      • Proteomic Analysis between U87MG and U343MG-A Cell Lines: Searching for Candidate Proteins for Glioma Invasion

        Background To investigate the molecular basis for invasion of malignant gliomas, proteomic analysis approach was carried out using two human glioma cell lines, U87MG and U343MG-A that demonstrate different motility and invasiveness in in vitro experiments. Methods High-resolution two-dimensional gel electrophoresis and matrix-assisted laser-desorption/ionization time-of-flight mass spectrometry analysis were performed. Results Nine distinct protein spots that were recognized with significant alteration between the two cell lines. Five of these protein spots were up-regulated in U87MG and four were up-regulated in U343MG-A. Conclusion Among these proteins, cathepsin D was shown to be one of the important proteins which are related with glioma invasion. However, further studies are necessary to reveal the exact role and mechanism of cathepsin D in glioma invasion.

        A new recombined porcine reproductive and respiratory syndrome virus virulent strain in China

        Porcine reproductive and respiratory syndrome (PRRS) is one of the most important swine diseases worldwide. In the present study, a new virulent strain of PRRS virus (PRRSV), GDsg, was isolated in Guangdong province, China, and caused high fever, high morbidity, and high mortality in sows and piglets. The genome of this new strain was 15,413 nucleotides (nt) long, and comparative analysis revealed that GDsg shared 82.4% to 94% identity with type 2 PRRSV strains, but only 61.5% identity with type 1 PRRSV Lelystad virus strain. Phylogenetic analysis indicated that type 2 PRRSV isolates include five subgenotypes (I, II, III, IV, and V), which are represented by NADC30, VR-2332, GM2, CH-1a, and HuN4, respectively. Moreover, GDsg belongs to a newly emerging type 2 PRRSV subgenotype III. More interestingly, the newly isolated GDsg strain has multiple discontinuous nt deletions, 131 (19 + 18 + 94) at position 1404–1540 and a 107 nt insertion in the NSP2 region. Most importantly, the GDsg strain was identified as a virus recombined between low pathogenic field strain QYYZ and vaccine strain JXA1-P80. In conclusion, a new independent subgenotype and recombinant PRRSV strain has emerged in China and could be a new threat to the swine industry of China.

      • Anti-tumor Activity and Apoptosis-regulation Mechanisms of Bufalin in Various Cancers: New Hope for Cancer Patients

        The induction of apoptosis in target cells is a key mechanism for most anti-tumor therapies. Bufalin is a cardiotonic steroid that has the potential to induce differentiation and apoptosis of tumor cells. Research on bufalin has so far mainly involved leukemia, prostate cancer, gastric cancer and liver cancer, and has been confined to in vitro studies. The bufadienolides bufalin and cinobufagin have been shown to induce apoptosis in a wide spectrum of cancer cell. The present article reviews the anticancer effects of bufalin. It induces apoptosis of lung cancer cells via the PI3K/Akt pathway and also suppressed the proliferation of human non-small cell lung cancer A549 cell line in a time and dose dependent manner. Bufalin, bufotalin and gamabufotalin, key bufadienolides, significantly sensitize human breast cancer cells with differing ER-alpha status to apoptosis induction by the TNF-related apoptosis-inducing ligand (TRAIL). In addition, bufadienolides induce prostate cancer cell apoptosis more significantly than that in breast epithelial cell lines. Similar effects have been observed with hepatocellular carcinoma (HCC) but the detailed molecular mechanisms of inducing apoptosis in this case are still unclear. Bufalin exerts profound effects on leukemia therapy in vitro. Results of multiple studies indicate that bufalin has marked anti-tumor activities through its ability to induce apoptosis. Large-scale randomized, double-blind, placebo or positive drug parallel controlled studies are now required to confirm the efficacy and apoptosis-inducing potential of bufalin in various cancers in the cliniucal setting.

        Dong Hai Bridge in Shanghai is the longest in world and the first sea-crossing project in china, the total length of which is 32.5km and the width of the bridge is 31.5m. In this paper the general concepts of design, construction and durability are introduced. 상하이의 동해대교는 전장 32.5km 폭 31.5m 의 세계 최장 교량이며, 중국에서는 최초의 해상 횡단 프로젝트이다. 이논문에서는 동해대교의 설계, 건설, 그리고 내구성에 대한 기본 개념을 소개한다.

        An Effective Guidewire Looping Technique for the Recanalization of Occlusive Segments of Infrapopliteal Vessels

        Objective: To determine the efficacy, safety and primary follow-up results of a guidewire looping technique for the treatment of infrapopliteal arteries. Materials and Methods: From October 2006 to May 2008, an intraluminal angioplasty of the infrapopliteal arteries was attempted in 200 consecutive patients. Altogether, 417 infrapopliteal lesions, with lengths varying from 2 cm to 32 cm, were treated as part of this study, including 305 lesions in the anterior tibial arteries, 89 in the posterior tibial arteries, and 23 in the peroneal arteries. The ‘U’-shaped guidewire technique was attempted in 393 lesions from 361 limbs. The tip of a hydrophilic 0.035-inch guidewire was formed into a ‘U’ shape with the aid of a 4-Fr catheter and collateral branch vessel to recanalize the completely occluded long segment lesions. Results: A successful angioplasty with at least one artery recanalized directly to the malleolar or dorsal foot was achieved in 322 limbs (89%). The looping technique had a success rate of 90% (352 of 393 lesions). After the procedure, the rest pain was relieved in 58 of 69 patients, while 207 of 245 limbs (85%) showed improvement for intermittent claudication. Complete wound healing was noted in 21 of 54 patients, while 20 of 54 patients showed an improvement in the wound size or depth. A total of 38 major immediate procedure-related complications were noted, including retroperitoneal hematoma, distal emboli, and vessel rupture. Conclusion: The results of this study suggests that the guidewire looping technique is a safe and effective method for the recanalization of the occluded lesions in infrapopliteal vessels.         Design and Analysis of Spider Bionic Flow Field for Proton Exchange Membrane Fuel Cell

