise and sensitive test to detect P. gingivalis is necessary for the early diagnosis of periodontitis. The objective of this study was to optimize a rapid visual detection system for P. gingivalis. First, we performed a visual membrane immunoassay using 3,3′,5,5′-tetramethylbenzidine (TMB; blue) and coating and detection antibodies that could bind to the host laboratory strain, ATCC 33277. Antibodies against the P. gingivalis surface adhesion molecules RgpB (arginine proteinase) and Kgp (lysine proteinase) were determined to be the most specific coating and detection antibodies, respectively. Using these two selected antibodies, the streptavidin-horseradish peroxidase (HRP) reaction was performed using a nitrocellulose membrane and visualized with a detection range of 10<sup>3</sup>-10<sup>5</sup> bacterial cells/ml following incubation for 15 min. These selected conditions were applied to test other oral bacteria, and the results showed that P. gingivalis could be detected without cross-reactivity to other bacteria, including Streptococcus mutans and Escherichia fergusonii. Furthermore, three clinical strains of P. gingivalis, KCOM 2880, KCOM 2803, and KCOM 3190, were also recognized using this optimized enzyme immunoassay (EIA) system. To conclude, we established optimized conditions for P. gingivalis detection with specificity, accuracy, and sensitivity. These results could be utilized to manufacture economical and rapid detection kits for P. gingivalis.

      • Exploring the Potential of an International Student Exchange Program Based on Experiential Learning: Focusing on the APEC Edutainment Exchange Program (AEEP)

        This paper explores the APEC Edutainment Exchange Program (AEEP) as a model for international student exchange. This program has been conducted in the APEC region since 2007 to respond to the increasing international demand for youth exchange. The AEEP is based on PBL (project based learning)- oriented experiential learning, and includes multinational team activities with support systems formed by human and physical resources. Accordingly, this paper reviewed prior research on international student exchange programs and experiential learning, as well as the AEEP. While the AEEP cases reviewed here focus on the participants in the Asia-Pacific, this paper demonstrates the potential of this program to be applied to similar situations elsewhere.

        Rapid and sensitive detection of Salmonella species targeting the hilA gene using a loop-mediated isothermal amplification assay

        Salmonella species are among the major pathogens that cause foodborne illness outbreaks. In this study, we aimed to develop a loop-mediated isothermal amplification (LAMP) assay for the rapid and sensitive detection of Salmonella species. We designed LAMP primers targeting the hilA gene as a universal marker of Salmonella species. A total of seven Salmonella species strains and 11 non-Salmonella pathogen strains from eight different genera were used in this study. All Salmonella strains showed positive amplification signals with the Salmonella LAMP assay; however, there was no non-specific amplification signal for the non-Salmonella strains. The detection limit was 100 femtograms (20 copies per reaction), which was ~1,000 times more sensitive than the detection limits of the conventional polymerase chain reaction (PCR) assay (100 pg). The reaction time for a positive amplification signal was less than 20 minutes, which was less than one-third the time taken while using conventional PCR. In conclusion, our Salmonella LAMP assay accurately detected Salmonella species with a higher degree of sensitivity and greater rapidity than the conventional PCR assay, and it may be suitable for point-of-care testing in the field.
