Characterization of Aldolase from Methanococcus jannaschii by Gas Chromatography

The products of reactions catalyzed by Methanococcus. jannaschii (Mj) aldolase using various substrates were identified by gas chromatography (GC). Although Mj aldolase is considered a fuculose-1-phosphate aldolase based on homology searching after gene sequencing, it has not been proven to be a fuculose-1-phosphate aldolase based on its reaction products. Mj aldolase was found to catalyze reactions between glycoaldehyde or D, L-glyceraldehyde and DHAP (dihydroxyacetone phosphate). Before performing GC the ketoses produced were converted into peracetylated alditol derivatives by sequential reactions, i.e., dephosphorylation, $NaBH_4$ reduction, and acetylation. By comparing the GC data of final products with those of standard alditol samples, it was found that the enzymatic reactions with glycoaldehyde, D-glyceraldehyde, and D, L-glyceraldehyde produced D-ribulose-1-phosphate, D-psicose-1-phosphate, and a mixture of D-psicose and L-tagatose-1-phosphate, respectively. These results provide direct evidence that Mj aldolase is a fuculose-1-phosphate aldolase.
정보공유 촉진요인이 협상성과에 미치는 영향 : 시뮬레이션 기법의 적용 A Test By Using Simulation Method

협상에 관한 기존 연구들은 협상자 양측 모두에게 이익이 되는 통합적 협상 결과를 위한 필요조건 중 하나로서 ‘협상당사자들이 협상상대로 얻게 되는 이익의 정보에 대해서 정확히 파악하는 것’을 들고 있다. 이러한 기존의 대표적 연구들은 어떠한 경우에 정보의 공유가 성공적으로 이루어질 수 있는지를 충분히 설명하고 있지 않다. 본 연구는 협상 당사자의 개인적 특성, 조직적 특성, 양자간의 경쟁상황 등이 정보공유와 협상성과에 미치는 영향을 역할수행에 의한 시믈레이션 방법으로 검증하였다. 첫째, 협상당사자간의 신뢰와 협상팀 구성원 간의 의사소통의 개방성은 협상당사자간의 정보공유에 유의한 영향을 미쳤고 협상당사자들의 핵심협상정보에 대한 인지와 협상팀 구성원간의 의견일치정도는 정보공유에 유의한 영향을 미치지 못하는 것으로 나타났다. 둘째, 협력적 협상상황하의 피험자들은 정보공유도 원활했을 뿐만 아니라 양자가 모두 이익을 얻는 통합적 결과를 얻었는데 반해 경쟁적 협상상황하의 피험자들은 양자가 일정한 양의 이익을 나누어 갖는 분배적 결과에 이르렀고 또한 원활한 협상정보공유를 이루지 않았다. This study examined the effects of negotiation situation and information sharing on the negotiation outcome and investigated the factors facilitating information sharing. It used negotiation simulation method to test the research hypotheses. Test result shows: (1) that trust between the negotiation parties and communication among the negotiation team members had positive effect on information sharing, but that understanding the key information of the negotiation situation or consensus among the negotiation team members did not have significant effect on information sharing; (2) and that the participants in cooperative negotiation situation shared more information and gained integrative outcome, while the participants in competitive negotiation situation did not share enough information and gained distributive outcome. Implications for managers and suggestions for further studies were discussed.
A Large Axiom Soybean SNP Genotyping Array: Development, Validation, and Genetic Mapping

An important worldwide plant source of dietary protein and oil, modern breeding and improvement of soybean is suffered from a narrow cultivated germplasm relative to other crop species likely because of underuse of wild soybeans as breeding resources. SNP genotyping array is regarded as a promising tool for dissecting wild and cultivated germplasms to find important adaptive genes by high-density genetic mapping and genome-wide association studies (GWAS). Here, we present the establishment of a large soybean SNP array and its use for diversity analysis and high density linkage mapping. More than 4 million high-quality SNPs identified from 16 high-depth and 31 low-depth soybean genome resequencing data were used to select 180,961 SNPs for the AxiomÒ SoyaSNP array. Our validation analysis for a set of 222 diverse soybean lines showed that a total of 171,161markers were of good quality for genotyping. Of the converted SNPs, 82.6% 82.6% SNPs had a marker spacing of less than 9 kb and 17.4% SNPs greater than 9 kb with the 297 inter-SNP spacings of >100 kb and with 812 kb of the largest spacing, thereby suggesting that our array is likely suitable for GWAS of soybean germplasm. This array is being used to construct high-density genetic map in populations generated from intermatings of two cultivated and two wild soybeans, with an objective to confirm large structural variations of chromosomes using the ultra-high-density maps
