In the modern era, molecular genetic techniques are crucial in ecological studies, as well as in the classification, typing, and phylogenetic analysis of prokaryotes. These techniques are mainly aimed at whole genome comparisons and PCRderived experiments, including amplifying the 16S rRNA and other various housekeeping genes used in taxonomy, as well as MLST (multilocus sequence typing) and MLSA (multilocus sequence analysis) of different taxonomic bacterial groups. The gene encoding threonine-tRNA ligase (thrS) is a gene potentially applicable as an identification and phylogenetic marker in bacteria. It is widely distributed in bacterial genomes and is subject to evolutionary selection pressure due to its important function in protein synthesis. In this study, specific primers were used to amplify a thrS gene fragment (~740 bp) in 36 type and 30 wild strains classified under family Bifidobacteriaceae. The full-length gene has not yet been considered as a possible identification, classification, and phylogenetic marker in bifidobacteria. The thrS sequences revealed higher sequence variability (82.7% of pairwise identities) among members of the family than that shown by 16S rRNA gene sequences (96.0%). Although discrepancies were found between the thrS-derived and previously reported whole genome phylogenetic analyses, the main phylogenetic groups of bifidobacteria were properly assigned. Most wild strains of bifidobacteria were better differentiated based on their thrS sequences than on their 16S rRNA gene identities. Phylogenetic confidence of the evaluated gene with respect to other alternative genetic markers widely used in taxonomy of bifidobacteria (fusA, GroELhsp60, pyrG, and rplB genes) was confirmed using the localized incongruence difference - Templeton analysis.

Temperature of Connections During Fire on Steel Framed Building

To study the global structural and thermal behaviour of buildings in fire, a research project was conducted including a fullscale test on a three storey steel frame building at Mittal Steel Ostrava before demolition. The main goal of the experiment was to verify the method for predicting joint temperatures and to improve it for the cooling phase. The fire compartment extending over a floor area of 24 m2 was built on the second floor. The fire load was 140 kg/m2 of wood and the ventilation was limited to an opening of 1,400×1,970 mm. This paper presents the time-temperature curves showing the development of the fire in the compartment and in the primary and secondary beams and its header plate connections. Comparisons are made between the test results and the temperatures predicted by the structural Eurocodes. The sensitivity of the connection behaviour to the estimated temperatures and associated degradation in material properties during the fire is demonstrated. The reaction of 2-ethynylpyridine (2EP) with stoichiometric equivalent of an alkyl halide RX (R is ethyl, nonyl, or hexadecyl and X is Br or I) gives a poly(N-alkyl-2-ethynylpyridinium halide) type ionic polymer that belongs to the family of π-conjugated polyelectrolytes (CPEs). Reaction conditions significantly influence configuration of the polymer main chains: polymerization in acetonitrile solution gives polymers with high content of cis units while bulk polymerization provides irregular cis/trans polymers. Increased regularity of the high-cis polymers is documented by the NMR and IR as well as SERS (surface enhanced Raman scattering) spectra measured on Ag-nanoparticles/ CPE systems. Both polymerization processes give polymers in which 2EP monomeric units are ionized only from ca one half, which exhibit good stability in air and good solubility in polar solvents such as MeOH, DMF, and DMSO and those with N-ethylpyridinium groups even in water.

The virtualized systems are today a very popular topic and their using plays the great role in current datacenters. The virtualization efficiency is a very important aspect in the real system deployment. Some studies have been published about this topic [1], mainly are based on various benchmarking techniques and are integrated into the specialized testing tools. Such a benchmark tool, which is able to simulate the behavior of a real computing system under the stress of different virtualization configurations, can e.g. well answer the question how many virtual machines could be simultaneously executed on it and how big the virtualization overhead is [2]. We developed and applied a new benchmark tool, which is able to measure the virtualization efficiency and overhead in the virtualization environment.

High powered computers and engineering computer systems allow designers to routinely simulate complex physical phenomena. The presented work deals with the analysis of two finite element method optimization techniques (First Order Method-FOM and Subproblem Approximation Method-SAM) implemented in the individual Design Optimization module in the Ansys software to analyze the behavior of real problems. A design optimization is a difficult mathematical process, intended to find the minimum or maximum of an objective function, which is mostly based on iterative procedure. Using optimization techniques in engineering designs requires detailed knowledge of the analyzed problem but also an ability to select the appropriate optimization method. The methods embedded in advanced computer software are based on different optimization techniques and their efficiency is significantly influenced by the specific character of a problem. The efficiency, robustness and accuracy of the methods are studied through strictly convex two-dimensional optimization problem, which is represented by volume minimization of two bars‟ plane frame structure subjected to maximal vertical displacement limit. Advantages and disadvantages of the methods are described and some practical tips provided which could be beneficial in any efficient engineering design by using an optimization method.
New Approach for Virtual Machines Consolidation In Heterogeneous Computing Systems

The energy consumption is one of the most important factors in the virtual machines deployment in the current data centres. Various studies proved that the energy aware management of the virtual machines can reduce the total energy consumption about tens of percents. We developed the new approach, based on the distributed algorithm, which is able to consolidate the virtual machines between various virtualization nodes without the central coordinator. The input data for this algorithm is collected online from the electronic wattmeters, which are placed before the energy input of each virtualization node.
