        Methodology of uncertainty analysis prediction based on multi-well data fusion

        During drilling activities, geological parameters of a well to be drilled (target well) can be predicted within a limited interval based on multi-well data fusion which aims at ensuring a drilling safety, enhancement of drilling efficiency, reduction of drilling cost as well as acquiring accurate measurements in respect to Oil and Gas protection layers. This work presents a method of uncertainty analysis prediction of pressures using fusion data (formation pressures) from adjacent multi-well. The Eaton method, effective stress theory, and mathematical confidence interval were the various methods used to establish the formation pressure matrix of the target well. The results revealed that due to the complexity and variability of the formations, data interpretation errors of the geological parameters were inevitable. Therefore, the probability density distribution function was established through stratigraphy, probability statistics, and information diffusion. Moreover, the real value of the wells’ formation pressure (target well) was within the distribution interval of multi-well data fusion. Hence, the developed method cannot only effectively reduce the interval of geological parameter of the target well but also enhance the accuracy of parameters prediction.

        Phosphorylation of DYNLT1 at Serine 82 Regulates Microtubule Stability and Mitochondrial Permeabilization in Hypoxia

        Hypoxia-induced microtubule disruption and mitochondrial permeability transition (mPT) are crucial events leading to fatal cell damage and recent studies showed that microtubules (MTs) are involved in the modulation of mitochondrial function. Dynein light chain Tctex-type 1 (DYNLT1) is thought to be associated with MTs and mitochondria. Previously we demonstrated that DYNLT1 knockdown aggravates hypoxia-induced mitochondrial permeabilization, which indicates a role of DYNLT1 in hypoxic cytoprotection. But the underlying regulatory mechanism of DYNLT1 remains illusive. Here we aimed to investigate the phosphorylation alteration of DYNLT1 at serine 82 (S82) in hypoxia (1% O2). We therefore constructed recombinant adenoviruses to generate S82E and S82A mutants, used to transfect H9c2 and HeLa cell lines. Development of hypoxia-induced mPT (MMP examining, Cyt c release and mPT pore opening assay), hypoxic energy metabolism (cellular viability and ATP quantification), and stability of MTs were examined. Our results showed that phosph-S82 (S82-P) expression was increased in early hypoxia; S82E mutation (phosphomimic) aggravated mitochondrial damage, ele-vated the free tubulin in cytoplasm and decreased the cellular viability; S82A mutation (dephosphomimic) seemed to diminish the hypoxia-induced injury. These data suggest that DYNLT1 phosphorylation at S82 is involved in MTs and mitochondria regulation, and their interaction and cooperation contribute to the cellular hypoxic tolerance. Thus, we provide new insights into a DYNLT1 mechanism in stabilizing MTs and mitochondria, and propose a potential therapeutic target for hypoxia cytoprotective studies.

        Exploring the Impact of Live Commercial Anchors on BusinessPerformance and Survival Strategy: Focusing on Resource Configuration and Environmental Choice 1

        Purpose – Through the analysis and research of different characteristics and related mechanisms of live commerce anchors, this paper aimed to help live commerce anchors to strengthen their characteristic advantages, beautify the live broadcast environment, improve resource allocation and enhance business performance, so as to help live commerce anchors to achieve better high-quality development. Research design, data, and methodology – This study was conducted by sample survey method on 361 live commercial anchors who have experience in using major online shopping malls for more than one year. Data analysis methods were frequency analysis, confirmatory factor analysis, reliability analysis, correlation analysis, and structural equation model analysis. Result – Hypothesis 1 through Hypothesis 5 were all supported. The results of this study suggest that professionalism and mutual entertainment of live commercial anchors have a significant impact on business performance, survival strategy, resource allocation and environment selection. The resource allocation and environment selection of live commercial anchors play a mediating role in the professionalism and mutual entertainment of live commercial anchors on business performance and survival strategy. Conclusion –The results implied that the professionalism and inter entertainment of live commercial anchors have a positive impact on business performance and survival strategy. In the new situation, live commercial anchors should further improve their professionalism and mutual entertainment, so as to achieve better development.