        Proton exchange membrane fuel cell (PEMFC) is a portable and clean power generation device. The structural arrangement of the flow field has a significant influence on the delivery efficiency of PEMFC. In this article, a new bionic flow channel is designed based on the inspiration of a spider shape. The branch channel width and branch corner are studied as the focus, and its simulation is carried out by the method of computational fluid dynamics (CFD). The results show that when channel width/rib width and corner of the branch are 1.5 and 130° , respectively, it is the best numerical combination and the cell comprehensive performance is excellent. The final model using this numerical combination is compared with the traditional flow channel model to verify the advancement of this scheme.

        Alterations of Amino Acid Level in Depressed Rat Brain

        Amino-acid neurotransmitter system dysfunction plays a major role in the pathophysiology ofdepression. Several studies have demonstrated the potential of amino acids as a source of neuro-specificbiomarkers could be used in future diagnosis of depression. Only partial amino acids such as glycineand asparagine were determined from certain parts of rats’ brain included hippocampi and cerebralcortex in previous studies. However, according to systematic biology, amino acids in different areaof brain are interacted and interrelated. Hence, the determination of 34 amino acids through entirerats’ brain was conducted in this study in order to demonstrate more possibilities for biomarkers ofdepression by discovering other potential amino acids in more areas of rats’ brain. As a result, 4 aminoacids (L-aspartic acid, L-glutamine, taurine and γ -amino-n-butyric acid) among 34 were typicallyidentified as potentially primary biomarkers of depression by data statistics. Meanwhile, anantidepressant called Fluoxetine was employed to verify other potential amino acids which were notidentified by data statistics. Eventually, we found L-α -amino-adipic acid could also become a newpotentially secondary biomarker of depression after drug validation. In conclusion, we suggested thatL-aspartic acid, L-glutamine, taurine, γ-amino-n-butyric acid and L-α-amino-adipic acid might becomepotential biomarkers for future diagnosis of depression and development of antidepressant.

        Numerical Analysis of the Effect of a Three-Dimensional Baffle Structure with Variable Cross-Section on the Parallel Flow Field Performance of PEMFC

        In this study, a 3D model of the proton exchange membrane fuel cell is established, and a new 3D baffle structure is designed, which is combined with the parallel flow field and then optimized by numerical simulation methods. The number of baffles and the cross-sectional trapezoidal base angle are taken as the main variables, and their impacts on the performance indexes of the cathode side are analyzed. The results show that the 3D baffle can facilitate the convection and diffusion mass transfer of reactants, improve the uniformity of oxygen distribution, enhance the drainage capacity, and make the cell performance superior; however, too small angle will lead to excessive local convective mass flux, resulting in the decrease of the overall uniformity of oxygen distribution and lowering the cell performance. Among them, the optimal number of baffles and angle are 9 and 58°, respectively, which improves the net output power density by 10.8% than conventional flow field.

        Fluid-structure coupling effects on periodically transient flow of a single-blade sewage centrifugal pump

        A partitioned fluid-structure interaction (FSI) solving strategy that depends on problem characteristics is applied to quantitatively obtain the coupling effects of a fluid-structure system in a single-blade centrifugal pump on the unsteady flow. A two-way coupling method is employed to realize strong FSI effects in the calculation procedure. The successful impeller oscillation measurement using two proximity sensors validated the FSI simulation accuracy in a complicated and practical fluid-structure system having a rotating component. The results show that the hydrodynamic force deviation can be observed in the results for the coupled versus uncoupled cases. Additionally,the coupled unsteady pressure is larger than the uncoupled value for every monitoring point at every impeller rotation position. Comparison results for different monitoring points under an overload condition and partial-load condition display the same regularities. To some extent, this interaction mechanism would affect the accuracy and reliability of the unsteady flow and rotor deflection analysis.

      • Fuzzy Task Assignment Model of Web Services Supplier in Collaborative Development Environment

        In view of collaborative development environment web services supplier in ability, cost, time supplier relations and component relevance information under uncertainty problems. The fuzzy multi-objective task assignment model of web services are built in a collaborative development environment. Using α cut sets and extension principle to simplify the fuzzy multi-objective assignment mode, we get the solution of simplified assignment model via the Genetic and simulated-annealing algorithm. In view of the model solution sets are fuzzy and uncertain membership function, a new centroid defuzzification method is used for the model solution. Thus the optimization allocation of component supplier’s fuzzy task is realized in collaborative development environment. Finally, the simulation results verify the feasibility of the proposed method, which can ensure the suppliers’ tasking in successive software project in collaborative development environment.
