A robust and simple-to-design multiplex DNA methylation assay based on MS-MLPA-CE-SSCP

<P>Aberrant DNA methylation is a potential diagnostic marker for complex diseases, such as cancer. With the increase in the number of genes known to exhibit disease-associated aberrant methylation, the need for accurate multiplex assays for quantifying DNA methylation has increased. Methylation-specific multiplex ligation-dependent probe amplification (MS-MLPA) is one method that has been highlighted in this context. However, two limitations make the custom design of MS-MLPA assays impractical: the need for long probes containing stuffer sequences and a reliance on only one restriction enzyme. Here, we developed a variation of MS-MLPA that employs a simpler probe-design process. To overcome the above-mentioned limitations, we used stuffer-free MS-MLPA probes that are subsequently analyzed using high-resolution capillary electrophoresis-based single-strand conformational polymorphism (CE-SSCP) instead of conventional length-dependent CE. Moreover, multiple methylation-sensitive restriction enzymes (<I>Hha</I>I, <I>Hpa</I>II, and <I>Aci</I>I) were used simultaneously; thus, probes satisfying desired criteria were available for all targets. Using this assay concept, we analyzed 17 genes associated with hepatocellular carcinoma. Our results showed that the custom-designed assay based on MS-MLPA-CE-SSCP provided robust multiplex quantification of DNA methylation levels.</P> <P>Graphic Abstract</P><P>We demonstrate a robust and simple method for the quantification of DNA methylation by using stuffer-free methylation-specific multiplex ligation-dependent probe amplification and high resolution conformation-sensitive separation. <IMG SRC='http://pubs.rsc.org/services/images/RSCpubs.ePlatform.Service.FreeContent.ImageService.svc/ImageService/image/GA?id=c3an01178j'> </P>
초등학생의 라이프스타일에 따른 소비자사회화 작용인과 윤리적 소비

본 연구는 초등 고학년의 라이프스타일 유형을 알아보고, 라이프스타일에 따른 소비자사회화 작용인과 윤리적 소비의 차이를 살펴보고자 서울 소재 초등학교에 재학 중인 5, 6학년 학생 548명을 대상으로 설문조사를 실시하였다. 초등학생 라이프스타일의 하위차원은 외모 지향, 노력/성취 지향, 디지털 지향, 사교 지향, 평범 지향으로 구분할 수 있었다. 초등학생의 소비자사회화 작용인은 친구, TV-정보획득, 학교, 인터넷, 부모-소비, TV-시청, 부모-금전관리의 7개의 요인으로 구분되었다. 초등학생 라이프스타일 유형은 ‘외모관심형’, ‘성취/사교형’, ‘평범추구형’, ‘디지털 무관심형’, ‘디지털 관심형’으로 구분되었다. 외모에 관심이 많은 외모관심형은 친구와 TV가 소비자사회화 작용인으로 나타났고, 성취와 사교에 관심이 많은 성취/사교형은 학교가 소비자사회화 작용인으로 나타났다. 컴퓨터나 스마트폰에 관심없는 디지털무관심형은 부모가 중요한 소비자사회화 작용인임이 확인되었다. 컴퓨터나 스마트폰을 많이 사용하는 디지털관심형은 인터넷이 가장 중요한 소비자사회화 작용인임이 확인되었다. 이와 같이 초등학생의 성향에 따라 소비자사회화 작용인의 차이가 있으므로 학교에서 소비자교육을 실시할 때 학생 특성에 따른 맞춤형 소비자 교육을 고려할 필요가 있다. ‘성취/사교형’의 윤리적 소비의 평균이 가장 높았고, ‘디지털 관심형’의 평균이 가장 낮은 것으로 나타나 적극적이고 사교적인 집단의 윤리적 소비 수준이 높다고 볼 수 있다. 소비자사회화 작용인 중 학교, 부모가 윤리적 소비에 정적으로 영향을 미치고, 인터넷은 부적인 영향을 미치는 것으로 나타났다. 이는 학교에서의 소비 교육과 부모와의 의사소통이 초등학생의 윤리적 소비에 미치는 영향이 크다는 것을 시사한다. 이에 학교나 지역사회에서 학부모 대상의 소비자 교육 프로그램을 개설하여 초등학생의 올바른 가치관 형성을 위한 소비자 교육을 실시할 필요가 있다. The purpose of this study is to examine the difference between consumer socialization agents and ethical consumption according to lifestyles. A survey was conducted on 548 elementary students in Seoul. The subdimensions of elementary students’ lifestyle could be classified into appearance orientation, effort/achievement orientation, digital orientation, social orientation, and ordinary orientation. The subdimensions of consumer socialization agents were classified into seven factors: friend, TV for information acquisition, school, internet, parental interaction about consumption, TV for viewing, parental interaction about financial management. The types of elementary students were divided into 5 types: appearance-interested, achievement/social, mediocrity pursuit, digital indifference, and digital interest type. Friends and TV appeared to be consumer socialization agent to the appearance-interested type, and school was found to be the agent to the achievement/socialization type. As described above, there is a difference in consumer socialization agents according to the lifestyle of elementary students, so when conducting consumer education in school, it is necessary to consider customized consumer education according to the characteristics of students. Parents are an important consumer socialization agent to the digital indifference type whereas Internet is a critical consumer socialization agent to the digital interest type. The average of the ethical consumption of the achievement/social type was the highest, and the average of the digital interest type was the lowest. Among consumer socialization agents, school and parental interaction have a significant postive influence on ethical consumption while Internet negatively affected. Therefore, it is necessary to conduct consumption education to form correct values of elementary students through parent education programs in schools or local communities.
