<P><B>Abstract</B></P><P>A reliable source of osteogenic cells is an essential factor for bone tissue engineering. In this study, human‐induced pluripotent stem cells (hiPSCs) without an embryoid body step were cultured in macrochanneled poly(caprolactone) (PCL) scaffolds prepared using a robotic dispensing technique, after which osteogenesis was promoted by the addition of exogenous osteogenic factors. The osteogenesis of the hiPSCs was demonstrated based on the detection of osteogenic molecules, such as osteopontin, using flow cytometry analysis, quantitative polymerase chain reaction and western blotting. Thereafter, the cell‐scaffold constructs were transplanted into the subcutaneous site of male athymic mice. At 4 weeks after implantation, histological assays (hematoxylin & eosin staining, Alizarin red staining, and osteocalcin immunostaining) were conducted to determine the bone induction of hiPSCs. The results indicated a production of pronounced levels of extracellular matrices and their mineral deposition within the cell‐scaffold implant, suggesting possible <I>in vivo</I> bone induction by the hiPSCs‐based tissue engineering approach. The results presented here provide useful information regarding the tissue engineering of bone utilizing hiPSCs in conjunction with cell‐supporting scaffolds. © 2012 Wiley Periodicals, Inc. <i>EML4-ALK</i> Fusion Gene in Korean Non-Small Cell Lung Cancer

<P>A fusion gene between echinoderm microtubule-associated protein-like 4 (<I>EML4</I>) and the anaplastic lymphoma kinase (<I>ALK</I>) has been identified in non-small cell lung cancers (NSCLCs). Although a few studies have evaluated <I>EML4-ALK</I> fusion genes in Korean NSCLCs, the prevalence of different <I>EML4-ALK</I> fusion variants has yet to be clearly assessed. Herein, we have examined the profiles of <I>EML4-ALK</I> fusion gene variants in Korean patients of NSCLCs. <I>EML4-ALK</I> fusion genes have been detected in 10 (6.0%) of 167 patients of NSCLCs and in 9 (7.4%) of 121 patients of adenocarcinoma. Of the 10 patients with fusion genes identified, 8 (80%) were E13;A20 (variant 1) and 2 (20%) were E6;A20, with an additional 33-bp sequence derived from intron 6 of <I>EML4</I> (variant 3b). These results indicate that the profiles of <I>EML4-ALK</I> fusion gene variants in Korean patients of NSCLC may differ from those in other ethnic populations. Herein, we describe for the first time the profiles of <I>EML4-ALK</I> fusion variants of Korean patients with NSCLCs.</P>
Hydrothermal Synthesis of La-Mn-hexaaluminates for the Catalytic Combustion of Methane

Hydrothermal synthesis by using urea hydrolysis at 1.0~3.0MPa and 120~130℃ was employed to prepare Mn-substituted hexaaluminate catalysts for methane combustion. The results from DTA-MS demonstrated that CO^2-_3 and OH^- anions co-exist in the hydrothermal reaction. XRD reveals that the components of carbonates of carbonates and hydroxides in the hydrothermal reaction are more favorable than those in the(NH_4)_2CO_3 co-precipitation for the formation of the Mn-substituted hexaaluminate phase. After calcination at 1,200℃ for 2h, LaMnAl_11O_19 is the major phase of the catalyst prepared by the hydrothermal synthesis method while LaAlO_3 is the major one of the catalysts prepared by NH_4OH and (NH_4)_2CO_3 co-precipitation. The catalyst prepared by hydrothermal synthesis has higher activity than that prepared by NH_4OH and (NH_4)CO_3 co-precipitation. The major reason is that more Mn^2+ ions have incorporated into the hexaaluminate lattice. The effect of drying methods on the formation of hexaaluminate phase was also discussed.
Genome-wide comparative analysis of the codon usage patterns in plants

Codon usage analysis has been a classical area of study for decades and is important for evolution, mRNA translation, and new gene discovery. Recently, genome sequencing has made it possible to perform studies of the entire genome in plant kingdoms. The base composition of the coding sequence, codon usage pattern, codon pairs, and related indicators of relative synonymous codon usage (RSCU), including the Fop, Nc, RSCU, CAI and GC contents, were analyzed. We found that the GC content of single-celled algae is the highest, whereas dicotyledons are the lowest. Moreover, the base composition of plants is similar within the same family. In addition, the GC content of the second base of the codon is lower than the first and third base. In conclusion, the codon usage characteristics are opposite in Gramineae, single-celled algae, fern and dicotyledon, moss, and Pinaceae. Furthermore, the degree of codon usage bias is decreasing with evolution. Therefore, we hypothesize that the lower the plants, the more that they must optimize codons and that higher plants no longer need to optimize codons.

In order to improve aerodynamic performance of multi-stage axial flow turbines used in aircraft engines, a one-dimensional aerodynamic design and optimization framework is constructed. In the method, flow path is generated by solving mass continuation and energy conservation with loss computed by the Craig & Cox model; Also real gas properties has been taken into consideration. To obtain an optimal result, a multi-objective genetic algorithm is used to optimize the efficiencies and determine values of various design variables; Final design can be selected from obtained Pareto optimal solution sets. A three-stage axial turbine is used to verify the effectiveness of the developed optimization framework, and designs are checked by three-dimensional CFD simulation. Results show that the aerodynamic performance of the optimized turbine has been significantly improved at design point, with the total-to-total efficiency increased by 1.17% and the total-to-static efficiency increased by 1.48%. As for the off-design performance, the optimized one is improved at all working points except those at small mass flow.

In order to study the mechanical property degradation rule of Q690 high-strength steel in the ocean wave splash area, the rusted specimen is obtained by indoor damp heat surrounding soaking environment corrosion test. The scanning electron microscopy, uniaxial tensile and cyclic loading tests of corroded steel are performed. This paper analyzes the steel from elastic modulus, yield strength, ductility and law of hysteresis degradation under diff erent corrosion cycles and establishes the constitutive model of HSS. The results show that the appearance of HSS is obviously aff ected by the corrosion cycle. With the increase of the corrosion cycle, reddish-brown fl aky corrosion products are formed on the surface of the steel and there are obvious corrosion pits. After 100 days of corrosion, the corrosion rate of steel reached 7.21%; the elastic modulus and yield strength under uniaxial tension load decreased by 10.2% and 4.9%. Under the eff ect of cyclic reciprocating load, the hysteretic energy is reduced by 15.3%; at the same time, the test curve is fi tted by the secondary plastic fl ow model and the Ramberg–Osgood model and it is found that the shape parameter S 1 and the cyclic strengthening coeffi cient K’ are reduced by 20.1% and 10.2%, While the shape parameter S 2 and the cycle strengthening index n’ have no obvious changes.
Aerodynamic topology optimization on tip configurations of turbine blades

The topology optimization (TopOpt) is adopted in this article to design tip geometries of a turbine blade. Its principle is to modify the porosity, which is one for fluid and is zero for solid. A CFD method coupled with source terms is implemented for numerical simulations. Moreover, to update the porosity, a quasi-sensitivity and an adjoint sensitivity analysis schemes are integrated with the method of moving asymptotes algorithm, respectively. The TopOpt is conducted to design the tip profile of a 2D tip model and of nine axial sections in a 3D turbine blade. Optimized tips are reconstructed and their performance is verified by experiments and CFD. Results indicate that the re-designed novel tip, which has a convexconcave-convex form and is different from flat and cavity tips, has good performance due to getting rid of flow separations above the tip. Hence, the optimized tip can lower total pressure loss in the gap region.