        Structural and biochemical analysis of Glyceraldehyde-3-Phosphate Dehydrogenase from Clostridium beijerinckii

        Clostridium beijerinckii is a promising industrial microorganism for its ability to produce butanol, acetone, and isopropanol using a wide range of substrates, including pentoses, hexoses, and starch, via fermentation. The ubiquitous and highly abundant glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is essential for most organisms’ energy and carbon metabolism, which plays a critical role in some industrial bacteria. It catalyzes the simultaneous oxidation and phosphorylation of D-glyceraldehyde-3-phosphate into 1,3-bisphosphoglycerate in the presence of inorganic phosphate and nicotinamide adenine dinucleotide (NAD+). We determined the crystal structure of the GAPDH from C. beijerinckii (C. beijerinckii GAPDH). C. beijerinckii GAPDH consists of an α-β-α domain which shares an evolutionarily conserved fold consisting of two juxtaposed domains, an N-terminal NAD+-binding domain (NBD) and a C-terminal catalytic domain (CD). These findings provide insight into the molecular mechanism of action and cofactorbinding of this important industrial bacterial enzyme.

        Chatbot Design Method Using Hybrid Word Vector Expression Model Based on Real Telemarketing Data

        In the development of commercial promotion, chatbot is known as one of significant skill by application of natural language processing (NLP). Conventional design methods are using bag-of-words model (BOW) alone based on Google database and other online corpus. For one thing, in the bag-of-words model, the vectors are Irrelevant to one another. Even though this method is friendly to discrete features, it is not conducive to the machine to understand continuous statements due to the loss of the connection between words in the encoded word vector. For other thing, existing methods are used to test in state-of-the-art online corpus but it is hard to apply in real applications such as telemarketing data. In this paper, we propose an improved chatbot design way using hybrid bag-of-words model and skip-gram model based on the real telemarketing data. Specifically, we first collect the real data in the telemarketing field and perform data cleaning and data classification on the constructed corpus. Second, the word representation is adopted hybrid bag-of-words model and skip-gram model. The skip-gram model maps synonyms in the vicinity of vector space. The correlation between words is expressed, so the amount of information contained in the word vector is increased, making up for the shortcomings caused by using bag-of-words model alone. Third, we use the term frequency-inverse document frequency (TF-IDF) weighting method to improve the weight of key words, then output the final word expression. At last, the answer is produced using hybrid retrieval model and generate model. The retrieval model can accurately answer questions in the field. The generate model can supplement the question of answering the open domain, in which the answer to the final reply is completed by long-short term memory (LSTM) training and prediction. Experimental results show which the hybrid word vector expression model can improve the accuracy of the response and the whole system can communicate with humans.

        Perspectives on Educative Teacher Performance Assessment (edTPA) from Teacher Candidates and College Supervisors

        As a response to the need of systematic teacher performance assessment, tecently the Educative Teacher Performance Assessment (edTPA) has been developed and implemented in over 600 teacher preparation programs across nearly 40 states in US (Stanford Center for Assessment, Learning, and Equity, 2015). However, little research has been conducted investigating ways teacher preparation programs improve the quality of their programs while helping their teacher candidates be well prepared for the edTPA. This study investigated perspectives of student teachers and their college supervisors regarding (1) the effectiveness of the teacher preparation program in a middle-sized public college in relation to edTPA completion and (2) the perceived value of the edTPA as preparation for becoming an effective teacher when New York State began to require successful completion of the edTPA for initial certification. Ninety-nine student teachers in both inclusive elementary and adolescence education programs and fourteen student teaching supervisors participated in the study. A teacher perception survey data including Likert Scale items and open-ended questions was collected as a main data source. Mixed methods were utilized to analyzed both quantitative and qualitative data during the academic year of 2014-2015.         Mechanical behavior and sealing performance of metal sealing system in roller cone bits

