        The influence of BRCA variants of unknown significance on cancer risk management decision-making

        Objective: To compare gynecological cancer risk management between women with BRCA variants of unknown significance (VUS) to women with negative genetic testing. Methods: Ninety-nine patients whose BRCA genetic testing yielded VUS were matched with 99 control patients with definitive negative BRCA results at a single institution. Demographics and risk management decisions were obtained through chart review. Primary outcome was the rate of risk-reducing bilateral salpingo-oophorectomy (RRBSO). Chi square tests, t-tests, and logistic regression were performed, with significance of p<0.05. Results: VUS patients were more likely to be non-Caucasian (p=0.000) and of Ashkenazi-Jewish descent (p=0.000). There was no difference in gynecologic oncology referrals or recommendations to screen or undergo risk-reducing surgery for VUS vs. negative patients. Ultimately, 44 patients (22%) underwent RRBSO, with no significant difference in surgical rate based on the presence of VUS. Ashkenazi-Jewish descent was associated with a 4.5 times increased risk of RRBSO (OR=4.489; 95% CI=1.484–13.579) and family history of ovarian cancer was associated with a 2.6 times risk of RRBSO (OR=2.641; 95% CI=1.107–6.299). Conclusion: In our institution, patients with VUS were surgically managed similarly to those with negative BRCA testing. The numbers of patients with VUS are likely to increase with the implementation of multi-gene panel testing. Our findings underscore the importance of genetic counseling and individualized screening and prevention strategies in the management of genetic testing results.

      • Distal Demonstrative Hitlo in Taiwanese Southern Min

        This article investigates the use of distal demonstrative Hitlo in Taiwanese Southern Min (TSM) from a discourse-pragmatic perspective. The analysis is based on a 5-hour corpus of spoken data, including daily conversations, radio interviews, TV drama series, and some random examples. A total of 172 tokens of Hitlos are identified in the data. They can be divided into six categories according to their functions: firstly, exophoric usage, those Hitlos which refer to an object non-linguistically which can be identified in the immediate situation; secondly, endophoric usage, those which refer to an element textually; thirdly, referent introducing function, those which can be used to introduce a new but identifiable referent into the conversation (the referent usually has topical importance); fourthly, hedging expression, those which serve as a marker of imprecision; fifthly, a condition introducing marker, those which function as an indicator of the coming of a conditional sentence; finally, pause fillers, those which help speakers to manage speech turn or indicate the mental states In addition, an interactive function which Hitlo is found to serve will be discussed. Moreover, a grammaticalizational process involving semantic bleaching which Hitlo is probably undergoing is revealed in general. Finally, a filled demonstrative principle, stating that it may be a universal phenomenon to use demonstratives as filled pause will be proposed.

        FNC inhibits non-small cell lung cancer by activating the mitochondrial apoptosis pathway

        Background: Previously, we published that 4'-azid-2'-deoxy-2'-fluorarabinoside (FNC), a novel cytosine nucleoside analog, has good anti-viral and anti-tumor activity. Objective: This study aimed to further explore the role and molecular mechanism of FNC in non-small cell lung cancer (NSCLC). Methods: FNC was tested in the NSCLC H460 cell line, the Lewis mouse model, and the H460 cell xenograft model. The effects of FNC were assessed by cell viability, transwell migration, and wound scratch analyses of cell migration and invasion. Apoptosis was assessed by flow cytometry. Proteins expression was assessed by western blot and immunohistochemistry staining (IHC). Results: FNC inhibits the proliferation and metastasis of H460 cells in a time- and dose-dependent manner. FNC treatment showed efficacy and low toxicity in the Lewis mouse lung cancer model as well as in the H460 cell xenograft model. Further, FNC induced H460 cell apoptosis through the activation of the mitochondrial pathway. Notably, FNC inhibited invasion by increasing E-cadherin protein and reducing the protein expression of VEGF, MMP-2, MMP-9, and CD31. Conclusion: FNC inhibits NSCLC by activating the mitochondrial apoptosis pathway and regulating the expressions of multiple proteins related to cell adhesion and invasion, highlighting its potential as an NSCLC therapeutic.

        Regeneration of hexamminecobalt(II) under the catalysis of activated carbon modified with ZnCl2 solution

        The simultaneous scrubbing of NO and SO2 can be accomplished by hexamminecobalt(II) solution. Activated carbon acts as a catalyst to regenerate hexamminecobalt(II), Co(NH3)62+, to sustain the NO removal efficiency. In this paper, ZnCl2 solution has been used to ameliorate the catalytic performance of coconut activated carbon in the regeneration of Co(NH3)62+. The effects of the modification condition on the catalytic performance of activated carbon have been investigated in a batch stirred cell. The change of the surface chemical characteristics of the activated carbon caused by ZnCl2 treatment was measured by determining the concentrations of acidic and basic groups as well as their points of zero charge (pHpzc)with Boehm titration and mass titration. The alteration of the surface physical characteristics of the carbons was determined with an ASAP2000 Surface Analyzer using N2 as the adsorbate. The experiments demonstrate that the catalytic performance of the coconut activated carbon may be improved when the carbon is treated by ZnCl2 solution with concentration above 0.30 mol l1. The best ZnCl2 concentration may be 0.50 mol l1. The optimal impregnation duration is 9 h. High activation temperature is propitious for the amelioration of the catalytic capability of carbon. 4 h may be the best time for the activation of activated carbon. In our experiment, the NO removal efficiency is maintained at a level of 73% when the regeneration of Co(NH3)62+ is under the catalysis of modified carbon while that is 57% with the regeneration of Co(NH3)62+ catalyzed by original carbon. It can be concluded that such modification can improve the catalytic performance of coconut activated carbon in the simultaneous removal of SO2 and NO with Co(NH3)62+ ammonia solution.

        Fault Detection Based on Graph Model for Dead Zone of Steam Turbine Control Valve

        The abnormal control valve dead zone in the steam turbine causes system oscillation severely, affecting the stable power generation of the power plant. In this study, a graph model is developed, and it can not only detect multiple faults at a time but also reduce the dependence on statistics. The graph topology of the steam turbine is built, including the process nodes and fault node indicating process variables and valve dead zone. Moreover, the graph convolution operation is conducted to classify the faulty and normal conditions. As a result, the simulation and experimental examples demonstrate that the accuracy of dead zone detection reaches 90% and 89% respectively, surpassing Multilayer Perceptron and Principal Component Analysis. The high-precision dead zone detection rate ensures that the method can be effectively applied to industry and improve economic benefits and operation safety.

        Resveratrol enhances the functionality and improves the regeneration of mesenchymal stem cell aggregates

        Mesenchymal stem cell (MSC)-based regeneration, specifically cell aggregate or cell sheet engineering, is a promising approach for tissue reconstruction. Considering the advantages of ease of harvest and lack of immune rejection, the application of autologous MSCs (i.e., patients’ own MSCs) in regenerative medicine has developed considerable interest. However, the impaired cell viability and regenerative potential following MSCs impacted by disease remain a major challenge. Resveratrol (RSV) exhibits reliable and extensive rejuvenative activities that have received increasing clinical attention. Here, we uncovered that resveratrol enhances the functionality and improves the regeneration of mesenchymal stem cell aggregates. Periodontal ligament MSCs (PDLSCs) from normal control subjects (N-PDLSCs) and periodontitis patients (P-PDLSCs) were investigated. Compared to N-PDLSCs, P-PDLSCs were less capable of forming cell aggregates, and P-PDLSC aggregates showed impaired osteogenesis and regeneration. These functional declines could be mimicked in N-PDLSCs by tumor necrosis factor alpha (TNF-α) treatment. Notably, a TNF-α-induced functional decline in N-PDLSC aggregates was rescued by RSV application. More importantly, in both N-PDLSCs and P-PDLSCs, RSV promoted cell aggregate formation and improved their osteogenic potential. Furthermore, as proven ectopically in vivo, the tissue regenerative capability of P-PDLSC aggregates was also enhanced after RSV treatment during aggregate formation in vitro. Finally, in a rat in situ regeneration model, we successfully applied both N-PDLSC aggregates and P-PDLSC aggregates to repair periodontal defects upon long-term functional improvements by RSV preconditioning. Together, our data unravel a novel methodology for using pharmacology (i.e., RSV)-based cell aggregate engineering to improve the functionality and facilitate the regeneration of MSCs from both healthy and inflammatory microenvironments, shedding light on improving the application of autologous MSC-mediated regenerative medicine.

        Gene Expression Profiles of HeLa Cells Impacted by Hepatitis C Virus Non-structural Protein NS4B

        By a cDNA array representing 2308 signal transductionrelated genes, we studied the expression profiles of HeLa cells stably transfected by Hepatitis C virus nonstructural protein 4B (HCV-NS4B). The alterations of the expression of four genes were confirmed by real-time quantitative RT PCR; and the aldo-keto reductase family 1, member Cl (AKR1C1) enzyme activity was detected in HCV-NS4B transiently transfected HeLa cells and Huh-7, a human hepatoma cell line. Of the 2,308 genes we examined, 34 were up-regulated and 56 were down-regulated. These 90 genes involved oncogenes, tumor suppressors, cell receptors, complements, adhesions, transcription and translation, cytoskeletion and cellular stress. The expression profiling suggested that multiple regulatory pathways were affected by HCV-NS4B directly or indirectly. And since these genes are related to carcinogenesis, host defense system and cell homeostatic mechanism, we can conclude that HCV-NS4B could play some important roles in the pathogenesis mechanism of HCV.

      • Vibration characteristics of functionally graded carbon nanotube-reinforced composite double-beams in thermal environments

        This paper presents an investigation on the free vibration characteristics of functionally graded nanocomposite double-beams reinforced by single-walled carbon nanotubes (SWCNTs). The double-beams coupled by an interlayer spring, resting on the elastic foundation with a linear layer and shear layer, and is simply supported in thermal environments. The SWCNTs gradient distributed in the thickness direction of the beam forms different reinforcement patterns. The materials properties of the functionally graded carbon nanotube-reinforced composites (FG-CNTRC) are estimated by rule of mixture. The first order shear deformation theory and Euler-Lagrange variational principle are employed to derive the motion equations incorporating the thermal effects. The vibration characteristics under several patterns of reinforcement are presented and discussed. We conducted a series of studies aimed at revealing the effects of the spring stiffness, environment temperature, thickness ratios and carbon nanotube volume fraction on the nature frequency.