        Metal sealing system is used frequently in the dynamic rotary seal of downhole tools of petroleum, natural gas and mining. Mechanical behavior and sealing performance of the metal sealing system in roller cone bit were analyzed in this paper. Influences of pressure difference, compression ratio, fluid pressure, angle and thermal load on the metal sealing system’s mechanical behavior and seal performance were studied. The results show that the effect of pressure difference should be considered under fluid pressure. The von Mises stress concentrates on the inside of O-rings under the influence of compression ratio. The low-stress area appears on the inside of O-rings under the effect of fluid pressure. The high-stress concentration area of metal sealing surface appears on the surface of metal rings under different angles. Sealing performance, compression ratio, fluid pressure and angle have the same change trend. Thermal load is not a single impact on the stress distribution and deformation, but together with fluid pressure affects the sealing performance. The dynamic O-ring more easily fails than the static O-ring. Those results can provide a new direction for the structural optimization of metal sealing system in roller cone bits.

        Phenol Removal Performance and Mechanism Using Catalytic Ozonation with the Catalyst of Cobalt-doped α-MnO2

        In this paper, Cobalt-doped α-MnO2 (i.e., Co-α-MnO2) were synthesized through hydrothermal method. Phenol was employed as targeted pollutants to investigate the catalytic ozonation performance of Co-α-MnO2. Results showed that Co-α-MnO2 significantly improved the phenol removal increased to 97.47 % after 40 min, which was 16.46 %, 38.92 % higher than that of α-MnO2 catalytic ozonation and single ozonation without catalyst. Additionally, the physicochemical properties of α-MnO2 and Co-α-MnO2 were analyzed using technologies such as XRD, TEM, BET and XPS. Compared to α-MnO2, Co-α-MnO2 has larger specific surface area (79.496 m2/g) and pore volume (0.0396 cm3/g), higher Mn3+ relative content (41.16 %) and adsorbed oxygen content (18.99 %). Also, the oxygen vacancy content, lattice defect content and surface hydroxyl content of Co-α-MnO2 are higher than that of α-MnO2, which could result in higher catalytic oxidation performance of Co-α-MnO2. The influence of masking agent showed that surface hydroxyl group, •OH and •O2− were involved in the catalytic ozonation of phenol. This study could help recognize the role of surface hydroxyl groups and active free radicals and demonstrate the contribution of reactive oxygen species on phenol removal in Co-α-MnO2 systems.

      • Identification of amino acid residues important for the arsenic resistance function of<i>Arabidopsis</i>ABCC1

        <P>The Arabidopsis ATP-Binding Cassette (ABC) transporter ABCC1 sequesters arsenic (As)-phytochelatin conjugates into the vacuole, thereby conferring As resistance. Here, we report the results of a screen for phosphorylation-dependent regulation sites of AtABCC1. Variants of AtABCC1 harboring mutations that replaced amino acid residues Tyr(682), Tyr(709), Tyr(822), Ser(846), Ser(1278), or Thr(1408) with alanine confer reduced resistance and decrease the intracellular As content relative to wild-type AtABCC1 when heterologously expressed in the SM7 yeast strain. This suggests that these mutations compromise the vacuolar sequestration of As by AtABCC1. Furthermore, through a phosphomimic mutant study, we found that phosphorylation of Ser(846) is required for the As resistance function of AtABCC1. Our analysis provides a first clue as to the phosphorylation-mediated regulation of AtABCC1 activity.</P>

      • Natural selection in the colloid world: active chiral spirals

        <P>We present a model system in which to study natural selection in the colloid world. In the assembly of active Janus particles into rotating pinwheels when mixed with trace amounts of homogeneous colloids in the presence of an AC electric field, broken symmetry in the rotation direction produces spiral, chiral shapes. Locked into a central rotation point by the centre particle, the spiral arms are found to trail rotation of the overall cluster. To achieve a steady state, the spiral arms undergo an evolutionary process to coordinate their motion. Because all the particles as segments of the pinwheel arms are self-propelled, asymmetric arm lengths are tolerated. Reconfiguration of these structures can happen in various ways and various mechanisms of this directed structural change are analyzed in detail. We introduce the concept of VIP (very important particles) to express that sustainability of active structures is most sensitive to only a few particles at strategic locations in the moving self-assembled structures.</P>
